Analysis of Benzo[a]pyrene and Bisphenol A in lakes of Kyonggi-Do Province (환경홀몬 Benzo[a]pyrene및 Bisphenol A의 경기도 일부호소에서의 요염실태 및 위해성 평가 방안)

  • Benzo(a)pyrene and bisphenol A have been classified as endocrine disrupting chemicals (EDCs), which have been of concern in toxicology and environmental sciences. Benzo(a)pyrene and bisphenol A were monitored by HPLC or GC-MS in Baekwoon and Ilwirl lakes of Kyonggi-Do province to investigate contamination levels of EDCs. During the period between June, 2000 and August, 2000, water samples were collected from four different sites of each lake once per month. Contamination levels of benzo(a)pyrene were 3.27~4.25 ppb in Ilwirl lake and 2.00~2.33 ppb in Baekwoon lake, respectively. Bisphenol A levels were detected with the range of 0.33~7.94 and 0.43~4.71 for Baekwoon lake and Ilwirl lake, respectively. pH levels were higher in Ilwirl lake than in Baekwoon lake, where the contamination was relatively lower. These data suggest that lakes in Kyonggi-Do province could be contaminated with EDCs and be subjected to the routine monitoring and water quality control.

  • Endocrine disrupting chemicals probably cause the cytological or/and morphological changes of germinal cells in gonad. Accordingly, this study was aimed to make sure that the effect of hormone-mimicking chemicals on gonad morphology such as decrease of germinal cells, inhibition of cellular maturation and change in the ratio of germinal cells in the different developmental phase can be observed by histopathological procedures and can be a useful bio-indicator for the evaluation of endocrine disruption by environmental chemicals. In this experiment, female Japanese medaka were exposured to quercetin, a phytoestrogen, at the concentration of 100 $\mu\textrm{g}$/L. quercetin showed the significant decrease in the number and rate of vitellogenic follicular oocytes in the treated group for 4 and 6 weeks. The weak development of yolk could be also observed. We could conclude that quercetin has anti-estrogenic or androgen-like potency by exerting the inhibition effect on oogenesis in fish female- gonad. From the result of this study, the applied methods and techniques can be evaluated to be a useful biomonitoring means for water pollution, expecting a good result of the subsequent study on apoptosis.

  • Endocrine disruptors have been implicated in carcinogenesis in animal studies, but carcinogenetic effects on human remain controversial. In order to examine the genotoxicity of two common endocrine disruptors, Bisphenol A and Diethylstilbestrol, cytogenetic endpoints including chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) analyses and DNA damage by single cell gel electrophoresis (SCGE) were assessed. The effects of Bisphenol A and Diethylstilbestrol on the frequencies of CA and MN were increased in a dose-dependent manner and that of Bispheol A was more significant by Kendall'$\tau$test. Bisphenol A and Diethylstilbestrol also increased the frequency of SCE. Bisphenol A and Diethylstilbestrol induced DNA damage in a dose-dependent manner and the DNA damage induced by Diethylstilbestrol in human blood lymphocytes was more significant.

  • To establish a test protocol for the rodent 20-day thyroid/pubertal assay, we dosed flutamide(fl), a non-steroidal androgen antagonist to intact male SD rats from postnatal day 33 for 20 days, and examined several reproductive endpoints for assessing the sensitivity of a list of parameters for detecting endocrine-related effects of endocrine-disrupting chemicals.(omitted)

  • Toxicology is under challenge from several new trends in science and technology, namely computer, the Internet, genome projects, genomic technologies, and combinatorial chemistry. These new trends will drastically change research style of toxicology. In addition to conventional uni cellular tests and animal tests using rodents, computer simulation, DNA chips (microarrays), in vivo tests using simple model organisms such as nematodesor flies become important routine screening tests. How to arrange these tests in tiers will become a new problem. Endocrine disruptors hypothesis is a good example for this kind of futuristic approach. Computer, particularly the Internet, is also enabling toxicologists and regulatory experts to collaborate more closely. The IPCS (International Program for Chemical Safety) which is ajoint project of WHO, ILO and UNEP, is a well-known international collaborative research for chemical risk assessments. The GINC project of IPCS is an effort to utilize the Internet for such collaborations. Some efforts were also made to establish regional collaboration network in East Asia under this project.
