Efficacy of Korean Red Ginseng Supplementation on Eradication Rate and Gastric Volatile Sulfur Compound Levels after Helicobacter pylori Eradication Therapy

  • This clinical study was performed to evaluate whether supplementation of proton pump inhibitor (PPI)-based triple therapy with Korean red ginseng can enhance Helicobacter pylori (H. pylori) eradication and reduce levels of halitosis-associated volatile sulfur compounds (VSCs) in the stomach. Seventy-six patients were randomized into an eradication regimen-only group (n=45) or an eradication regimen plus 10 weeks of Korean red ginseng supplementation group (n=31). The eradication regimen consisted of PPI b.i.d., clarithromycin 500 mg b.i.d., and amoxicillin 1 g b.i.d.. for seven days. Korean red ginseng supplementation commenced on the last day of the eradication regimen. $^{13}C$-urea breath test and halimeter measurements were performed prior to protocol repetition. By intention-to-treat analysis, the H. pylori eradication rate in the Korean red ginseng group (77.4%, 24 of 31) was higher than that in the control group (45.0%, 26 of 45). However, by per protocol analysis, the eradication rate in the Korean red ginseng group was significantly higher than that in the control group (92.3%, 24/26 vs. 69.4%, 26/38; p<0.05). H. pylori infection was significantly associated with increased VSC levels. However, VSC levels decreased significantly in the Korean red ginseng group (p<0.05). In conclusion, supplementation of triple therapy with Korean red ginseng increased the H. pylori eradication rate and led to significant reductions in VSC levels, suggesting the usefulness of this substance in combating H. pylori infection.

Optimal First-line Eradication Regimens for Helicobacter pylori Infection in Patients with Clarithromycin Resistance: A Pilot Study (국내 클래리스로마이신 내성균에서 가장 효과적인 헬리코박터 제균 치료법)

  • Background/Aims: Helicobacter pylori eradication rates using first-line treatment have decreased due to clarithromycin resistance. The aim of this study was to investigate optimal eradication regimens for patients with clarithromycin resistance in Korea. Materials and Methods: A total of 72 patients with confirmed clarithromycin resistance were enrolled from August 2015 to July 2017. Patients were randomized to a 7-day bismuth quadruple therapy (BQT) regimen or a 7-day metronidazole triple therapy (MTT) regimen. Eradication was confirmed using the $^{13}C$-urea breath test. Results: There were no differences in baseline characteristics between the groups. Intention-to-treat eradication rates were 77.8% for the BQT group and 66.7% for the MTT group (P=0.293). Per protocol eradication rates were 87.5% for the BQT group and 77.4% for the MTT group (P=0.292). Adverse events were more frequent in the BQT group. Conclusions: Eradication rates using MTT were comparable to those using BQT, and adverse events were less frequent in the MTT group. Thus, MTT may be considered as a first-line regimen for patients with clarithromycin resistance. Since this was a pilot study, a study with a large group is required.

Investigation of seroepidemiology of Mycoplasma hyopneumoniae infection and establishment of on-farm eradication protocol (Mycoplasma hyopneumoniae 감염의 혈청역학적조사 및 농장에서의 근절방안 설정)

  • The purposes of this study are to examine seroprevalence of Mycoplasma hyopneumoniae infection in pigs of different age groups, and retrospectively determine if nursery depopulation (ND) could influence the seroprevalence of M hyopneumoniae infection in nurseries. Sera of 4, 8, 12, 16, and 20 weeks old pigs from 7 farms were first selected from a serum bank to examine serologic profiles for M hyopneumoniae infections. Availability of representative sera in the serum bank was a major criterion for farm selection. The sera were tested for M hyopneumoniae antibodies by an enzyme linked immunosorbent assay (ELISA) using Tween-20 extracted antigen. Serum samples were also selected from 15 of 34 swine farms that previously participated in a ND study. In order to evaluate M hyopneumoniae infection following ND, ELISA was performed with sera of 8~10 weeks old nursery pigs collected prior to and after ND for up to 12 months from the 15 farms. Serological profiles showed positive ELISA titers for 2 of 7 farms at 8 weeks, 4 of 7 farms at 12 weeks, 6 of 7 farms at 16 weeks, 6 of 6 farms at 20 weeks of age. Prior to ND, 11 of the 15 farms had positive titers in sera of 8~10 weeks old pigs. Sera of 8~10 weeks old pigs collected from 7 of the 11 farms (63.6%) were ELISA antibody negative for up to 12 months following ND. In conclusion, seroconversion to M hyopneumoniae was detected commonly between 10~16 weeks of age, indicating the occurrence of natural infection during the nursery age. The ND appeared to be an effective method to prevent M hyopneumoniae infection within the nursery pig in some farms.

The Role of IMO for the Eradication of IUU Fishing (국제해사기구의 IUU 어업 퇴치 기여 및 전망)

  • The illegal, unreported and unregulated fishing can be properly regulated only if fishing activities, transhipment and landing of illegally caught fish, and processing of fish at sea involved in it are integrally managed. To do this, flag States, port States, coastal States, market States and relevant regional fisheries management organizations shall closely cooperate. In addition, fishing boats involved in IUU fishing often ignore requirements of safety of navigation so that they could be exposed to more causes of incidents and could have less chances to be rescued. Then, it seems to be necessary for the regulation on IUU fishing to include a certain consideration of strengthening safety of those ships. International Maritime Organization(IMO) has developed and implemented international rules for the safety of shipping. IMO has cooperated with Food and Agricultural Organization on IUU fishing since 2000 and the third joint meeting is scheduled in November 2015. This paper reviews the recommendations adopted in the previous meetings, and the measures actually taken as a result of the cooperation and, in particular, focus on the additional roles of IMO for the eradication of the IUU fishing.

Eliminating Potato Virus Y (PVY) and Potato Leaf Roll Virus (PLRV) Using Cryotherapy of in vitro-grown Potato Shoot Tips

  • Potato virus Y (PVY) and potato leafroll virus (PLRV) are among the most damaging potato viruses and prevalent in most potato growing areas. In this study, cryopreservation was used to eradicate PVY and PLRV using two cryogenic methods. Potato shoot tips proliferated in vitro were cryopreserved through droplet-vitrification and encapsulation-vitrification using plant vitrification solution 2 (PVS2; 30% glycerol + 15% dimethyl sulfoxide + 15.0% ethylene glycol + 13.7% sucrose) and modified PVS2. Both cryogenic procedures produced similar rates of survival and regrowth, which were lower than those from shoot tip culture alone. The health status of plantlets regenerated from shoot tip culture alone and cryopreservation was checked by reverse transcription-polymerase chain reaction. The frequency of virus-free plants regenerated directly from highly proliferating shoot tips reached 42.3% and 48.6% for PVY and PLRV, respectively. In comparison, the frequency of PVY and PLRV eradication after cryopreservation was 91.3~99.7% following shoot-tip culture. The highest cryopreserved shoot tip regeneration rate was observed when shoot tips were 1.0~1.5 mm in length, but virus eradication rates were very similar (96.4~99.7%), regardless of shoot tip size. This efficient cryotherapy protocol developed to eliminate viruses can also be used to prepare potato material for safe long-term preservation and the production of virus-free plants.

Elimination of respiratory pathogens in endemically infected swine herds by nursery depopulation (Nursery depopulation 기법에 의한 돼지 호흡기질병 상재돈군의 호흡기 병인체 전파방지에 관한 연구)

  • Recently new technologies for the establishment of high health herds are becoming efficient tools in the control of PRRS virus and secondary infections. Medicated early weaning(MEW) and nursery depopulation(ND) have shown to be one of the most successful procedures in the eradication and control of pathogens. Indirect evidence of the role of PRRSV in precipitating secondary infection comes from successful improvement in growth and in decreasing mortality on farms that have eliminated PRRSV through ND. Hence the present experiments were conducted in an effort to compare ND with MEW procedures as a means of eliminating PRRSV controlling secondary pathogens and improving performance of pigs in endemically infected swine herds. Following MEW and ND procedures practiced in the farms, some benefits obtained were as follows: 1. A decrease in PRRSV circulation in the nursery, but no entire elimination. 2. Decrease in the frequency of secondary bacteria and in the use of antibiotics. 3. Mycoplasma hyopneumoniae infection was prevented during the nursery stage. 4. ND protocol had a lower cost and management changes than MEW techniques. 5. Nursery performance was improved after the depopulation, cleaning and disinfection procedures, even though PRRSV still being cycled in the old nursery rooms. These studies revealed that the MEW and ND protocols are not always successful for PRRS virus elimination but it's great effect on control of secondary pathogens and improvement of performance make MEW and ND an efficient tools for the establishment of healthier and more efficient herds.

Bactericidal Efficacy of Oxidized Silver against Biofilms Formed by Curtobacterium flaccumfaciens pv. flaccumfaciens

  • Bacterial wilt is a re-emerging disease on dry bean and can affect many other crop species within the Fabaceae. The causal agent, Curtobacterium flaccumfaciens pv. flaccumfaciens (CFF), is a small, Gram-positive, rod-shaped bacterium that is seed-transmitted. Infections in the host become systemic, leading to wilting and economic loss. Clean seed programs and bactericidal seed treatments are two critical management tools. This study characterizes the efficacies of five bactericidal chemicals against CFF. It was hypothesized that this bacterium was capable of forming biofilms, and that the cells within biofilms would be more tolerant to bactericidal treatments. The minimum biocide eradication concentration assay protocol was used to grow CFF biofilms, expose the biofilms to bactericides, and enumerate survivors compared to a non-treated control (water). Streptomycin and oxysilver bisulfate had EC95 values at the lowest concentrations and are likely the best candidates for seed treatment products for controlling seed-borne bacterial wilt of bean. The results showed that CFF formed biofilms during at least two phases of the bacterial wilt disease cycle, and the biofilms were much more difficult to eradicate than their planktonic counterparts. Overall, biofilm formation by CFF is an important part of the bacterial wilt disease cycle in dry edible bean and antibiofilm bactericides such as streptomycin and oxysilver bisulfate may be best suited for use in disease management.

Interpretation of tube agglutination test for bovine brucellosis with turbidimetric readings and international unit

  • The tube agglutination test has been used for bovine brucellosis diagnosis in Korea since middle 1950s. The reported high specificity was its value in eradication program. However, the reading of reaction mostly depends on personal experience, thus here we report a way to improve accuracy and uniformity of reading. The tube agglutination was conducted according to the protocol provided by Korean Ministry of Agriculture and Forestry. The intensity of reaction was measured by spectrophotometer. The relationship between turbidity and percentage clearing was generally in direct proportion and linear. The correspondent percent transmittance at 75, 50, and 25% clearing were 91, 82, and 73, respectively. Then, the degree of percentage of clearing at given international unit was measured. With about 1.5 unit of serum, the maximum percentage clearing was observed. The international unit showing 25, 50, and 75 percentage clearing were 0.61, 0.83 and 1.35, respectively. Based on the information obtained using international standard serum, the calculation of international unit of test serum was available. According to the protocol for bovine brucellosis diagnosis which provided by Korean Ministry of Agriculture and Forestry, the available range of detectable international unit was between 15 and 538. And the corresponding international units for suspicious case ranged between 42 and 127. Of the 35 sera from B abortus infected cattle, about half of them had more than 538 international units. Collectively, the reading of turbidity using spectrophotometer and application to international unit improved accuracy and uniformity of reading.

Controlling by Effective Pruning of Twigs Showing Black Shoot Blight Disease Symptoms in Apple Trees (사과나무에서 가지검은마름병 억제를 위한 효율적 가지치기)

  • Black shoot blight disease caused by Erwinia pyrifoliae have damaged economic loss to apple and pear growers until now since it was firstly reported in 1995 in Korea. This study was performed to reduce economic loss by mandatory eradication of all infected trees in case of more 10% disease incidence per orchard as official control. It also aims to set up effective management protocol for this disease by examining how far bacterial pathogen is present from the border of symptomatic and asymptomatic regions in infected apple twigs. Colony-PCR using isolated bacterial cells instead of genomic DNA was used to identify bacterial pathogen, EpSPF/EpSPR primer designed in enterobacterial repetitive intergenic consensus (ERIC) region was selected as specific for E. pyrifoliae. As results of monitoring of this disease during April to October in 2014-2015 by colony-PCR, occurrence of this disease was frequent from mid-May to early-July, when daily average temperature was around $25^{\circ}C$. Moreover, bacterial cells were continuously detected only in symptomatic regions and also asymptomatic regions of less than 20 cm from symptomatic regions. Therefore, we concluded that pruning of infected twigs at the region of more than 20 cm from symptomatic regions might be effective to manage black shoot blight disease in apple trees.
