Tencel Dyeing by Natural Indigo Prepared from Dyer's Knotweed (천연인디고를 이용한 텐셀직물의 염색)

  • Natural indigo dye in powder form was prepared by modifying traditional Niram method, using $Ca(OH)_2$ instead of baked oyster powder for precipitating indigo dye. The prepared dye was applied to dyeing Tencel fabrics to investigate the effect of experimental conditions for the optimization of dyeing process. The indigo dye powder contained 15.2%(w/w) of indigo content and 0.757%(w/w) of indirubin content on the basis of HPLC analysis. Maximum dye uptake was obtained at $60^{\circ}C$ for 20min. Almost saturated dye uptake was obtained at 2g/L of sodium hydrosulfite concentration up to 4g/L of indigo dye and then slowly increased for further increase of sodium hydrosulfite. Whereas at higher indigo dye concentration(8g/L) more than 3g/L of reducing agent concentration was required for obtaining the maximum dye uptake. At the same indigo dye and reducing agent concentration, K/S value of the sample dyed without sodium hydroxide(pH 5.75) was 15.19, much higher than one dyed in alkaline condition(K/S 5.76). There was no difference in colorfastness ratings among samples with different color strength. However, more fading was occurred for the sample with low color strength.

A Fast Inter-layer Mode Decision Method inScalable Video Coding (공간적 스케일러블 비디오 부호화에서 계층간 모드 고속 결정 방법)

  • We propose a fast inter-layer mode decision method by utilizing coding information of base layer upward its enhancement layer inscalable video coding (SVC), also called MPEG-4 part 10 Advanced Video Coding Amendment 3 or H.264 Scalable Extension (SE) which is being standardized. In this paper, when the motion vectors from the base layer have zero motion (0, 0) in inter-layer motion prediction or the Integer Transform coefficients of the residual between current MB and the motion compensated MB by the predicted motion vectors from the base layer are all zero, the block mode of the corresponding block to be encoded at the enhancement layer is determined to be the $16{\times}16$ mode. In addition, if the predicted mode of the MB to be encoded at the enhancement layer is not equal to the $16{\times}16$ mode, then the rate-distortion optimization is only performed on the reduced candidated modes which are same or smaller partitioned modes. Our proposed method exhibits the complexity reduction in encoding time up to 72%. Nevertheless, it shows negligible PSNR degradation and bit rate increase up to 0.25dB and 1.73%, respectively.

A Study on Evaluation of Water Supply Capacity with Coordinated Weirs and Multi-reservoir Operating Model (댐-보 최적 연계운영을 통한 용수공급능력 평가에 관한 연구)

  • When we evaluate the water supply capacity of a river basin, it is a common practice to gradually increase the water demand and check if the water demands are met. This practice is not only used in the simulation approach, but also in the optimization approach. However, this trial and error approach is a tedious task. Hence, we propose a two-phase method. In the first phase, by assuming that the decision maker has complete information on inflow data, we use a goal programming model that can generate the maximum water supply capacity at one time. In the second phase, we simulate the real-time operation for the critical period by utilizing the water supply capacity given by the goal programming model under the condition that there is no foresight of inflow. We applied the two-phase method to the Geum-River basin, where multi-purpose weirs were newly constructed. By comparing the results of the goal programming model with those of the real-time simulation model we could comprehend and estimate the effect of perfect inflow data on the water supply capacity.

Optimization of Fermentation Conditions for CoQ10 Production Using Selected Bacterial Strains (CoQ10 생성 세균의 선별 및 발효조건 최적화)

  • Coenzyme Q10 (CoQ10) is an essential lipid-soluble component of membrane-bound electron transport chains. CoQ10 is involved in several aspects of cellular metabolism and is increasingly being used in therapeutic applications for several diseases. Despite the recent accomplishments in metabolic engineering of Escherichia coli for CoQ10 production, the production levels are not yet competitive with those by fermentation or isolation. So we tested several microorganisms obtained from the KCTC of Biological Resource Center to find novel sources of strain-development for CoQ10-production. Then we selected two strains, Paracoccus denitrificans (KCTC 2530) and Asaia siamensis (KCTC 12914), and tested to optimize the CoQ10 production conditions. Among the carbon sources tested, CoQ10 production was the highest when fructose was supplied about 4% concentration. Yeast extract produced the highest CoQ10 production about 2% concentration. The highest CoQ10 production was obtained at pH 6.0 for P. denitrificans and pH 8.0 for A. siamensis. And two strains showed the highest CoQ10 production at $30^{\circ}C$, but the highest DCW was obtained at $37^{\circ}C$. In the fed-batch culture, P. denitrificans yielded $14.34{\pm}0.473$ mg and A. siamensis yielded $12.53{\pm}0.231$ mg of final CoQ10 production.

A Development of a Seismic Vulnerability Model and Spatial Analysis for Buildings (건물에 대한 지진취약도 모델링 및 공간 분석)

  • The purpose of this study is to suggest a method of predicting seismic vulnerability and safety conditions of each building in a targeted area. The scope of this study includes 'developing a simulation model for precaution activities,' 'testing the validity of the developed model', From the facility point of view, target of this study is a local building system. According to the literature review, the number of earthquake prediction modeling and cases with GIS applied is extremely few and the results are not proficient. This study is conducted as a way to improve the previous researches. Statistic analyses are conducted using 348 domestic and international data. Finally, as a result of the series of statistical analyses, an adequate model is developed using optimization scale method. The ratio of correct expectation is estimated as 87%. In order to apply the developed model to predict the vulnerability of the several chosen local building systems, spatial analysis technique is applied. Gangnam-gu and Jongro-gu are selected as the target areas to represent the characteristics of the old and the new downtown in Seoul. As a result of the analysis, it is discovered that buildings in Gangnam-gu are relatively more dangerous comparing to those of Jongro-gu and Eunpyeong-gu.

Optimization of Wool Dyeing with Yellow Dye from Carthamus Tinctorius L. (홍화 황색소를 이용한 모염색의 최적화)

  • This study investigated the adsorption of safflower yellow dye on wool protein fiber and the optimum dyeing conditions to test color reproducibility. In addition, the effects of mordants on dye adsorption, color, fastness, and photofading rate were also studied. The prepared dye in powder form was characterized with UV-vis spectroscopy and FT-IR spectrometric analysis. The color of dyed fabrics was characterized by CIE $L^*a^*b^*$ coordinates, H V/C, and K/S values. The color reproducibility of the dyed wool fabrics was examined. The amount of dye adsorption increased and also, the shade of the dyed wool fabrics became deeper and more saturated with increasing temperature, time, and dye concentration. The maximum color strength was obtained at pH 3.0. The shade of dyed wool fabrics ranged from light yellow to dark mustard yellow as the pH of the dye bath shifted from alkaline to acidic. Color reproducibility was reliable with color differences in the range of 0.53~1.75. Fastness to dry cleaning was relatively good at 4/5 rating irrespective of mordanting. Fe and Cu mordants showed the least color change of the dyed wool fabrics after exposure to light. Mordants did not contribute to improve dye uptake and color fastness, although they made variations in color tone. Safflower yellow dye can be used satisfactorily without mordants and will not cause damage to the environment.

Isolation and Characterization of Various Strains of Bacillus sp. having Antagonistic Effect Against Phytopathogenic Fungi (식물 병원성 곰팡이에 길항작용을 갖는 다양한 Bacillus sp.의 균주 분리와 특성에 관한 연구)

  • This study was carried out to examine the antagonistic effect against phytopathogenic fungi of isolated strains from soil samples collected from Busan, Changwon, and Jeju Island: Botrytis cinerea, Colletotrichum acutatum, Corynespora cassiicola, Fusarium sp., Rhizoctonia solani, Phytophthora capsici, and Sclerotinia sclerotiorum. According to results of our studies, isolated strains showed an antagonistic effect against phytopathogenic fungi. Such an antagonistic effect against phytopathogenic fungi is seen due to the production of siderophores, antibiotic substances, and extracellular amylase, cellulase, protease, and xylanase enzyme activities. Extracellular enzymes produced by isolated strains were significant, given that they inhibited the growth of phytopathogenic fungi by causing bacteriolysis of the cell wall of plant pathogenic fungi. This is essential to break down the cell wall of plant pathogenic fungi and thus help plant growth by converting macromolecules, which cannot be used by the plant for growth, into small molecules. In addition, they are putative candidates as biological agents to promote plant growth and inhibit growth of phytopathogenic fungi through nitrogen fixation, indole-3-acetic acid production, siderophore production, and extracellular enzyme activity. Therefore, this study suggests the possibility of using Bacillus subtilis ANGa5, Bacillus aerius ANGa25, and Bacillus methylotrophicus ANGa27 as new biological agents, and it is considered that further studies are necessary to prove their effect as novel biological agents by standardization of formulation and optimization of selected effective microorganisms, determination of their preservation period, and crop cultivation tests.

Evaluation and Application of QUAL2E and QUAL2K Models in Anyang Stream (안양천에서 QUAL2E와 QUAL2K 모델의 적용 및 평가)

  • QUAL2K enhanced QUAL2E and applied in real fields efficiently incorporates denitrification process, sediment-water interaction process, bottom algae and detritus. Also, the CBOD of QUAL2K is divided into two real parts, one is slow CBOD(sCBOD) and another is fast CBOD(fCBOD). The simulation results of QUAL2E and QUAL2K models in Anyang Stream were compared and analyzed in water quality constituents of DO, BOD, Org-N, NH$_3$-N, NO$_3$-N, Org-P, Dis-P and Chl-a respectively. The similar results were shown in Org-N, NH$_3$-N, Org-P and Chl-a both QUAL2K and QUAL2E models. But the different results were revealed in DO, BOD, Dis-P and NO$_3$-N by the influence of new incorporating processes. DO was shown relatively low values in the effect of bottom algae. BOD which is influenced by particulate organic matter was revealed high values. NO$_3$-N was closed to the real values by the two processes of denitrification and sediment-water interaction. To evaluate the running results of QUAL2K and QUAL2E models, a simple statistical analysis was conducted. According to the statistical analysis, QUAL2K represented less relative error and coefficient of variation than QUAL2E in almost all of constituents. It was found that QUAL2K, which simulates the water quality more realistically, can be applied to control and manage the water problems of river or river-run reservoir effectively.

Isolation of Phytase Producing Pseudomonas fragi and Optimization of its Phytase Production (Acid Phytase를 생산하는 Pseudomonas fragi의 분리와 phytase의 생산조건)

  • A bacterial strain producing a high level of an extracellular phytase was isolated from livestock waste water, identified as a strain of Pseudomonas fragi and designated as Pseudomonas fragi Y9451. Under the phytase production medium, the activity of phytase reached the highest level after 120 hours of incubation. On the effect of carbon sources on the phytase production, the most favorable carbon source for phytase production was fructose. As for the effect of nitrogen sources, high levels of phytase activity were detected in the medium containing nutrient broth as the nitrogen source. Free $PO_4^{3-}$ inhibited phytase production with increasing concentration of $KE_2PO_4$ and phytate in the media. The addition of $CaCl_2$ and $MgSO_4$ also resulted in the inhibition of phytase production. To investigate the effect of aeration on the phytase production, different volumes of culture broth in Erlenmeyer flasks were incubated in rotary shaker at the speed of 200 rpm. As a result, a high level of phytase activity was detected at small volume of culture broth as compared to larger volume because of its more aerobic condition.

  • A two-loop Rankine cycle for engine waste heat recovery of gasoline vehicle has been investigated. Water-steam cycle as a high-temperature (HT) loop for exhaust gas heat recovery and R-134a cycle as a low-temperature (LT) loop for both heat recovery of the engine coolant and the residual heat from the HT loop were considered. Energy and exergy analysis was performed to investigate the performance of the system. Because two volumetric expanders are used for the HT and LT loop, the sizes of two expanders are very important for the optimization of the system. The effects of pressure ratio of the HT loop, considering the size of the HT expander, and the condensation temperature of LT loop on the performance of the system at a target engine condition were investigated. This study shows that about 20% of additional power from the engine waste heat recovery can be obtained at the target engine condition.
