Antimicrobial Effect of Pulsatilla Koreana Extracts on Food-Borne Pathogens (백두옹 추출물의 식중독성 미생물에 대한 항균효과)

  • This study was performed to investigate the antimicrobial effect of the Pulsatilla koreana extracts against food-borne pathogens. First, the Pulsatilla koreana was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Pulsatilla koreana was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Pulsatilla koreana extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Pulsatilla koreana showed the highest antimicrobial activity against Staphylococcus aureus, Salmonella enteritidis and Shigella dysenteriae. The Staphylococcus aureus and Shigella dysenteriae were inhibited by petroleum ether and chloroform extracts of Pulsatilla koreana as well as ethyl acetate extracts of Pulsatilla koreana. The synergistic effect has been found in combined extracts of Pulsatilla koreana and Portulaca oleracea as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Pulsatilla koreana against Staphylococcus aureus and Shigella dysenteriae. The ethyl acetate extract of Pulsatilla koreana showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 2,000 ppm. The 2,000 ppm of ethyl acetate extract from Pulsatilla koreana retarded the growth of S. aureus more than 12 hours and Shigella dysenteriae up to 9 hours.

  • Plectranthus amboinicus Benth (Lamiaceae) is a medicinal plant native to India, and its leaves are widely used in several traditional medicinal preparations. The purpose of this study was to detect and quantify phenolics present in ethyl acetate and acetone extracts of P. amboinicus leaves, and evaluate their antioxidant, antibacterial, antimutagenic and anticancer activities. The HPLC chromatograms of crude leaf extracts indicated the presence of phenolics like caffeic acid, coumaric acid, rutin, quercetin and gallic acid, which were present in the range of 0.01 - 1.41 mg/g in ethyl acetate and 0.03 - 1.93 mg/g in the acetone extract. The acetone extract showed statistically (p < 0.05) higher antioxidant activity ($IC_{50}$, 99.59 ${\mu}g/ml$) than ethyl acetate extract ($IC_{50}$, 149.96 ${\mu}g/ml$). Statistically (p < 0.05) higher antimutagenicity was shown by acetone extract (46.16%) as compare to ethyl acetate extract (12.16%) at 500 ${\mu}g/plate$ concentration. The acetone extract showed higher antibacterial activity than ethyl acetate extract, and both the extracts showed highest activity against B. cereus (375 and 625 ${\mu}g/ml$, respectively) and lowest activity against Y. enterocolitica (1000 and 1125 ${\mu}g/ml$, respectively). Both the extracts also showed inhibitory effect on cancer cell lines HCT-15 and MCF-7. These results suggest that the leaves of P. amboinicus possess various biological activities, and validate the traditional use of the leaves of P. amboinicus against cold, infection and ulceration.

Study on the Anti-oxidative, Anti-microbial and Anti-cancer effect of Bibangtalmyungsan. (비방탈명산(秘方奪命散)의 항산화 ${\cdot}$ 항암 ${\cdot}$ 항균 효과 연구)

  • Objectives : This study was carried out to evaluate anti-oxidative, anti-microbial and anti-cancer effect for clinical application of Bibangtalmyungsan (BTS) Results : 1. The oxidative effects were measured by polyphenol, DPPH radical scavenging activity. BTS water extract was showed more effective than ethanol extract and also various solvent fractions from BTS water extract showed effective in the following order : ethyl acetate fraction > butanol fraction > hexane fraction ${\fallingdotseq}$ chloroform fraction ${\fallingdotseq}$ aqueous fraction 2. The results of anti-microbial effects were as follows. 1) Antibacterial activities of BTS extracts against Gram's negative and positive bacteria were ineffective 2) Antifungal activities of the BTS extracts against Aspergillus spp. , Trichohyton mentagrophyte KTCC 1077 were not effective. 3. The result of anti-cancer effects were as followings: 1) BTS ethanol extract was more effective than water extract against Caco-2, Calu-6, but it had cytotoxic effect against NIH3T3. 2) On the Caco-2, effective only in ethyl acetate $fraction(IC_{50}:$ 35.81 ${\mu}g/ml)$. 3) On the Calu-6, the most effective in ethyl acetate $fraction(IC_{50}:$ 189.65 ${\mu}g/ml)$ and effective In butanol $fraction(IC_{50}:$ 299.74 ${\mu}g/ml)$ and hexane $fraction(IC_{50}:$ 345.13 ${\mu}g/ml)$ 4) On the SUN-601, $IC_{50}$ value was within 80 ${\mu}g/ml$ in ethyl acetate fraction and hexane fraction. 5) On the HCT-1l6, the most effective in ethyl acetate $fraction(IC_{50}:$ 82.94 ${\mu}g/ml)$ and effective in hexane $fraction(IC_{50}:$ 374.56 ${\mu}g/ml)$. 6) On the AML-2/WT, the most effective in ethyl acetate $fraction(IC_{50}:$ 41.44 ${\mu}g/ml)$ and effective In hexane $fraction(IC_{50}:$ 303.01 ${\mu}g/ml)$. 7) On the NIH3T3, effective only in ethyl acetate $fraction(IC_{50}:$ 203.42 ${\mu}g/ml)$, but it was more ineffective than other cancer cells. Conclusion : These result suggest that BTS has antioxidative, antifungal activities and cytotoxic effects against Caco-2, Calu-6, SUN-601, HCT-116, AML-2/WT and NIH3T3, especially ethyl acetate fraction from water extract has more effective in antioxidative and anticancer effects.

Isolation and Identification of an Antioxidant Substance from Ethanol Extract of Wild Grape (Vitis coignetiea) Seed (머루종자 에탄올 추출물로부터 항산화활성물질 분리 및 동정)

  • Antioxidant compound(s) were identified from the ethanol extract of wild grape (Vitis coignetiea) seed. Organic solvent fractions of n-hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of wild grape seed, among which ethyl acetate fraction showed the strongest reducing power. Ethyl acetate fraction was further purified through ODS column chromatography and HPLC, and isolated antioxidative active compound was identified through $^1H-NMR$ as (+)-catechin (52.7 g/100 g). (+)-Catechin and ethyl acetate fraction both showed approximately 80% scavenging effect. These results indicated (+)-catechin in the ethyl acetate fraction synergetically interacts with unknown antioxidative compound(s).

Study on the Antioxidant and Anticancer Effects of Extract of Stamens of Nelumbo nucifera and Kaempferol (연수 추출물과 Kaempferol의 항산화 및 항암작용에 관한 연구)

  • Objectives : The present study was performed to clarify the antioxidant and anticancer effects of extract of stamens of Nelumbo nucifera and Kaempferol. Methods : Antioxidant effect was measured by DPPH-radical scavenging activity for electron donating ability (EDA), superoxide dismutase (SOD)-like activity for SOD and lipid peroxidation. Anticancer effect was assessed by MTT absorbance for cytotoxicity. Results : Vitamin E, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera increased SOD-like activity and DPPH-radical scavenging activity dose-dependently. On the contrary, lipid peroxidation was time-dependently decreased. Furthermore, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera significantly decreased the growth rate of C6 glioma cells. Conclusions : These results suggest that ethyl acetate extract of stamens of Nelumbo nucifera may be a putative antioxidant or anticancer substance.

A Study of Antioxidant Effects of Pyracantha angustifolia(Franch.) C. K. Schneid Extract (피라칸타 추출물의 항산화 효능에 관한 연구)

  • In this study, Pyracantha angustifolia (Franch.) C. K. Schneid was extracted with 70% methanol at room temperature for 48 hrs and concentrated under reduced pressure to measure its total polyphenol contents; furthermore, the effect of electron donating ability was examined. Methylene chloride, ethyl acetate, and methanol were used to fractionate the extract to testify total polyphenol contents, electron donating abilities, the removal abilities of superoxide radical as well as hydrogen peroxide. The total polyphenol contents were $2007.30{\pm}109.28{\mu}g\;GAE/mL$ in 70% methanol extract, $273.39{\pm}10.19{\mu}g\;GAE/mL$ in methylene chloride fraction, $80.57{\pm}0.64{\mu}g\;GAE/mL$ in ethyl acetate fraction, and $1,160.87{\pm}44.71{\mu}g\;GAE/mL$ in methanol fraction. The total polyphenol contents showed significant differences (p<0.05) between the solvents. The electron donating ability was $79.07{\pm}7.31%$ for 70% methanol extract, $22.34{\pm}0.64%$ for methylene chloride fraction, $5.33{\pm}0.28%$ for ethyl acetate fraction, and $32.26{\pm}1.10%$ for methanol fraction. The electron donating abilities were significantly different(p<0.05) between the solvents. The removal ability of superoxide radical was $0.018{\pm}0.003$ for 70% methanol extract, $0.007{\pm}0.002$ for methylene chloride fraction, $0.0147{\pm}0.003$ for ethyl acetate fraction, and nothing for methanol fraction. The measurement of hydrogen peroxide decomposition was $0.022{\pm}0.0046$ for 70% methanol extract, $0.0027{\pm}0.0015$ for methylene chloride fraction, $0.0037{\pm}0.0012$ for ethyl acetate fraction, and $0.0009{\pm}0.0001$ for methanol fraction.

Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts (누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성)

  • This study investigated the antioxidant activities, whitening effects, and antimicrobial activities of fractions from Pleurospermum kamtschaticumin extract. Total polyphenolic contents of fractions from Pleurospermum kamtschaticumin extract were 116.44~382.73 mg/g GAE (gallic acid equivalent), with the highest value in the ethyl acetate fraction. Fractions of Pleurospermum kamtschaticumin showed the highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ($IC_{50}$=0.04 mg/mL) and ABTS radical scavenging activity (98.86% at 0.5 mg/mL), which was similar to ascorbic acid. Further, reducing power and FRAP activity were significantly high in the ethyl acetate and n-butanol fractions. The ethyl acetate and n-butanol fractions showed significantly high SOD-like activities (1 mg/mL, 86.93%, and 78.23%, respectively) compared to other fractions. Tyrosinase inhibition activities of the n-butanol fraction and 80% ethanol extract were 43.52% and 41.64%, respectively. Antimicrobial activity against Escherichia coli was only observed in the chloroform fraction (16.7 mm in 0.5 mg/disc). These results suggest that fractions from Pleurospermum kamtschaticumin extract show comparatively high antioxidant activity and thus could be used as a food additive or cosmeceutical ingredient.

Antimicrobial Effect of Citrus unshiu Markovich Extracts on Food-Borne Pathogens (청피 추출물이 식중독 유발 미생물의 증식에 미치는 영향)

Finally, the growth inhibition curve was determined using ethyl acetate extracts of Citrus unshiu Markovich against Bacillus cereus and Shigella sonnei. The ethyl acetate extract of Citrus unshiu Markovich showed strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. The 5,000 ppm of ethyl acetate extract from Citrus unshiu Markovich retarded the growth of Bacillus cereus more than 24 hours and Shigella sonnei up to 24 hours. The ethyl acetate extracts of Citrus unshiu Markovich have shown an antimicrobial effect against Bacillus cereus and Shigella sonnei.

Synergistic Antimicrobial Effect of Patrinia scabiosaefolia and Forsythiae fructus Extracts on Food-borne Pathogens (식중독 유발 세균의 증식에 미치는 패장과 연교 추출물의 상승 효과)

  • To investigate the antimicrobial effect of the Patrinia scabiosaefolia extracts against food-borne pathogens, we extracted the P. scabiosaefolia with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol, respectively. The antimicrobial activity of the P. scabiosaefolia extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of P. scabiosaefolia showed the highest antimicrobial activity against Escherichia coli and Shigella sonnei. Synergistic effect in inhibition was observed when P. scabiosaefolia extract was mixed Forsythiae fructus extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of P. scabiosaefolia against Staphylococcus epidermidis and Shigella sonnei. The ethyl acetate extract of P. scabiosaefolia had strong antimicrobial activity against S. sonnei at the concentration of 4,000 ppm. At this concentration, the growth of S. Sonnei was retarded more than 72 hours and up to 48 hours for S. epidermidis. These results suggest that the ethyl acetate extracts of P. scabiosaefolia can be used for the efficient material against the growth of S. epidermidis and S. sonnei.

Antioxidant and Antimicrobial Activities of Korean Mistletoe (Viscum album var. coloratum) Extracts against Food Poisoning Bacteria (한국산 겨우살이 (Viscum album var. coloratum) 추출물의 식중독 세균 증식 억제 및 항산화 활성)

  • This study was conducted to investigate the antimicrobial activities and antioxidant activities of the Korean mistletoe extract and its solvent fractions (e.g. n-hexane, ethyl ether, ethyl acetate, butanol). Ethyl ether fraction against Bacillus cereus showed stronger activities than benzoic acid (2.5 mg/mL). The MIC of korean mistletoe extract and slovent fractions were in the range of 6.25-25 mg/mL. The MIC (6.25 mg/mL) of ethyl acetate fraction onto Staphylocossus aureus was the lowest among them. Ethyl ether fraction which showed the strongest antioxidant activities by DPPH (1,1-diphenyl-2-picryl-hydrazyl) and FRAP (ferric ion reducing antioxidant power) methods had the highest total phenolic contents. It is suggested that Korean mistletoe could be utilized as natural preservative material through the study of the active compounds from ethyl ether fraction.
