Objectives : The objective of this study is to study the effects of hot aqueous extract and ethanol extract from Paeoniae Radix Rubra on nitric oxide(NO) and prostaglandin $E_2(PGE_2)$ production in macrophage. Methods : Paeoniae Radix Rubra were extracted in 2 ways. one was extracted with hot aqueous for 4 hr in $100^{\circ}C$ and the other one was extracted with 70% ethanol for 4 hr in $70^{\circ}C$. RAW264.7 cells, a mouse macrophage lines, were incubated with different concentrations of the extract for 30 min and then stimulated with LPS at indicated times. Cell toxicity was determined by MTT assay. The concentrations of NO and $PGE_2$ were measured by griess assay and enzyme immunoassay(EIA). Results : The hot aqueous and ethanol extracts of Paeoniae Radix Rubra significantly inhibited the NO productions in LPS-stimulated RAW264.7 cells. The hot water extract of Paeoniae Radix Rubra significantly inhibited the $PGE_2$ productions in LPS-stimulated RAW264.7 cells. Conclusions : Our results demonstrated that Paeoniae Radix Rubra extract is able to significantly inhibit the production of NO, $PGE_2$ expression. Hot aqueous extract of Paeoniae Radix Rubra has more effective anti-inflammation than ethanol extract.
In this study, we compared the anti-inflammatory activity of Pinus koraiensis cone bark extracts prepared by conventional extraction and microwave-assisted extraction (MAE). Water extracts and 50% ethanol extracts prepared using MAE were applied to RAW 264.7 cell at 5, 10, 25, and $50{\mu}g/mL$ of concentrations, and tested for cytoxicity. The group treated with $50{\mu}g/mL$ of 50% ethanol extracts showed toxicity. In order to investigate the inhibition of nitric oxide (NO) production in RAW 264.7 cells, extracts of water and ethanol were treated with 5, 10, and $25{\mu}g/mL$ concentrations. The inhibitory activity of water and 50% ethanol extracts groups were determined as 40% and 60% at $25{\mu}g/mL$ concentration, respectively. We found concentration dependent decreases on inducible NO synthase. The inhibitory effect against forming inflammatory cytokines, prostaglandin $E_2$, tumor necrosis factor-${\alpha}$, interleukin (IL)-6, and IL-$1{\beta}$, was also superior in the $25{\mu}g/mL$ treated group than the control group. According to these results, the water extracts and 50% ethanol extracts both inhibited inflammatory mediators by reducing the inflammatory response. Therefore, The MAE extracts of P. koraiensis cone bark can be developed as a functional ingredient with anti-inflammatory activity.
In this study, the anti-inflammatory effect of hyaluronidase (HAase) inhibition was determined from 92 species of oriental herbal medicine extracted with water and ethanol solvents because of their non-toxicity in the human body. The water extracts of Evodia officinalis (86.8%), Thuja orientalis (80.8%), Carthami semen (66.5%), Melia azedarach (74.7%), Siegesbeckia pubescens (61.3%), Saururus chinensis (49.15%) showed a relatively greater anti-inflammatory activity. The ethanol extracts of Ailanthus altissima and Saururus chinensis demonstrated the highest anti-inflammatory effect at above 90%. Saururus chinensis was selected for its high anti-inflammatory effect in both water and ethanol extract. Ethanol was more effective than water and optimal extraction conditions for phenolic compounds was determined to be extraction with 50% ethanol for 12 hours. The extracts from Saururus chinensis in optimal condition showed 70~80% anti-inflammatory effect when $100{\sim}250{\mu}g/mL$ phenolic concentration was treated. Concentration of above $500{\mu}g/mL$ decreased the inhibitory effect. The anti-inflammatory effect and extraction yield were increased by ultra-fine grind technology, indicating that this method can be used to increase the extraction yield of phenolic compounds from medicinal plants.

In this study, the inflammation of ethanol extracts from Caryopteris incana (CI) and fermented C. incana (FCI) on induced to lipopolysaccharide with Raw 264.7 cell was tested. The composition profile of L. plantarum was changed by fermentation, and confirmed by HPLC analysis. We performed the 3-[4,5-dimethylthiazol]-2-yl]-2,5-diphenyltetrazolium bromide assay to evaluate the toxicity of CI and FCI extracts. In cell viability, cell toxicity was not shown at 5, 10 and $15{\mu}g/mL$ of CI extracts and 10, 20, 30 and $40{\mu}g/mL$ of FCI extracts. The results of inducible nitric oxide synthase and cyclooxygenase-2 protein production were confirmed to be inhibitory in a concentration-dependent manner, respectively. Additionally, protein expression of nitric oxide and prostaglandin $E_2$ by CI and FCI extracts were also inhibited in a concentration-dependent manner. In the result of pro-inflammatory cytokine, $15{\mu}g/mL$ concentration of CI extracts was showed tumar necrosis factor $(TNF)-{\alpha}$ (57.3%), interleukin (IL)-6 (35.2%), and $IL-1{\beta}$ (48.0%), respectively. And $40{\mu}g/mL$ of FCI extracts was showed $TNF-{\alpha}$ (34.6%), IL-6 (32.1%), and $IL-1{\beta}$ (30.0%), respectively. These results suggest that FCI extracts showed better effect of anti-inflammatory than CI extracts. Therefore, it was found that both CI and FCI can be used as an excellent material for the development of new anti-inflammatory resource.

In the present study, we investigated the oral toxicity of Amomum tsao-ko Crevost et Lemaire, (Zingiberaceae) extract in Balb/c mice (BALB, n=60) for 3 weeks. Balb/c mice (10 mice/group, 6 group, $20{\pm}2g$, 6 weeks) were orally administered for 21 days, with dosage of 250, 500, 1000, 2000 mg/kg/day. Ethanol extract of A. tsao-ko did not affect any significant change of mortality, clinical signs, organs and body weights. Also, there were not significantly difference from the naive group (control) in hematological and serum biochemical examination. Consequently, these findings indicate that 3-week treatment with the ethanol extract of A. tsao-ko was not any toxic effects in Balb/c mice and the no-observed adverse effect level (NOAEL) for oral toxicity was determined to be 2000 mg/kg/day under our experimental conditions.

We investigated the anti-inflammatory activities of various organic solvent extracts with and without Lactobacillus plantarum fermentation of Aralia continentalis Kitagawa which has hypotensive effects in addition to excitatory effects on the central nervous system. It has been used to treat arthritis, colds, neuralgia, rheumatism, and itchy skin. Our extracts were tested for their anti-inflammatory potential on NO production and the expression of inflammatory factors in lipopolysaccharide-stimulated RAW264.7 macrophages. Extracts with and without L. plantarum fermentation were prepared using water, ethanol, hexane, ethyl acetate, and butanol. The RAW264.7 cells were tested for toxicity and the anti-inflammatory activity of each extract was determined at a concentration with no toxicity to the cells. The extracts used in this study significantly inhibited both the production of NO and the mRNA expression of COX-2 and iNOS, the major inflammatory factors. The production of inflammation-related cytokines $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ was also significantly reduced. These results suggest that the extracts involving fermentation by L. plantarum can inhibit cytokines by controlling the expression of inflammatory cytokine genes. It is considered that the water, ethanol, and butanol extracts after fermentation with L. plantarum could be useful as functional natural materials with anti-inflammatory effects.
BACKGROUND/OBJECTIVES: Adipogenesis is part of the cell differentiation process in which undifferentiated fibroblasts (pre-adipocytes) become mature adipocytes with the accumulation of lipid droplets and subsequent cell morphological changes. Several transcription factors and food components have been suggested to be involved in adipogenesis. The aim of this study was to determine whether mulberry leaf ethanol extract (MLEE) affects adipogenesis in 3T3-L1 adipocytes. MATERIALS/METHODS: The 3T3-L1 adipocytes were treated with different doses of MLEE for 8 days starting 2 days post-confluence. Cell viability, fat accumulation, and adipogenesis-related factors including CCAAT-enhancer-binding protein alpha ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$), $PPAR{\gamma}$ coactivator 1 alpha (PGC-$1{\alpha}$), fatty acid synthase (FAS), and adiponectin were analyzed. RESULTS: Results showed that MLEE treatments at 10, 25, 50, and $100{\mu}g/ml$ had no effect on cell morphology and viability. Without evident toxicity, all MLEE treated cells had lower fat accumulation compared with control as shown by lower absorbances of Oil Red O stain. MLEE at 50 and $100{\mu}g/ml$ significantly reduced protein levels of $PPAR{\gamma}$, PGC-$1{\alpha}$, FAS, and adiponectin in differentiated adipocytes. Furthermore, protein level of $C/EBP{\alpha}$ was significantly decreased by the treatment of $100{\mu}g/ml$ MLEE. CONCLUSION: These results demonstrate that MLEE treatment has an anti-adipogenic effect in differentiated adipocytes without toxicity, suggesting its potential as an anti-obesity therapeutic.
1) Higher yield of red color was observed by the isolated strain (Monascus D-7) than the type cultures in steamed rice medium. 2) In a case of Monascus purbigerus IAM 8004, best yield of color was obtained at Lin's submerged culture medium containing 1% wheat bran, 2% starch and 3% corn meal instead of rice powder as carbon source. However, in a case of isolated strain (M. D-7), good result was shown at 1% rice bran and 2% starch as a source of carbon in Lin's medium. 3) Good yields were obtained from both strains in Nishikawa's medium which was added with 3% defatted soybean flour. 4) There were no significant differences in pigment extractability among solvents. Extracted pigment was stable in wide range of pH and heat, whereas relatively unstable in sunlight. 5) Toxicological study of extracted pigment determined $LD_{50}$ at 0.2539g/20g, when injected in mouse. When injected in to mouse in 25% ethanol solution: considering the toxicity of ethanol, the toxicity of pigment itself is believed to be none.
Objectives: Wild ginseng pharmacopuncture is widely used in oriental medicine. However, there is no standard method for efficiently extracting the active ingredient. In this study, in order to determine an efficient extraction method, wild ginseng was extracted by the distillation and 70% ethanol reflux methods, respectively. In comparing each extract, the index compounds were analyzed, and antioxidant activity was measured. Methods: The index compounds, ginsenoside Rg1 and ginsenoside Rb1, were detected using high performance liquid chromatography (HPLC). Antioxidative activities of total phenolic compounds, DPPH (${\alpha}$, ${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) and FRAP (ferric reducing antioxidant power) were measured to compare their bioactivities. Since saponin is known to be hemolytic, the hemolytic activity of each extract was compared. Results: The index compounds were analyzed. Nothing was detected in the wild ginseng distilled extracts (WGDE). In the wild ginseng 70% ethanol reflux extracts (WGEE), ginsenoside Rg1 was 3.66 mg/g, and ginsenoside Rb1 was 16.70 mg/g. WGEE showed higher levels than WGDE in all antioxidative activities. In the hemolytic test, the extracts showed almost no toxicity, but WGEE showed lower toxicity than WGDE. Conclusions: In this study, it was concluded that WGEE is more advantageous than WGDE in the detection of index compounds and bioactivity. However, additional studies of additional extraction methods and other bioactivity tests are needed.

The critical role of folate in the remethylation pathway for methionine synthesis from homocysteine has been well documented. Hyperhomocysteinemia resulting from inadequate folate nutrition has been implicated in increased incidence of macrovascular diseases, colorectal cancer, neural tube defects, etc. Chronic exposure to ethanol impairs folate nutrition and one-carbon metabolism in the liver, which often results in fatty liver due to a defective remetylation process. This study was carried out to investigate the chronic effects of moderate levels of alcohol and dietary folate on plasma homocysteine levels, and on histopathology and biochemical functions of the liver. Rats were raised on experimental diets with three levels of folate (0, 2, 8 mg/kg diet), and 50% ethanol (1.8 ml/kg body weight) was administered intragastically by intubation tubes three times a week for 10 weeks. Plasma homocysteine concentrations were found to be significantly influenced by dietary folate intake and alcohol administration. Among all treatment groups, plasma homocysteine levels were the highest in the animals receiving a combined treatment of folate deficient diet and alcohol administration. Plasma homocysteine concentrations were negatively correlated with folate concentration in the plasma (p<0.01) and liver (p<0.05). Among alcohol treated rats, increase in plasma homocysteine values due to macrovascular and microvascular fatty changes and spotted necrosis were observed more frequently in folate-deficient animals diet than those on folate-adequate and folate supplemented diets in alcohol-treated rats. These results indicate that folate supplementation above the recommended level might be beneficial in the prevention of alcohol-related hyperhomocysteinemia and abnormal histologic changes in the liver.
