Enzyme Hydrolysis of Insoluble sericin (불용성 세리신의 효소 가수분해)

  • To hydrolyze insolule sericin the enzyme hydrolysis was used, and then obtained the results as given belows. When insoluble sericin was hydrolyzed by enzyme treatment, the solubility was best at pH 7, 60$\^{C}$ and was slightly increased both above 2 hours treatment and above 10% of enzyme concentration. As the results of electrophoresis, the distribution of molecular weight of sericin powder obtained by enzyme hydrolysis was very weak and showed in the wide range having no distinguishable band. Average degree of polymerzations (A.D.P.) of sericin hydrolyzed by enzyme were about 4.1∼6.3, average molecular weight were about 470∼730. The whiteness of the sericin powder obtained by enzyme hydrolysis was high and increased slightly with higher treatment concentration of enzyme. As the results of amino acid analysis, the amino acid analysis, the amino acid composition of the sericin powder from the enzyme treatment were similar to which located at near 230$\^{C}$ and 320$\^{C}$. The peak of near 230$\^{C}$ could not been found in the sericin powder obtained by enzyme hydrolysis.

  • In this work, colorants extraction process from gromwell was studied for making powder form of colorants by solving the high viscosity problem of gromwell extracts. In order to do that, sugar extracted together with colorants must be pre-extracted. For sugar decomposition, gromwell roots were pretreated with various enzyme solutions. The total sugar content of pre-extract with enzyme solution was measured. Accordingly, the effects of enzyme type and pretreatment condition on sugar decomposition were investigated to find appropriate enzyme(amylase, hemicellulase, pectinase) and enzyme activity (100~1000unit), pre-extracted time(3~24hr). Color characteristics and dye uptake of dyed fabrics were evaluated. Gromwell colorants were assessed for their potential antimicrobial activities, which possibly expand their end use as functional pigments. The efficiency of removing sugar was increased in the order of hemicellulase, pectinase, amylase, $H_2O$. Gromwell colorants powder yield was in the range of 4.4% to 9.8% depending on pretreatment enzyme. Gromwell colorants produced RP color on the silk and wool fabrics with good dye uptake. Antimicrobial activity of gromwell colorants will greatly increase its potentiality for applying as functional natural colorants in the future.

Effect of enzyme treatment on the DSC and TGA behavior of silkworm powder

  • Silkworm powder's thermal property is an important factor for its storage and marketing. This study examined the effect of edible enzyme on the thermal property of silkworm powder using Thermogravimetric Analysis (TGA) and Differential Scanning Calorimetry (DSC). Results of the TGA showed that regardless of the enzyme treatment, the weight loss patterns of silkworm powders exhibited 3 step thermal property deterioration at approximately $80^{\circ}C$, $280^{\circ}C$, and $480^{\circ}C$ due to water evaporation and thermal degradation. This is similar with the DSC which also resulted in all samples two endothermic peaks attributed also to water evaporation and thermal degradation. These results indicated that the use of enzyme such as protease and cellulase might not affect significantly the thermal properties of silkworm powder.

Physicochemical Components and Antioxidative Activity of Wheat Sprout Powder Prepared by the Enzyme and the Lactic Acid Bacteria (효소와 유산균 전처리 밀싹분말의 이화학적 성분 및 항산화 활성)

  • The purpose of this study was to evaluate the physicochemical components and antioxidant activities of wheat sprout powder prepared by the enzymatic hydrolysis and lactic acid bacteria treatment. The four kinds of pre-treatment were: no treatment (WP), treated with enzyme (WPE), treated with lactic acid bacteria (WPL), and treated with enzyme and lactic acid bacteria (WPEL) were applied to the wheat sprout powder. The WPEL had higher total free amino acid and essential amino acid content than the other samples. As for the volatile aroma of the wheat sprout powder, 29 types of compounds were identified in the WP and WPL, 28 types in the WPE, and 27 types in the WPEL, respectively. The total polyphenols and flavonoids contents, in the wheat sprout powder was enhanced with the enzyme and the lactic acid bacteria pre-treatment. The WPEL had highest DPPH radical scavenging activities. The overall acceptability was the highest at 6.24 points in the WPEL. Based on these observations, it was confirmed that the enzyme and lactic acid bacteria pre-treatment could improve the antioxidant activities and active component of the wheat sprout powder.

Quality Characteristics of White Pan Bread Added with Wheat Sprout Powder by Enzyme and Lactic Acid Bacteria Pretreatment (효소와 유산균으로 전처리한 밀싹분말을 첨가한 식빵의 품질 특성)

  • The purpose of this study was to evaluate the quality characteristics of white pan bread added with wheat sprout powder without treatment (WP) and wheat sprout powder with only enzyme treatment (WPE), only lactic acid bacteria treatment (WPL) and enzyme and lactic acid bacteria treatment (WPE&L). The three different powder concentration levels of 1%, 3%, and 5% were added to flour to produce the white pan bread. The bread volume and specific volume of the WPE&L group were the highest among all the addition groups. The bread weight, a-value, and b-value of the WP group was highest among all the addition groups, but the bread baking loss and the L-value of the WP group was the lowest among all the addition groups. The texture measurements indicated that the hardness, gumminess, and chewiness values of the bread were the highest in the WP group. The sensory evaluation test showed that bread in the WPE&L group with 3% wheat sprout powder was the best among all the samples studied. Based on our findings, we suggest that the enzyme and lactic acid bacteria pretreated wheat sprout powder is an effective ingredient for improving the overall quality of white pan bread.

  • This study was carried out to investigate the effects of added emulsifier and enzyme on the quality characteristics of Seolgiddeok with black rice powder, green tea powder, Lentines edodes powder, pumpkin powder during storage. Samlpes were divided into two groups, the emulsifier-enzyme added group and the control, and stored fur 4 days at 2$0^{\circ}C$. Moisture content was measured by the air oven method; the Hunter's cole. values by Lightness (L), redness (a), yellowness (b) value; the degree of gelatinization by the enzyme digestion method, and expressed as glucose content by Somogi-Nelson method. The degree of retrogradation was calculated as the retrogradation rate using the degree of gelatinization value, and total cell and mold counts were also measured. Texture and sensory characteristics were also investigated. The results are as follows: The emulsifier-enzyme group was significantly different from the control from each examination and the retrogradation of pumpkin Seolgiddeok with emulsifier-enzyme was delayed the most among samples tested.

Effect of Enzyme-Treated Radish Leaves on Lipid Metabolism in Rats Fed a High-Fat Diet

  • The purpose of this study was to investigate the effect of enzyme-treated radish (Raphanus sativus L.) leaves on lipid metabolism in rats fed a high fat diet. Rats were divided into four experimental groups which were composed of a high fat diet group (HF group), a high fat diet with 10% radish leaf powder-supplemented group (MA group), a high fat diet with 5% enzyme-treated radish leaf powder-supplemented group (MB group) and a high fat diet with 10% enzyme-treated radish leaf powder-supplemented group (MC group). Total dietary fiber content of enzyme-treated radish leaves were greater than untreated radish leaves. Body weight gain and food efficiency ratio (FER) of the HF group increased compared to the MA, MB and MC groups. The serum total cholesterol, LDL-cholesterol and atherogenic index contents in the radish leaf powder-supplemented groups were lower than that of the HF group, while those values for the MB and MC groups were significantly lower than that of the HF group. The serum HDL-cholesterol contents of the MB and MC groups increased compared to the HF group. The hepatic triglyceride contents of the MA, MB and MC groups decreased compared to the HF group. In fact, the hepatic triglyceride contents of the MB and MC groups were significantly lower than the MA group. The hepatic total cholesterol contents of the MB and MC groups significantly decreased compared to those of the HF group. The fecal total cholesterol contents of the MA, MB and MC groups significantly increased compared to those of the HF group. These results indicate that supplementation with enzyme-treated radish leaves increase the useful fiber contents. Furthermore, it may have a pronounced impact on lipid metabolism of serum and liver in rats fed a high fat diet.

Effects of Medicinal Enzyme Powder on Intestinal Mobility, Lipid Level, and Blood Parameters of Loperamide-Induced Constipation in Rats (약선효소 분말식이가 Loperamide의 투여로 유발된 변비 쥐의 장 운동과 지질 함량 및 혈액학적 성분 변화에 미치는 영향)

  • This study was aimed at investigating whether dietary therapy using medicinal enzyme powder is effective in reducing constipation caused by loperamide in rats. Nine-week-old male Sprague Dawley were subdivided into 4 groups: normal diet group (C), loperamide treatment and normal diet (CL), medicinal enzyme powder diet (E), and loperamide treatment and medicinal enzyme powder diet (EL). Constipation was induced by subcutaneous injection of loperamide (1.5 mg/kg) 3 days prior to sacrifice. The treatment with loperamide led to an increase in weight gain, a decrease in the number and wet weight of fecal pellets, and a decrease in intestinal motility. The administration of the medicinal enzyme powder significantly reduced weight gain but increased intestinal mobility compared with the loperamide-treated group. The treatment with loperamide in the normal diet group reduced the activities of both suggesting that constipation may be involved in the low level of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT). Additionally, the loperamide treatment in the medicinal enzyme powder diet group increased the level of GOT, but reduced the level of GPT. Loperamide treatment also reduced cholesterol and increased the atherogenic index (AI) and cardiac risk factors (CRFs). Interestingly, the treatment with the medicinal enzyme powder effectively attenuated both the increase in AI and the reduction in high density lipopretein (HDL)-cholesterol, caused by the treatment with loperamide. Although there were no significant differences in the blood protein level, including hemoglobin and hematocrit, between the normal diet group and the loperamide-treated group, the administration of the medicinal enzyme powder to the loperamide-treated group effectively increased the levels of both hemoglobin and hematocrit. Collectively, the results demonstrate that the medicinal enzyme powder can help to combat the negative events caused by constipation.

Effects of Acorn Supplementation on Lipid Profiles and Antioxidant Enzyme Activities in High Fat Diet-Induced Obese Rats (고지방 식이로 유도된 비만흰쥐의 체내 지질패턴 및 항산화효소 활성에 도토리 급여의 효과)

  • This study was performed to investigate the effect of acorn supplementation on the lipid profile and redox antioxidant enzyme activities in obese rat. Obesity in the rats was induced by feeding diet contained 10% lard and 0.5% cholesterol for 4 week. After 4 weeks, rats were divided into the following 5 groups; high fat diet (Control), high fat diet plus 10% Acorn powder (APlO%), high fat diet plus 20% Acorn powder (AP20%), high fat diet plus 0.2% Acorn extract (AE0.2%), high fat diet plus 0.5% Acorn extract (AE0.5%). Total food intake and food efficiency ratio (FER) was not significantly different by acorn powder and extract supplementation. But, body weight was decreased by 20% acorn powder. Acorn powder and extract supplementation for 4 weeks tend to decrease total cholesterol and triglyceride level on the serum and hepatic tissue. There was no significant difference in hepatic glutathione (GSH) content among all the groups. The hepatic GST activity in acorn supplemented groups was lower than that of control. Glutathione peroxidase and catalase activities were higher in acorn supplemented groups than that of control. Hepatic TBARS levels of experimental groups were also significantly lower than that of control group. Our finding suggest that acorn powders and extract might have potential role for improving lipid profiles and antioxidant enzyme activities in obese rats.

Modification of the characteristics of silkworm powder by treatment with alkaline protease

  • Enzymatic modification of proteins is often used to increase the biological activity of materials. Silkworm powder has been investigated as a functional food resource, but no study has been performed on its modification by commercial food enzyme. Therefore, this study aimed to determine the feasibility of such modification of silkworm powder by alkaline protease. The activity of the enzyme was confirmed using an azocasein assay. Subsequently the silkworm powder was hydrolyzed by enzymatic treatment. UV visible spectrometry showed that the supernatant of silkworm powder subjected to enzymatic treatment had a stronger absorption band than the untreated powder. SDS-PAGE electrophoresis showed that the molecular weight of silkworm powder decreased on enzymatic treatment. Thus the results indicate that commercial enzymes might be used to modify the characteristics of silkworm powder.
