The objective of this study was to produce calves by transfer of embryos derived from slaughter house(SHD) and ultrasound-guided ovum pick-up (OPU). At 60 hrs after injection of 400 mg FSH dissolved in 25% polyvinylpyrrolidone(PVP) by single dose, ultrasound-guided follicular oocyte aspiration was ferformed. Day-7 and day-8 blastocysts produced by in vitro maturation (IVM), fertilization (IVF) and culture(IVC) of the oocytes derived from SHD and OPU were nonsurgically transferred into recipients. The results obtained were as follows. The cleavage rate and the development rate to blastocysts were not significantly (P<0.05) different between the oocytes obtained by SHD (72.9% vs. 34.1%) and OPU (75.9% vs. 38.4%). The oocyte recovery rate from the number of follicles by ultrasound-guided aspiration were not significantly (P<0.05) different between Holstein (61.7%) and Hanwoo (60.1%), but the rate of oocytes useful for IVF was significantly (P<0.05) higher in Hanwoo (69.3%) than Holstein (59.6%). The cleavage rate and the development rate to blastocysts was not significantly (P<0.05) different between Holstein (74.9% vs. 39.2%) and recipients on day 8 of estrus cycle resulted in 13 pregnancies (34.2%). One of them was sacrificed during gestation period due to mastitis and another was aborted spontaneous. The resulting 14 calves were morphologically normal at birth. Seventy eight fresh OPU-IVF embryos were transferred into 21 recipients on day 8 of estrus cycles, resulting in pregnancy of 12 recipients (41.4%). Two of them were sacrificed during gestation period due to mastitis and the other two were aborted. Nevertheless, the 11 OPU-calves have been born normally.
Serial ultrasonography was performed to measure the normal appearance of uterine during estrous cycle and to determine whether the unterine appearance was related to the sex hormone, progesterone and estrogen. The uterine appearances, shape, diameter and echogenicity were daily monitored with ultrasonography in 9 Miniature Schnauzer dogs undergoing II estrous cycles. During proestrus and estrous, the uterus became hypoechoic but developed hyperechoic luminal echo. In the longitudinal view, the shape of the uterus occasionally changed from rectangular to coiled or serpentine, compared to other stages of the cycle. The diameter of the uterus during proestrus and estrous was larger (range: 0.60${\sim}$0.86 mm) than other stages (range: 0.48${\sim}$0.62 mm) of the cycle. The rising estrogen concentrations (range: 14.51${\sim}$16.86 pg/ml) in plasma during proestrus correlated with changes in the uterus (p<0.05). Progesterone concentrations were 0.08${\sim}$0.15 ng/ml at the onset of proestrus, but rose 1.06${\sim}$1.26 ng/ml at the end of proestrus. There was no relation to progesterone concentration from onset of estrus (p>0.05). There was dramatical changes in normal uterus and sex hormone during estrous cycle. Especially, the appearance, shape and diameter of uterus were related to plasma estrogen concentration during proestrus, correlated with other stages of the cycle.
The present study exarnines the physiological alteradons in prolactin (PRL) messenger ribonucleic acid (mRNA) and serum PRL levels during the rat estrous cycle and the effed of naloxone, an endogenous oploid peptide receptor antagonist, on PRL gene expression during the rat estrous cycle. Adult female rats exhibiting at least two consecutive 4-day estrous cycles were used in this study. A single injection of naloxone (2mg/kg b.w.) or saline was given sc 30 mm prior to decapitation. Animals were sacrificed at 10:00 h of each stage of the estrous cycle, and at 2-h intervals from 10:00 h to 20:00 h during the proestrus. PRL mRNA and serum PRL levels were determined by a RNA-blot hybridization with the rat PRL cDNA probe and by a PRL radjoimmunoassay, respectively. PRL mRNA and serum PRL levels were not dramatically altered in the morning of each stage of diestrus I, II and proestrus, and naloxone failed to modify the two parameters. During estrus naloxone clearly suppressed serum PRL levels, but it was unable to modify PRL mRNA levels. A more detailed examination of the proestrus stage revealed that PRL mRNA and serum PRL levels were fluctuated as a function of time: PRL mRNA levels reached a maximum level at 12:00 h and gradually decreased until 18:00 h. PRL mRNA levels then rose at 20:00 h. No difference of PRL mRNA levels between the control and naloxone-treated groups was observed. Changes in serum PRL levek during proestrus were conversely related to changes in PRL mRNA: serum PRL levels were low from 10:00 h to 14:00 h, then increased and reached a maximum level at 16:00-18:00 h. Following then, serum PRL levels were decreased. Naloxone was effective in suppressing the charaderistic afternoon surge of PRL from 16:00 h to 20:00 h. These data clearly showed that alterations in PRL mRNA levels were conversely correlated with changes mn serum PRL levels on proestrus, indicating a differential regulation of PRL gene expression and secretion.
This study was conducted to investigate the ovarian cycle changes of the mare according to the season. Twenty four Jeju crossbred horses(Thoroughbred ${\times}$ Jeju horse) raised in Subtropical Livestock Research Institute, National Institute of Animal Science, RDA were used to identify follicles and corpus luteum with ultrasonography once a week(May 2016~June 2017). Blood samples of experimental horses were collected twice a week for analysis of P4 hormone levels. The mares were considered to have resumed ovarian cyclicity on the day of ovulation if they followed by regular ovarian cycles. Only 13 cases(61.9%) of the total 21cases showed normal ovarian cycle, and 8 cases (38.1%) showed delayed ovarian cycle. Three cases(16.7%) in October, 5 cases(27.8%) in November and 5 cases(27.8%) in December(27.8%) ceased the heat and the remaining 5 cases(27.8%) showed that the estrus was maintained in winter. Horses that stopped estrus ceased the heat until March of next year, and 27.8% were continued the heat during non-breeding season. Eleven cases(61.1%) of 18 cases in April and 2 cases(11.1%) of 18 cases in May returned the estrus.
The present study was undertaken to identify changes of plasminogen activators (PAs) in porcine oviductal epithelial cells (POECs) during the estrous cycle classified with post-ovulatory stages (Post-Ov), early to mid-luteal stages (Early-mid L) and pre-ovulatory (Pre-Ov) stages. The urokinase-type plasminogen activator (uPA) was only observed on day 5 and day 7 of culture in the POECs on all the estrous cycles and gradually increased according to increasing culture times, but not Early-mid L. In POECs-conditioned medium, uPA, tissue-type (tPA) and tPA-PA inhibitor (tPA-PAI) activity were observed at all culture times during estrous cycles. The uPA activity of POECs-conditioned medium on Post-Ov stage were significantly (p<0.05) decreased during prolonged cultures. On the other hand, the tPA activity of POECs-conditioned medium at Post-Ov stage was significantly (p<0.05) higher on day 5 than compared to the other days. Although was higher on day 1 at Post-Ov stage, the tPA-PAI activity of POECs-conditioned medium was significantly (p<0.05) higher on day 7 at all stage than that of day 5 of the culture. Taken together, these results suggest that uPA, tPA and tPA-PAI are produced by POECs, and the variations of the PAs activity are regulated in the different stages of the estrous cycle.
Plasma progesterone concentrations in 44 suckling Korean native cows were determined to monitor ovarian activity postpartum by radioimmunoassay. Blood samples were collected in 3 day intervals from 15 to 80 days postpartum. The ovaries and uterus were examined in 6 day intervals by rectal palpation. Results are summaried as follows: 1. The cows were qualified into four categories; Type I(normal): cyclic changes in plasma progesterone concentrations appear within 45 days postpartum(35 cows, 79.5%), Type II(cycle delayed): delayed resumption of ovarian activity by 55 days postpartum(5 cows, 11. 4%), Type III(cycle ceased with low progesterone): plasma progesterone concentrations remained low(${\leq}1ng/ml$) until 80 days postpartum(3 cows, 6.8%), Type IV(cycle ceased with high progesterone): plasma progesterone concentrations after 30 days remained high(${\geq}4.0ng/ml$) until 80 days postpartum(1 cow, 2.3%). 2. Out of the cows classified Type II and III, 7 cows had inactive ovaries and a cow had follicular cyst by rectal palpation. The cow of Type IV was diagnosed as bearing persistent corpus luteum by rectal palpation. 3. About 82% of the cows showed significant rises in plasma progesterone concentrations prior to 50 days postpartum and the cows of Type II and III resumed cycles in 31. 6(range 17~55) days after calving. However, 43.2% resumed cycles in 50 days postpartum by estrus signs. These results indicated that plasma progesterone concentrations assessed by radioimmunoassay can be utilized for monitoring postpartum ovarian activity and would be helpful for the early detection of ovarian dysfunction in the Korean native cow.
Effects of short-term treatment with somidobove (recombinantly produced bovine somatotropin, BST) on estrous cyclicity and fertility were studied in dairy buffaloes. Twenty buffaloes of Nili-Ravi breed calving during the same season were assigned to either control (n=8) or treated group (n=12). The buffaloes of treated group received single infection (prolonged release) of 320 mg of somidobove on day-60 postpartum. The mean values for interval to first postpartum estrus, first service conception rate, services per conception, service period and calving interval for the treated group were 96.4 days, 66.7%, 1.70, 164 days and 473 days, respectively. The corresponding values for the control group were 92.5 days, 62.5%, 1.87, 135 days and 439 days. Means of all variables did not differ between control and treated group (p > 0.05). Three buffaloes of the control and four buffaloes of the treated group did not conceive at first service. Out of these, two buffaloes of control and one buffalo of treated group exhibited normal estrous cycles. It is concluded from these data that short term BST-treatment has no adverse effect on reproductive functions of dairy buffaloes.
Animal in dustry in Korea urgently needs the domestic introduction and the industrial a, pp.ication of embryo transfer technique. Namely, this technique can be utilized effectively, as means of the improvement of livestocks, as means of the increase of meat production, as means of substitute for the livestock import, and dissemination of new breed. However, as this technique avaliable in our country is remaining initial stage, we can not make use of the technique industrially unless we make much improvement as follows; induction of superovulation, non-surgical recovery of embryos, synchronization between the estrus such cycles of donor and recipient, non-surgical transfer of embryos, etc. Simultaneously, the basic studies such as harvesting oocytes from ovary, in vitro culture of oocytes, in vitro capacitation of spermatozoa, cloning by culture of blastomeres and transfer of nuclei, sexing embryo, etc. should not be neglected in order to make the technique of embryo transfer more simple and convenient. For the success of these studies, universities, national and public institutes, large scale cattle farms, and small scale cattle farms should cooperate each other. For instance, universities undertake basic researches, and the national and public institutes a, pp.y the results of the researches to animal industry along with cooperation by large scale cattle farms. By the help of the cooperative organizations, the technique relevant to our environment and farm condition may be able to be finalized, and to be a, pp.ied to samll scale cattle farm. Consequently, being served to stimulate animal productivity, this technique can be contributed to the development of livestock industry in Korea.
This study used adult wistar-based rats to observe the sexual cycle as a morphological characteristic of vaginal epithelial cells by vaginal smearing, and investigated the fetal number through mating with male rats of the same strain. The target animal was a 12 to 13-week-old Wistar-based mature unlighted rat (weight 220 g to 240 g), room temperature 23 ± 2℃, 14 hours artificial lighting (05:00 to 19:00 hours), 10 hours Adapted individuals were used for rearing for at least 2 weeks under the conditions of the darkroom (19:00 to 05:00). The feed was managed for free feeding of pellet feed for animals and water. The vaginal smearing method was used for the experiments by observing the sexual cycle every morning and confirming that the normal sexual cycle of 4 or 5 days was repeated at least 2 cycles or more. As a result, the proestrus was found to have few red blood cells, the cells and nuclei were rather large and round, and many nucleated cells were identified. In the case of the estrus, the cells were large and the nuclei were not stained, and most of the keratinocytes were found. In addition, in the metestrus and diestrus, there were many white blood cells, and it was confirmed that nucleated epithelial cells and keratinocytes were significantly reduced. The pregnancy period was 21 ± 1.8 days, and the number of live births per delivery was 11.9 on average. The number of fetuses on the 8th and 10th days of pregnancy were 15.2 ± 0.4 and 15.4 ± 0.3, respectively. On the contrary, the number of fetuses on the 12th day of pregnancy was 12.9 ± 0.6, which was significantly (p < 0.05) decreased compared to the 10th day of pregnancy, and the number of fetuses was similar until delivery. As a result of investigating the change of body weight according to the birth weight and growth stage after delivery, the birth weight of female and male was 9.2 ± 2.0 g and 9.8 ± 2.5 g, respectively. After that, until the 16th day, the female and the male showed similarly moderate weight gain, and then showed a rapid weight gain until the 21st day of lactation. With reference to the results of this study, it is expected to be used as basic data for determining the mating time of rodents and controlling pregnancy and fetal number.
Background: Rabbit breeding with high performance imported ones would be of benefit for genetic diversity and improvement of performance in domestic rabbit breeds. The rearing of more productive rabbit breeds could be pathway to improve the productivity and reduce the production cost. Maternal nutritional status exert a great influence on reproductive functions of does, which may expand from conception, through gestation and parturition and development of kits to puberty. Methods: Four breeds of rabbit were evaluated for their parturition, weaning and pubertal differences among the rabbit population in Ibadan, Nigeria. The breed consist of Fauve De Bourgogne (FDB), Chinchilla (CHA), British Spot (BS) and New Zealand White (NZW) rabbits. A total of 60 bucks and 360 does consisting of 15 bucks and 90 does per breed were mated in 6 mating cycles, three each of natural mating and artificial insemination. All does were synchronized for estrus with 20 IU pregnant mare serum gonadotropin 48 h prior to mating. The does after parturition were assessed for milk yield (g) and kit survival rate (%) till weaning, weight changes of kits from birth to puberty. At puberty, the pubertal age (days) and weight (g) of the offspring were assessed. Result: Results obtained reveals that British Spot doe had highest milk yield among the breeds which significantly increased growth of kit and weight at weaning in British Spot rabbits. Survival rates of Chinchilla kits were significantly (p < 0.05) higher than Fauve de Bourgogne, British Spot and New Zealand White kits. Puberty attainment of the rabbits indicates that British spot does and Fauve de Bourgogne bucks are early maturing. Conclusion: Chinchilla shows high kit survivability and British spot has highest milk yield among the four breeds of rabbit.
