Alkalophilic Bacillus circulans가 생산하는 Cyclodextrin Glucanotransferase 의 정제와 효소반응특성 (Purification and Characterization of Cyclodextrin Glucanotransferase Excreted from Newly Isolated Alkalophilic Bacillus circulans)

  • 토양으로부터 역가높은 CGTase를 분비하는 호알칼리성 미생물을 분리하였으며, 동정 결과 Bacillus circulans로 판정되었다. 배양액 중의 CGTase를 ammonium sulfate 침전, DEAE-Sephadex 그리고 Sephadex G-100 column chromatography로 분리, 정제하여 단일 단백질 band를 얻었다. 정제된 CGTase의 분자량은 약 93,000, 최적 pH와 온도는 6.0, $50^{\circ}C$였으며, pH와 온도안정성은 5.5-11, $65^{\circ}C$까지였다. Soluble starch를 기질로 할 때의 $V_{max}$$K_{m}$ 값은 각각 0.16$\mu$mole $\beta$-CD/min, 14.3mg soluble starch/mi이였고 24시간 반응액의 $\alpha$-:$\beta$-:${\gamma}$-CD 의 생성비율은 1:8.1:1.9로서 $\beta$-CD를 우선적으로 합성하였다. 기질로 glucose와 maltose를 사용하였을 때 CD합성작용이 없었으며, sweet potato 그리고 cornstarch를 사용하였을 때 가장 높은 CD합성작용을 보였다. 어느 수준 이상의 과다한 CGTase 첨가경우에는 $\alpha$-CD생성이 급격히 증가하였다. 또한 정제된 CGTase는 stevioside에로의 당전이성을 갖고 있었다.

Three Protein Kinases from the Etiolated Oat Seedlings Phosphorylate Oat Phytochrome A In Vitro

  • Phosphorylation of phytochrome may play important functional roles to control plant photomorphogenesis. Many attempts have failed to identify the protein kinase that phosphorylates phytochrome in vivo. It has been reported that a polycation-stimulated protein kinase activity was associated with the purified phytochrome. However, it is not known if the kinase activity is an intrinsic property of phytochrome or whether it comes from a contaminant of the purified phytochrome. In the present study, three protein kinases that phosphorylate phytochrome have been identified from etiolated oat seedlings. A polycationstimulated protein kinase that had very similar enzymatic properties with that associated with the purified phytochrome was identified in the cytosolic extract. It phosphorylated several contaminant proteins in the kinase preparation as well as phytochrome and had a broad substrate specificity. A CK II-type protein kinase phosphorylated phytochrome and the exogenously added casein. It is likely that this kinase may not be a feasible candidate for the kinase phosphorylating phytochrome in vivo since the content of the kinase seemed to well exceed the content of phytochrome in the etiolated oat seedlings. Another protein kinase that had unique enzymatic properties phosphorylated phytochrome very specifically and seemed to be present in a small quantity in the etiohlted seedlings. It is expected that one of three kinases may be responsible for the phytochrome phosphorylation in vivo.

효소분해가 감쥬스의 이화학적 특성에 미치는 영향 (Effect on Enzymatic Hydrolysis on the Physicochemical Properties of Persimmon Juice)

  • 마쇄한 감 puree에 2종류의 상업적 다른 탄수화물 분해효소인 Viscozyme과 Celluclast를 처리하여 얻은 감쥬스의 추출수율, 점도, 색, 적정산도, 당에 미치는 영향을 조사하였다. Arabinase, cellulase, xylanase, hemicellulase와 ${\beta}-glucanase$의 효소활성을 가진 Viscozyme으로 $50^{\circ}C$에서 10분간 가수분해할 때 추출수율, L값, 환원당은 현저히 증가하였고 점도는 감소하였다. 감쥬스의 추출수율은 Viscozyme으로 처리시 42.8%에서 80%로 증가하는 반면 Celluclast는 73.3%로 증가하였다. 한편 관능적 특성중 감맛과 감내, 주홍색과 주황색은 Viscozyme으로 60분간 가수분해한 쥬스에서 크게 향상되었다.

Enzymatic Properties of Cytochrome Oxidase from Bovine Heart and Rat Tissues

  • Cytochrome oxidase was purified from bovine-heart mitochondria and its enzymatic properties were examined. The purified cytochrome oxidase was identified by its absorption spectrum and chromatogram through gel filtration. The specific activity, purification degree and yield of purified cytochrome oxidase were 18 nmol/mg/ml/min, 24.83 fold and 0.93%, respectively. The activity of the enzyme assayed by a ferrocytochrome $c-O_2$ system was optimized at $25^{\circ}C$ and pH 6.5. Examining the effect of nonionic detergents established that cytochrome oxidase was deactivated by Triton X-100. The oxidase was activated by Tween 80 and deactivated by Tween 20. The Michaelis constant and maximum velocity of the oxidase for ferrocytochrome c were 0.032~0.044 mM and 0.019~0.021 mM/min, respectively. After adaption to basal diet for a week, experimental diets containing 6 mg Cu/kg, or zero mg Cu/kg, or 12 mg Cu/kg were fed to a control group, a copper-free group and a copper-rich group of Sprague-Dawley rats, respectively, for 4 weeks. The specific activities assayed for the ferrocytochrome $c-O_2$ system of isolated cytochrome oxidase from the rat liver of control, copper-free, and copper-rich group were 1.00, 1.19, and 0.878 nmol/mg/ml/min, respectively. Their degrees of purification were 11.38, 10.82 and 8.78 fold, respectively. The specific activities for liver and heart mitochondrial cytochrome oxidase of copper-free/copper-rich groups assayed using the ferrocytochrome $c-O_2$ system were 81.4% and 96.4%/64.1% and 61.1%, respectively, compared with those of the control.

전통 이화주의 양조와 관련된 미생물 및 효소적 특성 (Microbial and Enzymatic Properties Related to Brewing of Traditional Ewhaju)

  • 이화주의 전통적 배경을 조사하여 주품으로써의 위치를 채조명하며 전래된 리화주 양조방법을 현지답사, 확인하고 전통적인 방법으로 누룩을 만들어 리화주를 양조하여 누룩과 제조중인 리화주에 대하여 미생물 및 효소적 실험을 수행한 결과는 다음과 같다. 리화주 누룩의 미생물은 Aspergillus 와 oryzae와 Hanse-nula sp. 가 주종이었으며, 균수는 각각 1.2$\times$$10^6$ CHU/g 이었고 기타 미생물은 희석 배양에서도 생육되지 않았다. a-amylase 활성은 누룩이 30.7, 이화주는 담금 직후 19.3,숙성 100일에 21.2, 1년간 숙성된 것도 20.3이었으며, 0-amylase활성은 누룩이 34.4, 이화주 담금 직후 18.8, 숙성 100일에 19.8, 1년간 숙성된 것은 19.9이었다. 저장성에 있어서도 가열처리나 보존제의 첨가 없이 장기저장이 가능할 뿐 아니라 자장후에도 amylase의 활성도가 상당히 높아서 소화를 촉진할 수도 있는 저알코올성 전통주로서 개발 가치가 있다고 생각된다.

Effect of C- or D-Domain Deletion on Enzymatic Properties of Cyclodextrin Glucanotransferase from Bacillus stearothermophilus NO2

  • To analyze the role of the C and D domains in the cyclization activity of cyclodextrin glucanotransferase (CGTase), two plasmids, pKB1ΔC300 and pKB1ΔD96, were constructed in which DNA regions encoding 100 and 32 amino acids, respectively, from the C and D domains of B. stearothermophilus NO2 CGTase were deleted. The mutated CGTase from the pKBlΔC300 produced much lower amounts of ${\alpha}$-, ${\beta}$-, and $\gamma$-cyclodextrin (CD) than the parental CGTase. However, the mutated CGTase from the pKBlΔD96 showed a similar production pattern of CDs to wild-type CGTase. The production ratios of the ${\alpha}$-, ${\beta}$- and $\gamma$-CDs were not affected by the deletions, when compared to those of parental CGTase. The optimum temperature of the mutated CGTase from the pKBlΔC300 was decreased from $60^{\circ}C$ to $55^{\circ}C$. The optimum pH of the mutated CGTase from the pKB1D96 was shifted from 6.0 to 7.0. The thermostability of the two mutant CGTases were not changed. From these results, it is suggested that the C and D domains are not related to cyclization activity directly because mutant-enzymes deleted C or D domains still possessed their activity. However, they are important for other enzymatic properties such as productivity and pH optimum as a partition of CGTase tertiary structure.

  • 국내 연안의 대표적인 갈조류인 다시마로부터 효과적인 알긴산을 추출하기 위해 다시마 추출액의 전처리 추출공정인 열수 추출공정, 당 또는 염첨가에 의한 추출 및 효소 첨가추출공정에 따른 다시마 추출액의 물성변화를 조사하였다. 열수 추출 및 효소 첨가 추출공정의 경우 추출시간이 증가할수록 점성이 증가함을 보였으며, 추출시간 l시간 이후부터는 비교적 완만한 증가치를 보였다. 당 또는 염첨가에 의한 추출공정시 첨가된 농도가 증가할수록 점성이 증가하였으며, EDTA-2Na의 경우는 첨가농도에 관계없이 매우 낮은 정성치를 나타내었다.

  • This study was conducted to improve yield and functional properties of autoclave-treated salmon frame extracts (SFETA) using commercial enzymes (Alcalase 2.4 L FG, Flavourzyme 500 MG, Neutrase 0.8 L and Protamex 1.5 MG). Yield and angiotensin I converting enzyme (ACE) inhibitory activity of all enzymatic hydrolysates improved compared to those of control (undigested extracts), which were the highest in hydrolysates incubated with Protamex 1.5 MG for 4 hrs (P4-treated hydrolysates) and 2 hrs (P2-treated hydrolysates), respectively. However, antioxidant activities of all enzymatic hydrolysates showed less than 29%. According to the trichloroacetic acid soluble-N, volatile component intensity and sensory evaluation, when compared to control, taste of P4-treated hydrolysates improved, while its fish odor strongly smelt. Therefore, for efficient use of P4-hydrolysates, the fish odor should be improved by Maillard reaction of extracts or pre-treatment of salmon frame.

산처리에 의한 찹쌀 전분의 성질 변화 (Properties of Lintnerized Waxy Rice Starches)

  • 신선찰벼와 한강찰벼 전분을 산처리하여 이호학적 성질, differential scanning calrorimetry 및 효소에 의한 가수분해율의 차이를 비교 검토하였다. 산 가수분해율은 한강찰벼가 약간 높았으며, 산처리에 따라 가용성 선형물질이 분해되어 ${\lambda}_{max}$와 청가는 감소하였고 또 물결합능력, 용해도 및 결정도는 증가하였으나 팽윤력은 감소하였다. 요오드 반응에 의한 최대흡수파장은 한강찰벼가 높았으나 최대흡수파장 및 680nm에서 흡광도는 시료간에 차이가 없었다. 물결합 능력과 용해도는 신선찰벼가 높았으나 팽윤력은 한강찰벼가 높았다. 가열에 의한 광투과도는 신선찰벼가 낮아서 열에 대한 안정성이 높았다. KSCN에 의한 겔부피는 신선찰벼가 커서 용출물질량이 많았다. 산처리중 전분의 결정형태는 A형이 유지되었으며 상대결정도는 신선찰벼가 높았다. DSC특성값은 산처리에 따라 감소하였으며, glucoamylase에 의한 가수분해율은 산처리에 따라 증가하여 산처리 48시간에 신선찰벼는 98%, 한강찰벼는 96%로 전체적으로 신선찰벼가 높았다.

Enzymatic Determination of Somatic Cells by Using Transparisation in Raw Milk

  • The transparisation technology for milk and milk products could be applied widely and very importantly to various determination because transparisation can economize the cost and increase with precision in the milk payment system. Component of butanone or Triton in transparisation solvent would inhibit the growth of bacteria and method. Enzymatic determination of leucocytes were proposed to evaluate milk quality as mastitis in the milk payment system, this can be easily applied to simplify automation of the determation with the lowest investment cost in milk pay system. The significance of this technique, it can be used in the quality control of raw milk and milk products, milk payment system, and programming of national dairy project. Transparisation technology is used in somatic cell counting by enzymic methods. The range of deviation for this method is 16% in 74 samples. But the deviation is increased to 20% when the Infoss method is used. It is affected by the percentage of epithelial cells and white blood cells in somatic cells from different animals and the stages of aging. NAgase activity has an obvious correlation with white-blood cells in milk. In the case of mastitis the white-blood cells is 90-95% in somatic cells in milk, it is showing greater precision in measuring the state of mastitis. In conclusion the enzymic method of somatic cell counting is a relatively simple and easy method of measurement and can be easily practiced. And the importance of this method is also worth utilizing for indirect counting of Somatic cells by use of synthetic substrates to NAgase. In the future, with the further development of the research in this field, it will b possible to automatize the measurement.
