양제근 추출물 및 분획의 항산화 활성과 Tyrosinase 저해 활성 (Antioxidant and Tyrosinase Inhibitory Activity of Extract Rumex japonicus HOUTT Root and Its Fractions)

  • Background: We investigated the antioxidative and tyrosinase inhibitory activities of 70% ethanol extract, and its fractions, of the root of Rumex japonicus HOUTT. Methods and Results: The total phenolic compound contents of the 70% ethanol extract and ethyl acetate fraction were 168.99 mg/g and 651.78 mg/g, respectively. The antioxidant activity was compared through the DPPH radical and nitric oxide (NO) scavenging assays. The ethyl acetate fraction showed the highest DPPH radical and NO scavenging abilities, which confirmed the antioxidant activity. Specifically, the ethyl acetate fraction showed a higher DPPH radical scavenging ability than ascorbic acid. These results were related to the total phenolic compound content of the ethyl acetate fraction. Moreover, in the tyrosinase inhibition assay, the ethyl acetate fraction exhibited stronger inhibitory activity than arbutin, which was used as the positive control. The cell viability of L929 cells was analyzed by MTT assay after treatment with 70% ethanol extract and all fractions; no changes in viability were observed, which demonstrated the nontoxic nature of the extract and fractions. Conclusions: These results suggested that the extract from the root of R. japonicus and its ethyl acetate fraction could be a novel resource for the development of a cosmetic with antioxidant and tyrosinase inhibitory activity.

마치현 에틸아세테이트 분획물의 뇌세포 보호효과 (Neuroprotective Effect of Ethyl Acetate Fraction of Portulaca oleracea L.)

  • Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.

  • Cactus (Opuntia ficus-indica var saboten Makino) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. We previously found that the ethanol extract of cactus stem showed anti-inflanunatory action. This investigation was designed to isolate the active fraction of anti-inflanimatory action from cactus stem by solvent extraction and colunm chromatography. Carrageenan-induced paw edema in rats and acetic acid-induced writhing test in mice were used as animal models to search anti-inflammatory and analgesic activities. respectively. The ethanol extract of cactus stem was consecutively extracted with hexane, ethyl acetate, and n-butanol. The hexane fraction was the most effective in carrageenan-induced paw edema, and then was separated in colunm chromatography of silica gel by the elution with hexane/ethyl acetate mixture. The most effective fraction 1 was separated in a second colunm chromatography by eluting with hexane/diethyl ether mixture. The most effective fraction 1-5 was obtained, and separated in a third column chromatography by eluting with hexane/chloroform mixture. It produced the most effective fraction 1-5-1. Moreover, fraction 1-5-1 showed an inhibitory effect on acetic acid-induced writhing in the doses of 30mg/kg and 60mg/kg,p.o.,indicating that it also contained analgesic activity.

Effects of Ethylacetate Fraction of Persimmon Leaves on Experimentally-induced Gastric Mucosal Damage and Gastric Ulcers in Rats

  • The protective effects of the ethylacetate fraction of persimmon leaves(PEF) against experimentally induced gastric mucosal damage and gastric ulcers were evaluated in ratss. In prophylatic study, 100 mg/kg ethylacetate fraction of persimmon leaves (PEFH) exhibited a total protection of 73.8% and 65.7% against HCl-ethanol and 0.2N NaOH-induced gastric mucosal membrane lesions, respectively, which was superior to cimetidine 50 mg/kg, a commonly used anti-ulcer drug. PEFH showed excellent anti-ulcer effects against pylorus ligation induced gastric ulcers, compared to the control group, however, 50 mg/kg ethylacetate fraction of persimmon leaves (PEFL) and PEFH did not affect ulcers induced by water immersion stress, and that is inferior to cimetidine 50 mg/kg. In conclusion, the results suggest that the ethylacetate fraction of persimmon leaves can be used both in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.

Effect of Butanol Fraction of Panax ginseng Head on Gastric Lesion and Ulcer

  • From our previous result that Panax ginseng head extract had inhibition of gastric damages, the extract was fractionated. Among the hexane, chloroform, butanol and water fractions, butanol fraction Showed the most potent inhibition of HCl.ethanol-induced gastric lesion, aspirin-induced gastric ulcer, acetic acid-induced ulcer and Shay ulcer. Butanol fraction showed significant increase in mucin secretion, and inhibited malondialdehyde (MDA) and $H^{+}/K^{+}ATPase$ activity in the stomach. This results indicate that the effectiveness of the fraction on gastric damages might be related to inhibition of acid secretion, increment of mucin secretion and antioxidant property.

야관문(Lespedeza cuneata) 종자의 열수 및 Ethanol 추출물의 항산화 효과 (Antioxidative Activity of Hot Water and Ethanol Extracts of Lespedeza cuneata Seeds)

  • 본 연구에서는 야관문의 열수 및 ethanol추출조건에 따른 항산화활성의 변화를 비교평가 하고자 하였다. 야관문에 존재하는 항산화물질은 용매분획 과정 중에서 열수추출조건에 비해 ethanol 추출조건에서 항산화 성분이 잘 이행되었고, 열수추출조건에서는 상대적으로 활성이 낮게 나타났다. Silica gel adsorption column chromatography 상의 chloroform-methanol의 용매계에서 각 획분에 대해 DPPH-radical scavenging 효과를 측정한 결과, 열수 추출한 경우는 methanol함량이 50% 부근을 중심으로 활성이 나타났다. 반면 ethanol 추출한 경우는 methanol함량이 50-60% 부근을 중심으로 활성이 나타나, 열수 추출물과 ethanol 추출물의 항산화활성 물질의 성질은 약간의 차이를 나타냄을 알 수 있었다. Sephadex LH-20의 gel filtration으로 분획한 결과 두가지 처리조건 모두 bed volumn에 대한 elution volumn의 비(Ve/Vt)가 1.25-1.52의 용출범위(1.1-2.2 g)에 활성이 인정되었다. 열수추출 및 ethanol 추출 조건의 경우 항산화 활성물질의 HPLC retention time은 각각 7.25분 그리고 7.41분으로 나타났지만 두 peak의 HPLC상의 거동이 일치하여 동일한 성분일 것으로 추정하였다.

각종 변이원에 대한 쇠비름 추출물의 돌연변이 억제 효과 (Inhibitory Effects of Poturaca oleracea L. Extract on the Mutagenicity of Various Mutagen)

  • This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), , 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene (B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Poturaca oleracea L. only Inhibited mutagenic activity of MNNG in Salmonella typhimurium TA100, On 4NQO, the ethanol extract 100-1,600$\mu\textrm{g}$/plate of Porturaca oleracea L. showed a slight inhibitory effect of 13-48%, 4-47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6-86% in TA100, And the treatment of 1,600$\mu\textrm{g}$/plate of ethanol extract of Porturacea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85-93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether. chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TA100 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B (a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain Induced by Trp-P-1, respectively.

연잎 에탄올 추출물의 항산화 효과 (Antioxidative Activity of Ethanol Extract from Lotus (Nelumbo nucifera) Leaf)

  • 본 연구에서는 연잎 추출분획물의 항산화 활성을 비교, 검토하고자 하였다. $70\%$ ethanol로 추출한 추출수율은 $8.16\%$였으며 이를 hexane, chloroform, ethyl acetate, butanol, water로 순차 분획한 수율은 butanol층이 $36.12\%$로 가장 높은 수율을 보였으나 대체적으로 극성이 높을수록 높은 분획수율을 나타내었다. 분회물간 총 페놀함량 측정결과 ethyl acetate 분획물의 총 페놀함량이 가장 높게 나왔으며 전자공여능 측정결과 또한 ethyl acetate 분획물에서 가장 우수한 결과를 나타내었다. Ethyl acetate 분획물의 전자공여능은 BHA, BHT와 비교한 결과 BHA와 유사한 결과를 나타내었으며 BHT보다 우수한 값을 나타내었다. 항산화 활성이 높게 나타난 ethyl acetate 분획물을 취하여 TBA가에 의한 지질과산화를 측정한 결과 BHA가 가장 우수한 활성을 보여 주었으며 ethyl acetate 분획물은 BHT보다도 활성이 떨어졌으나 이는 ethyl acetate 분획물이 단일물질로 분리정제가 이루어지지 않은 것으로 malonaldehyde 이외 다른 성분이 BHA와 반응하여 발색한 것으로 추측되어 졌다. AV와 POV 측정 결과 BHA가 가장 우수한 활성을 보여주었으나 ethyl acetate 분획물과 유의적인 차이는 없었으며 BHT에 비해서는 우수한 활성을 나타내었다.

  • The ameliorating effect of the MeOH extract of the root of Platycodon grandiflorum on the ethanol-induced cognitive dysfunction in mice was investigated. The total MeOH extract was devided into two fractions, saponin rich fraction and non-saponin fraction. The mice with repeated administration of the total MeOH extract and those with saponin rich fraction both showed a markedly prolongεd latency time upon the step-through test (passive avoidance performance) performed after acute ethanol intoxication, while those with non-saponin rich fraction did not. The results suggest that the extracts from the root of Platycodon grandiflorum exert a beneficial effect on ethanol-induced memory impairment in mice and also suggest that these effects of the extracts might be ascribed to the saponin constituents.

Inactivation Mechanism of Bacillus subtilis Spores by Ethanol Extract of Torilis japonica Fruit

  • To confirm the antimicobial mechanism of Torilis japonica, antimicrobial profile was observed on various spore conditions by combining 0.1% (3 mM) torilin with antimicrobial activity and 0.27% water fraction with germinants. A 75% ethanol extract of T. japonica fruit reduced Bacillus subtilis ATCC 6633 spore counts by 3 log cycles and reduced the vegetative cells to undetectable level (by about 6 log cycles) (both in terms of CFU/mL). Further fractionating the ethanol extract into n-hexane and water fractions revealed that the former reduced the spore count by 1 log cycle whereas the latter had no effect. The antimicrobial active compound was isolated and purified from the hexane layer, and identified as torilin ($C_{22}H_{32}O_5$). The water fraction of the ethanol layer did not show antimicrobial activity, whereas the antimicrobial effect of 0.1% (3 mM) torilin was significantly enhanced in the presence of the water fraction (0.27%). This result can be explained by synergistic effects of the water fraction containing considerable amounts of germinants such as L-alanine and K+ ions that triggered germination.
