활엽수 알칼리 선추출물 내 헤미셀룰로오스의 분리 및 정제 (Isolation and Purification of Hemicelluloses in Alkali Pre-extractives from Mixed Hardwood)

  • In this study, hemicelluloses were pre-extracted from mixed hardwood chips using sodium hydroxide solution and then they were isolated from pre-extractives by using various solvents. Isolation was conducted by precipitating hemicelluloses with isopropanol, ethanol, 1,4-dioxane, dimethylsulfoxide (DMSO) and potassium hydroxide (KOH), respectively. The precipitate yield on alkali pre-extractives was the highest when hemicelluloses were isolated by DMSO and then precipitated with ethanol. Most precipitates were yellow colored. The efficiency of isolation was evaluated by analysing the characteristics of isolated hemicelluloses. Isolation using DMSO, KOH and 1,4-dioxane ($80^{\circ}C$) showed rather high efficiency. The highest total separation efficiency was about 77% when alkali pre-extractives were reacted with KOH and precipitated with ethanol. The quantity and purity of isolated hemicelluloses were affected by the solvent type.

Comparison of sericin produced through laboratory- and plant-scale extraction

  • In this study, the structural characteristics of sericin recovered from wastewater released from the silk textile industry (Plant sericin) were comparatively analyzed with those of sericin extracted from a silkworm cocoon produced in a laboratory (Lab sericin). To prepare Plant sericin, ethanol was added to wastewater (i.e., a sericin aqueous solution) after the degumming process to remove nonprotein materials, affording a sericin precipitate. To prepare Lab sericin, nonprotein materials were removed from a silkworm cocoon and sericin was subsequently extracted from the cocoon. Lab sericin and Plant sericin exhibited similar solution viscosities, gel strengths, and crystallinity indices, indicative of the similar molecular weights (MWs) of the two sericin samples. In the case of sericin powder, Plant sericin was more crystalline than Lab sericin due to its treatment with ethanol. The findings of this study revealed that sericin recovered from industrial wastewater can be used equally as its MW is similar to that of sericin obtained through laboratory-scale extraction.

목이버섯으로부터 추출한 항혈전물질의 제품화와 동물실험을 통한 항혈전활성 검증 (Production of Antithrombotic Material Extracted from Auricularia auricular-judae and the Verification of Its Antithrombotic Activity via Animal Test)

  • 목이버섯으로부터 항혈전물질을 추출하여 제품화하기 위한 대량생산공정을 다음과 같이 확립하였다. 즉, 건조된 목이버섯을 분쇄하여 75% ethanol로 추출하여 원심분리한 침전물에 증류수에 첨가한 후 $76^{\circ}C$에서 2시간 추출한 후 압착여과하여 여과액을 얻는다. 이를 감압농축한 후 여기에 최종 농도가 80%가 되도록 ethanol을 첨가하여 추출한 침전물을 동결건조한다. 제품의 소비지 기호도 조사를 통해 부재료의 종류와 첨가량을 결정하여 배합비를 목이버섯 추출물 90.5%, 고과당 2.0%, $\beta$-cyclodextrin 1.5%, 올리고 당 2.0%, 배 농축액 4.0%로 결정하였다. 제품을 포장하여 25, 37, $45^{\circ}C$에서 저장하였을 경우 저장 8주에서도 수분활성도, 수분함량, pH, 산도에 큰 변화가 없었다. 미생물 생균수는 $25^{\circ}C$$37^{\circ}C$에서 저장하였을 경우 저장기간이 지남에 따라 약간 증가한 반면, $45^{\circ}C$에서 저장하였을 경우에는 저장 2개월 후에는 대부분 사멸하였다. 제품을 쥐에 정맥투여하였을 경우 1,000 mg/kg 투여 시 APTT 활성, TT 활성, PT 활성, FIB 활성이 대조구와 비교하여 유의적으로 높았으며 500 mg/kg을 경구투여하였을 경우 항혈소판활성이 아스피린과 동일한 수준을 나타내었다.

Fabrication and Micropatterning of a Hybrid Composite of Amorphous Calcium Carbonate and Poly(ethylenimine)

  • Amorphous calcium carbonate (ACC) can readily be prepared using ethanol as the reaction medium and ammonium carbonate as the source of carbon dioxide. Other additives, or any elaborate pH control are not needed to form the initial calcium carbonate precipitate. Ammonia generated from ammonium carbonate maintains the reaction medium in a neutral or weakly basic condition, retarding the crystallization of ACC, while ethanol itself inhibits the dissolution of ACC. The ACC prepared in this way provides a rare opportunity to fabricate molded biomimetic crystals in vitro, but the ACC is too fragile to be fabricated into proper shapes. The malleability of ACC is, however, greatly enhanced by incorporating poly(ethylenimine) (PEI). The ACC/PEI composite can then be fabricated, using a proper mold or template, into mechanically durable biomimetic crystals of definite shape. The ACC in the ACC/PEI composite can further be transformed into vaterite by heating under N2 atmosphere, while the native ACC simply converts into calcite.

고려홍삼 다당체 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향 (Effect of Korean Red Ginseng Polysaccharide on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid)

  • This study was devised to observe the inhibitory effects of ethanol treatment precipitate (crude acidic polysaccharide) from Korean red ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. The yields of crude acidic polysaccharide from Korean red ginseng was 63.5%. In vitro, at the concentration of 500rg /ml, the inhibition rate of lipolysis by the crude acidic polysaccharide of Korean red ginseng was 38.8% and the total inhibitory activity per gram of ginseng material was 4,928 nit. In vivo, the red ginseng polysaccharide(40mg/ml in saline soon.) 16ul/g of body weight was injected to the sarcoma-180 bearing mice once In 3 days until death. The effects against the extension of life span was little but body weight gain of sarcoma-180 bearing mice decreased significantly by administration of Korean red ginseng polysaccharide compared to those of the control group.

영지버섯 다당체의 Nitric Oxide 생성능 및 생성기전 연구 (Nitric Oxide Production Ability and its Formation Mechanisms in Macrophage TIB 71 Cell Line by Polysaccharide Extracted from Ganoderma lucidum)

  • This study was carried out to get infomation on the nitric oxide production ability and its formation mechanisms of polysaccharides extracted from Ganoderma lucidum(PSG) by using murine macrophage cell line. The cultured mycelial cells of Ganoderma lucidum were extracted by alkali, and than neutralized by acid. The extract were passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and PSG was obtained. The immunomodulating effects of PSG on macrophage were performed by using murine macrophage cell line ATCC TIB 71 cells with PSG 0.5mg. PSG alone could not induce the production of nitrite, but it had a significant potential effect on nitrite secretion when the cells were primed and triggered with BCG and Interferon(IFN)-${\gamma}$. Also it was prominent by using calcium channel blocker(verapamil) and adenylate cyclase activator(forskolin).

  • To investigate a possibility of producing the antitumor component by shake culture method, the mycelia of Flammulina velutipes were cultured in flasks on a shaker at $26{\sim}28^{\circ}C$ at 180 rpm for seven days. The extract of the mycelia was concentrated under vacuum. The precipitate obtained by adding a three-fold volume of ethanol was centrifugated and freeze-dried after dialysis. The fraction was tested against sarcoma 180 in the mice. The inhibition ratio of the fraction against the tumor was 68.0% at the dose of 20 mg/kg/day for the period of ten days and the tumors in three of the mice were completely regressed. The results showed, therefore, that the antitumor component was produced by the shake culture method.

영지버섯에서 분리한 항암성 다당체에 관한 연구 (Studies on the Polysaccharide Extracted from Ganoderma Incidum)

  • The cultured mycelial cells of Ganoderma lucidum was extracted by alkali, and then neutralized by acid. The extract was passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and then PSG(polysaccharides) was obtained. PSG was composed of 82.2% polysaccharide, 0.7% protein and 17.1% uronic acid. Sugar conjugates of its hydrolysates were produced using with fluorescent compound(7-amino-1,3-naphthalene disulfonic aicd : 7-AGA), and then fluorescent labeled sugar conjugates were separated by reverse phase high perfomance liquid chromatography. Hydrolysates of PSG were composed of sixteen amino acids and 95.7% glucose, 2.7% xylose, 1.6% fucose and tract amount of galactose and mannose. The immunomodulating effects of PSG on macrophage were perfomed using murine macrophage cell line ATCC TIB 71 cells. PSG augumented the phagocytic activity of TIB 71 cells against fluorescent latex beads.

기호음료 성분의 아질산염 소거작용 (The Nitrite-scavenging Effects by the Component of Traditional Tea Materials)

  • 전통기호음료성분의 기능특성을 조사하기 위한 연구의 일환으로 결명자, 들깨, 대추, 모과, 오미자, 오갈피 및 생강 추출물을 여러가지 획분으로 분획하여 아질산염 소거작용을 조사한 결과, 본 실험에 사용한 시료는 모두 아질산염 소거작용이 있었으며, 수용성 획분을 에탄올 가용성 획분이 에탄올 침전획분으로 분획하였을 때, 결명자, 들깨, 오갈피, 생강의 경우에는 에탄올 가용성 획분이 에탄올 침전획분보다 아질사염 소거작용이 뛰어났으며 대추, 모과, 오미자의 경우에는 그 반대의 경향을 나타내었다. 시료 추출물 중에 함유되어 있는 환원성 물질의 관여도 클 것으로 생각되어 전자공여작용을 조사해 본 결과, 기호음료 원료에서 추출한 수용성 획분의 전자공여 작용은 오갈피>들깨>모과>생강>대추>오미자>결명자의 순으로 나타났으며, 에탄올 침전획분과 에탄올 가용성 획분의 전자공여효과도 아질산염 소거효과와 경향이 일치하였다.

Cordyceps miiitaris에 함유된 혈당강하 성분이 간세포 Glucokinase활성에 미치는 영향 (Cordyceps militaris Increases Hepatic Glucokinase Activities)

  • 본 연구에서 는 P japonica와 c. mifitaris에서 혈당강하 기능성 성분을 분리하고 간세포내 glucokinase활성을 측정하여 항당뇨 작용 기전을 규명 하고자 하였다. 동충하초에서 분리 된 fraction A의 간세포내 glucokinase 활성은 대조군과 비교하여 C. militaris는 371.4%, P. japonica는 379%로 증가하였다. Fraction B의 활성은 C. miliraris 314.2%, P. japonim는 147.4% 증가하여 C. milifaris에서 추출된 fraction B는 P. japonin와 비교했을 때 2배 이상의 glucokinase의 활성 증가를 나타냈다. 이상의 결과를 근거로 C. militaris의 혈당 강하 작용을 in vivo에서 검증한 결과 C. militaris 첨가군의 glucokinase활성은 대조군에 비해 1300% 이상의 활성 증가를 나타냈다. 결론적으로 C. militaris추출물은 간세포 내 glucokinase활성을 현저히 증가시킴으로써 혈중 포도당의 급격한 상승을 억제할 수 있을 것으로 판단된다.
