Analysis of Endospore-forming Bacteria or Nitrogen-fixing Bacteria Community Isolated from Plants Rhizosphere in Dokdo Island (독도서식 식물근권에서 분리한 포자형성세균과 질소고정세균의 군집구조 분석)

  • Bacteria were isolated from roots of plants belonging to family Solanaceae and Gramineae, inhabited in Dokdo island. Fifty six endospore-forming bacteria grown on tryptic soy broth (TSB) agar medium and 23 nitrogen-fixing bacteria (NFB) grown on nitrogen free agar medium were isolated, respectively. The isolates were partially identified by analyzing the 16S rDNA and categorized into phylogenetic groups. The 16S rDNA sequences of each identified isolates were compared with sequences of each type strains to analyze phylogenetic relationship by phylogenetic tree. As a result, endospore-forming bacteria and nitrogen-fixing bacteria were classified into 4 and 6 lineage groups, respectively. Among these isolated, 18 were presumed to be novel species candidates based on the similarity (lower than 98%) analysis of the l6S rDNA sequences.

Attachment of Pasteuria penetrans Endospores to Meloidogyne spp. Juveniles Affected by Temperatures and the Nematode species (선충기생세균(Pasteuria penetrans) 내생포자의 뿌리혹선충(Meloidogyne spp.) 유충 부착에 대한 온도와 선충종의 영향)

  • A greenhouse soil infested with an obligate nematode parasitic bacterium, Pasteuria penetrans, was used to test the effect of temperatures on the endospore attachment to root-knot nematode, Meloidogyne arenaria, juveniles (J2). Freshly hatched J2s were inoculated to the soil in petri dish and incubated under different temperatures of $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$ for 7 days. The endospore attachment rates were 100% in all the temperatures, while the number of endospores attached per J2 was highest in $25^{\circ}C$ with 28.3 endospores/J2 followed by 20.2, 18.6, and 13.6 in $30^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$, respectively. When the soil was pre-treated under different temperatures before the J2 inoculation, the endospore attachment rates significantly decreased from 60% in room temperature to $25^{\circ}C$, 31.7, 8.3, 5.0, and 0% after the soil incubation in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$ for 10 days, respectively. The endospore numbers attached per J2 were 3.5, 4.3, 1, 1, and 0 when the soil was pre-treated in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$, respectively, which were lower than 5.3/J2 of room temperature treated soil. The P. penetrans isolate in the soil showed nematode species-specific endospore attachment characteristics with 100% attachment rate only on M. arenaria J2s while the rates were 0% on M. hapla and M. incognita J2s.

Dominance of Endospore-forming Bacteria on a Rotating Activated Bacillus Contactor Biofilm for Advanced Wastewater Treatment

  • The bacterial diversity inherent to the biofilm community structure of a modified rotating biological contactor wastewater treatment process, referred to as the Rotating Activated Bacillus Contactor (RABC) process, was characterized in this study, via both culture-dependent and culture-independent methods. On the basis of culture-dependent methods, Bacillus sp. were found to exist in large numbers on the biofilm (6.5% of the heterotrophic bacteria) and the microbial composition of the biofilms was quite simple. Only three phyla were identified-namely, the Proteobacteria, the Actinobacteria (High G+C Gram-positive bacteria), and the Firmicutes (Low G+C Gram-positive bacteria). The culture-independent partial 16S rDNA sequence analysis revealed a considerably more diverse microbial composition within the biofilms. A total of eight phyla were recovered in this case, three of which were major groups: the Firmicutes (43.9%), the Proteobacteria (28.6%), and the Bacteroidetes (17.6%). The remaining five phyla were minor groups: the Planctomycetes (4.4%), the Chlorobi (2.2%), the Actinobacteria (1.1%), the Nitrospirae (1.1%), and the Verrucomicrobia (1.1%). The two most abundant genera detected were the endospore-forming bacteria (31.8%), Clostridium and Bacillus, both of which are members of the Firmicutes phylum. This finding indicates that these endospore-forming bacteria successfully colonized and dominated the RABC process biofilms. Many of the colonies or clones recovered from the biofilms evidenced significantly high homology in the 16S rDNA sequences of bacteria stored in databases associated with advanced wastewater treatment capabilities, including nitrification and denitrification, phosphorus accumulation, the removal of volatile odors, and the removal of chlorohydrocarbons or heavy metals. The microbial community structures observed in the biofilms were found to correlate nicely with the enhanced performance of advanced wastewater treatment protocols.

Isolation and Characterization of Ultra-Violet and Gamma-radiation Resistant Bacteria from Natural Habitats (자연 생태계로 부터 자외선 및 방사선 내성 박테리아의 분리 및 특성 연구)

  • Attempts to isolate the naturally occurring ultra-violet resistant bacteria from environmental sources were made. The isolates, designated No.29, 100, and 107, among numbers of bacterial isolates revealed a remarkable resistance to UV ray, whose degree of resistance in dose/response kinetics was comparable to that of an endospore-former, Bacillus subtilis. In a range of 100-300 $Jm^{-2}$/min of UV irradiation, the isolates exhibited 500-1000 fold resistance compated with E. coli. The isolated appeared to possiss cell-bound pigment of organge or crimson-red. The isolate 29 is spherical in pairs or tetrads, whereas the isolates 100 and 107 are rod. All are Gram-gositive bacteria and seemed to be non-endospore-bearer. A number of biochemical studies pursued on the isolates suggested that they are quite different to each other. Electron microscopic examination and the physiological characters of the isolate 29 suggested that this UV resistant spherical bacterium might be one species of Deinococcus, probably Deinococus radiophilus. Since there is no documents on UV resistant, Gram-positive, non-sporeformer bacillus so far, the isolates 100 and 107 might be turned out as new kinds of UV resistant bacteria occurring in nature by further investigation.

Effects of Quicklime Treatment on Survival of Bacteria and Structure of Bacterial Community in Soil (생석회 처리가 토양 세균의 생존과 군집구조에 미치는 영향)

  • When quicklime is added into soil for various purposes, abrupt changes in soil chemistry may affect essential ecological functions played by indigenous bacterial communities in soil. The magnitude of influence was estimated by observing changes in abundance and diversity of soil bacteria after quicklime treatment. When several soil samples were treated up to 20% (w/w) quicklime, plate count of viable cells ranged $10^2{\sim}10^3$ CFU $g^{-1}$, showing a reduction of more than $10^4$ times from viable counts of the untreated sample. Diversity of the bacterial isolates that survived after quicklime treatment was analyzed by conducting $GTG_5$ rep-PCR fingerprinting. There were only two types of fingerprints common to both 5% and 20% quicklime samples, implying that bacteria surviving at different strength of quicklime treatment differed depending on their tolerance to quicklime-treated condition. Isolates surviving the quicklime treatments were further characterized by Gram staining and endospore staining. All isolates were found to be Gram positive bacteria, and 85.4% of them displayed endospores state. In conclusion, most bacteria surviving quicklime treatment appear to be endospores. This finding suggests that most of ecological functions of bacteria in soil are lost with quicklime treatment.

The Preventive Effect of Bacillus polyfermenticus KJS-2 and Angelica gigas Nakai Extract on Triton WR-1339-induced Hyperlipidemia (Bacillus polyfermenticus KJS-2와 참당귀 추출물의 triton WR-1339 유발 고지혈증에 대한 예방효과)

  • The purpose of this study was to investigate the effects of Bacillus polyfermenticus KJS-2 (B. polyfermenticus KJS-2) and Angelica gigas Nakai extracts (AGNE) on hyperlipidermia. The purity of the major decursin and decursinol angelate (D/DA) in the AGNE were analyzed at 78%. Increased concentrations of AGNE (0.1-20 mg/ml) showed a higher 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase inhibition activity. Endospore-forming B. polyfermenticus KJS-2 ($1{\times}10^9CFU/ml$) exhibited good bile tolerance (0.3 mm) on an agar plate. An animal study was carried out using different groups, including a normal control, positive control (atorvastatin), negative control (triton WR-1339), AGNE group, B. polyfermenticus KJS-2 group, AGNE + B. polyfermenticus KJS-2 group, and Atorvastatin + AGNE + B. polyfermenticus KJS-2 group to determine the effect of hyperlipidemia. There were no significant changes in body weight, kidney weight, or liver weight except for the liver weight of the triton WR-1339-treated group. Groups with AGNE and B. polyfermenticus KJS-2 had increased HDL-cholesterol and decreased total cholesterol and triglycerides. The liver histopathological results also showed that all AGNE and B. polyfermenticus KJS-2-treated groups contained lower fat accumulation in the liver tissues. The findings of this study verified that AGNE and Endospore-forming B. polyfermenticus KJS-2 combination materials have a hyperlipidemic effect.

The Bacterial Community Structure in Biofilms of the RABC Process for Swine Butchery Wastewater Treatment (돼지 도축폐수 처리를 위한 RABC 공정의 생물막 세균군집 구조)

  • Culture-independent microscopic observations and 16S rDNA analyses were applied to describe the bacterial community inherent to the biofilm structure of the RABC (Rotating Activated Bacillus Contactors) process for swine butchery wastewater treatment. The ratios of Gram-positive bacterial counts to total bacterial counts of the RABC process were significantly increased in the last aeration tank as well as returned sludge, while those of the existing A2O (Anaerobic-Anoxic-Oxic) process maintained constant from aeration tanks to returned sludge. Totally nine phyla were recovered by 16S rDNA analysis, two of which were major groups: the Proteobacteria (64.1%) and the Actinobacteria (18.4%). The third major group was the endospore-forming Firmicutes (5.4%). The remaining six minor groups are the Bacteroidetes (3.3%), the Chlorobi (2.2%), the Nitrospirae (1.1%), the Chlorofleix (1.1%), the Acidobacteria (1.1%), and the Fusobacteria (1.1%). The ratio of endospore-forming bacteria was 19.4%, which was composed of the members of the Firmicutes phylum (5.4%) and the Intrasporangiaceae family (14.0%) of the Actinobacteria phylum. Nitrifying and denitrifying related- and phosphorus accumulating related-sequences were composed of 6.5% and 5.4% of total community, respectively, these could mean the high capacity of the RABC process to remove odor compounds and reduce eutrophication by efficient removing inorganic nutrients.

Mass Cultivation and Characterization of Multifunctional Bacillus velezensis GH1-13 (복합기능성 Bacillus velezensis GH1-13 균주의 대량배양 최적화 및 특성)

  • Bacillus genus are found abundantly in various sites and their secondary metabolites were used as potential agents in agriculture, notably plant growth promoting and bio-control. The objective of this study was to develop the culture conditions of GH1-13 strain including higher cell growth, stable endospore-forming and enhancement of potential agents which are related with plant growth promoting and phytopathogen suppression. The optimal carbon and nitrogen sources were determined by glucose and soy bean flour, respectively, then resulted in $7.5{\times}10^9cells/mL$, $6.8{\times}10^9\;endospore\;cells/mL$ and sporulation yield of 90% after 30 h cultivation in 500 L submerged fermenter at $37^{\circ}C$, pH 7.0. Cells and cell-free supernatant of GH1-13 strains showed the potent antifungal activity against phytopathogenic fungi of Colletotrichum gloeosporioides. It was also confirmed that indole-3-acetic acid (IAA) production of GH1-13 strain was greatly increased by addition of 0.3% tryptophan.

  • A new rod-shaped endospore-forming bacterium isolated from the hindgut flora of the termite, Reticulitermes speratus kyushuenesis Morimoto is described. The isolate stained Gram positive, but the KOH test and the test for L-alanine aminopeptidase were negative. The length of a single cell varies from 2.5-9.0 $\mu $m, and the cell is about 0.5-0.7$\mu $m thick. The isolate had a high cellulolytic ability and was identified as Bacillus amyloliquefaciens.

  • The family Bacillaceae에 속하는 모든 미생물은 endospore를 형성하는 것이 그 특성이며 포자형성은 일련의 생화학적 반응은 물론 형태ㆍ구조적 변화를 수반하는 질서 정연한 원시적인 생물분화의 일종이다. 따라서 포자형성 발아에 관한 연구가 많이 이루어졌고 또 현재도 활발히 진행되고 있다. 산업미생물학도로서 흥미있는 점은 산업적으로 유용한 몇가지 생화학물질이 포자형성 과정과 특별히 관련되어 생산된다는 점이며 이중 몇 가지 효소와 항생물질 그리고 독소에 관하여 간략히 고찰해 보고자 한다.
