Stemness and Proliferation of Murine Skin-Derived Precursor Cells under Hypoxic Environment

  • Skin-derived precursors (SKPs) have potential to differentiate to various cell types including osteoblasts, adipocytes and neurons. SKPs are a candidate for cell-based therapy since they are easily accessible and have multipotency. Most mammalian cells are exposed to a low oxygen environment with 1 to 5% $O_2$ concentration in vivo, while 21% $O_2$ concentration is common in in vitro culture. The difference between in vitro and in vivo $O_2$ concentration may affect to the behavior of cultured cells. In this report, we investigated the effect of hypoxic condition on stemness and proliferation of SKPs. The results indicated that SKPs exposed to hypoxic condition for 5 days showed no change in proliferation. In terms of mRNA expression, hypoxia maintained expression of stemness markers; whereas, oncogenes, such as Klf4 and c-Myc, were downregulated, and the expression of Nestin, related to cancer migration, was also downregulated. Thus, SKPs cultured in hypoxia may reduce the risk of cancer in SKP cell-based therapy.

  • As modern society has been progressed, the demolition method of concrete structure that satisfy the condition of safety, economic efficiency, and environment-friendliness is required. This study investigated problems of existing demolition methods and developed modified method minimizing cooling water and sludge for demolition job. It was also verified the validity of this method based on the finite element methods. A analysis parameters as a number, depth and size of boring, and self weight were introduced for this study, and gave optimal condition for the demolition job and analysis.

Effects of Inorganic Aerosols on the Gas-Phase Reactions of the Hydroxyl Radical with Selected Aliphatic Alkanes

  • Effects of inorganic aerosols on the kinetics of the hydroxyl radical reactions with selected aliphatic alkanes have been investigated using the relative rate technique. The relative rates in the absence and presence of aerosols were determined for n-butane, n-pentane, n-hexane, n-octane, and n-decane. P-xylene was used as a reference compound. Inorganic aerosols including (NH$_4$)$_2$SO$_4$, NH$_4$NO$_3$, and NaCl aerosols at two different aerosol concentrations that are typical of polluted urban conditions were tested. Total surface areas of aerosols were 1400 (Condition I) and 3400 $\mu$$m^2$ cm$^{-3}$ (Condition II). Significant changes in the relative rates in the presence of the inorganic aerosols were not observed for the n-butanel/$.$OH, n-pentanel/$.$OH, n-hexanel/$.$OH, n-octanel/$.$OH, and n-decanel/$.$OH reactions versus p-xylenel/$.$OH reaction. These results suggest that the promoting effects depend on the semiconducting property of the aerosols and the nature of the organic compounds.

An Analysis of the Effect of Climate Change on Nakdong River Flow Condition using CGCM ' s Future Climate Information (CGCM의 미래 기후 정보를 이용한 기후변화가 낙동강 유역 유황에 미치는 영향분석)

  • For the assessment of climate change impacts on river flow condition, CGCM 3.1 T63 is selected as future climate information. The projections come from CGCM used to simulate the GHG emission scenario known as A2. Air temperature and precipitation information from the GCM simulations are converted to regional scale data using the statistical downscaling method known as MSPG. Downscaled climate data from GCM are then used as the input data for the modified TANK model to generate regional runoff estimates for 44 river locations in Nakdong river basin. Climate change is expected to reduce the reliability of water supplies in the period of 2021~2030. In the period of 2051~2060, stream flow is expected to be reduced in spring season and increased in summer season. However, it should be noted that there are a lot of uncertainties in such multiple-step analysis used to convert climate information from GCM-based future climate projections into hydrologic information.

  • Context aware computing is an emerging paradigm to achieve ubiquitous computing environments by enabling computer systems to understand their situational contexts. A context aware system uses context to provide relevant information and services to the user depending on the user's task. In this paper, we propose an ontology based context aware modeling methodology that transmits low level contexts acquired by directly accessing various sensors in the physical environments to high level contexts. With these high level contexts, context aware application can provides proactive and intelligent services using ECA (Event Condition Action) rules. We implemented a presentation service in smart office environment.

Analytical Solution of Multi-species Transport Equations Coupled with a First-order Reaction Network Under Various Boundary Conditions (다양한 경계조건을 가진 일차 반응 네트워크로 결합된 다종 오염물 거동 해석해)

  • In this study, analytical solution of multip-species transport equations coupled with a first-order reaction network under constant concentration boundary condition or total flux boundary condition is obtained using similarity transformation approach of Clement et al. (2000). The study shows the schematic process about how multi-species transport equations with first-order sequential reaction network is transformed through the similarity transformation approach into independent and uncoupled single species transport equations with first-order reaction. The analytical solution was verified through the comparison with popular commercial programs such as 2DFATMIC and RT3D. The analytical solution can be utilized in nuclear waste sites where radioactive contaminants and their daughter products occur and in industrial complex cities where chlorinated solvent such as PCE, TCE, and its biodegradation products produces. In addition, it can help the verification of the developed numerical code.

Total Coliforms Viability in Effluent of Wastewater Treatment Plant Discharged into the Ocean (해양으로 배출된 하수처리수내 총대장균군의 활성 평가)

  • Difference of total coliforms (TC) survival time of mixed samples (effluent were mixed by seawater or freshwater) were examined in various conditions. The time taken to achieve a 90% reduction in the TC concentration ($T_{90}$) of effluent, high initial TC concentration and low initial TC concentration samples indicated 143.9, 121.9 and 89.6 hours at $25^{\circ}C$, respectively. At $4^{\circ}C$, log removal rate after 336 hours were 0.96, 1.04, and 1.30. TC survival time of effluent-inoculated seawater sample was longer than that of effluent-inoculated freshwater sample in laboratory. At outdoor condition, TC inactivation curves of effluent-inoculated seawater or freshwater samples showed similar patterns. And both of them were greatly influenced by climate condition. There was not enough evidence that TC survived longer in freshwater than sea water. It was unlikely that the salinity of sea water contributed to the inactivation process of TC.

Biological Removal of a VOC Mixture in a Two-stage Bioreactor (이단미생물반응조에서 혼합 VOCs의 생분해 특성)

  • A two-stage bioreactor, which consists of a biotrickling filter module and a biofilter module in series, was investigated for the enhanced treatment of a VOC mixture, toluene and methyl ethyl ketone (MEK). Throughout the experiments, the overall inlet loading rate was maintained at approximately $43g/m^3/hr$, but the inlet ratios of the VOCs were modified. The experimental results showed that the different ratios of the VOC mixture resulted in changes of overall removal efficiencies, elimination capacities (ECs) and microbial accumulation on the surface of each packing material. The ratio of inlet toluene to MEK at 50 : 150 was found to be most effective in terms of the overall removal efficiency, because, at this condition, MEK (i.e., the hydrophilic compound) was mostly removed in the biotrickling filter module and the following biofilter module was used to remove toluene. It was also found that when the inlet loading rate of the VOC mixture was serially increased stepwise within short-term periods, the ECs for toluene dropped significantly but the ECs for MEK increased at the ratio of the VOC mixture. These results implied that substrate interaction and/or substrate preferable utilization might have an effect on the biological removal of each compound in the two-stage bioreactor; therefore, the bioreactor should be operated in the condition where the substrate interaction could be minimized in order to maximize overall performance of the two-stage bioreactor.

A Study of Vitality Measurements of the Natural Monuments - Focused Natural Monuments Zelkova in Kyongsangdo - (천연기념물 노거수 활력도 측정에 관한 연구 -경상도 천연기념물 느티나무를 중심으로-)

  • As a result of this study, vitality of natural monuments (large trees) looks relatively good but they require continuous management. Also, the management for condition recovery and condition maintenance, and minimum measures are needed at least to avoid natural calamity. Major points induced are as follows : The first, although most of natural monument large trees showed favorable growth environment, most of them are aged and have dense twigs with requirement for weed trimming to remove complicated and unnecessary branches. In addition, as most of large trees grow alone in a large space with potential natural calamity, thus, minimum measures are required at least. Secondly, change in electric resistance in surveyed large trees was relatively favorable, however, large trees with soil covering showed high electric resistance and were found to have low vitality. As shown, this study was conducted to find out the measurement techniques of natural monuments, Basic foundation is suggested through the measurement of electric resistance, and the vitality of natural monuments; more accurate investigation will be remained for further discussion.

Effects of Acid Rain Treatment on Height Growth of Several Landscape Tree Species, pH Value and $Al^{3+}$ Concentration in Soil: Comparison after 5 Years [I] (人工酸性雨 處理가 5年後 몇 가지 造景樹種의 樹高生長, 土壤 酸度와 可溶性 알루미늄의 濃度에 미치는 影響 [I])

  • To identify the long-term influence of acid rain treatment on tree growth, acid rain of various composition (pH 2.0, pH 4.0 and pH 5.6 as control) was applied to several landscape trees for five months (April through August, 1991). Tree height, pH values and $Al^{3+}$ concentration in soil were investigated. Acid rain treatments seemed to promote height growth in the first year (1991), but have become an inhibiting factor over five years. All of coniferous species and most broad-leaved species, except Acer ginnala, showed opposite trends in height responses to acid rain treatments between the first (1991) and last (1996) year. In contrast, Acer ginnala showed similar trends to acid rain treatments in the height growth between 1991 and 1996. This result suggested that Acer ginnala has a characteristic adapability to acid rain stress. pH values of surface soil were lower than those of 30 cm soil depth. This fact suggested that acid rain treatments made surface soil acidic condition. In addition, physiological characteristics (photosynthesis, stomatal condition and biomass) have to be investigated to identify the relationship between long-term effects of $AL^{3+}$ concentration and growth.
