Intelligent Motion Planning System(IMPS) is presented for a robot to achieve an efficient path toward the given target point in two dimensional unknown environment is constructed with unrestricted obstacle shapes. IMPS consists of three components for making intelligent motion. These components are real-time motion planning algorithm based on a discontinous boundary method, fuzzy neural network decision system for heuristic knowledge representation, and world modeling with forgetting and reinforcing memory cells. First of all, in real-time motion planning algorithm, the behavior-based architectural method is used to generate subgoal. A behavior generates a subgoal independently by using the method of discontinuous boundary in sensed area. The discontinuous boundary method is a new proposed fast obstacle avoidance algorithm. The second component is fuzzy neural network decision system for accomplishing the subgoal. The heuristic rules are imbedded on the fuzzy neural network to make an intelligent decision. The last one is a forgetting, reinforcing memory technique for the construction of external world map. The activation values of all activated memory cells in grid space are decreased monotonically and after all they are burned out. Therefore, after sufficient journey, robot can have a stationary world map even if the dynaic obstacles exist. Using the IMPS, several simulations show the efficient achievement of target point in unknown enviroment with obstcles of various shapes.
This study was designed to investigate the productivity of L-methionine by the method of protoplast fusion between Brevibacterium flavum ATCC 14067 and Corynebacterium glutamicm ATCC 13032, and then L-methionine production was performed to continuous fermentation using the immobilized fusant cells. Mutants B. flavum K 104($thr\;met\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}\;as\;genetic\;marker$) and C. glutamicum B 70($thr\;Hos\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}as\;genetic\;marker$) were isolated by MNNG treatment. On the other hand, protoplast of mutants were formed to treat with lysis solution containing $500{\mu}g/ml$ of lysozyme. The ratios of protoplast formation and regeneration were 99% and $64{\sim}66%$ respectively. Fusion frequency between B. flavum K 104 and C. glutamicum B 70 showed the $3.5{\times}10^{5}$ in the 35% polyethylene glycol(PEG6000) containing 3% PVP solution. The productivity of L-methionine by fusant BFCG 37 immobilized with sodium alginate was 0.89 g/l the batch fermentation and was $18.75mg/^{1}hr\;^{1}$ on the continuous fermentation at $30^{\circ}C$ for 72 hr.
Recently, many researches into distributed processing on Internet, and multimedia data processing have been performed. Rapid and convenient multimedia services supplied with high quality and high speed are to be needed. In this paper, we design and implement clip-based video retrieval system on the Web enviroment in real-time. Our system consists of the content-based indexing system supporting convenient services for video content providers, and the Web-based retrieval system in order to make it easy and various information retrieval for users in the Web. Three important methods are used in the content-based indexing system, key frame extracting method by dividing video data, clip file creation method by clustering related information, and video database construction method by using clip unit. In Web-based retrieval system, retrieval method ny using a key word, two dimension browsing method of key frame, and real-time display method of the clip are used. In this paper, we design and implement the system that supports real-time display method of the clip are used. In this paper, we design and implement the system that supports real-time retrieval for video clips on Web environment and provides the multimedia service in stability. The proposed methods show a usefulness of video content providing, and provide an easy method for serching intented video content.
For screening thermostable $\alpha$-amylase from thermophiles, various samples from extreme environments such as hot spring and sewage near them, and compoat, wereexamined microbial growth in enrichment culture medium at 55$\circ$C on the assumption that enzymes from thermophiles are inevitable thermostable. One strain showing higher $\alpha$-amylase activity was pure cultured and designated as Bacillus sp. TR-25 from the results of morphological, cultural and physiological characteristics. The most important carbon sourses for the enzyme production were soluble starch, dextrin, potato starch and corn starch. Glucose and fructose had a catabolite repression on the enzyme production. The good nitrogen sources for the enzyme production were yeat extract, nutrient broth, tryptone, corn steep liquor and ammonium sulfate. The enzyme production was accelerated by addition of CaCl$_{2}$. $\cdot $ H$_{2}$O. The optimal medium composition for the enzyme production was soluble starch 2.0%, yeast extract 0.55, CaCl$_{2}$ $\cdot $ 2H$_{2}$O 0.015, Tween 80 0.001%, pH8.0, respectively. In jar fermenter culture, this strain shows a rapid growth and required cheaper carbon and nitrogen source. These properties are very useful to fermentation industry. The $\alpha$-amylase of this strain demonstrated a maximum activity at 80$\circ$C, pH 5.0, respectively. And calcium ion did not improve thermostability of the enzyme. At 10$0^{\circ}C$, this enzyme has 235 of relative activity. Transformation was carried out by thermophilic Bacillus sp. TR-25 genomic DNA. As a result, the transformant has increased thermostable $\alpha$-amylase activity.
In order to examine the traits of sluice gate water control, halophyte community formation and their inter-relations in Saemangeum, both water level condition and halophyte community formation were analyzed periodically and spatially on the topographic map with Surfer, Saemageum Spatial Analysis System, and related field reports. The traits of water level condition are that average water level in the growing period of halophytes was similar to annual average water level, annual low level and high level appeared in the growing period, and water level was usually maintained within a range of -1.0m~0.5m above mean sea level, but it has changed more frequently year by year. Routine water level control, natural disaster prevention, construction, and civil appeal took major percentages of the reasons for sluice gate's opening and shutting. Since 2007, not only the overall control frequency of sluice gate but also its control frequency for construction and natural disaster prevention have increased outstandingly. Halophyte community had formed at a rate of 1,209ha/year in the 4,315 ha land in 2008, 6.3 times larger than in 2005, and 2,382 ha above around 1.0m was estimated to be artificially vegetated, 89.1 % of the 2,673ha-size sown area. High water level was found to be a more possible determinant than average water level or low water level in halophyte community formation and it was thought to be secondary factors whether tillage was conducted or/and whether surface sealing formed.
Activated sludge samples were collected from a municipal sewage treatment plant and used for enrichment of microbial consortia with aniline as the sole carbon and nitrogen source. Threc aniline-degrading bacteria were obtained lrom microbial consortia and an isolate which has excellent aniline degradability was selected for this study. The isolate was Gram-negative, and identified and designated as Delfha sp. JK-2 on the basis of various physiological and biochemical tests. 10 mM aniline was completely degraded within 24 hours after inoculation of the culture. Ammonium ion was liberated in the medium transiently during the incubation and disappeared when aniline was completely degraded. Addition of glucose as a supplementary source to aniline minimal media showed significant decrease in aniline degradat~on rate for the strain Effective degradation of aniline was achieved by the addition of 0.5% nitrate as a nitrogen source, and resulted in approximately 80% higher aniline degradation compared to the absence of nitrate. Phylogenetic analysis based on 16s [DNA sequence revealed that the strain was closely related to De@ia acidovorans, with 96% overall similarity. The 16s [DNA sequence of JK-2 was also found to be closely related to those of six other clonal types, including Acidovoru, Aquaspirillum. Xylophilus, Variovorm, and Rhodofernr.
Phytoplasmas were detected consistently in 42 mulberry cultivars showing dwarf disease using DNA analysis by amplification with phytoplasma universal primer pairs P1/P7 (about 1.8 kb and R16F2n/R2 (about 1.2 kb). The point mutation from 42 cultivars of mulberry tree was detected by single-strand conformation polymorphism (SSCP) analysis. The SSCP profiles were clearly observed from all of cultivars in 8% polyacrylamide gel, electrophoresizing for and running 8-15 hrs. at 150V, $10^{\circ}C$. The MD and JWB phytoplasma PCR products was mixed and electrophoresis was performed to detect their polymorphism. In this results, the SSCP profiles of all bands of MD and JWB were analyzed on single lane and were distinct in their each of band patterns. The SSCP analysis was possible to detect of 1.8 kb and 1.2 kb nucleotide size and near close band patterns were distinct by mix of two samples. Previously, it was only possible to detect of point mutation under 600 bp nucleotide sequence by SSCP analysis but this modification of SSCP technique was possible to detect clearly SSCP band patterns of about 1.8 kb and 1.2 kb nucleotides.
This study was performed to investigate the bioconcentration of IBP, methidathion and piperophos. The BCFs(bioconcentration factor), depuration rate constants for three pesticides in zebrafish(Brachydanio rerio) were measured by OECD guideline 305. The concentration of test pesicides were one-hundredth and one-thousandth concentration of 96-hrs L $C_{50}$ in accordance with OECD guideline 305. The results obtained are summarized as follows: The average BCF values of IBP were 5.31(n=4) and 7.30(n=4) at one-hundredth and one-thousandth concentration of 96-hrs L $C_{50}$ . The average BCF values of methidathion were 8.72(n=4) and 11.25(n=4), the average BCF values of piperophos were 34.30(n=4) and 42.60(n=4). Depuration rate constants of IBP were 0.09( $h^{-1}$ ) and 0.08( $h^{-1}$ ), half-life of IBP were 7.70 and 8.66 at each tested concentration. The concentrations of IBP in zebrafish at low and high concentrations rapidly decreased after 12(0.243$\mu\textrm{g}$/g) and 12 hours(0.040$\mu\textrm{g}$/g). Depuration rate constants of methidathion were 0.40( $h^{-1}$ ), half-life of methidathion were 1.73 at one-hunderdth and of 96-hrs L $C_{50}$ , repectively. The concentrations of methidathion in zebrafish at high concentrations rapidly decreased after 6 hours(0.18 $\mu\textrm{g}$/g). Depuration rate constant of low concentration was no measured because methidathion in zebrafish was depurated in 6 hours. Depuration rate constants of piperophos sere 0.15( $h^{-1}$ ) and 0.44( $h^{-1}$ ), half-life of piperophos were 4.62 and 1.58 at each tested concentration. The concentrations of piperophos in zebrafish at los and high concentrations rapidly decreased after 12(0.26$\mu\textrm{g}$/g) and 6 hours(0.015 $\mu\textrm{g}$/g). It was suggested that high BCF of piperophos was due to high Kow(octanol-water partition coefficient). The possibility of bioconcenration was not likely to be high because of its $K_{DEP}$(depuration rate constant) in the evniroment. It was suggested that low BCF of methidathion showed lowest Kow as well as the most rapid $K_{DEP}$. Therefore, the possibility of bioconcentration was not occured in the enviroment. It was suggested that the BCF dtermined for IBP was lower than that of other pesticides due to high Sw(water solubility), show $K_{DEP}$. Therefore, IBP revealed little bioconcentration effect on in aquatic ecosystem.ystem.

A sunday system with a horizontal bin-type composter was constructed and operated to evaluate its composting performance for four days for each test in October, 1992. A sundry system is one of popular systems for composting livestock manure, of which main benefit is to utilize unlimited, clean, and free solar radiation. A rectangular concrete bin(composter) with dimension of 300cm(length) X90cm(width) X60cm(height) was bedded alternatively with four lanes of aeration pipes and heating pipes, and was insulated at three walls with 50mm styrofoam. Each aeration pipe of a diameter of 25mm had 4mm perforated holes at every 15cm longitudinally, and supplied air of about 2m$^3$/min to the composter to maintain aerobic condition . A stirrer rotating at 1 rpm made one round trip every 20 minutes on the conveying chain along the the length of the composter. Five tests (Test 1~Test 5) were implemented to evaluate the composting effectiveness of a sundry system with a horizontal bin-type composter. Treatments of two levels of the mixture ratio of swine manure and paper sludge cakes(manure : paper sludge cakes= 1 : 4 and 1 : 2) and two levels of the water content(W/C ; 70% and 50%) were made to test the significance of the physicochemical properties for decomposition of the mixture materials. Temperature, C/N ratio, water content, microbial activity of the composting materials were taken measurements to evaluate its performance with the lapse of composting time for tests. A small-scale sundry system with a bin-type composter did not appear to be an appropriate system for composting livestock manure. Since heat generation by the composting materials could not overcome heat loss due to areation in a small-scale composter, a proper thermal enviroment could not be maintained to propagate massively thermopilic microorganism relatively in a short period of time. Different from the result of Chol et al.(1992) 6), a temperature variation of the composting materials did not show the peak clearly and C/N ratio didn't lower with time as expected. Mesophilic microoragnism seemed to play an important role for decomposition of the mixture materials. A sundry system with a bin-type composter may be good for a large-scale livestock farm household which may produce enough animal manure. Therefore a decision should be made very carefully to choose a system for composting livestock waste.
