A gas chromatography/mass spectrometric assay method was developed for the determination of propylene oxide adduct, N-(3-hydroxypropyl)valine. Adduct was released from hemoglobin by alkaline hydrolysis and extract at pH 8 with ethyl ether. The dried extract was completely derivatized with N-Methyl-N-(trimethylsilyl) trifluoroacetamide (MSTFA)/TMS-I (100:3). The detection limits of the assay were 0.08 ng/g for N-(3-hydroxypropyl)valine based upon assayed hemoglobin of 0.1 g. The method was applied to the determination of propylene oxide adduct formed in young female Sprague-Dawley rats after treatment for 1, 2 and 3 weeks with 0.008 % propylene oxide via the drinking water. An adduct was detected by proposed procedure. The structure of the adduct could be assigned to N-(3-hydroxypropyl)valine.

Artemisia, as one of the largest genera in the tribe Anthemideae of the Asteraceae comprises an important part of Iranian flora. While cytotoxic and apoptotic properties have already been reported for some species of the genus there is not any report on cytotoxic effects of A. ciniformis. Petroleum ether (40-60), dichloromethane, ethyl acetate, ethanol and ethanol-water (50:50) extracts of the aerial parts of A. cinformis were subjected to cytotoxic and apoptotic evaluations on two cancer human cell lines (K562 and HL-60) and on J774 normal cells. Among multiple extracts evaluated for cytotoxicity, dichloromethane ($CH_2Cl_2$) and petroleum ether (PE) extracts were shown to possess the highest anti-proliferative effects on HL-60 and K562 cells with $IC_{50}$ values of 31.3 and $25.5{\mu}g/ml$ respectively. Apoptosis induction verified by sub-G1 peaks was seen in flow cytometry histograms. Increase in the amount of Bax protein, formation of DNA fragments, and cleavage of PARP to 24 and 89kDa sub units all confirmed induction of apoptosis by A. cinformis extracts. Taken together according to the result of the present study some extracts of A. cinformis could be considered as sources for natural cytotoxic compounds and further mechanistic and phytochemical studies are recommended to fully understand the underlying mechanisms of cnacer cell death as well as identification of responsible phytochemicals.
Allelopathic effects on some crops(rice, barley, wheat, Chinese cabbage, leaf mustard, onion, welsh onion, tobacco, red pepper) for extracts of Phytolacca americana and Armoracia rusticana were investigated in MS solid medium. Germination percentage, radicle length, hypocotyl length, plant height, fresh weight, dry weight, and seedling vigor were rapidly inhibited by methanol extract from Phytolacca americana according to increase of the concentration in test plants. Autotoxicity of Phytolacca americana began to appear at concentrations greater than 50$\mu$l methanol extracts per 20ml medium. The fresh weight and dry weight of Phytolacca americana were also decreased by its own methanol extract according to increase of the concentration. Initial amounts of growth of all crops were also inhibited by methanol extracts from Armoracia rusticana, but the inhibitory effects were lowered than those of methanol extracts of Phytolacca americana. Of four fractions, ethyl acetate, petroleum ether, $1^{st}H_2O, 2^{nd}H_2O$, reextracted from methanol extracts of Phytolacca americana, the ethyl acetate fraction showed the highest allelopathic effects on germination percentage and initial amounts of growth. Chlorophyll contents of rice, barley and Chinese cabbage were more inhibited in the ethyl acetate fraction than in the other fractions. Free proline content of Chinese cabbage was increased 31.2 times in the 100$\mu l$ ethyl acetate fraction, and the contents of rice and barley were also increased according to concentration levels in the ethyl acetate fraction.

In previous reports the authors have screened the inhibition effects of marine algae extracts on halitosis, and demonstrated that a brown algae, Eisenia bicyclis (' Daehwang') possess not only strong deodorant effect bug also considerable anticariogenic activities. In this study, we screened antibacterial effects of various marine algae, and measured minimum inhibitory concentration (MIC) value of them against mutans streptococci in vitro. Among the 27 species of marine algae, $80\%$ ethanol extract of dried sea-mustard, Laminaria japonica ('Dasima') showed the strongest inhibition activity against Streptococcus mutans KCTC 3300. The extracts of Ulva lactuca ('Galparae'), Codium fragile ('Cheonggak'), Ecklonia cava ('Gamtae'), E. stolonifera ('Gompi') and Undalia Pinnatifida ('Miyeok') showed slightly weaker inhibitory potency than L. japonica. Differences of MIC values in $80\%$ ethanol extract of some species of marine algae were observed depending on test bacterial species, i.e., S. mutans KCTC 3300 or S. sobrinus KCTC 3307. Eighty percent ethanol extract of dried L japonica was fractionated with diethyl ether, chloroform, ethyl acetate, n-buthanol and water successively, The ether-soluble fraction had inhibitory effect on S. mutans KCTC 3300, however the inhibitory effects were not found in the other fractions. The MIC values of $80\%$ ethanol extract and ether fraction were 180 and 105 $\mu$g/mL respectively, while no significant inhibition activity of water-soluble fraction was found even when the fraction was added up to 5,500 $\mu$g/mL.
The autofluorescent cells in the penetration area of powdery mildewed leaves of barley had been reported. The present experiments were performed in order to obtain the fluorescence-inducing extract fraction from the conidia. The preparations were made by extractions and residue which were extracted from the conidia of Erysiphe graminis hordei race I with water, ethanol, and ethyl ether. Bioassaying was carried out on the culled-leaf surface of incompatible Turkey 290 and compatible Kobingataki varieties by placing the drops of extract solutions. Fluorescence-elicitor activity was shown only in tile ethanol-extract fraction to both varieties. However, fluorescence-eliciting rate was more rapid on the leaves of incompatible variety Turkey 290 than on tile those of compatible variety Kobingataki; Turkey 290 less than 8 hours, Kobingataki less than 16 hours.
This study was performed to investigate the antimicrobial effect of the Forsythiae Fructus extracts against food-borne pathogens. First, Forsythiae Fructus was extracted with methanol at room temperature and the methanol extracts were fractionated by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Forsythiae Fructus extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Forsythiae Fructus showed the highest antimicrobial activity against Salmonella paratryphimurium and Salmonella typhimurium. A synergistic effect in inhibition was observed when Forsythiae Fructus extract was mixed with Ulmus davidiana Japonica extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Forsythiae Fructus against Shigella flexneri and Salmonella paratyphimurium. The aqueous extract of Forsythiae Fructus had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 10,000 ppm. At this concentration, the growth of Shigella fexneri was retarded for more than 24 hours and for up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Forsythiae Fructus efficiently inhibited Staphylococcus epidermidis and Shigella flexneri.

Fruits of Sonneratia apetala (Buch.-Ham.), (English: mangrove apple, Bengali: keora) both seeds and pericarps, are largely consumed as food besides their enormous medicinal application. The fruit seeds have high content of nutrients and bioactive components. The seeds powder of S. apetala was successively fractionated using n-hexane, diethyl ether, chloroform, ethyl acetate, and methanol. The fractions were used to evaluate antibacterial, anti-diarrhoeal, analgesic, and cytotoxic activities. Methanol fraction of seeds (MeS) stronly inhibited Escherichia coli strains, Salmonella Paratyphi A, Salmonella Typhi, Shigella dysenteriae, and Staphylococcus aureus except Vibrio cholerae at $500{\mu}g/disc$. All the fractions strongly inhibited castor oil induced diarrhoeal episodes and onset time in mice at 500 mg extract/kg body weight (P<0.001). At the same concentration, MeS had the strongest inhibitory activity on diarrhoeal episodes, whereas the n-hexane fraction (HS) significantly (P<0.05) prolonged diarrhoeal onset time as compared to positive control. Similarly, HS (P<0.005) inhibited acetic acid induced writhing in mice at 500 mg extract/kg, more than any other fraction. HS and diethyl ether fractions of seed strongly increased reaction time of mice in hot plate test at 500 mg extract/kg. All the fractions showed strong cytotoxic effects in brine shrimp lethality tests. Gas chromatography-mass spectrometry analysis of HS led to the identification of 23 compounds. Linoleic acid (29.9%), palmitic acid (23.2%), ascorbyl palmitate (21.2%), and stearic acid (10.5%) were the major compounds in HS. These results suggest that seeds of S. apetala could be of great use as nutraceuticals.

The present study was conducted to prepare seaweed extracts suppressing mutagenic activity of 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine ( PhIP ) ana 2-amino-3,8-dimethylmidazo[4,5-f]quinoxaline (MeIQx) derived from cooked meat produce. The tumor initiation activity of PhIP and MeIQx was assayed with Ames method using Salmonella typhimurium TA98 in the presence of S-9 mixtures before and after addition of methanol-solubles of seaweed, such as, Phaeophyta; Undaria pinnatifida, Ecklonia stolonifera Ecklonia cava, Laminaia japonica Sargassum fulvellum, Sargassum hornezi, Sargassum miyabei, Sargassum thunbergii, Agarum cribrosum and Hizikia fusifomis, Rhodophpei Porphyra yezoensis, Grateloupia eiliptiut Lomentraria catenata, Ploemium telfairiae and Glarilaria verrucosa, Chlorophyta; Codium fragile, Enteromorpha compresa and Ulva pertusa. Among seaweed tested, Phaeophyta was shown the higher desmutagenic activity than Rhodophyta and Chlorophyta. E. stolonifera, E. cava and S. miyabei, among Phaeophyta exerted the stronger desmutagenic activity (above $90\%$/2 mg). The ethyl acetate, diethyl ether and chlomform extracts except water extracts from E. stolonifera exhibited a high desmutagenic activity. The ethyl acetate extract of E. stolonifera which showed highest activity was fractionated with Sephadex LH20 column chromatography to give active fraction A-7, which showed desmutagenicity of $90\%$/mg against PhIP and $80\%$/mg against MeIQx. The active fraction had the absorbance at 207.7 and 232nm.

Acanthopanax trifoliatus (L) Merr (AT) is commonly used as an herbal medicine and edible plant in some areas of China and other Asian countries. AT is thought to have anticancer effects, but potential mechanisms remain unknown. To assess the anticancer properties of AT, we exposed prostate cancer cells to AT extracts and assessed cell proliferation and signaling pathways. An ethanol extract of AT was suspended in water followed by sequential extraction with petroleum ether, ethyl acetate and n-butanol. PC-3 cells were treated with different concentrations of each extract and cell viability was determined by the MTT and trypan blue exclusion assays. The ethyl acetate extract of the ethanol extract had a stronger inhibitory effect on growth and a stronger stimulatory effect on apoptosis than any of the other extracts. Mechanistic studies demonstrated that the ethyl acetate extract suppressed the transcriptional activity of NF-${\kappa}B$, increased the level of caspase-3, and decreased the levels of phospho-Erk1/2 and phospho-Akt. This is the first report on the anticancer activity of AT in cultured human prostate cancer cells. The results suggest that AT can provide a plant-based medicine for the treatment or prevention of prostate cancer.
This study was performed to investigate the antimicrobial activities of the Plagiorhegama dubium extracts against food-borne pathogens. The methanol extract was partitioned into petroleum ether, chloroform, ethyl acetate, and methanol portions. The antimicrobial activity of the P. dubium extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of P. dubium showed the highest antimicrobial activity against Staphylococcus aureus and Escherichia coli. The synergistic effect has been found in combined extracts of P. dubium and Hedyotis diffsa as compared to each extracts alone. Finally, the growth inhibition curve was determined using methanol extracts of P. dubium against S. aureus and E. coli. The methanol extract of P. dubium showed strong antimicrobial activity against S. aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate portion from P. dubium retarded the growth of S. aureus more than 36 hours and E. coli up to 60 hours. The methanol extracts of P. dubium has been shown the antimicrobial activity against S. aureus and E. coli.
