Antioxidant Activities of the Mycelium and Culture Broth of Phellinus igniarius and Agrocybe cylindracea (말똥진흙버섯 및 버들송이버섯의 균사체 및 배양액의 항산화활성)

  • The fractions obtained from the mycelia and their culture broth of Agrocybe cylindracea and Phellinus igniarius were tested for the antioxidation activities. In electron donating ability test, strong activities more than 70% were observed in ethyl acetate and butanol fractions of A. cylindracea mycelium, butanol fraction of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether and ethyl acetate fractions of P. igniarius culture broth. Lipid peroxidation inhibition was observed in diethyl ether, butanol and water fractions of A. cylindracea mycelium, and water fraction of A. cylindracea induced the decrease in uric acid production. Activities of $50{\sim}60\;%$ of nitrite scavenging ability were observed in diethyl ether fraction of A. cylindracea mycelium, ethyl acetate and water fractions of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether, chloroform, butanol and water fractions of P. igniarius culture broth. In melanin biosynthesis inhibition activity measurement, water fraction of A. cylindracea mycelium and butanol fraction of P. igniarius mycelium showed twice higher activities than control. From these results, the fungi of P. igniarius and A. cylindracea had strong antioxidant activities.

Antioxidative Activity of Gynostemma pentaphyllum Makino Extracts (덩굴차(Gynostemma pentaphyllum Makino) 추출물의 항산화 효과)

  • This study was carried out to examine the antioxidant activity of Gynostemma pentaphyllum Makino. Using the DPPH method, we found that free radical scavenging activity was strong in all the fractions except the water fraction of the water extract (GPW) and ethanol extract (GPE) of G pentaphyllum Makino. Pseudo-SOD activity was highest in the diethyl ether fraction of the ethanol extract, while the other fractions of the ethanol and water extracts were lower. For xanthine oxidase inlribition activity, the diethyl ether fraction and butanol fraction of GPW, and the diethyl ether fraction and butanol fraction of GPE, showed activities over 80%. Nitrite scavenging ability was strong (over 60%) in all the GPW and GPE fractions. The diethyl ether fraction and butanol fraction of GPW had more effective nitrite scavenging abilities than the other extract fractions. These results suggest that the extracts of G pentaphyllum Makino can be used as a functional material in a tea or drink.

Antioxidant and Antimicrobial Activities of Korean Mistletoe (Viscum album var. coloratum) Extracts against Food Poisoning Bacteria (한국산 겨우살이 (Viscum album var. coloratum) 추출물의 식중독 세균 증식 억제 및 항산화 활성)

  • This study was conducted to investigate the antimicrobial activities and antioxidant activities of the Korean mistletoe extract and its solvent fractions (e.g. n-hexane, ethyl ether, ethyl acetate, butanol). Ethyl ether fraction against Bacillus cereus showed stronger activities than benzoic acid (2.5 mg/mL). The MIC of korean mistletoe extract and slovent fractions were in the range of 6.25-25 mg/mL. The MIC (6.25 mg/mL) of ethyl acetate fraction onto Staphylocossus aureus was the lowest among them. Ethyl ether fraction which showed the strongest antioxidant activities by DPPH (1,1-diphenyl-2-picryl-hydrazyl) and FRAP (ferric ion reducing antioxidant power) methods had the highest total phenolic contents. It is suggested that Korean mistletoe could be utilized as natural preservative material through the study of the active compounds from ethyl ether fraction.

The Effect of AS Aqua-acupuncture on the Diabetic Rats Induced by Streptozotocin (가시오가피(五加皮) 약침(藥鍼)이 항당뇨(抗糖尿) 및 신장보호활성(腎臟保護活性)에 미치는 영향(影響))

  • Objective ; In order to study the effect of Acanthopanax senticosus(AS) aqua-acupuncture manufactured with water soluble fraction and ether soluble fraction on the streptozotocin induced diabetic rats Methods ; The fractions of AS aqua-acupuncture were carried out on corresponding bilateral loci of Bisu(BL20) everyday for 4 weeks. The experimental animals were divided into control group and AS groups(AS water fraction group and AS ether fraction group). Thereafter the levels of serum glucose, total choleterol, HDL, triglyceride, AST, ALT, creatinine, BUN, urinary albumin excretion, index of kidney hypertrophy, heart rate, mean blood pressure and fibronectin in glomeruli and tubular cells were measured. Results ; The increased serum total cholestrol, triglyceride levels, HDL and urinary albumin excretion, the index of kidney hypertrophy, the mean blood pressure and the amount of fibronectin in glomeruli and tubular cells were significantly decreased in the AS groups, showing more significant decrease in the AS water fraction group as compared with the control group. In the serum ALT, AST, creatinine and BUN levels, there were no significant changes in the AS groups as compared with the control group. Conclusion ; According to the above results, it reveals that Acanthopanax senticosus water soluble and ether soluble fraction have the antidiabetic effect, the antilipidemic effect and the inhibitory effect of renal damage. Also, the results showed that Acanthopanax senticosus water soluble fraction is more effective than ether soluble fraction.

Separation of Ether-Soluble Neutral Extract of a Commercial High-Grain Feed Stimulating Hay Intake in Cattle

  • Appetite stimulant ether-soluble neutral extract of a high-grain compound feed was separated into four fractions with an open column of silica gel chromatograph using four carrier solutions containing n-pentane and diethyl ether as 100:0 (Fraction A), 90:10 (Fraction B), 75:25 (Fraction C) and 0:100 (Fraction D). The stimulative effects of the fractions were examined by comparing the intake of hay (with or without the fractions) in two-choice test bioassays with cattle. The Fractions A, B and C of the neutral extract stimulated (p<0.05) hay consumption in cattle, whereas Fraction D had no effect on selective feeding when compared with the control fraction. Furthermore, Fractions A, B and C had higher (p<0.05) feeding stimulative indices (FSI) than that of the Fraction D. The results suggest that chemical stimulants to increase palatability of hay are present in the neutral Fractions A, B and C of high-grain concentrate.

Hypouricemic Activity of Actinidia polygama (개다래의 고요산혈증 개선 활성)

  • Ethanol extract of Actinidia polygama Max. (AP) was examined for hypouricemic activity in potassium oxonateinduced hyperuricemic animal model. Ethanol extract of AP lowered the plasma uric acid level by 20% when compared to the hyperuricemic control group. AP ether fraction at a dose of 100 mg/kg showed 27% reduction, whose efficacy was comparable to the probenecid-treated group. AP ether fraction-treated group also showed 23% reduction of urate in urine and this result suggests that AP ether fraction has anti-gout activity probably due to xanthine oxidase inhibition.

(no cell survived) A double dilution of the fraction(10ug/ml) still showed strong activity.(0.2 cells survived) The ether fraction contained strong activity. Of the fraction which have veen obtained by a prcparative silicagel TLC I, II, III & IV were cytotoxic, but V and VI were inactive Howevcr, it is worthwhile to note that no one of these has no strong Cytotoxicity as the pertoleum ether fraction itself. This observation says that the active fractions could be additive or synergic to each other. The most active fraction, IV, was scrached out from the TLC and its UV-spectrum taken. The absorption maxima were 207 and 272 nm, indicating the presence of a flavanone strucuture.

The Effect of Ginseng on the Hepatic Glutathione S-Transferase Activity (인삼이 간 Glutathione S-Transferase 활성에 미치는 효과)

  • The investigation aimed to study the effect of ginseng on the hepatic glutathion S-transferase activity. The ginseng methanol extract was administered to rats and mice for 10 days and their hepatic gluthatione S-transferase activities were measured by the method of Habig et al. Glutathione S-transferase activities in the rat treated with 100 and 500mg/kg ginseng methanol extract were increased by 13.4% and 17.10%, respectively and their increases were statistically significant. Similar results were also found in the mouse treated with ginseng 100mg/kg methanol extract. To investigate components of the extract which induce the enzyme, the methanol extract was fractionated into ether and butanol fraction and their effect on the enzyme was compared. Glutathione S-transferase activities in the rat treated with ether fraction were increased by 13.1%, similar to that obtained with ginseng methanol extract, whereas, butanol fraction did not show any increase in the enzyme activities. In the rats treated with maltol, one of the components in ether fraction, 5mg/kg for 10 days, activity of glutathione S-transferase was increased by 7.89%, but its increase was not significantly different from control group. Therefore, it may be concluded that ginseng methanol extract and its ether soluble fraction had effect on the elevation of glutathione S-transferase activities, whereas, butanol fraction of ginseng methanol extract had no effect on the enzyme.

강원도산 참당귀와 일본산 일당귀의 생리 활성 성분 탐색

  • The ethanol extracts from Angelica gigas Nakai and Angelica acutiloba Kitagawa were fractionated to diethyl ether and aqueous partitions. Both partitions had strong antimutagenic effect on the MNNG (N-methyl-N-nitro-N-nitrosoguanidine) by Ames mutagenicity test. Diethyl ether fractions exhibited the greatest antimutagenic effect suppressing the mutagenicity of MNNG with inhibition of 78-80%. The ethanol extracts from both species showed the inhibitory effect on the growth of several human cancer cell lines. Especially, the diethyl ether fraction from ethanol extracts was most effective on human hepatocellular carcinoma cells, inhibiting 90-95% of cell growth. However, the aqueous fractions had least inhibition activity on many cancer cells. There was little cytotoxicity on human normal liver cell by ethanol extracts. Diethyl ether fraction from Angelica gigas Nakai ethanol extract had cytotoxicity less than 20% on human normal liver cells, compared with that from Angelica acutiloba Kitagawa ethanol exract. The adding of 0.5 (g/l) of diethyl ether fractions of Angelica gigas Nakai or Angelica acutiloba Kitagawa increased immune activity by enhacing human B and T cells up to three to four times. It was proven that diethyl ether fraction (0.7 g/1) from Angelica gigas Nakai could control blood pressure by suppressing angiotensin converting enzyme activity up to 98%. From TLC, it was appeared that both of diethyl ether partitions had umbelliferon, known to one of active substances from Angelica gigas Nakai and Angelica acutiloba Kitagawa.
