A single large dose of ethanol as well as chronic ethanol consumption produces alcoholic fatty liver in human and experimental animals. We examined the effects of YH439, a potential hepatoprotective agent, on alcoholic fatty liver generation in adult female rats. In rats treated with YH439 (250 mg/kg, po) 4 hr prior to a single dose of ethanol (6 g/kg, po), a significant decrease in hepatic triglyceride accumulation was observed. YH439 also has an inhibitory effect on hepatic triglyceride and cholesterol accumulation induced by repeated ethanol treatments for one week. Because it has been known that induction of alcoholic fatty liver is associated with lipid peroxidation and/or hepatic glutathione depression, the effect of YH439 on these parameters was determined in the livers of rats treated with ethanol. Coadministration with YH439 inhibited MDA formation and gIutathione depression induced by acute or repeated ethanol administration. In order to determine the effect of YH439 on ethanol metabolism in vivo, disappearance of ethanol from blood was measured. In rats treated with a single dose of ethanol (6 g/kg, po), the ethanol concentration in blood reached a peak approximately 120 min following the treatment which declined linearly for 18 hrs. YH439 had no effect on the decline of blood ethanol concentration regardless of the dose of ethanol given to rats. These results in this study suggest that YH439 has an inhibitory effect on fatty liver generation induced by acute or repeated ethanol consumption through a mechanism not directly related to the rate of ethanol metabolism in vivo.

This study was performed to investigate effects of ethanol and dietary protein levels on protein metabolism in 15-week-old male rats given a normal diet. Rats were divided into 8 groups : control group (165 protein, 16 PC) and 8%(8PE), 16%(16PE) and 24% protein groups (24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight gain, organ weight, serum glucose concentration and liver protein concentrations were not affected by either ethanol or dietary protein levels. Serum total protein concentrations after 10 weeks were significantly increased in the 16 PE and 24PE groups compared with that of control group. Serum albumin concentrations after 10 weeks were significantly increased in all the ethanol-administered groups than that of control group. Fecal and urinary nitrogen metabolism wee not affected by ethanol.

Objective: The purpose of this study was to determine the effects of feeding ethanol on growth performances, carcass characteristics, and lipid metabolism of finishing Korean cattle (Hanwoo) steers. Methods: Thirty (30) Hanwoo steers (average 25.1 months of age, body weight 660.1 kg) were assigned to three treatments: control (0% ethanol), E-3 (1.44% ethanol for 3 months), or E-5 (0.72% ethanol for 2 months followed by 1.44% ethanol for 3 months). The animals were allotted by treatment group into six pens and fed concentrate and perennial ryegrass. Ethanol (30%, v/v) was supplemented into drinking water twice a day to meet final concentrations based on average water consumption of finishing Hanwoo steers. Results: There were no statistical differences among the groups in final body weight, average daily gain, or carcass yield grade indices such as cold carcass weight, fat thickness, and loin area. The marbling score tended (p = 0.228) to increase with the highest score (6.7) in the E-5 group followed by 6.3 and 6.0 in E-3 and control groups, respectively. The appearance frequencies of quality grades of $1^{{+}{+}}$ (the best), $1^{+}$, 1, and 2, were; 30%, 50%, 0%, and 20% for control, 10%, 80%, 10%, and 0% for E-3, and 10%, 80%, 0%, and 10% for E-5 groups, respectively, indicating improvements of quality grades by feeding ethanol. Concentrations of serum glucose tended to decrease whereas those of insulin and non-esterified fatty acid to increase by feeding ethanol (E-3 and E-5; p>0.05). Conclusion: Feeding ethanol directly into drinking water of finishing Hanwoo steers stimulated lipogenesis in intramuscular adipose tissue (marbling) and thereby improved carcass quality grade. The serum metabolites results supported the hypothesis of lipolysis of existing adipose tissue, such as abdominal fats, and lipogenesis in intramuscular adipocytes.
Disturbance of antioxidant system is very common in chronic alcoholics and herbal or natural products with antioxidant activity have been used for its treatment. This study was to investigate the effect of Vitis vinifera extract(V), Schisandra chinensis extract(S), Taraxacum officinale extract(T), Gardenia jasminoides extract(G), Angelica acutiloba extract(A) and Paeonia japonica extract(P), and their combinations on the antioxidant and ethanol oxidation system. Male Sprague-Dawley rats were subjected to Lieber-DeCarli ethanol liquid diet(ED) and were then given different herbal extract mixtures for 6 weeks including VST(V 100+S 150+T 150mg/kg/day), VSG(V 100+S 150+G 150mg/kg/day), VTG(V 100+T 150+G 150mg/kg/day), and VAP(V 100+A 150+P 150mg/kg/day). When the activity of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were compared between ED only group and herbal extracts treatment group, the differences were statistically significant. Phase I and II(glutathione-S-transferase, phenol sulfatransferase) enzyme activities were found to be significantly higher in the VAT treatment group compared to the ED group. Herbal extracts not only repressed the ethanol-induced elevation of malondialdehyde level, but also protected against ethanol-induced decrease in glutathione content, glutathione reductase, glutathione peroxidase, catalase and superoxide dismutase activities. The administration of the herbal extracts was found to be effective in eliminating lipid-peroxides induced by long-term consumption of alcohol by activating various enzyme systems and physiological active compound formation system. After a chronic consumption of alcohol, Angelica Radix protected the liver via activating the ethanol-metabolism enzyme system, and Paeoniae Radix via activating the ethanol-metabolism enzyme and the phase I, II-metabolism enzyme system. Taraxaci Herba was also effective in liver protection via activating the ethanol-metabolism enzyme system and the phase I, II-metabolism enzyme system, Gardeniae Fructus via activating the phase II-metabolism enzyme system and the anti-oxidation system enzyme, and Schisandra Fructus and a grapestone via activating the anti-oxidation system. Our data suggest that these herbal extracts may be useful as a health functional food or new drug candidate for fatty liver and hepatotoxicity induced by chronic alcohol consumption.
The effect of dietary carnitine on ethanol-induced fattv liver and hvpertriglyceridemia was examined in an animal model. Consistent with literature, ethanol fed at 5g/Kg of b.w. to rats produced a significant increase in hepatic concentrations of total lipid, kislycerine, phospholi-pid, free cholesterol and esteriaed cholesterol as well as elevated plasma concentrations of triglvceride. It was when the ethanol diet was supplemented with D.L. camitine that there was a singini-cant reduction in the accumulation of lipids in the ethanol-compromised liver. Dietary cacti-tine was also effective in ameliorating ethanol-induced hypeuriglyceridemia. Total protein con-tents in the plasma was not varied among the groups. Ethanol의 대사과정에 관여하는 영향중에 특징적인 것으로 과유지방혈증(hyperlipidemia) 까 지방간(fatty liver)을 거쳐 간경변에 이르는 간에 관계하는 일련의 증상들을 들 수 있다. 본 실험에서는 만성적 ethanol의 지방대사 장해에 대한 D.L.-carnitine의 효과에 대해 고찰하였다. 실험용 횐쥐를 사용하여 실험군(ethanol group)에게 체중 kg당 5g의 ethanol(30% in saline)을 투여하여 알콜유발성 지방간과 과유지방혈증을 일으키고, 그 실험군 흰쥐들에게 carnitine(0.4 mg/g of body weight)을 첨가하여 그 효과를 관찰하였다. 그 결과 carnitine을 첨가하여 투여한 흰쥐들에서 ethanol처리군과 비교하여 볼 때 간과 혈장에서 지방축적이 현저히 감소하는 것을 관찰할 수 있었다.
Citrus sunki Hort. ex Tanaka is one of several Jeju-native citrus fruits. A number of biological properties for citrus fruits have been suggested, however little is known about those of C. sunki. The present study tested whether fermented product of C. sunki peel (FSP) might affect the activities of enzymes regulating ethanol metabolism. Effects on body weight gain as well as on fatty liver formation were also investigated. The activities of alcohol dehydrogenase and aldehyde dehydrogenase were stimulated remarkably by FSP. Excessive ethanol-induced cytotoxicity was also prevented by FSP in HepG2 cells. FSP decreased the weight gain and fatty liver formation induced by a high-fat diet in mice. From these results, FSP might be a potent source of neutraceuticals useful for preventing ethanol-induced health problems.
This study was conducted to investigate the physicochemical properties of the ethanol extracts and soluble dietary fiber from Cassia tora L. seed. The proximate composition of Cassia tora, soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract, molecular mass distribution, sugar contents and viscosity of soluble fiber from Cassia tora were analyzed. Cassia tora contains 12.6% of moisture, 5.2% of ash, 13.4% of crude protein, 7.2% of crude fat, 8.8% of insoluble fiber and 48.3% of soluble fiber. The effects of extract condition on soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract were investigated. The soluble solid contents were higher in 70% or 50% ethanol extracts than those in 100% ethanol extracts and showed highest value in grind sample extracts. In Hunter's color value, 100% ethanol extracts and whole Cassia tora sample extracts were higher in L and b value, but on the contrary, were lower in a value, than those of the other. The highest emodin and rhein contents were observed in 70% and 50% ethanol extracts, respectively, and showed higher value in room temperature extracts than in heating extracts. The molecular mass of soluble fiber from Cassia tora seed was estimated by gel filtration chromatography. Most soluble fiber(80%) exhibited a molecular mass range of between 50~2000 kDa. The major sugars of soluble fiber from Cassia tora seed were identified as xylose, mannose and galactose. The apparent viscosity of 0.5% soluble fiber from Cassia tora seed was 33 mPas showing a higher value than pectin or xanthan gum.
This study was performed to investigate effect of dried powder or ethanol extracts of onion flesh and peel intakes on lipid metabolism, antioxidative and antithrombogenic capacities in l6-month -old rats. Total of 40 Sprague-Dawley male rats of l6-month-old and weighing 816$\pm$6g were blocked into 5 groups according to body weight and raised for three months with control and experimental diets containing 5% (w/w) of dried powders of onion flesh or peel or ethanol extracts from equal amount of each dried powder. Contents of total flavonoids and total dietary fibers in peel powder were highest among onion preparations. Body weight gain and epididymal pad fat weight were lower in peel powder group than other groups. Plasma total lipid, triglyceride and total cholesterol concentrations of onion-containing groups were lower than control group. Above all, peel ethanol extract intake decreased them most remarkably. Plasma HDL-cholesterol concentrations in onion-containing groups were higher than control group, especially that of flesh powder group was the highest among groups. Liver total lipid, triglyceride and total cholesterol concentrations were not significantly different among all experimental groups. However, liver total lipid and triglyceride concentrations were tended to be lower in onion-containing groups than control group. Thiobarbituric acid reactive substances (TBARS) concentrations in LDL + VLDL fraction was not significantly affected by onion intakes, However peel powder group showed the lowest concentration, Plasma TX $B_2$ concentrations in onion flesh powder, peel powder and peel ethanol extract groups were lower than control group, while plasma 6-keto-PG $F_{1a}$ concentrations in these same groups were higher than control group. Clotting time was tended to be increased in peel ethanol extract group. In conclusion onion diets seemed to improve lipid metabolism and antithrombogenic capacity while effect on antioxidative was not significant.t.
Excessive ethanol intake is known to induce a number of physiological symptoms, including headache, dizziness and vertigo. In this study, we investigated the attenuation effect of sprouted peanut extract (SPE) on ethanol-induced hangover in male Sprague-Dawley rats. The animals were divided into five groups: the control group, which was administered ethanol only; the ethanol plus SPE experimental groups, which were administered ethanol and 100, 200, or 400 mg SPE/kg b.w.; and the positive control group, which was administered ethanol plus DAWN808$^{(R)}$, a commercial product. SPE-suspended water was delivered to rats via gavage 15 h and 30 min before the administration of ethanol. Blood was collected from the tail 0, 1, 3, and 5 h after ethanol administration. The results showed that serum ethanol concentrations were significantly lower in SPE treated groups than in the control group. Furthermore, hepatic alcohol and acetaldehyde dehydrogenase activities were enhanced by SPE in a dose dependent manner. These results suggest that SPE could be useful in attenuating hangover after alcohol consumption.
This study was designed to observe the effect of ethanol extracts from fish flour on the nucleic acid metabolism in rats. Young rats, weighing 75-85g were used as the experimental animals and diet used were 8 kinds; diet supplemented with 10% fish flour, diets which were supplemented with the extracts and or remainders of fish flour after extracting by either 76% or 96% ethanol to the rice diet, respectively, and diet supplemented with 6% casein. After feeding corresponding diet for 40 days, RNA and DNA contents, and DNase activities in the liver, kidney and braid were determined. The results obtaioed from this study are summarized as follows: 1. The RNA contents of the ethanol-treatment groups are, in the liver and kidney, similar to, and in the brain, generally higher than, that of the control group. 2. The DNA contents of each organ show no difference between ethanol-treatment groups and control group, but in the liver, of ethanol extrat groups are lower than casein group. 3. the DNase activity of each organ in the ethanol-treatmeut groups, is generally lower than the control group. The above results indicate that ethanol extracts from fish flour have influence on the nucleic acid metabolism.
