The Effects of Mori folium on Insulin Resistance and Adipose Tissue Inflammation in an Experimental Mouse Model of Obesity (상엽(桑葉)이 비만 유발 생쥐의 인슐린 저항성 및 지방세포 염증에 미치는 영향)

  • Objective: This study was undertaken to investigate the effects of Mori folium on insulin resistance and adipose tissue inflammation in an experimental mouse model of obesity.Methods: Obesity was induced in C57BL/6 mice by feeding them a high-fat diet. The mice were divided into four groups (n=6): a normal diet, high-fat diet, high-fat diet with 40 mg of Mori folium, and high-fat diet with 800 mg of Mori folium groups. After 13 wk, the body weights, fasting blood glucose and fasting serum insulin levels, insulin resistance (homeostatic model assessment) levels, oral glucose tolerance test levels, epididymal fat and liver weights, and gene expression of tumor necrosis factor-α, interleukin-6, and interferon-γ were measured. In addition, adipose tissue macrophages were analyzed by fluorescence-activated cell sorting.Results: Mori folium significantly reduced blood glucose levels, oral glucose tolerance levels, and liver weights. It also reduced adipose tissue macrophage numbers and tumor necrosis factor receptor-α gene expression.Conclusions: These results show that Mori folium has insulin resistance reduction and anti-inflammatory effects in an experimental mouse model of obesity.

The Hypolipidemic Effect of Allium Hookeri in Rats Fed with a High Fat Diet (삼채가 고지방식이를 급여한 흰쥐의 고지혈증 개선 효과에 미치는 영향)

  • This study evaluated the effect of Allium hookeri roots on lipid metabolism of the serum, liver, and adipose tissues induced by a high-fat diet in male Sprague-Dawley rats. The rats were divided into four groups with 8 rats per group for 4 weeks: the normal-diet group (N), the high-fat diet (HFD) group, the HFD containing 3% Allium hookeri (HFD-A3) group, and the HFD containing 5% Allium hookeri (HFD-A5) group. The results showed that the body weight gain and food intake of rats in the HFC-A3 and HFC-A5 groups were significantly decreased compared with those in the HFD group. The epididymal adipose tissue weight in the HFD-A5 group was significantly decreased compared with those in the HFD group, and adipose tissue weights of liver and mesenteric adipose tissues in the HFD-A3 and HFD-A5 groups were significantly decreased compared with those in the HFD group. Serum triglyceride, total cholesterol and LDL cholesterol concentrations, atherogenic index, and cardiac risk factor were significantly lower in the HFD-A3 and HFD-A5 groups than in the HFD group. Serum lipid profiles, as well as ALT and AST activities did not show any difference in all groups. Serum ALP and LDH activities were suppressed in the HFD-A5 group compared with those in the HFD group. The hepatic triglyceride and total cholesterol levels of rats in the HFD-A5 group was significantly lower than those in the HFD group. Moreover, triglyceride and total cholesterol in the epididymal and mesenteric adipose tissues were significantly lower in the HFD-A5 group than in the HFD group. These results demonstrated that the intake of Allium hookeri showed a hypolipidemic effect, changing the lipid metabolsim of a high-fat diet induced rats.

Incorporation of $Acetate-1-^{14}C$ into Lipid of Rats Fed High Fat Diet (고지방식이(高脂肪食餌)에 따른 $Acetate-1-^{14}C$이 흰쥐의 체내지질(體內脂質)에의 편입도(編入度))

  • The long-term effects of vegetable and animal high fat diet on the lipid metabolism were investigated in male weaning rats. The rats were fed one of four semipurified diet ad libitum : control diet supplied 12% of calories as fat(control group), low fat diet supplied 3% of calories as fat (3% F group), 45% corn oil diet supplied 45% calories from corn oil (45% C group) and 45% butte. fat diet supplied 45% calories from butter fat (45 % B group). Incorporation of $acetate-1-^{14}C$ into lipid of liver, serum and adipose tissue as well as exhalation of $^{14}CO_{2}$ from $acetate-1-^{14}C$ were observed in rats fed for 4,8 and 12 weeks. The weigh of epididymal adipose tissue of rats, fed 45% corn oil and 45% butter fat . from 4 weeks to 8 weeks were higher, but not different at 12 weeks, compared with those of control group. The weight of abdominal adipose tissue appeared to be similar to those of epididymal adipose tissue. Incorporations of $acetate-1-^{14}C$ into lipid of liver were remarkably decreased in high fat diet groups, especially in 45% C group, but in 3% F group were increased more than those of control group. Incorporations of $acetate-1-^{14}C$ into epididymal adipose tissue in 3% F, 45% C and 45% B group at 8 weeks were remarkably increased but not different at 12 weeks, compared with those of control group. The incorporation of {acetate-1-}^{14}C into abdominal adipose tissue appeared to be similar to those of epididymal adipose tissue. Incorporations of $acetate-1-^{14}C$ into lipid of serum in 45% C and 45% B group were reduced reasonably at 4 and 8 weeks of diet as compared with those of control group. Exhalation of $^{14}CO_{2}$ was increased to maximum level at 10 minutes after injection of $acetate-1-^{14}C$. Expiration of $^{14}CO_{2}$ in 45% C and 45% B group were higher than those in 3% F and control group for initial 5 minutes after injection, but expirations of $^{14}CO_{2}$ did not have significant difference among groups of diet since 10 minutes.

UCP2 KO mice exhibit ameliorated obesity and inflammation induced by high-fat diet feeding

  • Uncoupling protein 2 (Ucp2) was first introduced as a member of Uncoupling protein family and a regulator of ROS formation; however, its role in adipose tissue is not fully understood. In the present study, we have investigated the role of Ucp2 against high-fat diet (HFD)-induced obesity in epididymal white adipose tissue (eWAT) and browning of inguinal white adipose tissue (iWAT). Diet-induced obesity is closely related to macrophage infiltration and the secretion of pro-inflammatory cytokines. Macrophages surround adipocytes and form a crown-like-structure (CLS). Some reports have suggested that CLS formation requires adipocyte apoptosis. After 12 weeks of HFD challenge, Ucp2 knockout (KO) mice maintained relatively lean phenotypes compared to wild-type (WT) mice. In eWAT, macrophage infiltration, CLS formation, and inflammatory cytokines were reduced in HFD KO mice compared to HFD WT mice. Surprisingly, we found that apoptotic signals were also reduced in the Ucp2 KO mice. Our study suggests that Ucp2 deficiency may prevent diet-induced obesity by regulating adipocyte apoptosis. However, Ucp2 deficiency did not affect the browning capacity of iWAT.

Suppression of Adiposity and Improvement of Fat Metabolism in High-fat Diet-induced Obese Mice Treated with an Inonotus obliquus Extract (고지방식이 유도 비만 마우스에서 차가버섯(Inonotus obliquus) 추출물의 체중 증가 억제 및 지방대사 개선 작용)

  • Using high-fat diet-induced obese (DIO) mice, the mechanism of anti-adiposity and anti-obesity actions produced by Inonotus obliquus water extract (IOE) was investigated. Significant reduction in body weight in DIO mice orally administrated with IOE for 8 weeks compared to IOE-non-treated control mice was observed, which was attributed to the reduction of epididymal and mesenteric adipose tissue, but not the liver and skeletal muscle. Adiponectin synthesis in epididymal adipose tissue (EAT) and AMPK phosphorylation in the liver were significantly increased in IOE-treated DIO mice. Gene expression analysis showed that IOE-treated DIO mice had higher expression levels of lipogenic genes in EAT and fatty-acid oxidative genes in the liver, but lower expression levels of hepatic pro-inflammatory cytokines compared to IOE-non-treated controls. Our findings confirm a therapeutic potential of Inonotus obliquus for reducing adiposity and ameliorating hyperlipidemia to treat metabolic disorders.

A Comparative Study on Anti-Obesity Efficacy of Cydonia oblonga Miller Fruit Extract in Diet-Induced Obesity Animal Models (식이유도 비만 동물모델에서 마르멜로추출물의 항비만 효능 비교 연구)

  • Objectives: The objective of this study was to explore the anti-obesity effect of Cydonia oblonga Miller fruit extract (COME) and to compare its anti-obesity efficacy with Garcinia cambogia extract (GCE) in diet-induced obese mice. Methods: Five-week-old male C57BL/6 were allocated into four groups: control diet (CD), high-fat diet (HFD), HFD + 400 mg/kg body weight (BW)/day COME (H+C), or HFD + 400 mg/kg BW/day GCE (H+G) groups. COME or GCE was administered once a day by oral gavage for eight weeks. Body weight, body fat percentage, fat weight, and biochemical parameters in serum were measured. The expressions of transcription factors and their target genes in epididymal adipose tissues were analyzed by reverse transcription polymerase chain reaction. Results: COME reduced body weight, weight gain, body fat percentage, total white adipose tissue weight, adipocyte size, and serum levels of insulin and leptin in high-fat diet-induced obese C57BL/6 mice. COME suppressed the mRNA expressions of CCAAT/enhancer binding proteinα, peroxisome proliferator-activated receptorγ, sterol-regulatory element-binding protein-1c, fatty acid synthase, and adipocyte protein 2 and increased carnitine palmitoyl transferase 1 mRNA expression in epidydimal adipose tissues. The anti-obesity efficacy of COME was found to be similar to that of GCE at the same dose. However, COME more effectively decreased adipose tissue weights, epididymal adipocyte size, serum insulin and leptin compared to GCE. Conclusions: These results demonstrated that COME is not toxic and exhibits anti-obesity efficacy at a level similar to that of GCE, suggesting that COME may be applicable as an anti-obesity agent.

Effect of Cheongpesagan-tang on the Change of Inhibitory Effect against Lipase Activity and Weight Loss, Plasma and UCP1, 2 mRNA Expression in db/db Mouse (태음인 청폐사간탕이 lipase 활성저해 및 db/db mouse의 체중감량, 혈장지질, UCP 1, 2 발현에 미치는 영향)

Objectvies This experimental study was designed to investigate the effect of cheongpesagan-tang extract on the obstruction of the lipase activity and weight, plasma, UCP1, 2 mRNA in db/db mouse. Material and Methods: The body weight loss, food intake, feeding efficiency ratio, weight of the internal organs (liver, kidney, epididymal fat, brown adipose tissue), plasma glucose, triglyceride, total cholesterol, white adipose tissue, adipocyte size distribution, expression of UCP1, 2 mRNA were measured in db/db mouse administered Cheongpesagan-tang extract for 6 weeks. These were then compared with those of control groups administered the diet. 2. Results 1) Inhibitory effect against lipase activity was Kilgyung(81.7%), Nabokja (73.1%), Seungma(73.0%), Daewhang (68.4%), Kalgeun (55.3%), Kobon(34.5%), Hwanggeum(4.2%). 2) In the sample group, the body weight was significantly decrease than that of control group. 3) In the sample group, the weight of epididymal fat showed significantly decrease than that of control group. 4) In the sample group, triglyceride showed significantly decrease than that of control group. 5) In the sample group, distribution of adipose tissue showed significantly larger than that of control group. 6) In the sample group, UCP1, 2 mRNA in BAT showed significantly increase than that of control group. 3. Conclusions These results show that cheongpesagan-tang has an effect on the treatment of obesity.

The Effects in Metabolism and Adipose Tissue Inflammation Induced by the Massa Medicata Fermentata on Obese Type 2 Diabetes Mouse Model (신국(神麯) 투여가 비만형 제 2형 당뇨병 동물모델의 대사인자와 지방조직 염증반응에 미치는 영향)

  • Objectives: Recent data have revealed that the plasma concentration of inflammatory mediators is increased in the insulin-resistant states of obesity and type 2 diabetes. The purpose of this study was to investigate the antidiabetic and anti-obesity effect of Massa Medicata Fermentata on obese type 2 diabetes mice. Methods: In order to examine the effects of Massa Medicata Fermentata, obese type 2 diabetes mice induced by Surwit's high fat, high sucrose diet. Mice were divided into 4 groups of ND (normal diet), HFD (high fat and high sucrose diet), Met (high fat and high sucrose diet with metformin) and MMF (high fat and high sucrose diet with Massa Medicata Fermentata) and investigated over 8 weeks. Diabetic and obese clinical markers, including body weight, glucose level, lipid level, leptin concentration, epididymal fat pad and liver weights and adipose tissue macrophage (ATM) were determined. Results: Compared with the HFD group, body weight, fructosamine, triglyceride, epididymal fat pad weight and ATM were significantly reduced in the MMF group. Conclusions: From the above results, the intake of Massa Medicata Fermentata may be effective in anti-hyperglycemia and anti-obesity by the attenuation of glucose and lipid levels and also inflammation state. Massa Medicata Fermentata may be beneficial for controlling diabetes mellitus type 2 in humans.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

Deletion of adipose triglyceride lipase abolishes blood flow increase after β3-adrenergic stimulation in visceral adipose tissue of mice

  • Dynamic changes in adipose tissue blood flow (ATBF) with nutritional status play a role in the regulation of metabolic and endocrine functions. Activation of the sympathetic nervous system via β-adrenergic receptors (β-AR) contributes to the control of postprandial enhancement of ATBF. Herein, we sought to identify the role of each β-AR subtype in the regulation of ATBF in mice. We monitored the changes in visceral epididymal ATBF (VAT BF), induced by local infusion of dobutamine, salbutamol, and CL316,243 (a selective β1-, β2-, and β3-AR agonist, respectively) into VAT of lean CD-1 mice and global adipose triglyceride lipase (ATGL) knockout (KO) mice, using laser Doppler flowmetry. Administration of CL316,243, known to promote lipolysis in adipocytes, significantly increased VAT BF of CD-1 mice to a greater extent compared to that of the vehicle, whereas administration of dobutamine or salbutamol did not produce significant differences in VAT BF. The increase in VAT BF induced by β3-AR stimulation disappeared in ATGL KO mice as opposed to their wild-type (WT) littermates, implying a role of ATGL-mediated lipolysis in the regulation of VAT BF. Different vascular reactivities occurred despite no significant differences in vessel density and adiposity between the groups. Additionally, the expression levels of the angiogenesis-related genes were significantly higher in VAT of ATGL KO mice than in that of WT, implicating an association of ATBF responsiveness with angiogenic activity in VAT. Our findings suggest a potential role of β3-AR signaling in the regulation of VAT BF via ATGL-mediated lipolysis in mice.
