Effect of pre-treatment with hot water extract of marine brown alga Sargassum polycystum C.Ag. (100 mg/kg body wt, orally for period of 15 days) on HCI-ethanol (150 mM of HCI-etha-not mixture containing 0.15 N HCI in $70\%$ v/v ethanol given orally) induced gastric mucosal injury in rats was examined with respect to lipid peroxides, antioxidant enzyme status, acid/pepsin and glycoproteins in the gastric mucosa. The levels of lipid peroxides of gastric mucosa and volume, acidity of the gastric juice were increased with decreased levels of antioxidant enzymes and glycoproteins were observed in HCI-ethanol induced rats. The rats pre-treated with seaweed extract prior to HCI-ethanol induction reversed the depleted levels of antioxidant enzymes and reduced the elevated levels of lipid peroxides when compared with HCI-ethanol induced rats. The levels of glycoproteins and alterations in the gastric juice were also maintained at near normal levels in rats pre-treated with seaweed extract. The rats given seaweed extract alone did not show any toxicity, which was confirmed by histopathological studies. These results suggest that the seaweed extract contains some anti-ulcer agents, which may maintain the volume/acidity of gastric juice and improve the gastric mucosa antioxidant defense system against HCI-ethanol induced gastric mucosal injury in rats.
Excessive drinking causes 'alcohol hangover' within 8-16 hours. The cause of 'hangover' has not been elucidated exactly until now, but it is reported that it is caused by the creation of blood ethanol and acetaldehyde as ethanol metabolites. In this study vinegar extract of wood (VE) or OC-1, to which the powder extract of green tea leaves extract is added, was administered to the rats 30 minutes before the oral administration of ethanol (3 g/kg) and the blood ethanol and acetaldehyde concentration was measured in order to evaluate the efficacy of the beverage material for detoxification. As a result, the blood ethanol concentration in the group of the VE-1(vinegar crude extract) and VE-2 (double diluted solution) is statistically lower (P,0.05) than the exclusive alcohol administered control group. The blood acetaldehyde concentration of all groups of VE and OC-2, which is the double dilution of OC-1, is statistically low after 7 hours following ethanol administration. Especially, the AUC value of OC-2 group is statistically low compared to the control group. Accordingly, it indicates the conclusion that VE and OC-1, reducing the blood ethanol and acetaldehyde concentration which are two leading factors of 'hangover' after drinking, and worthwhile to be developed as beverage materials to eliminate 'hangover'.
This study was conducted to evaluate the hypotensive properties of the extract of Chinese balloon flower (Platycodon grandiflorum)'s root. In the studies for calcium channel-blocking using Xenopus oocytes, the ethanol-extract ($26.2{\pm}5.2%$) showed higher activity than water-extract. Twenty female rats were fed 25, 35, and 45 mg/kg BW/day of the ethanol-extract for 14 days to observe the changes in blood pressures and heart pulses. Ethanol-extract decreased the systolic, diastolic, and mean blood pressures of the rats. Especially, the rats fed with 45 mg/kg BW/day of the ethanol-extract showed significant decreases in the blood pressures. These results suggested that a decrease in blood pressures was due to the extension of a blood vessel with calcium channel-blocking by ethanol-extract of Chinese balloon flower. Forty %-ethanol showed the highest efficiency for ethanol-extraction of Chinese balloon flower.
Objective : This experimental study was performed to investigate the effects of Houttuyniae Herba extract on anti-inflammation and anti-oxidation. Methods : The cytotoxicity of Houttuyniae Herba water extract and ethanol extract about viability of Raw 264.7 cell were tested using a colormetric tetrazolium assay(MTT assay). We investigated the inhibitory effects of Houttuyniae Herba water extract and ethanol extract on Propionibactrium acnes using paper disk diffusion method. To investigate the anti-inflammation effects of Houttuyniae Herba water extract and ethanol extract on LPS-induced macrophage Raw 264.7 cell, we used ELISA kit. We evaluated anti-oxidation effects of Houttuyniae Herba water extract and ethanol extract on HaCaT cell by Enzyme recycling method. Results : 1. In Houttuyniae Herba water extract and ethanol extract, cell toxicity depended on the density and wasn't difference between two extracts. 2. Houttuyniae Herba water extract and ethanol extract has not the significant inhibition effect of Propionibactrium acnes. 3. Concentration of 50, $100{\mu}g/m{\ell}$ Houttuyniae Herba water extract inhibited the production of NO in the Raw 264.7 cell stimulated with LPS. 4. All extracts except for $20{\mu}g/m{\ell}$ Houttuyniae Herba water extract showed anti-oxidation effect by decreasing the DPPH radicals. Conclusion : These results indicate that Houttuyniae Herba extract has anti-inflammation and anti-oxidation effects. If further study is performed, the use of Houttuyniae Herba extract will be valuable and benificial in the therapy of Propionibactrium acnes.
The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. The aim of this study was to investigate the effect of ethanol extract of Fagopyrum esculentum on the melanogenesis. To determine whether ethanol extract of Fagopyrum esculentum suppress melanin synthesis in cellular level, B16F10 melanoma cells were cultured in the presence of different concentrations of Fagopyrum esculentum ethanol extract. In the present study, we examined the effects of Fagopyrum esculentum ethanol extract on cell proliferation, melanin contents, tyrosinase activity, expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2). Cell proliferation was slightly increased by treatment with ethanol extract of Fagopyrum esculentum $(25-200 {\mu}g/m{\ell}).$ The ethanol extract of Fagopyrum esculentum effectively suppressed melanin contents at a dose of $100 {\mu}g/m{\ell}).$ It was observed that the color of cell pellets was totally whitened compared with the control. The ethanol extract of Fagopyrum esculentum inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. Using western blot analysis, the ethanol extract of Fagopyrum esculentum dose-dependently decreased tyrosinase and TRP-1 protein levels, and tyrosinase and TRP-1 were detected in similar manner. ${\alpha}-MSH$ leads to a stimulation of melanin synthesis through increase of tyrosinase activity, melanin contents and cytoplasmic dendricity. In this study, ethanol extract of Fagopyrum esculentum down-regulated the ${\alpha}-MSH$-induced tyrosinase activity, melanin contents and cytoplasmic dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase and TRP-1 was increased after incubation with a-MSH. The treatment of ethanol extract of Fagopyrum esculentum decreased the ${\alpha}-MSH$-induced expression levels of tyrosinase and TRP-1. These results suggest that the ethanol extract of Fagopyrum esculentum exerts its depigmenting effects through the suppression of tyrosinase, TRP-1 and cytoplasmic dendricity. And it may be a potent depigmetation agent in hyperpigmentation condition.
Objectives : In this study, the antioxidant activities of the 80% ethanol and hot water extracts of Euphorbia supina Rafinesque were investigated. Methods : We measured total phenol contents, flavonoid contents, 2,2-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging assay. The production of reactive oxygen species was measured in LPS-induced RAW 264.7 cells using flow cytometry system. Results : The content of phenol in the hot water extract was $65.529{\pm}0.462mg/g$ and $126.932{\pm}1.894mg/g$ in the 80% ethanol extract, and that of flavonoid in the hot water extract was $16.063{\pm}0.471mg/g$ and $29.159{\pm}1.963mg/g$ in the ethanol extract. The 80% ethanol extract also showed higher DPPH and ABTS radical scavenging activities ($90.8{\pm}1.0%$ and $92.5{\pm}0.7%$) than the hot water extract ($81.5{\pm}0.5%$ and $91.5{\pm}0.2%$). The production of reactive oxygen species(ROS) was reduced dose-dependently by 80% ethanol and hot water extract at concentration of 1, 10 and $100{\mu}g/m{\ell}$ of RAW 264.7 cells. Conclusion : According to these results, the 80% ethanol extract of Euphorbia supina Rafinesque has a good anti-oxidative effects than the hot water extract. Thus, the 80% ethanol extract of Euphorbia supina Rafinesque may serve as useful natural antioxidants.
Objectives : This study aimed to evaluate the effect of Wongisaengmaek-san (extracted with water or 50% ethanol) on antioxidative activity and recovery of fatigue induced by weight-loaded forced swimming exercise. Methods : Antioxidant activity of Wongisaengmaek-san was evaluated in terms of total amount of polyphenol, 1.1-diphenyl-2-picrylhvdroxyl (DPPH) radical scavenging activity and xanthine oxidase inhibition. The anti-fatigue effect of Wongisaengmaek-san was investigated using an acute weight-loaded forced swimming test by monitoring swimming test times and blood biochemical parameters creatinine. BUN, lactic acid dehydrogenase (LDH), free fatty acid (FFA) and lactic acid (LA). Results : 1. 50% EE (ethanol extract) had 2.3 times higher amount of total polyphenol compared to water extract. 2. It was identified that 50% ethanol extract showed enhancement of xanthine oxidase inhibition and DPPH scavenging effect in vitro. 3. According to anti-fatigue effect in the scale of 5% weight loaded mouse swimming test, both water and 50% ethanol extract showed significant improvement of swimming time elongation respectively, and 50% ethanol extract induced a more positive result. 4. After swimming for 1% weight-loaded mouse. water extract and 50% ethanol showed significant anti-fatigue effect with manifestation of blood sampling among both of the intervention: 50% ethanol extract showed a greater result. Conclusions : 50% ethanol extract of Wongisaengmaek-san has more effective antioxidant activity and anti-fatigue than water extract.
In Korea and China, cherry elaeagnus (Elaeagnus multiflora Thunb.) has been used traditionally to treat cough, diarrhea, itching, and foul sores. Therefore, in this study, the ethanol and water extracts of cherry elaeagnus leaves were examined for their antioxidant activities. The ethanol extract of the cherry elaeagnus leaves contained more phenolics than the water extract. All the cherry elaeagnus leaf extracts had higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability than ascorbic acid at concentrations of $250-1,000\;{\mu}g/mL$. The ethanol extract also showed higher superoxide dismutase (SOD)-like activity compared to the water extract. Furthermore, the SOD-like activity of the ethanol extract amounted to 89% of that of ascorbic acid at a concentration of $500\;{\mu}g/mL$. The nitrite scavenging ability and xanthine oxidase inhibitory (XOI) activity of the ethanol extract were higher than those of the water extract. In particular, the ethanol extract had higher XOI activity than ascorbic acid at a concentration of $1,000\;{\mu}g/mL$.

This study aimed to investigate the effects and epidermis penetration system with polymer micelle of Red Pinus densiflora extract. In the antioxidant test, the total concentration of polyphenol compounds was determined to be $137.5163{\pm}7.70mg/g$ in ethanol extract, $133.956{\pm}1.57mg/g$ in hydrothermal extract. The DPPH radical scavenging effects were $95.29{\pm}0.15%$ in ethanol extract at 1,000 mg/L. Elastase inhibition rates were $100.00{\pm}2.85%$ in ethanol extract at 2,000 mg/L. The antimicrobial effect of the ethanol extraction was higher than that of hydrothermal extractions. In the epidermal permeability experiment, it was confirmed that the permeation of the polymer micelle containing the Red Pinus densiflora's ethanol extract and cell penetrating peptides was remarkable. Here, we confirmed that ethanol extract of Red Pinus densiflora displayed excellent the effects in antioxidant test and epidermis penetration system with polymer micelle. As a result, Red Pinus densiflora extract has potential to be used as a safe and natural cosmetic material in the future.
