염화칼슘 제설제 고농도 처리에 따른 토양개량제와 참억새 식재 처리가 염류저감 및 생육개선에 미치는 영향 (Effects of Soil Amendments and Planting Miscanthus sinensis on Salt Reduction and Growth Improvement in Substrate irrigated with High Concentration of Calcium Chloride Deicing Salts)

  • Contamination of soil by deicing salt is among the important environment problems due to their toxicity and negative impact to human health and the environment. One of the effective methods for cleaning the soil from deicing salts is desalination using soil amendment-phytoremediation continuum treatment. The purpose of this study was to determine how much of the pH, EC control and Ca2+, Na+, Mg2+, and K+ taken up soil amendments and Miscanthus sinensis, and to evaluate the effect of salt reduction and growth improvement as affected by soil amendment in high concentration of calcium chloride (CaCl2) deicing salts. Results indicated that the addition of soil amendments was decrease the EC and pH, also significantly reduce the leaching of Ca2+, Na+, Mg2+, K+, a chloride ions related deicing salts, compared to the control for CaCl2 10 g/L treatment. It also resulted in an enhanced plant growth and higher plant height, leaf length, leaf width, number of leaves, fresh weight and dry weight in Hydroball treatment + Miscanthus sinensis planting continuum treatment compared to the treatment that planted Miscanthus sinensis only. Therefore, we concluded that soil amendments might be attributed to an accumulation of deicing slats in the roadside soil, resulting in the improvement of Miscanthus sinensis growth.

Isolation and Characterization of Pb-Solubilizing Bacteria and Their Effects on Pb Uptake by Brassica juncea: Implications for Microbe-Assisted Phytoremediation

  • The aim of this study was to isolate and characterize lead (Pb)-solubilizing bacteria from heavy metal-contaminated mine soils and to evaluate their inoculation effects on the growth and Pb absorption of Brassica juncea. The isolates were also evaluated for their plant growth-promoting characteristics as well as heavy metal and salt tolerance. A total of 171 Pb-tolerant isolates were identified, of which only 15 bacterial strains were able to produce clear haloes in solid medium containing PbO or $PbCO_3$, indicating Pb solubilization. All of these 15 strains were also able to dissolve the Pb minerals in a liquid medium, which was accompanied by significant decreases in pH values of the medium. Based on 16S rRNA gene sequence analysis, the Pb-solubilizing strains belonged to genera Bacillus, Paenibacillus, Brevibacterium, and Staphylococcus. A majority of the Pb-solubilizing strains were able to produce indole acetic acid and siderophores to different extents. Two of the Pb-solubilizing isolates were able to solubilize inorganic phosphate as well. Some of the strains displayed tolerance to different heavy metals and to salt stress and were able to grow in a wide pH range. Inoculation with two selected Pb-solubilizing and plant growth-promoting strains, (i.e., Brevibacterium frigoritolerans YSP40 and Bacillus paralicheniformis YSP151) and their consortium enhanced the growth and Pb uptake of B. juncea plants grown in a metal-contaminated soil. The bacterial strains isolated in this study are promising candidates to develop novel microbe-assisted phytoremediation strategies for metal-contaminated soils.

Mycorrhiza 처리가 Ardisia pusilla의 생육 및 내건성에 미치는 영향 (Effect of Mycorrhiza on Plant Growth and Drought Resistance in Ardisia pusilla)

  • 균근을 이용한 Ardisia pusilla의 내건성 증진효과를 알아보기 위하여 내생균근 Glomus spp.을 접종하여 생육상에서 30일 동안 재배한 후 수분스트레스처리를 실시하였다. 수분스트레스처리는 5일간 무관수 상태로 두었다가 관수하는 방법으로 60일 동안 반복 처리하였다. 식물생육은 균근처리시 대조구에 비해 증가하였으며, 관수시 스분스트레스의 회복력도 빨랐다. 식물체내 양분함량은 지하부의 Fe, Mn, 및 Cu의 함량의 경우 균근 접종 식물이 대조구에 비해 현저하 높았으나 이에 반하여 프롤린 함량은 대조구에서 균근 접종식불에 비해 높게 나타났다.

식물생장촉진근권세균 Bacillus subtilis YGB36을 이용한 고추 탄저병의 생물학적 방제 (Control of Red Pepper Anthracnose Using Bacillus subtilis YGB36, a Plant Growth Promoting Rhizobacterium)

  • 고추는 한국의 주요 경제 작물 중 하나이지만 재배과정에서 Colletotrichum acutatum에 기인한 탄저병이 많이 발생한다. 이의 방제를 위해 길항균인 Bacillus subtilis YGB36을 선발하여 16S rRNA 염기서열 및 생리·생화학적 분석을 통해 Bacillus subtilis로 동정하고 이를 생물학적 방제제로 개발하고자 실험을 진행하였다. In vitro screening에서 YGB36은 Cylindrocarpon destructans에 강한 항균활성을 나타내었고, cellulase, protease, amylase, siderophore 생산 및 phosphate solubility를 보유하고 있었다. 이 균주의 배양액(106 cfu/ml) 및 배양여액은 in vitro 실험에서 C. acutatum의 포자발아를 강하게 억제하였고, 고추 과실을 이용한 실내실험에서도 이 배양액이 탄저병 병반의 진전을 억제하였으며, 방제가는 65.7%로 나타났다. 아울러 YGB36배양액은 고추 종자의 발아 및 초기 뿌리 생장을 촉진하였고, 온실에서의 식물 생장을 촉진하였다. 기존 화학농약과의 혼용 가능성 조사 결과, YGB36은 실험에 사용한 모든 살충제에서 생존에 영향을 전혀 받지 않았으며, 살균제 21종 중 11종에서 잘 생존하였다. 따라서 YGB36균주는 고추 탄저병에 대한 생물농약으로서의 잠재적 가치가 상당한 것으로 판단된다.

애기장대 AtPGR 단백질의 병 저항성에 관한 생리적 특성 분석 (Physiological Characterization of an AtPGR from Arabidopsis Involved in Pathogen Resistance)

  • 식물체에서 당 조절 인자의 병 저항성 생리적 특성을 살펴보길 위하여, 원형질막에 존재하는 glucose 조절인자인 애기장대 AtPGR 유전자의 과발현 및 RNAi 형질전환체를 사용하였다[3]. AtPGR 유전자는 병원균 처리에 의하여 전사 발현양이 증가하였을 뿐만 아니라, JA와 SA 처리 시에도 AtPGR 전사 발현양이 증가함을 확인하였다. 과발현 형질전환체를 이용하여 병 저항성을 살펴본 결과, AtPGR 유전자는 병원균에 대해 저항성을 유도함을 알수 있었다. 또한 병원균 유도 증가 유전자로 알려진 PDF1.2 및 PR1 유전자 발현 양상을 qPCR을 통해 살펴본 결과, AtPGR 유전자는 PDF1.2 유전자를 SA 경로 하에서는 증가시키는 반면, JA 경로 하에서는 발현 증가량을 감소시키는 경향이 있음을 나타내었고, PR1 유전자의 발현은 JA 경로를 통해 조절할 것으로 생각되어진다. 이러한 결과를 바탕으로, AtPGR 유전자는 glucose 뿐만 아니라 병원균 반응에도 관련되어져 있음을 알 수 있다.

Botrytis cinerea hypovirulent strain △BcSpd1 induced Panax ginseng defense

  • Background: Gray mold, caused by Botrytis cinerea, is one of the major fungal diseases in agriculture. Biological methods are preferred over chemical fungicides to control gray mold since they are less toxic to the environment and could induce the resistance to pathogens in plants. In this work, we try to understand if ginseng defense to B. cinerea could be induced by fungal hypovirulent strain △BcSpd1. BcSpd1 encodes Zn(II)2Cys6 transcription factor which regulates fungal pathogenicity and we recently reported △BcSpd1 mutants reduced fungal virulence. Methods: We performed transcriptomic analysis of the host to investigate the induced defense response of ginseng treated by B. cinerea △BcSpd1. The metabolites in ginseng flavonoids pathway were determined by UPLC-ESI-MS/MS and the antifungal activates were then performed. Results: We found that △BcSpd1 enhanced the ginseng defense response when applied to healthy ginseng leaves and further changed the metabolism of flavonoids. Compared with untreated plants, the application of △BcSpd1 on ginseng leaves significantly increased the accumulation of p-coumaric acid and myricetin, which could inhibit the fungal growth. Conclusion: B. cinerea △BcSpd1 could effectively induce the medicinal plant defense and is referred to as the biological control agent in ginseng disease management.

Anti-Invasive and Anti-Angiogenic Effects of Xanthohumol and Its Synthetic Derivatives

  • Invasion and metastasis is the main cause of cancer mortality. Angiogenesis is a prerequisite for the tumor growth and metastasis. Matrix metalloproteinases (MMPs) are the key enzymes playing in the invasive growth and metastasis of cancer as well as angiogenesis. Xanthohumol, a prenylated chalcone of the Hop plant (Humulus lupulus L), has been reported to suppress cancer invasion and angiogenesis. In the present study, we investigated the antiinvasive effects of xanthohumol (1) and its synthetic derivatives, 4'-O-methylxanthohumol SEM ether (2), xanthohumol C (3), and xanthohumol C MOM ether (4) in relation to MMP expression in HT-1080 human fibrosarcoma cells. The compound 1 and its derivative, 3 and 4, significantly inhibited serum-induced HT-1080 cell invasion, and 12-O-tetradecanoylphorbol-13-acetate (TPA)-enhanced activity and expression level of MMP-2 and MMP-9 in a concentration-dependant manner. In addition, they inhibited TPA-enhanced expression of MT1-MMP with relatively weak inhibition in tissue inhibitor of metalloproteinase (TIMP)-1 and TIMP-2 level. The compound 1 significantly decreased the cell viability, whereas the derivatives, 2 and 3 showed no cytotoxicity, and compound 4 showed slight cytotoxicity in the cells. Furthermore, in a chick chorioallantoic membrane (CAM) assay, the derivatives 3 and 4 dose-dependently suppressed vascular endothelial growth factor (VEGF)-induced angiogenesis, which is similar to that of compound 1. Taken together, the results indicate that compounds 3 and 4 may be valuable anti-angiogenic agents in the treatment of chronic diseases such as cancer and inflammation working through suppression of MMP-2 and MMP-9.

Enhanced Carboxymethylcellulase Production by a Newly Isolated Marine Bacterium, Cellulophaga lytica LBH-14, Using Rice Bran

  • The aim of this work was to establish the optimal conditions for production of carboxymethylcellulase (CMCase) by a newly isolated marine bacterium using response surface methodology (RSM). A microorganism producing CMCase, isolated from seawater, was identified as Cellulophaga lytica based 16S rDNA sequencing and the neighborjoining method. The optimal conditions of rice bran, ammonium chloride, and initial pH of the medium for cell growth were 100.0 g/l, 5.00 g/l, and 7.0, respectively, whereas those for production of CMCase were 79.9 g/l, 8.52 g/l, and 6.1. The optimal concentrations of $K_2HPO_4$, NaCl, $MgSO_4{\cdot}7H_2O$, and $(NH_4)_2SO_4$ for cell growth were 6.25, 0.62, 0.28, and 0.42 g/l, respectively, whereas those for production of CMCase were 3.72, 0.54, 0.70, and 0.34 g/l. The optimal temperature for cell growth and the CMCase production by C. lytica LBH-14 were $35^{\circ}C$ and $25^{\circ}C$, respectively. The maximal production of CMCase under optimized condition for 3 days was 110.8 U/ml, which was 5.3 times higher than that before optimization. In this study, rice bran and ammonium chloride were developed as carbon and nitrogen sources for the production of CMCase by C. lytica LBH-14. The time for production of CMCase by a newly isolated marine bacterium with submerged fermentations reduced to 3 days, which resulted in enhanced productivity of CMCase and a decrease in its production cost.

Single and mixed chelants-assisted phytoextraction of heavy metals in municipal waste dump soil by castor

  • The phytoextraction of some toxic heavy metals from municipal waste dump soil by castor plant (Ricinus communis) was tested under natural and single or mixed chelant-assisted scenarios in pot microcosms. A sandy loam with total metal contents (mg/kg): Cd (84.5), Cu (114.5), Ni (70.3), Pb (57.8), and Zn (117.5), was sampled from an active dumpsite in Calabar, Nigeria and used for the study. Castor (small seed variety) was grown under natural phytoextraction or single/binary chelant (citric acid, oxalic acid, and EDTA) applications (5-20 mmol/kg soil) for 63 days. Castor exhibited no visual phytotoxic symptoms with typically sigmoid growth profiles at the applied chelant doses. Growth rates, however, decelerated with increase in chelant dose. Post-harvest biomass yields were higher under chelant application than for natural phytoextraction. Both root and shoot metal concentrations (mg/kg) increased quasilinearly and significantly ($p{\leq}0.05$) with increase in chelant dose, furnishing maximum levels as: Cd (55.6 and 20.9), Cu (89.5 and 58.4), Ni (49.8 and 19.6), Pb (32.1 and 12.1), and Zn (99.5 and 46.6). Ranges of translocation factors, root and shoot bioaccumulation factors were 0.21-3.49, 0.01-0.89 and 0.01-0.51, respectively. Overall, the binary chelant treatments were less toxic for R. communis growth and enhanced metal accumulation in shoots to a greater extent than the single chelant scenarios, but more so when EDTA was present in the binary combination. This suggests that the mixed chelants could be considered as alternative treatments for enhanced phytoextraction and revegetation of degraded waste dump soils.

Micropropagation and Mass Production of Adventitious Roots of Polygonatum odoratum via the Culture of Seedling Explnnts

  • Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.
