Chemical Composition and Antimicrobial Activity of Essential Oil from Cones of Pinus koraiensis

  • The essential oil from the cones of Pinus koraiensis was prepared after removing the seeds, and its chemical composition analyzed using gas chromatography-mass spectrometry (GC-MS). Hydrodistillation of the P. koraiensis cones yielded 1.07% (v/w) of essential oil, which was almost three times the amount of essential oil extracted from the needles of the same plant. Moreover, the antimicrobial activities of the oil against the growth of Gram-positive bacteria, Gram-negative bacteria, and fungi were evaluated using the agar disc diffusion method and broth microdilution method. Eighty-seven components, comprising about 96.8% of the total oil, were identified. The most abundant oil components were limonene (27.90%), ${\alpha}$-pinene (23.89%), ${\beta}$-pinene (12.02%), 3-carene(4.95%), ${\beta}$-myrcene (4.53%), isolongifolene (3.35%), (-)-bornyl acetate (2.02%), caryophyllene (1.71%), and camphene (1.54%). The essential oil was confirmed to have significant antimicrobial activities, especially against pathogenic fungal strains such as Candida glabrata YFCC 062 and Cryptococcus neoformans B 42419. Therefore, the present results indicate that the essential oil from the cones of Pinus koraiensis can be used in various ways as a nontoxic and environmentally friendly disinfectant.

DNCB로 아토피피부염을 유발한 NC/Nga mice에서 사이프러스 에센셜 오일의 효능에 관한 연구 (A Study on the Skin Protection Effects of Cypress Essential Oil on the DNCB-induced Atopic Dermatitis in NC/Nga Mice)

  • Objectives : The application of DNCB (1-chloro-2,4-dinitrochlorobenzene) can cause cellular immunity allergic reaction such as erythema or edema on NC/Nga mice and combinational function of cells participating in immunity increase inflammatory mediator. In this study, the effects of cypress essential oil on NC/Nga mice have been assessed. Methods : Male SPF NC/Nga mice aged 8 weeks have been used for atopic dermatitis induction skin. 1% DNCB was applied on ears and backs of which hair was removed using clipper on $1^{st}$ day and 0.4% DNCB was applied three times a week for 3 weeks. In this study, cypress essential oil has been treated 1 time a day for 3 weeks after application of DNCB to induce atopic dermatitis on skin for further experiments. Results : This study shows that inhalation or application of cypress essential oil reduced edema in ears and the thickness of epidermis induced by DNCB treatment. And it can be known that treatment of cypress essential oil inhibited mast cell proliferation and reduced IgE level similar to that of the negative control especially when cypress essential oil was inhaled by the mice. Synthetic oil showed the effects lower than those of cypress essential oil. Conclusions : Inhalation or direct application of cypress essential oil on skin reduced IgE level in blood and prohibits the proliferation of mast cell, from which it can be known that cypress essential oil can be effectively used to reduce the symptoms of atopic dermatitis.

침엽수 잎으로부터 효율적인 정유 추출법 탐색 및 정유성분 분석 (Efficient Extraction Methods and Analysis of Essential Oil from Softwood leaves)

  • 편백나무와 화백나무의 잎으로부터 시료전처리, 물과 잎의 혼합비, 추출시간, 채취시기 등 정유 추출에 적합한 조건을 구명하고 잎으로부터 추출된 정유조성성분을 GC-MS로 분석하였다. 정유추출에 적합한 전처리법으로는 믹서기로 잎을 분쇄하는 방법이 좋았으며, 증류수와 잎의 혼합비율은 그다지 영향을 미치지는 않았으나 증류수 1 L당 100 g의 잎을 혼합하였을 때 추출효율이 가장 좋았다. 정유추출시간은 시간이 경과함에 따라 추출효율은 증가하였지만 탄화향이 심해 5시간이 적합한 것으로 나타났다. 계절에 따른 정유수율은 편백나무는 겨울이 4.5%, 화백나무는 가을에 채취한 시료가 5.3%의 높은 수율을 보였다. 편백나무정유와 화백나무정유 조성분은 차이를 보였으며, 주성분은 monoterpene류였다. 편백나무정유의 주성분은 limonene, terpinene-4-ol, ${\gamma}$-selinene 및 $\alpha$-cedrene였으며, 화백나무정유의 주성분은 $\alpha$-pinene, myrcene, limonene, bomyl acetate, $\beta$-caryophyllene, longifolene 및 $\beta$-cedrene이었다.

Susceptibility of Oral Bacteria to Essential Oil of Artemisia capillaris Thunb.

  • Objective : The aim of this work is to investigate the antibacterial activity of the essential oil obtained from Artemisia capillaris (A. capillaris), as the development of microbial resistance to antibiotics make it necessary to constantly look for new and active compounds effective against pathogenic bacteria. Methods : The crushed materials of A. capillaris (1 kg) were subjected to steam distillation for 3 h, using a modified Clevenger type apparatus in order to obtain essential oil. Diethyl ether was the extracting solvent kept at 25°.... The essential oil was analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The essential oil and the composition were tested for antimicrobial activities against 15 different genera of oral bacteria. Results and Conclusion : The components of the essential oil identified were: β-pinene (9.36%), camphor (3.32%), 1,8­cineole (4.38%), artemisia alcohol (3.32%), β-caryophyllene (11.08%), γ-cadinene (4.23%), and capillene (32.74%). The essential oil of A. capillaris exhibited considerable inhibitory effects against all oral bacteria tested, while their major components demonstrated various degrees of growth inhibition.

석청포 정유의 진정 및 진통효과 (Sedative and analgesic effects of essential oil of Acorus gramineus Soland in mouse)

  • The rhizomes of the Acorus gramineus Soland have been used as sedatives, analgesics, stomachics and anthelmintics in chinese medicine. It is known that the rhizomes of the Acorus gramineus Soland contains the essential oil about 0.5~0.8% and this essential oil contains asarone about 86%. The asarone possess many pharmacological properties similar to those of reserpine and chlorpromazine. Sedative and analgesic effects of essential oil of Acorus gramineus Solaad in mouse was observed. The essential oil of Acorus gramineus Soland decreased the frequency of ambulation on mouse in proportion of concentration. ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride) and opioids receptor antagonist(naloxone hydrochloride) were markedly decreased in frequency of ambulation. The essential oil of Acorus gramineus Soland decreased writhing syndrome in mouse and ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride), opioids receptor antagonist(naloxone hydrochloride) were not increased above effects. In conclusion, these experimental results show that the essential oil of Acorus gramineus Soland have sedative and analgesic effects, but it did not antagonized ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride) and opioids receptor antagonist(naloxone hydrochloride).

형개의 생육시기와 식물체 부위별 정유성분 (Variation of Essential Oil Components in Stages and Organs of Schizonepeta tenuifolia Briquet)

  • 형개의 잎, 줄기 및 화수의 정유함율 및 정유구성성분을 생육시기별로 비교 분석하여 조사한 결과를 요약하면 다음과 같다. 1. 형개의 정유함율은 잎이 줄기나 화수에 비하여 월등히 높았으며 영양성장기부터 개화 후반결관까지 생육이 진전됨에 따라 잎과 줄기의 정유함율은 증가하였다. 2. 정유성분의 구성은 잎과 화수의 경우는 d-menthone과 l-pulegone이 56~71%의 함유 비율을 나타내었으며, 줄기는 21~26%의 함유비율을 보이고 잎과 화수에 거의 나타나지 않는 두 물질이 확인 되었다. 3. 잎의 정유성분구성은 생육후기로 갈수록 d-menthone은 생육초기에 많았다가 후기에 증가하는 반면에 l-pulegone은 점차 감소하여 두가지 주정유성분의 함유비율이 서로 반대로 나타났다. 4. 형개의 정유 성분 평가를 위하여 주요 성분의 생약 약리학적인 검토가 필요하다.

Variation of the Essential oil and Main Component from Different Origin of Levisticulum officinale Koch

  • The essential oils were isolated by hydrodistillation and their composition determined capillary GC method with standards. The essential oil content showed significant differences between the two populations on the vegetative organs. The essential oil level of the leaves and roots was considerably higher in the Korean population at full flowering and waxy ripening stage but essential oil content of the roots was significantly higher in the Hungarian taxon at leaf rosette stage. We observed the essential oil accumulation tendency was mianly dependent on plant organs and intra-specific taxon during the vegeation period. Butylidene-phthalide was proved to be the main component of the oil in both population roots (50.9-73.3%), while dimethyl-acetate was showed as a major compound on the over-ground parts (56.7-62.0%). The qualitative composition of the essential oil in the reproductive organs concerning the identified compounds was the same as the vegetative parts with the main component ${\alpha}-phellandrene$ (4.8-28.1%) and butylidene-phtalide (9.7-16.1%), The quantitative composition showed some changes during the ontogenesis phases. Most characteristic ones are the decreasing proportion of dimethyl-acetate (from 7.3% to 1.1%) and the appearance of ${\alpha}-pinene$ (from 0.5% to 1.5%) only after fruit setting in both population.

Combined Effects of the Essential Oil from Eucalyptus globulus with Ketoconazole against Candida and Trichophyton Species

  • We have evaluated the combined antifungal effects of the essential oil from Eucalyptus globulus or its main component 1,8-cineole with ketoconazole. Checkerboard microtiter tests were used to analyze their effects against three Candida and six Trichophyton species. The susceptibility of the Trichophyton species to E. globulus essential oil differed distinctly. The fractional inhibitory concentration indices (FICIs) against the tested Candida species ranged between 0.09 and 0.38 for ketoconazole combined with E. globulus essential oil or 1,8-cineole, indicating significant synergism between ketoconazole and the oil samples. Similar experiments using Trichophyton species resulted in FICIs between 0.28 and 0.63, indicating relatively weaker combined effects than those observed with Candida species. Thus, the data reported here show that the anti-Candida effects of ketoconazole can be significantly improved in the presence of E. globulus essential oil or 1, 8-cineole.

전호(Anthriscus sylvestris) 뿌리 정유의 항균 및 항산화 작용 (Antibacterial and Antioxidant Activities of the Essential Oil from the Roots of Anthriscus sylvestris)

  • To develop a new effective and safe natural antibiotics and antioxidant the essential oil was extracted from the roots of Anthriscus sylvestris by steam distillation. Its composition was analyzed by GC-MS. The activities of the essential oil fraction and its main components were evaluated against antibiotic-susceptible and -resistant strains of some food-born bacteria. In addition the synergism was examined with this oil combined with antibiotic by checkerboard titer test. The antioxidant activities were determined by in 1,1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging activity test and reducing power assay. The essential oil fraction of A. sylvestris revealed significant inhibiting activities against antibiotic-susceptible and -resistant species of Vibrio and Shigella with MICs ranged from 1.00~4.00 mg/ml. It showed synergistic or additive effects when it was combined with amphicillin or trimethoprim/sulfamethoxazole (1 : 9). Additionally, the essential oil fraction of A. sylvestris exhibited significant DPPH free radical scavenging activity and the reducing power.

Terpenoid 분석을 통한 취나물류의 향기지표물질 비교 (A Comparison of Volatile Flavor Characteristics of Chwi-namuls by Terpenoid Analysis)

  • A comparison of essential oils composition of Aster tataricus L. (gaemichwi), Ligularia fischeri (gomchwi), Solidago virga-aurea var. asiatica Nakai (miyeokchwi), and Aster scaber (chamchwi) was performed by gas chromatography and mass spectrometry for the identification of volatile flavor characteristics in chwi-namuls. The essential oils were extracted by the hydro distillation extraction method. One hundred volatile flavor components were identified from gaemichwi essential oil. ${\alpha}$-Pinene (11.5%) was the most abundant compound, followed by myrcene (8.9%) and ${\beta}$-pinene (7.5%). Ninety-one volatile flavor components were identified from the essential oil of gomchwi. Aromadendrene (14.8%) was the most abundant component, followed by ${\beta}$-caryophyllene (7.6%) and 1-methyl-4-(1-methylethylidene)-cyclohexene (7.3%). Ninety-five volatile flavor constituents were detected in the essential oil of miyeokchwi, moreover, spathulenol (15.7%) was the most abundant component. Ninety-six volatile flavor constituents were detected in the essential oil of chamchwi. Epi-bicyclosesquiphellandrene (21.9%) was the most abundant component, followed by ${\beta}$-caryophyllene (9.5%) and ${\delta}$-terpinene (8.9%). The essential oil composition of gaemichwi was characterized by a higher contents of pinenes. The essential oil composition of gomchwi can be easily distinguished by the percentage of aromadendrene. Spathulenol and epi-bicyclosesquiphellandrene were regarded as the characteristic odorants of miyeokchwi and chamchwi, respectively.
