  • This paper was proposed drive strategy Induction Motor using the fuzzy algorithm of auto-tuning PID control. In fuzzy control, the 1M driving system is controlled in trial and error without the mathematical modeling and using fuzzy lookup table the real time control is possible. Also, ,dividing the fuzzy rules in several zone, the stability and response of the 1M driving system is improved. The parameters of 1M are varying according to the environmental conditions, the variance of the parameters is affected with the driving characteristics of 1M. Using the fuzzy algorithm of the driving system which has the auto tuning control function for high performance, high accuracy of the driving system, a designed and proposed through the comparision with the PID control method and the driving characteristics is reviewed and analyzed.

  • Sulfur, an important component of plants, is regulated by a variety of stresses in sulfate assimilation and metabolism. Increase has been observed in the expression of O-acetylserine(thiol)lyase (OASTL) through two-dimensional electrophoresis with the shoot tips of Arabidopsis infected by beet severe curly top geminivirus (BSCTV). With the three- to six-fold increases in the transcript expression of OASTL, serine acetyltransferase (SAT) and $\gamma$-glutmylcysteine synthetase (GSH) were induced over the mock-inoculated organization in each organization through real-time RT-PCR analysis. The expression of those genes might affect the accumulation of anthocyanin in symptomatic tissues and the induction of abnormal callus-like structures formed by additional cell divisions as typical disease symptoms of BSCTV-infected Arabidopsis. This is the first report to describe the collaborative induction of OASTL, SAT, and GSH in virus-infected plants. The changed expressions of OASTL, SAT, and GSH in Arabidopsis infected with BSCTV raises new aspects regarding the biological function of symptomatic tissues related to sulfate metabolism.

A Study on Analysis between Accidents Caused by Human Errors and Personal Characteristics of Railway Drivers (철도기관사들의 개인적 특성과 인적오류사고 발생에 대한 비교 분석)

  • To verify the effect of driver's personal characteristics of driver on the accident frequency through railway accidents caused by human errors and the relationship with aptitude test. To prove the relevance between the driver's personal characteristics and human error accidents. Accident data from 2010 to 2011 was analyzed which collected from a train crew department in K national corporation, and 31 drivers gave an personal interview from Sep. 2011 to Nov. 2011 who had controlled a train alone and caused an accident. Compared between driver's personal characteristics and accident rate, and accident induction possibility surveyed from normal person and disqualified in aptitude tests. Accidents was occurred with the age 40s (27%) and 50s (25%), and with the experience between 15 years and 20 years (38%) and over 20 years (30%). Because more aged, more experienced, it can be seen in the correlation between driver's age and accidents induction caused by human errors like illusion. First of all it must be checked whether working conditions and environmental factors are human error-prone. Most accidents occur when received civil complaints or manager at the riding. Therefore accidents can be prevented when investigated through subsequent surveys how often human error happens, even though no accident, and safety device installed based on the error frequency.

  • Mulberry silkworm is classified into uni, bi and multivoltine based on the frequency of diapause incidence. The variation in the incidence of diapause in bivoltine silkworm provides a unique opportunity to study the process of evolution of adaptive plasticity towards seasonal variations. The diapause expression in bivoltine silkworm is highly variable and is determined by environmental factors experienced by the maternal generation. Diapause in natural populations is functionally associated with the overwintering mechanism that facilitates survival in harsh winter conditions. In contrast, under standard commercial rearing conditions, the domesticated bivoltine silkworm is known to enter diapause in every generation. This paper presents a short review of the literature dealing with the role of temperature, photoperiod, diapause hormone and its receptor in diapause induction. Also, we briefly review the incidence of non-diapause eggs in bivoltine silkworm under controlled conditions.

In vitro culture of adventitious root from Rhodiola sachalinensis (기내배양을 통한 홍경천(Rhodiola sachalinensis)의 부정근 생산)

  • Rhodiola sachalinensis is one of the most popular oriental medicines in East Asia. It is a perennial herb, belonging to the family Crassulaceae, which is mainly distributed in mountains at the altitudes of 1700-2500 m in Baek-Du mountain. Cultivation of this species for the production of medicine materials is not easy in nature, because of restrict habitats. In vitro production of roots can be applicable for the production of medicinal materials. Here, we investigated the optimal conditions for induction and proliferation of adventitious roots in in vitro culture system. Leaf, stem and root segments from R. sachalinensis were cultured on Murashige and Skoog(MS) medium supplemented with the various concentrations of IBA(Indole-3-butyric acid)(0.5, 1.0, 3.0, and 5.0 mg/L) and sucrose(10, 30, 50, and 100 g/L). The optimal explant for adventitious root induction was leaf segment. Induction of adventitious roots was highest on MS medium supplemented with 5.0 mg/L IBA and sucrose 30 g/L. In liquid culture, fresh weight of adventitious roots was highest(15.65 g) on 1/3 strength MS liquid medium supplemented with 5.0 mg/L IBA with 30 g/L sucrose. These results revealed the first attempt for the production of adventitious roots in R. sachalinensis.

Effects of Organic Germanium on Metallothionein Induction in Liver and Kidney of Cadmium and Mercury Intoxicated Rats (유기 게르마늄의 투여가 카드뮴 및 수은에 중독된 흰쥐 간장 및 신장조직의 metallothionein 형성에 미치는 영향)

  • This study was initiated to investigate the effects of organic germanium on cadmium and mercury intoxication. The effect was determined by the metallothionein induction in liver and kidney. Male rats (Sprague-Dawley) were treated with CdCI$_{2}$ (2mg/kg), HgCI$_{2}$ (1 mg/kg) and organic germanium (GE-132) (100 mg/kg) in single and in combination via intraperitoneal injection or intragastric administration every other days for 17 days. Experimental animals were sacrificed after 7, 12 and 17 days treatment. The serum transaminase activities (SGOT, SGPT), concentration of metal and metallothionein, metal-binding capacity of metallothionein in liver and kidney were determined and pathomorphological observations were undertaken. The combined treatment of GE-132 and CdCI$_{2}$ significantly decreased the increment of serum transaminase activities in rats treated with CdCI$_{2}$ only, but the combined treatment of GE-132 and HgCI$_{2}$ did not affect to activities of transaminases induced by mercury only. The concentration of metals (Cd and Hg) except Ge in the liver and kidney of rats increased with the time of treatment. Mercury concentration in kidney of rat treated with HgCI$_{2}$ only was significantly higher than the combined treatment of GE-132 and HgCI$_{2}$. The combined treatment of GE-132 and CdCI$_{2}$ significantly increased the concentration of metallothionein in liver compared to the CdCI$_{2}$ only, although the concentration of cadmium in liver were not significantly different between two groups. This indicates that GE-132 decreased toxicity of cadmium in liver by promoting metallothionein induction. There were no significant differences in metallothionein concentration in liver and kidney of rats between the combined treatment of GE-132 and HgCI$_{2}$ and HgCI$_{2}$ only. Metal-binding capacity of metallothionein varied with each time intervals in liver and kidney of metals treated rats except the liver of the combined treatment of GE-132 and CdCI$_{2}$. This finding explains the concentration of metallothionein in liver keeps abreast with the concention of metal. Furthermore, the combined treatment of GE-132and CdCl$_{2}$ revealed pathologically less changes in liver tissue than CdCl$_{2}$ only; the damages of liver cell, such as lobular necrosis and portal inflammation, were relieved and appeared more later. From the above results, organic germanium is considered to have some beneficial effect on the protection of liver from the cadmium intoxication.

Induction of Petal Color Mutants through Gamma Ray Irradiation in Rooted Cuttings of Rose (장미 삽목묘의 감마선 처리에 의한 화색 돌연변이체 유기)

  • This study was carried out to establish a system for mutation breeding by irradiation of gamma-ray in $Rosa$ $hybrida$ Hort. The rooted cuttings of two roses, 'Spidella' and 'Cabernet' were irradiated with different gamma-ray doses (0, 30, 50, 70, 90, 110, 130, 150 and 170 Gy) from a $^{60}Co$ source to reveal an optimal dose for induction of mutants. The irradiated plants were planted in a greenhouse, and investigated on the appearance of petal color mutants and shoot growth by gamma ray dose. The 50% lethal doses ($LD_{50}$) of plant were 110 Gy for 'Spidella' and 150 Gy for 'Cabernet', respectively. The 50% decrease dose of shoot length was observed at 70-90 Gy dose for 'Spidella', and 110 Gy dose for 'Cabernet'. Solid, chimeric and mosaic petal mutants with various colors were induced from pink petal of 'Spidella' and red petal of 'Cabernet' when 30-170 Gy dose was irradiated. The mutants obtained from 'Spidella' had white, ivory, pinky ivory, light pink and deep pink petal colors. The mutants obtained from 'Cabernet' had pink, deep pink, purple red (magenta), orange red and purple petal colors. It was suitable to irradiate 70-90 Gy dose for 'Spidella' and 90-110 Gy dose for 'Cabernet' for the induction of various mutants considering plant survival rate, shoot growth and mutant occurrence rate.

In vitro Induction of Hepatic Cytochrome P450 (CYP) with Exposure to $\beta$-naphthoflavone in Marine Fishes ($\beta$-naphthoflavone(BNF)에 in vitro 노출시킨 해산 어류의 간장 미크로좀 중 cytochrome P450 (CYP) 유도)

  • Cytochrome P450 (CYP) induction was determined in microsomes of three aquacultured fish species (Sebastes schlegeli, Paralichthys olivaceus and Pagrus major) and two wild fish species (Mugil cephalus and Stephanolepis cirrhifey) in vitro exposed to $\beta$-naphthoflavone (BNF). The microsomes of five fish were exposed to BNF (5 mM or 10 mM) in dimethylsulfoxide at $30^{\circ}C$ for 9 hr. The CYP contents in most fish increased according to exposure duration for 3 or 5 hour, and then decreased, while steady increase of CYP was observed in P. major for 9 hour. The induction of CYP contents in aquacultured fish species (207~422%) were higher than those in wild fish species (206~207%).

Effect of Environmental Factors on In Vivo Folding of Bacillus macerans Cyclodextrin Glycosyltransferase in Recombinant Escherichia coli

  • Effect of environmental factors on the expression of soluble forms of Bacillus macerans cyclodextrin glycosyltransferase in recombinant Escherichia coli BL21(DE3)pLysE:pTCGT1 were investigated. The amount of soluble CGTase produced in the cell was measured by determining its enzymatic activity. The soluble fractionof the enzyme was increased by lowering the culture temperature to $30{\circ}C$ and medium pH to 5.8 compared to the enzyme production in LB medium at $37^{\circ}C$ and pH7.0. Addition of 0.2 M NaCl enhanced enzyme expression levels at the expense of cell growth. Glycine betaine that was added after 3 h of induction protected not only the cell growth from hig osmotic pressue but also hepld in vivo folding of CGTase in recombinant E. coli. Addition of 1 mM $CaCl_2$ was also effective in the expression of soluble CGTase, resulting in 15 U/ml of the enzyme activity.

Relationships between Blossom-End Rot in Pepper and Calcium Fertilization during the First Fruit Setting Period

  • Blossom-end rot is probably the most recognizable Ca deficiency that affects fruit production including pepper. We investigated the induction of blossom-end rot during the period of rapid vegetative growth and fruit setting, and the effect of Ca fertilization on the suppression of blossom-end rot in pepper plants grown in a plastic-film house. During the first fruit setting period, more than 60% of pepper fruits larger than 8 cm showed the symptoms of blossom-end rot. Under the condition of transpiration occurring at a much faster rate, growing leaves of pepper plants could be the greater sinks for Ca than developing fruits. The incidence of blossom-end rot was significantly decreased after two weeks of Ca fertigation. Calcium application during the rapid vegetative growth and fruit setting period could be suggested as a preventive step to overcome the local Ca deficiency inducing blossom-end rot of pepper fruits.
