Lack of any Association between Blood Groups and Lung Cancer, Independent of Histology

  • Introduction: Lung cancer, the leading cause of cancer deaths, is divided into 2 main classes based on its biology, therapy and prognosis: non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC). Many cases are at an advanced stage at diagnosis, which is a major obstacle to improving outcomes. It is important to define the high risk group patients for early diagnosis and chance of cure. Blood group antigens are chemical components on erythrocyte membranes but they are also expressed on a variety of epithelial cells. Links between ABO blood groups with benign or malignant diseases, such as gastric and pancreas cancers, have been observed for a long time. In this study, we aimed to investigate any possible relationship between lung cancer histological subtypes and ABO-Rh blood groups. Materials and Methods: The files of 307 pathologically confirmed lung cancer patients were reviewed retrospectively. Cases with a serologically determined blood group and Rh factor were included and those with a history of another primary cancer were excluded, leaving a total of 221. The distribution of blood groups of the lung cancer patients were compared with the distribution of blood groups of healthy donors admitted to the Turkish Red Crescent Blood Service in our city in the year 2012. Results: There was no significant difference between patients with lung cancer of either type and the control group in terms of distribution of ABO blood groups and Rh factor (p: 0.073). There was also no relationship with non small cell cancer histological subtypes. Conclusions: In this study, we found no relationship between the ABO-Rhesus blood groups and NSCLC and SCLC groups. To our knowledge this is the first analysis of ABO blood groups in SCLC patients.

Role of CXCR7 and Effects on CXCL12 in SiHa Cells and Upregulation in Cervical Squamous Cell Carcinomas in Uighur Women

  • CXCR7 is involved in tumor development and metastasis in multiple malignancies. However, the function and molecular mechanisms of action of CXCR7 in human cervical cancer are still unclear. In the present study a loss of-function approach was used to observe the effects of recombinant CXCR7 specific small interfering RNA pBSilence1.1 plasmids on biological behavior including proliferative activity and invasive potential, as indicated by MTT assays with the cervical cancer SiHa cell line in vitro. Reverse transcription polymerase chain reaction and Western blotting revealed that CXCR7 was downregulated in transfected compared with control cells, associated with inhibited cell growth, invasiveness and migration. The expression of CXCR7 and CXCL12 was also determined immunohistochemically in 152 paraffin-embedded, cervical squamous cell carcinoma (CSCC) and cervical intraepithelial neoplasia (CIN), or normal cervical epithelial to assess clinico-pathological pattern and CXCR7 status with respect to cell differentiation and lymph node metastasis in Uighur patients with CSCC. CXCR7 and CXCL12 expression was higher in cervical cancer than CIN and normal cervical mucosa, especially in those with higher stage and lymph node metastasis. CXCL12 appeared to be positively regulated by CXCR7 at the post-transcriptional level in CSCC. We propose that aberrant expression of CXCR7 plays a role in carcinogenesis, differentiation and metastasis of CSCC, implying its use as a potential target for clinical biomarkers in differentiation and lymph node metastasis.

Expression Patterns of Cancer Stem Cell Markers During Specific Celecoxib Therapy in Multistep Rat Colon Carcinogenesis Bioassays

  • The purpose of this study was to investigate the role of colon cancer stem cells (CSCs) during chemically-induced rat multi-step colon carcinogenesis with or without the treatment with a specific cyclooxygenase-2 inhibitor drug (celecoxib). Two experiments were performed, the first, a short term 12 week colon carcinogenesis bioassay in which only surrogate markers for colon cancer, aberrant crypt foci (ACF) lesions, were formed. The other experiment was a medium term colon cancer rat assay in which tumors had developed after 32 weeks. Treatment with celecoxib lowered the numbers of ACF, as well as the tumor volumes and multiplicities after 32 weeks. Immunohistochemical proliferating cell nuclear antigen (PCNA) labeling indexes LI (%) were downregulated after treatment by celecoxib. Also different cell surface antigens known to associate with CSCs such as the epithelial cell adhesion molecule (EpCAM), CD44 and CD133 were compared between the two experiments and showed differential expression patterns depending on the stage of carcinogenesis and treatment with celecoxib. Flow cytometric analysis demonstrated that the numbers of CD133 cells were increased in the colonic epithelium after 12 weeks while those of CD44 but not CD133 cells were increased after 32 weeks. Moreover, aldehyde dehydrogenase-1 activity levels in the colonic epithelium (a known CSC marker) detected by ELISA assay were found down-regulated after 12 weeks, but were up-regulated after 32 weeks. The data have also shown that the protective effect of celecoxib on these specific markers and populations of CSCs and on other molecular processes such as apoptosis targeted by this drug may vary depending on the genetic and phenotypic stages of carcinogenesis. Therefore, uncovering these distinction roles of CSCs during different phases of carcinogenesis and during specific treatment could be useful for targeted therapy.

Resveratrol Exerts Differential Effects in Vitro and in Vivo against Ovarian Cancer Cells

  • Epithelial ovarian cancer represents the most lethal gynecological cancer, and the high mortality rate makes this malignancy a major health concern. Poor prognosis results from an inability to detect ovarian cancers at an early, curable stage, as well as from the lack of an effective therapy. Thus, effective and novel strategies for prevention and treatment with non-toxic agents merit serious consideration. Resveratrol, obtained from grapes, berries, peanuts and red wine, has been shown to have a potent growth-inhibitory effect against various human cancer cells as well as in in vivo preclinical cancer models. The objective here was to evaluate potential antitumor effects of resveratrol in both in vitro and in vivo NuTu-19 ovarian cancer models. In vitro an invasion assay was performed. After 48 h, the numbers of viable cells that invaded the extracellular matrix layer were reduced by 94% with resveratrol in comparison to control. For the in vivo anti-tumor assessment, 10 rats were injected with NuTu-19 cells into the ovarian bursa. Thereafter, half were provided with a diet mixed with a dose of 100 mg resveratrol/kg body weight/day for 28 days. Following sacrifice, anticancer effects were assessed by histological evaluation of ovarian as well as surrounding tissues, and immunohistochemical detection of cell proliferation and apoptosis, but there were no observable differences between the control and resveratrol-treated groups for any of the biological endpoints. While resveratrol is effective in suppressing the in vitro cellular invasion of NuTu-19 ovarian cancer cells, these effects do not appear to impact on in vivo NuTu-19 ovarian cancers in rats.

Elevated Expression of Nuclear Protein Kinase CK2α as a Poor Prognosis Indicator in Lymph Node Cancerous Metastases of Human Thyroid Cancers

  • Aim: To investigate the expression of protein kinase $CK2{\alpha}$ ($CK2{\alpha}$) in human thyroid disease and its relationship with thyroid cancer metastasis. Materials and Methods: Using immunohistochemistry we measured the expression of $CK2{\alpha}$ in 76 benign and malignant human thyroid cancer tissues, including 10 pairs of papillary carcinoma tissues with or without lymph node cancerous metastasis and similarly 10 pairs of lymph nodes. Results: The expression of $CK2{\alpha}$ was found to be higher in thyroid carcinoma cases (papillary carcinoma, follicular carcinoma, anaplastic carcinoma and medullary carcinoma) than in ones such as chronic lymphocytic thyroiditis, nodular goiter and adenoma. These findings were also confirmed by RT-PCR and Western blotting. More strikingly, elevated expression of $CK2{\alpha}$ in thyroid papillary carcinoma tissues was not only significantly associated with lymph node cancerous metastasis and clinical stage of thyroid cancers; but also correlated with epithelial-mesenchymal transition (EMT) and high tenascin C (TNC) expression. In addition, EMT and high TNC expression in thyroid carcinoma tissues was significantly associated with lymph node cancerous metastasis. Conclusions: Elevated expression of nuclear $CK2{\alpha}$ is a poor prognosis indicator in lymph node cancerous metastasis of human thyroid cancers.

중이염의 병인에 관한 실험적 연구 (Experimental Study on the Pathogenesis of Otitis Media - Histopathological and Biochemical Characteristics of Middle Ear Mucosa Infected with Pseudomonas aeruginosa in Rats -)

  • 백서를 사용한 동물실험을 통하여 조직병리학적 및 생화학적 측면에서 중이염의 병인에 관한 연구를 시행하였다. 기본적인 백서고실골포의 해부도 및 정상상피세포의 분포를 함께 보고하는 바이다. 록농균중이염은 $10^{9}$/ml농도의 Pseudomonas aeruginosa SL 45를 백서의 고실 골포에 주입하여 유발시키었다. 점골막층의 조직병리소견은 3 일에서 3주까지 급성기, 4주부터 만성기의 소견을 보였으며, 12주까지 관찰하였다. 중이강내의 염증조직으로 부터 PZ-peptidase, LDH, Lysozyme의 활성도를 측정한 결과 이에 상응하는 혈청보다 높은 값을 얻었으며, 이효소들의 활성도의 혈청에 대한 조직의 비는 PZ-peptidase는 13-38, LDH는63-177, lysozyme은 18-94의 결과를 얻었다. 또한, 조직내에서 PZ-peptidase의 역할 및 이들 세 enzymes의 기원에 관해 문헌고찰 및 그 가능성에 대해서 살펴보았다.

소 수정란에서 Green Fluorescent Protein 유전자 검색 및 PCR에 의한 성감별 (Screening of Green Fluorescent Protein Gene and Sexing by PCR in Bovine Embryos)

  • The efficiency of transgenic livestock production could be improved by early screening of transgene-integration and sexing of embryos at preimplantational stages before trasferring them into recipients. We examined the effciency of multiplex PCR analysis for the simultaneous confirmation of the trasgene and sex during the preimplantational development of bovine embryos and the possibility of green fluorescent protein(GFP) gene as a non-invasive marker for the early screening of transgenic embryos. The GFP gene was microinjected into the male pronuclei of bovine zygotes produced in vitro. The injected zygotes were co-cultured in TCM-199 containing 10% FCS with boving oviductal epithelial cells in a 5% CO2 incubator. Seventeen(13.0%) out of 136 gene-injected bovine zygotes developed by multiplex PCR analysis and the expression of GFP was detected by observing green fluorescence in embryos under a fluorescent microscope. Eight(67%) of 12 embryos at 2-cell to blastocyst stage were positive in the PCR analysis, but only two(11.8%) of 17 blastocysts expressed the GFP gene. Their sex was determined as 7 female and 5 male embryos by the PCR analysis. The results indicate that the screening of GFP gene and sex in bovine embryos by PCR analysis and fluorescence detection could be a promisible method for the preselection of transgenic embryos.

크��토스포리디움증의 실험실적 진단법 (Techniques for laboratory diagnosis of cryptosporidiosis)

  • Diagnosis of cryptosporidiosis is currently confirmed by the detection of the oocysts or endogenous stages in fecal or tissue samples. Various conventional staining methods and serodiagnostic techniques have been reported, but the latter has far been limited to a few laboratories. Cryptosporidium has recently been reported in mice and chiekens in Korea, but there has been no report on staining methods to the oocysts. The present study was performed by light and scanning electron microscopic observations, and discussed with staining properties of four conventional methods such as dichromate solution floatation method, Carbol fuchsin stain, Auramine-O stain and Giemsa stain method. Cryptosporidial oocysts were isolated from the laboratory mouse. In tissue sections of duodenum, jejunum, ileum, cecum and upper colon, numerous very small, basophilic bodies were observed on the border of mucosal epithelial cells. In scanning electron microscopic observations, a few of developmental stages of Cryptosporidium were seen. Two types of thick and thin-walled oocysts were recognized in the intestinal contents. Mean size of its were $5.19{\pm}0.23{\times}4.31{\pm}0.32{\mu}m$ and $5.14{\pm}0.25{\times}4.27{\pm}0.4{\mu}m$, respectively. Carbol fuchsin and Auramine-O stain methods are recommended as the satisfactory ones for the identification of Cryptosporidium oocysts. Giemsa stain was also recommended as available in the laboratory, because a few of developmental stage fo Cryptosporidium could be seen by it.

식도암에서 경열공 식도절제술의 적용에 대한 고찰 (Transhiatal Esophagectomy in Esophageal Cancer)

  • 식도암에 대한 주된 치료법은 외과적 절제로서, 식도암 수술 후의 성적은 불량한 가운데에서도 지난 20여년 간 지속적으로 개선되어 왔다. 그리고 예후가 불량한 식도암에서의 수술은 근본적으로 보존적인 치료로서의 의미가 커서, 수술에 의한 이병률과 사망률이 낮고 연하기능과 더 나아가서 생존이 장기간 유지될 수 있다면 소기의 성과를 얻었다고 할 수 있을 것이다. 현재 식도암에 대하여 시행되고 있는 식도절제술은 크게 식도열공을 통한 비개흉적 식도절제술과 개흉적 식도절제술로 대별할 수 있으며, 전자의 큰 장점은 수술시간이 단축되어 수술에 의한 이병률과 사망률이 낮으며, 호흡기계에 대한 장해도 적어 입원기간이 단축된다는 점이다. 이 수술의 적응으로는 이론의 여지가 있으나 크게 다발성의 표재성 병변(T1)과 절제가 가능하나 전신상태가 불량하여 개흉술에 위험이 따르는 경우라고 할 수 있을 것이다. 저자들은 식도암환자에 대한 경열공 식도절제술식의 적용기준과 술식, 그리고 성적 등에 관하여 문헌고찰과 함께 기술하였다.

Expression and Prognostic Significance of lncRNA MALAT1 in Pancreatic Cancer Tissues

  • Background: Long non-coding RNAs (lncRNAs) have been recently observed in various human cancers. However, the role of lncRNAs in pancreatic duct adenocarcinoma (PDAC) remains unclarified. The aim of this study was to detect the expression of lncRNA MALAT1 in PDAC formalin-fixed, paraffin embedded (FFPE) tissues and to investigate the clinical significance of the MALAT1 level. Methods: The expression of MALAT1 was examined in 45 PDAC and 25 adjacent non-cancerous FFPE tissues, as well as in five PDAC cell lines and a normal pancreatic epithelium cell line HPDE6c-7, using qRT-PCR. The relationship between MALAT1 level and clinicopathological parameters of PDAC was analyzed with the Kaplan-Meier method and Cox proportional hazards model. Results: The relative level of MALAT1 was significantly higher in PDAC compared to the adjacent normal pancreatic tissues (p=0.009). When comparing the MALAT1 level in the cultured cell lines, remarkably higher expression of MALAT1 was found in aspc-1 PDAC cells compared with the immortal pancreatic duct epithelial cell line HPDE6c-7 (q=7.573, p<0.05). Furthermore, MALAT1 expression level showed significant correlation with tumor size (r=0.35, p=0.018), tumor stage (r=0.439, p=0.003) and depth of invasion (r=0.334, p=0.025). Kaplan-Meier analysis revealed that patients with higher MALAT1 expression had a poorer disease free survival (p=0.043). Additionally, multivariate analysis indicated that overexpression of MALAT1, as well as the tumor location and nerve invasion, was an independent predictor of disease-specific survival of PDAC. Conclusion: MALAT1 might be considered as a potential prognostic indicator and may be a target for diagnosis and gene therapy for PDAC.
