A Super-Absorbent Polymer Combination Promotes Bacterial Aggressiveness Uncoupled from the Epiphytic Population

  • Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.

Evaluation of Fermentation Quality of a Tropical and Temperate Forage Crops Ensiled with Additives of Fermented Juice of Epiphytic Lactic Acid Bacteria (FJLB)

  • This study aimed to examine the fermentation quality of a tropical Elephant grass (Pennisetum purpuereum) and temperate Italian ryegrass (Lolium multiflorum) forages ensiled additive of fermented juice of epiphytic lactic acid bacteria (LAB) and to determine what factor affects the fermentation characteristics of the crops. In both species cell walls neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents in silages were consistently decreased (p<0.05) with the addition of FJLB at ensiling more then Control treatment. The FJLB additive increased number of LAB (cfu) and lactate concentration in the silages in both species. The Control treatment without additive underwent a clostridial type of fermentation with traces of propionic, iso-butyric, n-butyric acids contents with higher (p<0.01) levels of volatile basic nitrogen (VBN %TN) and had appreciable decreased of nutrient in silages. FJLB treatment improved silage nutritive value with little contents of VBN %TN, ethanol and very small amount of dry matter (DM) and hemicellulose losses (p<0.05) between 2 to 5% and 7 to 3% respectively, in Elephant grass and Italian ryegrass species. The results in this study indicates that while among the factors affecting silage fermentation butyric type of fermentation was more pronounced in tropical elephant grass compared to the temperate Italian ryegrass, FJLB additive revealed a better silage fermentation products in both species.

Ultrastructure of the Epiphytic Sooty Mold Capnodium on Walnut Leaves

  • Cellular aspects of sooty mold on walnut leaves were investigated by using light and electron microscopy. A black coating developed on the adaxial leaf surface of a walnut tree. No infestations were found on the abaxial leaf surface with peltate glandular trichomes. Light microscopy showed that fungal complexes from the leaf surface were composed of brown conidia and hyphae. Conidia, with longitudinal and transverse septa, were variable in length ranging from 10 to $30{\mu}m$, and commonly found in clusters, forming microsclerotia. Neither epidermal penetration nor hyphal entrance to host tissues was observed. Based on their morphological characteristics, the fungal complexes were assumed to be Capnodium species. An electron-dense melanized layer was present on the cell wall of multi-celled conidia. Concentric bodies in the fungal cytoplasm had an electron-translucent core surrounded by an electron-dense margin with a fibrillar sheath. Chloroplasts without starch granules in the palisade mesophyll cells of sooty leaves had electron-dense stromata and swollen plastoglobuli. These results suggest that the epiphytic growth of fungal complexes can be attributed to the melanized layer and concentric bodies against a water-deficient environment on the leaf surface. Ultrastructural characteristics of the sooty leaves indicate typical features of dark-adapted and non-photosynthetic shade leaves.

수목착생지의류(樹木着生地衣類)를 이용한 울산지역(蔚山地域)의 대기환경평가(大氣環境評價) (Estimation of Air Pollution Using Epiphytic Lichens on Forest Trees around Ulsan Industrial Complex)

  • 대기오염이 심한 것으로 판단되는 울산지역의 석유화학공단과 온산공단을 중심으로 수목착생지의류를 이용한 대기환경의 오염정도를 평가하기 위하여 공단 주변의 산림에서 지의류의 출현종수와 피도, 생육한계분포농도에 따른 분포특성과 대기청정도지수(IAP)를 조사한 결과, 조사지점에서 출현한 지의류는 총 16종류였으며, 그중 Lepraria sp.(30.85%)과 Lecanora strobilina(26.18%), Parmelia austrosinensis(13.42%) 등이 우점하고 있었다. 석유화학공단과 온산공단 주변 조사지점에서는 지의사막대(地衣砂漠帶)의 형성과 더불어 공단으로부터 멀어질수록 출현종수가 증가했다. 조사지점별 평균피도는 I-V계급으로 오염물질이 공단이 위치하는 해안가로부터 내륙으로 유입됨을 추측할 수 있으며, 공단으로부터 멀어질수록 평균피도계급도 증가하였다. $SO_2$ 농도에 대한 지의류의 종별 생육한계분포농도에 따른 분포특성을 Cladonia sp.과 Dirinaria applanata, Parmelia austrosinensis, Lepraia sp., Lecanora strobilina를 대상으로 살펴 본 결과, 오염에 대한 민감정도에 따라 분포형태가 다르게 나타났다. 특히, 대기오염에 내성 종인 Lepraria sp.과 Lecanora strobilina는 I부터 V의 피도계급으로 가장 폭넓게 분포하였으며, 분포형태가 비슷한 Lecanora strobilina도 대기오염에 강한 종임을 추측할 수 있었다. 대기청정도지수(IAP)는 0-64.3으로 6계급으로 구분하여 조사한 결과, 조사지점별 IAP 등치선도는 피도 등치선도와 비슷한 형태로 공단으로부터 멀어질수록 IAP가 높아졌다. IAP와 지의류 출현종의 분포는 IAP가 5-10으로 낮은 부분에서는 대기에 저항성 종으로 알려진 Lepraria sp.과 Lecanora strobilina가 출현하고 있었으며, IAP 5-10 사이부터는 Parmelia austrosinensis와 Dirinaria applanata가 IAP 10이상부터 오염에 비교적 약한 종으로 말려진 Cladonia sp.를 비롯하여 Candelaria concolar와 Parmelia borreri 등이 출현하였고, 조사지점의 IAP와 지의류의 출현종수는 정의 상관관계 (r=0.9308)를 나타내었다.

해초에 부착하는 부착생물 군집의 생태학적 특성 - I. 잘피 ($Zostera$ $marina$ L.)의 성장에 따른 부착생물의 군집 변화 (Ecological Characteristics of the Epiphytes on Seagrass - I. Variations of the Epiphytic Community and Biomass Related to the Host Plant ($Zostera$ $marina$ (eelgrass))

  • 본 논문은 잘피의 성장과 부착생물과의 관계, 그리고 부착생물간의 관계를 이해하고자 실시된 연구로서 여수시 돌산읍 율림리에서 1998년 7월에서 1999년 7월까지 총 13개월 동안 실시되었다. 연구 결과를 토대로 다음과 같은 세 가지 결론을 얻을 수 있었다. 1) 잘피 잎의 면적과 부착 생물의 현존량과는 정상관관계를 나타냄으로써 부착가능 면적이 넓어지게 되면 부착생물의 현존량이 증가함을 확인할 수 있었다. 이것은 해초의 외적인 성장이 증가할수록 부착생물이 부착할 수 있는 시간적 가능성이 증가할 뿐만 아니라 부착이 가능한 면적이 확대되는 기회를 제공해 줄 수 있다. 2) 부착생물의 건중량에 대한 무기물량 및 유기물량의 비율과 엽록소 양과의 상관관계를 살펴볼 때, 부착조류의 경우 석회홍조류나 규조류와 같이 무기물의 비율이 높은 조류의 현존량이 녹조류나 갈조류와 같은 조류보다 상대적으로 높은 것으로 나타났다. 3) 독립영양지수 (AI, Autotrophic Index)를 통해 살펴본 부착생물의 군집은 약 2개월의 종속영양군집이후 1개월의 독립영양군집이 나타나는 현상을 보였으나, 독립영양생물을 위주로 한 군집의 유지기간이 짧고 지수의 값이 높은 것을 고려할 때, 유기물질 또는 미생물 등을 위주로 한 종속영양생물의 군집이 우점한다고 유추할 수 있었다.

Morphology and molecular characterization of the epiphytic dinoflagellate Amphidinium massartii, isolated from the temperate waters off Jeju Island, Korea

  • Amphidinium massartii Biecheler is an epiphytic and toxic dinoflagellate. Prior to the present study, A. massartii has been reported in the waters off the Mediterranean, Australian, USA, and Canadian coasts. We isolated Amphidinium cells from the coastal waters of Jeju Island, Korea and their morphology and rDNA sequences indicated that they were A. massartii. Herein, we report for the first time the occurrence of A. massartii in the waters of the temperate region in the northwestern Pacific Ocean. The large subunit (LSU) rDNA sequences of the Korean strains were 0.7% different from those of an Australian strain of A. massartii CS-259, the closest species, but were 4.1-5.8% different from those of the other Australian strains and the USA strains of A. massartii and from those of Amphidinium sp. HG115 that was isolated from subtropical Okinawan waters. In phylogenetic trees based on LSU, internal transcribed spacer, small subunit rDNA, and cytochrome b sequences, the Korean strains belonged to the A. massartii clade, which was clearly divergent from the A. carterae clade. The morphology of the Korean A. massartii strains was similar to that of the originally described French strain and recently described Australian strain. However, we report for the first time here that scales were observed on the surface of the flagella. In conclusion, the Korean A. massartii strains have unique rDNA sequences, even though they have a very similar morphology to that of previously reported strains. This report extends the known range of this dinoflagellate to the temperate waters of the northwestern Pacific Ocean.

Effect of storage time and the level of formic acid on fermentation characteristics, epiphytic microflora, carbohydrate components and in vitro digestibility of rice straw silage

  • Objective: The study aimed to evaluate the effect of storage time and formic acid (FA) on fermentation characteristics, epiphytic microflora, carbohydrate components and in vitro digestibility of rice straw silage. Methods: Fresh rice straw was ensiled with four levels of FA (0%, 0.2%, 0.4%, and 0.6% of fresh weight) for 3, 6, 9, 15, 30, and 60 d. At each time point, the silos were opened and sampled for chemical and microbial analyses. Meanwhile, the fresh and 60-d ensiled rice straw were further subjected to in vitro analyses. Results: The results showed that 0.2% and 0.6% FA both produced well-preserved silages with low pH value and undetected butyric acid, whereas it was converse for 0.4% FA. The populations of enterobacteria, yeasts, moulds and aerobic bacteria were suppressed by 0.2% and 0.6% FA, resulting in lower dry matter loss, ammonia nitrogen and ethanol content (p<0.05). The increase of FA linearly (p<0.001) decreased neutral detergent fibre and hemicellulose, linearly (p<0.001) increased residual water soluble carbohydrate, glucose, fructose and xylose. The in vitro gas production of rice straw was decreased by ensilage but the initial gas production rate was increased, and further improved by FA application (p<0.05). No obvious difference of FA application on in vitro digestibility of dry matter, neutral detergent fibre, and acid detergent fibre was observed (p>0.05). Conclusion: The 0.2% FA application level promoted lactic acid fermentation while 0.6% FA restricted all microbial fermentation of rice straw silages. Rice straw ensiled with 0.2% FA or 0.6% FA improved its nutrient preservation without affecting digestion, with the 0.6% FA level best.

덕유산 엽상지의식물의 집락분석 (Cluster Analysis of the Foliose Lichens in Mt. Duckyoo)

  • The epiphytic lichen communities were analysed in terms of cluster analysis on forty two stands and eight environmental variables in Mt. Duckyoo. Ordination of stand and species by principal component analysis (PCA) and sum of square algorithm (SSA) gave similar results. Species cluster showed three groups(I, II, III) and stand revealed three groups (A, B, C). Interaction of stand and species cluster was interpreted by analysis of concentration technique. The results indicated a significant cluster structure at the level of different environment variable.

Additive Effects of Green Tea on Fermented Juice of Epiphytic Lactic Acid Bacteria (FJLB) and the Fermentative Quality of Rhodesgrass Silage

  • Two experiments were carried out on a laboratory scale. The first involved a study of the effect of green tea on characteristics of fermented juice of epiphytic lactic acid bacteria (FJLB). FJLB was treated with 50 g/L of green tea products as follows: new shoot powder (FJLB+N), leaf powder (FJLB+L), commercial powder (FJLB+P), sterilized new shoot powder (FJLB+SN), sterilized leaf powder (FJLB+SL) or sterilized commercial powder (FJLB+SP). FJLB without any additive was also prepared (Untreated FJLB). After incubation, the number of microorganisms in FJLB were studied. Subsequently, these FJLB were applied at 10 ml/kg to chopped rhodesgrass to study their effects on fermentation. Compared with untreated FJLB, the addition of green tea increased (p<0.05) lactic acid bacteria (LAB) and also aerobic bacteria counts in FJLB. At 60 d of ensiling, all the FJLB treated silages were well preserved, pH and butyric acid content were lower (p<0.001) and lactic acid was higher (p<0.001) than that of the control. Lactic acid content was significantly higher (p<0.001) with treated FJLB than with untreated FJLB. FJLB treated with sterilized green tea decreased (p<0.001) the pH and the lactic acid content was higher (p<0.001) than that in the unsterilized green tea silages.
