A series of 3-[1-(4-(2-methylpropyl) phenyl) ethyl]-1,2,4-triazole-5-thione (I) and its bicyclic condensed derivatives 6-benzylidenethiazolo[3,2-b]-1, 2,4-triazole-5(6H)-ones (IIa-IIf) were investigated for the prevention of ethanol-induced oxidative stress in liver and brain of mice. Administration of ethanol (0.1 mL/mice, p.o.) resulted in a drop of total thiol groups (T-SH) and non-protein thiol groups (NP-SH), and an increase in thiobarbituric acid reactive substances (TBARS) in both liver and brain tissue of mice (p<0.001). Among the compounds investigated (at a dose of 200 mg/kg, p.o.), I and IId ameliorated the peroxidative injury in these tissues effectively. Compounds IIa, IIc and IIe improved the peroxidative tissue injury only in brain. These findings suggest that certain condensed thiazolo-triazole compounds may contribute to the control of ethanol-induced oxidative stress in an organ selective manner.
The present study, antioxidant and anti-inflammatory effects were measured of 6 endemic plants in Indonesian extracted by hot water or ethanol. The Nipa Fruticans Wurmb ethanol extract (NFWE) and Orthosiphon aristatus ethanol extract (OAE) showed the highest polyphenol and flavonoid contents of 203.70 and 33.70 ㎍/mg, respectively. Antioxidant activity of OAE was highest in DPPH radical scavenging activity (77.49% at 10 ㎍/mL) and ABTS+ radical scavenging activity (93.36% at 10 ㎍/mL). FRAP activity was significantly higher in NFWE than other extracts. Anti-inflammatory effects of 6 endemic plants in Indonesian extracted by hot water or ethanol were examined using nitric oxide (NO) inhibition assays. In the LPS-induced BV2 cells, OAE showed the highest inhibition of NO production without toxicity. The results of this study, suggest that OAE is a potential functional raw material for antioxidant and anti-inflammatory.

The increased concern for the security of the oil supply and the negative impact of fossil fuels on the environment, particularly greenhouse gas emissions, has put pressure on society to find renewable fuel alternatives. Compared to the traditional biofuel, ethanol, higher alcohols offer advantage as gasoline substitutes because of their higher energy density and lower hygroscopicity. For this reason, microbial fermentation is known as potential producers for sustainable energy carriers. In this study, bacterial responses including cellular and molecular toxicity were studied in three different microorganisms, such as Escherichia coli, Clostridium acetobutylicum, and Saccharomyces cerevisiae. In this study, it was analyzed specific stress responses caused by ethanol and buthanol using four different stress responsive genes, i.e. fabA, grpE, katG and recA. The expression levels of these genes were quantified by semi-quantitative reverse transcription-PCR. It was found that four genes have shown different responsive patterns when E. coli cultures were under stressful conditions caused by ethanol and buthanol, respectively. Therefore, in this study, the stress responsive effects caused by these alcohols and the extent of each stress response can be analyzed using the expression levels and patterns of different stress responsive genes.
Alnus japonica cortex has been used as antidiarrhea, antihemorrhage and the remedy of indigestion. This study was performed to investigate the effectiveness of the methanol extracts of the Alnus japonica cortex on the gastric lesion and ulcer. The methanol extract was fractionated with hexane, chloroform and butanol, followed by bioassay on antigastritic and antiulcer activity. The methanol extract showed low acute toxicity with minimum lethal dose of more than 5000 mg/kg, p.o. in mice. The chloroform and the butanol fraction reduced gastric lesion in HCI. ethanol induced gastritic model. On gastric secretion in pylorus ligated rat, the hexane and chloroform fraction decreased the volume and acidity. The butanol fraction had significant inhibitory effects on aspirin and Shay's ulcer. The butanol fraction showed a tendency to inhibit the decrease of mucin secretion due to ingestion of absolute ethanol.
The comparison of topic effects of methanol and ethanol on rats and frogs was performed. Intraperitoneal administration in graded doses of absolute alcohols showed specific toxicities on animals, suggesting that these substances had different effects on species specificity of animals. Throughout the experiment, the following results were obtained and summerized here. 1. In rats $LD_{50}$ dose of absolute methanol was $2.65{\mu}l/g$ of body weight, while that of absolute ethanol was $1.85{\mu}l/g$ of body weight. 2. The $LD_{50}$ dose of methanol in frogs was $17.2{\mu}l/g$ of body weight, while that of ethanol was $10.9{\mu}l/g$ of body weight.

Alcohol drinking during pregnancy can result in abnormal fetal development including fetal alcohol syndrome (FAS). The molecular mechanisms of FAS, however, is not completely elucidated. In the present study, we evaluated the developmental toxicity of ethanol and its metabolite, acetaldehyde using post implantation whole embryo culture and determined changes of gene expression by ethanol treatment by cDNA microarray.(omitted)
A number of toxicants have been incriminated as a causing hepatic disease. Among many detrimental injury, alcohol has been noted for hepatitis, fatty liver, fibrosis, and hepatic cirrhosis. The purpose of this study was to develop animal model for hepatic fibrosis in pigs fed ethanol, and to search for a new anti-fibrogenic agent via this model. Twelve male Landrace pigs were divided into 3 groups of 4 animals each. Group 1, 2 and 3 were fed with active ceramic water only, ceramic water + liquid diet containing 15% ethanol and normal tap water + liquid diet containing 15% ethanol for 12 weeks, respectively. At week 12, all pigs were immediately sacrificed for collection each tissue and blood. Serologically, serum ALT and AST levels were significantly reversed in group 2, as compared to group 3. They were normal range in pigs of group 1. Microscopically, macrovesicular lipid droplets and moderate hepatocellular necrosis were evident in the tap water + ethanol fed group 3. However, the active ceramic water treated group 1 showed normal architecture. Moreover, in group 2, mild fatty changes and necrosis were observed in hepatocytes. Collagen fibers were increased in spaces surrounding periportal and interlobular connective tissues in the group 3 of tap water + ethanol, but collagen synthesis and its thickness of fibrotic septa connecting portal tracts were markedly reduced in the group 2 of ceramic water + ethanol. Myofibroblasts were detected mainly in the interlobular connective tissues of pig liver of group 3 treated ethanol and tap water. Few to no myofibroblasts were observed in groups 1 and 2. CYP2E1 was not or rarely detected in group 1 fed ceramic water. However, group 2 showed slightly activation of CYP2E1 in the area of pericentral vein, while CYP2E1 was significantly activated in group 3 fed tap water and ethanol. Based on the above data, we believe that we have developed a unique alcohol induced fibrosis model in pig, which will be useful in developing anti-fibrotic agents and drugs. Furthermore, the active ceramic water used in our study had an inhibitory and may be protective against ethanol induced hepatic toxicity and fibrosis.
Background; To explore the efficacy of aspartate as NAD regenerating agent for ethanol and acetaldehyde oxidation, we performed crossover challenge in two groups of volunteers by coadministration of various doses of aspartate, asparagine and ethanol. Methods; 18 healthy male volunteers were randomly divided into two groups. 6 volunteers of the first group were administered 5 gm monosodium aspartate(MSA), 5 gm asparagine or placebo with 100 ml of $40^{\circ}$ whiskey by the 3 way-crossover design, while 12 volunteers of the other group were administered placebo, 1, 2 or 5 bottles of $Aspar^(circledR)$ containing 1 gm of MSA per bottle with 100 ml of $40^{\circ}$ whiskey by the 4 way-crossover design. Ethanol(and acetaldehyde) concentrations in venous blood drawn at 0, 0.25, 0.5, 1, 2, 4, 8th hour after ethanol ingestion were analysed by gas chromatogaphy. Subjective symptoms, liver function tests and psychomotor function tests were also performed during the study periods. Result; Plasma concentration and AUC of acetaldehyde in asparagine and MSA trials on ethanol ingestion were significantly lower than those of placebo trial in the 1st group. Plasma ethanol concentration of 5 bottle $Aspar^(circledR)$ trial was significantly lower than that of placebo trial in the 2nd group. Improvement of subjective symptoms or psychomotor performance by the treatment was not statistically significant. Conclusion; Aspartate and asparagine may be prospective candidates for acceleration of ethanol metabolism and prevention of ethanol toxicity.
The effects of ethanol extracts from Asparagus cochinchinensis, Polggonatum officinale, Poria cocos, Panax ginseng and Acanthopanax sessiliflorum on the toxicity of polychlorinated biphenyls(PCBs) were examined and following results were obtained. By the administration of these extracts, PCBs intoxicated rats showed the normalizing tendency in body weights and various hematological parameters-counts of red blood cell and white blood cell, hemoglobin content and hematocrit value. These extracts ameliorated the elevated levels of serum cholesterol, total lipids, $_{8}-GOT-,\;_{8}-GPT$ and s-alkaline phosphatase activities of PCBs intoxicated rats.

This study was carried out to investigate the biological effect of deer's antler velvet on the testicular toxicity of rats exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD ). Thirty male rats were divided into three equal groups. The control group received vehicle (DMSO/acetone/soybean oil mixture) and saline; single dose of 50 $/mu$g/kg body weight TCDD was injected intraperitoneally into the single TCDD-treated and test group. Test group received ethanol extract of antler velvet (EAV) at daily dose of 20 mg/kg body weight for 5 weeks from one week before TCDD exposure. Decrease in body weight increment was less remarkable in test group compared with that of TCDD-treated group. TCDD-induced decrease in testicular weight, microtubular diameter and Johnson's score, and lesion were significantly alleviated by the treatment of EAV. This result led us to the conclusion that antler velvet can attenuate TCDD-induced testicular toxicity in rats.
