소성 처리된 굴 패각을 활용한 PLA/PBAT 복합필름의 항균 포장재 적용 연구 (Application of PLA/PBAT Composite Films Containing Calcined Oyster Shell Powder for Antimicrobial Packaging)

  • 최근 플라스틱 포장 폐기물 및 굴 패각이 야기하는 환경문제와 장기간 식품의 품질 보존을 위한 액티브 패키징이 주목받고 있다. 이에 따라 본 연구에서는 PLA/PBAT 복합필름에 OSP의 함량을 서로 달리하여 이축 압출기로 PLA/PBAT-OSP 복합필름을 제조하였고 항균 포장재로써 적용가능성을 확인하였다. 이를 위해 화학적 특성, 표면 특성, 열적 특성, 기계적 특성 및 항균성을 평가하여 분석하였다. PLA/PBAT-OSP 복합필름의 고분자 매트릭스 내에서 OSP의 물리적 분산을 확인하였으며, OSP의 함량이 증가함에 따라 항균성이 증가하였다. 특히 OSP 5%, OSP 10% 필름에서는 99% 이상의 항균성을 나타내어 제조된 필름이 우수한 항균성을 가진다는 것을 확인할 수 있었다. 그러나 OSP의 함량의 증가는 PLA/PBAT-OSP 매트릭스 내의 OSP 응집을 일으켜 기계적 물성의 저하 및 표면의 거칠기 증가가 나타났으며, 이는 필름의 표면 에너지 증가로 이어졌다. 결론적으로 PLA/PBAT-OSP 복합필름은 미생물로 인한 식품의 부패를 막기 위한 포장재로 적용될 수 있지만, 이를 위해 OSP의 분산성과 필름의 기계적 물성을 향상시키기 위한 추가 연구가 필요할 것으로 보인다.

눈다랑어(Thunnus obesus)를 활용한 고령친화식품용 연화식 함박스테이크, 완탕 및 토마토스프의 개발 및 품질특성 (Development and Quality Characteristics of Softened Hamburger Steak, Wantang, and Tomato Soup for Senior-friendly Seafoods Using Bigeye Tuna Thunnus obesus)

  • This study was developed to prepare hamburger steak (HS), wantang soup (WS), and tomato soup (TS) for senior-friendly seafood's (SFS) using bigeye tuna Thunnus obesus, and to investigate their quality characteristics. All data were presented in the order of HS, WS, and TS. The hardness values were 298.9×1.03, 47.1×1.03 and 14.9 ×1.03 N/m2, respectively. The viscosity of TS was 2,856 mPa·s. The protein contents were 13.8, 11.7, and 9.7 g/100 g, respectively. The fat-soluble vitamins content were as follows: vitamin A 21.88, 5.03, and 23.72 ㎍RAE/100 g; vitamin D 1.15, ND, and 1.81 ㎍/100 g, respectively. The water soluble vitamins contents were as follows: vitamin C 47.22, 32.83 and 37.01 mg/100 g; vitamin B2 0.17, 0.11 and 0.10 mg/100 g; vitamin B3 34.87, 34.76 and 54.62 mgNE/100 g. The Ca contents were 15.9, 16.8, 28.9 mg/100 g, and the K contents were 383.8, 167.4, and 300.0 mg/100 g, respectively. The dietary fiber was 0.04, 0.07 and 0.08 g/100 g, respectively. Escherichia coli was not detected in any of the products. These results suggest that the products should be classified as follows, based on their appropriateness as SFS: HS, WS and TS.

Development and Evaluation of a Next-Generation Sequencing Panel for the Multiple Detection and Identification of Pathogens in Fermented Foods

  • These days, bacterial detection methods have some limitations in sensitivity, specificity, and multiple detection. To overcome these, novel detection and identification method is necessary to be developed. Recently, NGS panel method has been suggested to screen, detect, and even identify specific foodborne pathogens in one reaction. In this study, new NGS panel primer sets were developed to target 13 specific virulence factor genes from five types of pathogenic Escherichia coli, Listeria monocytogenes, and Salmonella enterica serovar Typhimurium, respectively. Evaluation of the primer sets using singleplex PCR, crosscheck PCR and multiplex PCR revealed high specificity and selectivity without interference of primers or genomic DNAs. Subsequent NGS panel analysis with six artificially contaminated food samples using those primer sets showed that all target genes were multi-detected in one reaction at 108-105 CFU of target strains. However, a few false-positive results were shown at 106-105 CFU. To validate this NGS panel analysis, three sets of qPCR analyses were independently performed with the same contaminated food samples, showing the similar specificity and selectivity for detection and identification. While this NGS panel still has some issues for detection and identification of specific foodborne pathogens, it has much more advantages, especially multiple detection and identification in one reaction, and it could be improved by further optimized NGS panel primer sets and even by application of a new real-time NGS sequencing technology. Therefore, this study suggests the efficiency and usability of NGS panel for rapid determination of origin strain in various foodborne outbreaks in one reaction.

Effect of microencapsulation of egg yolk immunoglobulin Y by sodium alginate/chitosan/sodium alginate on the growth performance, serum parameters, and intestinal health of broiler chickens

  • Objective: Egg yolk immunoglobulin (IgY) is an antibiotic alternative to prevent and fight intestinal pathogenic infections. This study aimed to investigate the effects of sodium alginate/chitosan/sodium alginate IgY microcapsules on the growth performance, serum parameters, and intestinal health of broiler chickens. Methods: One-day-old broilers (Ross 308) were divided into five treatments, each with 10 replicates of five chickens. The dietary treatments were maintained for 28 days and consisted of a basal diet (NC), basal diet + 500 mg chlortetracycline/kg diet (CH), basal diet + 50 mg non-microencapsulated IgY/kg diet (NM), basal diet + 600 mg low levels microencapsulated IgY/kg diet (LM), and basal diet + 700 mg high levels microencapsulated IgY/kg diet (HM). Results: Throughout the 28-day trial period, the NM, LM, HM, and CH groups increased average daily gain compared with the NC group (p<0.05), and the HM group reduced feed conversion ratio compared with the CH group (p<0.05). The LM and HM groups increased relative organ weights of thymus and spleen compared with the CH and NM groups (p<0.05). The HM group improved the duodenal, jejunal and ileum villi height (VH) and villus height to crypt depth ratio (VH:CD) compared with the CH and NM groups (p<0.05). Compared with the CH group, the HM group increased serum immunoglobulin (IgA), immunoglobulin G (IgG), superoxide dismutase, total antioxidant capacity, and glutathione peroxidase levels (p<0.05), and decreased serum malondialdehyde levels (p<0.05). Compared with the NC group, the NM, LM, HM, and CH groups reduced colonic Escherichia coli and Salmonella levels (p<0.05). and the HM group promoted the levels of lactic acid bacteria and bifidobacteria compared with the CH group (p<0.05). Conclusion: Microencapsulation could be considered as a way to improve the efficiency of IgY. The 700 mg high levels microencapsulated IgY/kg diet could potentially be used as an alternative to antibiotics to improve the immune performance and intestinal health, leading to better performance of broiler chickens.

초음파 및 유기산 처리에 따른 재배유형별 마늘의 저장 중 품질변화 (Alterations in qualities of different cultivation types of garlic during storage: Changes assessed by ultrasonic and organic acid treatment)

  • 본 연구에서는 초음파 및 유기산 처리에 따른 재배유형별 마늘의 저장 중 품질 특성 조사를 위해 초음파(60℃, 5 min)와 유기산(citric acid 1.0%, fumaric acid 0.5%, 각각 5 min) 처리 후 냉장(4℃) 온도에서 28일간 저장하면서 품질 특성을 조사하였다. 총균수는 한지형과 난지형 모두 대조구와 citric acid 처리 시 저장 기간이 증가할수록 총균수도 증가하였다. 대장균은 한지형과 난지형 모두 대조구에서 저장 7일부터 검출되기 시작하였으며, 그 수준은 식품공전의 신선 편의 식품 법적인 기준 2.0 log CFU/g을 초과하였다. 곰팡이도 대장균과 마찬가지로 저장 7일부터 검출되기 시작하였으나 초음파 처리 시에는 저장 기간 중 모든 재배유형별 마늘에서 검출되지 않았다. Fumaric acid 0.5%를 고려했을 때 총균수는 0.06±0.13으로 한지형과 난지형이 동일하였고, 곰팡이는 한지형 0.68±0.25와 난지형은 증식하지 않는 등 낮은 수준을 유지하고 있었으며, 대장균은 검출되지 않아 미생물 억제에 효과적이었다. 무기질 변화 분석 결과 모든 재배유형별 마늘에서 Fe와 Zn은 검출되지 않았으며 성분이 많은 함유량은 K, P, Mg, Ca, Na 순으로 이었다. 경도는 전반적으로 저장 기간이 증가할수록 감소하는 경향을 보였는데, fumaric acid 처리구에서는 모든 재배유형에서 경도에 유의적인 차이가 없어 조직변화 측면에서 fumaric acid 0.5%, 5분 처리가 가장 효과적인 것으로 나타났다. 색도는 L값은 감소, a값과 b값은 증가하여 육안으로 보았을 때 녹변현상과 일치하였다. 따라서 본 연구에서 활용한 전처리 조건에서는 일부 기간 미생물 감소, 무기질 및 경도, 색 변화와 같은 품질을 유지하는 방안으로 fumaric acid 0.5%, 5분 처리가 마늘의 미생물 저감과 품질 유지에 효과적인 것으로 나타났다. 그러나 채소류의 미생물, 색, 경도 등 품질을 향상하고 유지할 수 있는 다양한 물리적, 화학적인 방법들이 추가로 연구되어야 할 것으로 사료된다.

Effect of dietary inclusion of Bacillus-based probiotics on performance, egg quality, and the faecal microbiota of laying hen

  • Objective: Our study examined the impact of propriety blends of Bacillus strain probiotics on the performance, egg quality, and faecal microflora of laying hens. Methods: A total of 183 Institut de selection Animale (ISA) brown laying hens aged 23 weeks with an average body weight of 1,894±72 g were randomly allocated into 3 groups as control (corn-soybean meal based diet, CON), 0.5 g/kg Enterosure probiotics (ET1, 3×108 colony-forming unit [CFU]/kg feed), and 5 g/kg Enterosure probiotics (ET2, 3×109 CFU/kg feed) administered in mashed form. At the completion of each phase hen day egg production (HDEP), average egg weight (AEW), feed intake, and faecal microbiota were evaluated. Results: HDEP and AEW were higher (p<0.05) in the ET2-supplemented diet in phase 3 (week 9 to 12) compared with CON. Egg mass (EM) was higher (p<0.05) in phase 2 at ET2, and also higher (p<0.05) in phase 3 at the ET1 and ET2-supplemented diets compared with CON. Feed conversion ratio was lower (p<0.05) in phase 3 at the ET1 and ET2-supplemented diets, with ET2 being the lowest compared with ET1 and CON. Yolk colour was higher (p<0.05) in the ET-supplemented diets at phase 3 compared with CON. Bifidobacterium spp. was higher (p<0.05) in the ET2- supplemented diet compared with CON in phase 2, while in In phase 3, Lactobacillus spp. and Bifidobacterium spp. were higher (p<0.05) in the ET-supplemented diets compared with CON. Coliforms were lower (p<0.05) in the ET-supplemented diets compared with CON in phase 3. Conclusion: The propriety blends of Bacillus strain probiotics supplements at 0.5 g/kg and 5 g/kg could improve the production and quality of eggs with more significance at 5 g/kg for HDEP, AEW and EM, which was achieved via the increase of beneficial microbiomes such as Lactobacillus spp., Bifidobacterium spp., and the decrease of pathogenic microbiomes like Escherichia coli and Coliforms which was speculated to improve gut barrier function and the reproductive hormone.

Cytokine-like Activity of Liver Type Fatty Acid Binding Protein (L-FABP) Inducing Inflammatory Cytokine Interleukin-6

  • It has been reported that fatty acid binding proteins (FABPs) do not act only as intracellular mediators of lipid responses but also have extracellular functions. This study aimed to investigate whether extracellular liver type (L)-FABP has a biological activity and to determined serum L-FABP levels in patients with end-stage renal disease (ESRD). We isolated L-FABP complementary deoxyribonucleic acid (cDNA) from the Huh7 human hepatocarcinoma cell line and expressed the recombinant L-FABP protein in Escherichia coli. A549 lung carcinoma and THP-1 monocytic cells were stimulated with the human recombinant L-FABP. Human whole blood cells were also treated with the human recombinant L-FABP or interleukin (IL)-1α. IL-6 levels were measured in cell culture supernatants using IL-6 enzyme-linked immunosorbent assay (ELISA). Human recombinant L-FABP induced IL-6 in a dose-dependent manner in A549, THP-1 cells, and whole blood cells. The blood samples of healthy volunteers and patients with ESRD were taken after an overnight fast. The serum levels of L-FABP in healthy volunteers and ESRD patients were quantified with L-FABP ELISA. The values of L-FABP in patients with ESRD were significantly lower than those in the control group. Our results demonstrated the biological activity of L-FABP in human cells suggesting L-FABP can be a mediator of inflammation.

생합성된 galactosylated 2-phenoxyethanol의 피부 투과도 (Skin permeability of the biosynthetic galactosylated 2-phenoxyethanol)

  • 화장품 소재는 안전성이 무엇보다 중요시된다. 화장품에 사용되는 기존의 보존제인 PE에 대장균 효소 𝛽-gal을 이용하여 안전성이 증가된 PE-gal을 생합성하였다. 화장품 소재로 사용하기 위해 생합성된 생성물인 PE-gal의 피부 흡수도를 기존의 보존제인 PE와 비교하기 위해 Franz Diffusion cell Assay 시스템을 이용하여 경피투과도를 측정하였다. 같은 질량농도의 시료를 사용하였을 때 PE의 Flux 값, Kp 값은 시간이 지날수록 증가하는 것으로 나타났으나 PE-gal은 투과도를 측정할 수 있을 만큼 투과되지 못하였다. 이는 PE의 피부투과도가 생합성된 PE-gal 배당체보다 높다는 것을 나타낸다. Marzulli 등에 따라 Kp 값을 이용하여 투과 정도를 확인하였을 때 PE의 투과속도는 1mg/mL의 농도에서 느림(slow)으로 측정되었다. 따라서 배당체 형태의 PE-gal은 PE에 비해 경피 투과도가 현저히 낮게 나타났다.

  • 본 연구는 마우스 대식세포 유래 Raw264.7 세포주에서 황기와 산초 1:1 혼합물(AZM-1:1)의 면역 증진 효능을 탐색하였다. Raw264.7 세포에 100-400 ㎍/mL의 A ZM-1:1 처치는 세포 생존율의 변화 없이 inducible nitric oxide synthase mRNA의 발현 증가와 함께 nitric oxide의 생성을 통계적으로 유의하게 증가시켰다. 더불어 A ZM-1:1은 처치 농도 의존적으로 cyclooxygenase-2 mRNA의 유도와 함께 세포 배양액 중 prostaglandin E2의 함량을 증가시켰다. 또한, AZM-1:1은 tumor necrosis factor-α, interleukin-1β, interleukin-6 및 monocyte chemoattractant protein-1의 전사를 촉진하였다. Immunoblot 분석을 통하여 AZM-1:1은 mitogen-activated protein kinase의 인산화를 증가시키고, inhibitory-κBα의 인산화를 매개한 분해를 촉진하며, p65의 인산화를 증가시킬 수 있음을 확인하였다. AZM-1:1의 처치는 녹색 형광으로 표지된 대장균 파편의 탐식작용을 촉진하였다. 따라서, 이상의 결과는 A ZM-1:1가 대식세포를 포함한 내재면역을 증진시키는 기능성 식의약 소재가 될 수 있음을 나타낸다.

Metabolic Regulation of Longevity and Immune Response in Caenorhabditis elegans by Ingestion of Lacticaseibacillus rhamnosus IDCC 3201 Using Multi-Omics Analysis

  • Probiotics, specifically Lacticaseibacillus rhamnosus, have garnered attention for their potential health benefits. This study focuses on evaluating the probiotic properties of candidate probiotics L. rhamnosus IDCC 3201 (3201) using the Caenorhabditis elegans surrogate animal model, a well-established in vivo system for studying host-bacteria interactions. The adhesive ability to the host's gastrointestinal tract is a crucial criterion for selecting potential probiotic bacteria. Our findings demonstrated that 3201 exhibits significantly higher adhesive capabilities compared with Escherichia coli OP50 (OP50), a standard laboratory food source for C. elegans and is comparable with the widely recognized probiotic L. rhamnosus GG (LGG). In lifespan assay, 3201 significantly increased the longevity of C. elegans compared with OP50. In addition, preconditioning with 3201 enhanced C. elegans immune response against four different foodborne pathogenic bacteria. To uncover the molecular basis of these effects, transcriptome analysis elucidated that 3201 modulates specific gene expression related to the innate immune response in C. elegans. C-type lectin-related genes and lysozyme-related genes, crucial components of the immune system, showed significant upregulation after feeding 3201 compared with OP50. These results suggested that preconditioning with 3201 may enhance the immune response against pathogens. Metabolome analysis revealed increased levels of fumaric acid and succinic acid, metabolites of the citric acid cycle, in C. elegans fed with 3201 compared with OP50. Furthermore, there was an increase in the levels of lactic acid, a well-known antimicrobial compound. This rise in lactic acid levels may have contributed to the robust defense mechanisms against pathogens. In conclusion, this study demonstrated the probiotic properties of the candidate probiotic L. rhamnosus IDCC 3201 by using multi-omics analysis.
