Investigation of the Geoelectrical Response at the Hydrocarbon-impacted Zone (유류 오염대의 전기적 물성 특성 연구)

  • A physical model experiment with GPR and 3-D resisitivity survey were conducted to investigate the geoelectrical response of hydrocarbon-impacted zone, so called smeared zone, on the geophysical data. The results from the experiment show that GPR signals were enhanced when LNAPL was present as a residual saturation in the water saturated system (${\varepsilon}_r$ = 21) due to less attenuation of the electromagnetic energy through the medium, compared to when the medium was saturated with only water (${\varepsilon}_r$ = 21). 3-D resistivity data obtained from the former gas station site demonstrate that the highly contaminated zones could be imaged with low resistivities attributed to the biodegradation of petroleum hydrocarbons at the aged, hydrocarbon-impacted sites. The study results also show that the geophysical methods, as a non-invasive sounding technique, can be a very useful tool for mapping hydrocarbon-contaminated zones.

Substrate Utilization Patterns During BTEX Biodegradation by an o-Xylene-Degrading Bacterium Ralstonia sp. PHS1

  • The biodegradation of BTEX components (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene) individually and in mixtures was investigated using the o-xylene-degrading thermo-tolerant bacterium Ralsronia sp. strain PHS1 , which utilizes benzene, toluene, ethylbenzene, or o-xylene as its sole carbon source. The results showed that as a single substrate for growth, benzene was superior to both toluene and ethylbenzene. While growth inhibition was severe at higher o-xylene concentrations, no inhibition was observed (up to 100 mg $l^-1$) with ethylbenzene. In mixtures of BTEX compounds, the PHS1 culture was shown to degrade all six BTEX components and the degradation rates were in the order of benzene, toluene, o-xylene, ethylbenzene, and m- and p-xylene. m-Xylene and p-xylene were found to be co-metabolized by this microorganism in the presence of the growth-supporting BTEX compounds. In binary mixtures containing the growth substrates (benzene, toluene, ethylbenzene. and o-xylene), PHS1 degraded each BTEX compound faster when it was alone than when it was a component of a BTEX mixture, although the degree of inhibition varied according to the substrates in the mixtures. p-Xylene was shown to be the most potent inhibitor of BTEX biodegradation in binary mixtures. On the other hand, the degradation rates of the non-growth substrates (m-xylene and p-xylene) were significantly enhanced by the addition of growth substrates. The substrate utilization patterns between PHS1 and other microorganisms were also examined.

  • Previous experiment was performed under abiotic condition. Steady-state abiotic experiments in the sand-tank reactor with air flowing through the reactor headspace demonstrated that oxygen supply through the water table interface into the saturated zone was enhanced when an LNAPL (dodecane) pool was present at the water table. Biotic condition was considered in this study. Biotic experiments performed after inoculating the reactor with Pseudomonas putida mt-2, which does not grow on dodecane, indicated that the enhanced oxygen supply in the presence of the LNAPL pool also enhanced biodegradation of a solute (glucose) plume passing beneath the LNAPL pool at steady-state.

  • Oxygen uptake characteristics of soil microcosm added by hydrocarbon degrading bacteria screened from polluted site in Korea was studied. The degradation of TPH was enhanced by additon of nononionic surfactants. The amount of oxygen consumed was decreased at higher concentration. The degradation rate of hydrocarbon was decreased by increasing the hydrcarbon concentration.

Effects of Slow Release Fertilizer and Dispersant on Biodegradation of Oil Contaminated in Sand Seashore Mesocosms (지속성 영양염제와 유분산제가 해변모래에 오염된 유류의 생분해에 미치는 영향)

  • To evaluate the effects of slow release fertilizer and chemical dispersant on oil biodegradation, mesocosm studies were conducted on sand seashore. The rapid removal rates (85%) of aliphatic hydrocarbons and the simultaneous decreases of n-$C_{17}$/pristane (69%) and $n-C_{18}/phytane$ (61%) ratios by the addition of slow-release fertilizer (SRF) within 37 days of experiment indicated that SRF could enhance the oil degrading activity of indigenous microorganisms in sand mesocosm. Although the growth of heterotrophic bacteria and petroleumdegrading bacteria in the mesocosm treated with $Corexit 9527^{R}$ was stimulated, the biological oil removal based on the ratios of $Corexit 9527^{R}$ and $n-C_{18}/phytane$ was inhibited. Removal rates of aliphatic hydrocarbons (56%), and n-$C_{17}$/pristane (27%) and $n-C_{18}/phytane$ (17%) ratios by the addition of chemical dispersant $Corexit 9527^{R}$ were similar or lower than those values of control (50, 60, 46%), respectively. The biodegradation activity, however, when simultaneously treated with SRF and $Corexit 9527^{R}$, was not highly inhibited and even recovered after the elimination of chemical dispersant. From these results it could be concluded that the addition of SRF enhanced the oil removal rate in oligotrophic sand seashore and chemical dispersant possibly inhibit the oil biodegradation. Hence, in order to prevent the unrestrained usage of chemical dispersant in natural environments contaminated with oil, the National Contingency Plan of Oil Spill Response should be carefully revised in consideration of the application for bioremedaition techniques.

  • Rusted iron could retain activity to redox-sensitive pollutants in batch reactor. Flow-through columns packed with permeable reactive iron filings (Fe$^{0}$ ) between soil and sand layers were used to evaluate the applicability of bio-enhanced iron barriers to treat explosives-contaminated groundwater. One column was bioaugmented with municipal anaerobic sludge to evaluate the enhancement of biodegradation. Military contaminants (RDX, HMX, TNT, 2,4DNT, 2,6DNT), which coexist in soils at military sites, were completely removed in the bioaugmented Fe$^{0}$ layer after 8 months of operation. Overall, this research suggests that Fe$^{0}$ barriers can effectively clean up groundwater contaminated with military explosives, and that treatment efficiency can be enhanced by bioaugmentation.

Adsorption and Degradation of Herbicide Napropamide in soils (토양중 제초제 Napropamide의 흡착 및 분해 특성)

  • This study was carried out to investigate the adsorption and the degradation of herbicide napropamide in soils tinder laboratory conditions. The adsorption distribution coefficients(Kd) of napropamide in 3 soils varied from 5.38 to 11.39. With higher content of organic matter in soils, the extent of adsorption was high. The rapid degradation of napropamide took place after a lag period. The time to 50% loss of napropamide was approximately 65, 45 and 30 days in soil incubated at 10, 20 and $30^{\circ}C$, respectively. The time to the 90% loss varied from about 90 days at $10^{\circ}C$ to about 45 days at $30^{\circ}C$. In the soil pre-incubated with napropamide, the herbicide degraded without a lag time, and the rate of degradation was also accelerated. This should be a evidence on enhanced biodegradation of napropamide in soil with the repeat-application.

Release of Organic Matter and Behavior of Nitrogen in the Degradation of Sewage Sludge Using Ultrasound (초음파를 이용한 하수 슬러지 분해에서 유기물 방출과 질소 거동)

  • The sewage sludge with concentrated MLSS, ranging from 5967 to 8400mg/L was degraded by ultrasound. In this study, ultrasound treatment was used to investigate the behavior characterization of SBOD, sludge biodegradation, C/N ratio, TN, turbidity and sludge morphology. From the experimental results of C/N ratio change and TN removal, the optimal irradiation time was found to be 10 minutes. The results showed the relative index of sludge biodegradation(SBOD/TCOD) was enhanced to 0.76 from the initial value of 0.013 at the 5967mg/L MLSS, during the 60minutes treatment. Throughout this research, the results provide useful engineering reference data for reuse of sewage sludge using ultrasound.

Isolation of a Poly (L-lactide) Degrading Bacterium and Improvement of its Degradation Capacity (Poly(L-lactide)분해 세균의 분리 및 활성 증진)

  • A thermophilic bacterium capable of poly (L-lactide)(PLLA) degradation was isolated from cultivating soil in Korea. The isolate was Gram positive rod-shaped bacterium, and was identified as Geobacillus caldoxylosilyticus based on the 16S rDNA sequence analysis. The strain proved to be a new PLLA degrading bacterium which has not been reported in the open literatures yet. The degradation activity of the strain was assessed in a sterilized compost inoculated with the strain under controlled compost condition at $58^{\circ}C$ for 40 days. The strain mineralized 66%, 57%, 41% and 40% of PLLA5000, PLLA11000, PLLA34000 and PLLA256000 whose weight average molecular weights were 5000, 11000, 34000 and 256000, respectively. Incorporation of 0.1% each of gelatin, yeast extract and ammonium sulfate in the compost containing PLLA256000 as a nutritional supplement raised the biodegradation activity by 27%, 13% and 10%, respectively. Increase of the inoculum size from $10^9cfu\;g^{-1}\;to\;10^{10}cfu\;g^{-1}\;and\;10^{11}cfu\;g^{-1}$ also enhanced the biodegradation activity by 14% and 20%, respectively.
