Expression and Purification of Extracellular Solute-Binding Protein (ESBP) in Escherichia coli, the Extracellular Protein Derived from Bifidobacterium longum KACC 91563

  • Bifidobacterium longum KACC 91563 secretes family 5 extracellular solute-binding protein via extracellular vesicle. In our previous work, it was demonstrated that the protein effectively alleviated food allergy symptoms via mast cell specific apoptosis, and it has revealed a therapeutic potential of this protein in allergy treatment. In the present study, we cloned the gene encoding extracellular solute-binding protein of the strain into the histidine-tagged pET-28a(+) vector and transformed the resulting plasmid into the Escherichia coli strain BL21 (DE3). The histidine-tagged extracellular solute-binding protein expressed in the transformed cells was purified using Ni-NTA affinity column. To enhance the efficiency of the protein purification, three parameters were optimized; the host bacterial strain, the culturing and induction temperature, and the purification protocol. After the process, two liters of transformed culture produced 7.15 mg of the recombinant proteins. This is the first study describing the production of extracellular solute-binding protein of probiotic bacteria. Establishment of large-scale production strategy for the protein will further contribute to the development of functional foods and potential alternative treatments for allergies.

  • In this paper, we propose a method for generating luma signal mapping function to improve the coding efficiency of luma signal mapping methods in LMCS. In this paper, we propose a method to reflect the cognitive and perceptual features by multiplying the attention map of convolutional neural networks on local spatial variance used to reflect local features in the existing LMCS. To evaluate the performance of the proposed method, BD-rate is compared with VTM-12.0 using classes A1, A2, B, C and D of MPEG standard test sequences under AI (All Intra) conditions. As a result of experiments, the proposed method in this paper shows improvement in performance the average of -0.07% for luma components in terms of BD-rate performance compared to VTM-12.0 and encoding/decoding time is almost the same.

Fast Inverse Transform Considering Multiplications (곱셈 연산을 고려한 고속 역변환 방법)

  • In hybrid block-based video coding, transform coding converts spatial domain residual signals into frequency domain data and concentrates energy in a low frequency band to achieve a high compression efficiency in entropy coding. The state-of-the-art video coding standard, VVC(Versatile Video Coding), uses DCT-2(Discrete Cosine Transform type 2), DST-7(Discrete Sine Transform type 7), and DCT-8(Discrete Cosine Transform type 8) for primary transform. In this paper, considering that DCT-2, DST-7, and DCT-8 are all linear transformations, we propose an inverse transform that reduces the number of multiplications in the inverse transform by using the linearity of the linear transform. The proposed inverse transform method reduced encoding time and decoding time by an average 26%, 15% in AI and 4%, 10% in RA without the increase of bitrate compared to VTM-8.2.

Heterologous Expression of the Hot Pepper ABA 8'-Hydroxylase in Escherichia coli for Phaseic Acid Production

  • The CYP707A family genes encoding ABA 8'-hydroxylase catabolize abscisic acid (ABA), a plant stress hormone that plays an important role in stress condition, such as drought, heat, cold and salinity. Phaseic acid (PA) is a catabolic product of ABA. Recent studies have shown that PA is important for the physiological functions in plants. It is also a neuroprotective molecule that protects against ischemic brain injury in mice. To obtain enzymes for the PA production, four CaCYP707A genes (CaCYP707A1, CaCYP707A2, CaCYP707A3 and CaCYP707A4) were isolated from hot pepper. They were heterologously expressed in Escherichia coli. Among them, CaCYP707A2 showed significantly higher expression levels in both the membrane fraction and the soluble fraction. Preferred redox partners were investigated to improve the efficiency of CaCYP707A2's catalytic reaction, and NADPH-cytochrome P450 reductase (CPR) from hot pepper (CaCPR) was preferred over other redox partners (i.e., rat CPR and ferredoxin reductase/ferredoxin). The production of 8'-hydroxy ABA and PA by ABA hydroxylation activity was confirmed in CaCYP707A2 from both membrane and soluble fractions. Therefore, CaCYP707A2 is the first identified plant CYP protein that is expressed a soluble form in cytosolic fraction having stable activity. Taken together, we propose a new CYP707A protein with industrial applications for PA production without additional modifications in E. coli heterologous expression.

  • Research into vision-based end-to-end autonomous driving systems utilizing deep learning and reinforcement learning has been steadily increasing. These systems typically encode continuous and high-dimensional vehicle states, such as location, velocity, orientation, and sensor data, into latent features, which are then decoded into a vehicular control policy. The complexity of urban driving environments necessitates the use of state representation learning through networks like Variational Autoencoders (VAEs) or Convolutional Neural Networks (CNNs). This paper analyzes the impact of different image state encoding methods on reinforcement learning performance in autonomous driving. Experiments were conducted in the CARLA simulator using RGB images and semantically segmented images captured by the vehicle's front camera. These images were encoded using VAE and Vision Transformer (ViT) networks. The study examines how these networks influence the agents' learning outcomes and experimentally demonstrates the role of each state representation technique in enhancing the learning efficiency and decision- making capabilities of autonomous driving systems.

Optimization of Retrovirus Mediated-Gene Transfer into Hematopoietic Stem Cells (Retrovirus를 이용하여 조혈모세포에 유전자를 전달하기 위한 최적화)

  • In this study, optimal conditions to infect CD34 positive cells containing hematopoietic stem cells obtained from cord blood and bone marrow were found using two different retroviral vectors expressing human growth hormone (hGH) and $\beta$-galactosidase. CD34 positive cells were successfully infected with recombinant retroviruses only when the CD34 positive cells were co-cultured with packaging cells secreting recombinant retroviruses. To find the highest infection efficiency for the gene transfer, CD34 positive cells from cord blood were co-cultured with packaging cells secreting recombinant retroviruses encoding E. coli lacZ gene. The highest infection efficiency was obtained when CD34 positive cells were cultured for 3 days, and then co-culturing was done for another 2 days. When CD34 positive cells from bone marrow were co-cultured with packaging cells secreting recombinant retroviruses encoding hGH gene, the maximum amount of hGH was also secreted at the same conditions found above, i.e. 3 days of culture and 2 days of co-culture. These results show that there are optimal conditions for the gene transfer into hematopoietic stem cells regardless of sources of target cells or retroviral vectors used to infect.

Adaptive Search Range Decision for Accelerating GPU-based Integer-pel Motion Estimation in HEVC Encoders (HEVC 부호화기에서 GPU 기반 정수화소 움직임 추정을 고속화하기 위한 적응적인 탐색영역 결정 방법)

  • In this paper, we propose a new Adaptive Search Range (ASR) decision algorithm for accelerating GPU-based Integer-pel Motion Estimation (IME) of High Efficiency Video Coding (HEVC). For deciding the ASR, we classify a frame into two models using Motion Vector Differences (MVDs) then adaptively decide the search ranges of each model. In order to apply the proposed algorithm to the GPU-based ME process, starting points of the ME are decided using only temporal Motion Vectors (MVs). The CPU decides the ASR as well as the starting points and transfers them to the GPU. Then, the GPU performs the integer-pel ME. The proposed algorithm reduces the total encoding time by 37.9% with BD-rate increase of 1.1% and yields 951.2 times faster ME against the CPU-based anchor. In addition, the proposed algorithm achieves the time reduction of 57.5% in the ME running time with the negligible coding loss of 0.6%, compared with the simple GPU-based ME without ASR decision.

Regulation of GFP Expression Using the Tetracycline Inducible Retroviral Vector System (Tetracycline Inducible Retrovirus Vector System에 의한 GFP 유전자의 발현 조절)

  • One of the critical problems to be solved in transgenic animal production is uncontrollable constitutive expression of foreign genes, which usually results in serious physiological disturbances in the transgenic animal. To circumvent this problem, we constructed and tested two retrovirus vectors designed to express the GFP(green fluorescent protein) gene under the control of the tetracycline-inducible promoters. To maximize the GFP gene expression at turn-on state, WPRE(woodchuck hepatitis virus posttranscriptional regulatory element) sequence was introduced into the retrovirus vectors at downstream region of either the GFP gene or the sequence encoding rtTA(reverse tetracycline-controlled transactivator). Transformed cells were cultured in the medium supplemented with or without doxycycline(tetracycline derivative) for 48 hours, and induction efficiency was measured by comparing the GFP gene expression level using fluorometry and western blotting. Higher GFP expression was observed from the vector carrying the WPRE sequence at 3' side of the GFP gene, while tighter expression control(up to 20 fold) was obtained from the vector in which the WPRE sequence was placed at 3' side of rtTA sequence. The resulting tetracycline inducible vector system may be used in transgenic animal production and gene therapy.

Improved Bi-directional Symmetric Prediction Encoding Method for Enhanced Coding Efficiency of B Slices (B 슬라이스의 압축 효율 향상을 위한 개선된 양방향 대칭 예측 부호화 방법)

  • A bi-directional symmetric prediction technique has been developed to improve coding efficiency of B-slice and to reduce the computational complexity required to estimate two motion vectors. On the contrary to the conventional bi-directional mode which encodes both forward and backward motion vectors, it only encodes a single forward motion vector, and the missing backward motion vector is derived in a symmetric way from the forward motion vector using temporal distance between forward/backward reference frames to and from the current B picture. Since the backward motion vector is derived from the forward motion vector, it can halve the computational complexity for motion estimation, and also reduces motion vector data to encode. This technique always derives the backward motion vector from the forward motion vector, however, there are cases when the forward motion vector is better to be derived from the backward motion vector especially in scene changes. In this paper, we generalize the idea of the symmetric coding with forward motion vector coding, and propose a new symmetric coding with backward motion vector coding and adaptive selection between the conventional symmetric mode and the proposed symmetric mode based on rate-distortion optimization.

  • In this paper, a method of rate control for constant bitrate (CBR) coding of High Efficiency Video Coding (HEVC) is addressed. The existing rate control of H.264/AVC may not provide exact rate control in the case of hierarchical coding structure since it doesn't consider the characteristics of the hierarchical coding structure. It is expected that a rate control is added to the reference software called HM for CBR encoding in the near future. More accurate rate control may be required in a hierarchical structure of random access (RA) mode defined in the common test condition of HM. In this paper, we propose a method of rate control based on quadratic Rate-Distortion (R-D) model considering temporal layers and frame types in hierarchical coding structure for efficient rate control. In the consideration of the trade-off relationship between the bit fluctuation and the average PSNR, both of frame and coding unit (CU) are set as the basic unit of rate control. The performance of the proposed rate control method is verified by simulations along with the trade-off relationships for the both cases of basic unit.
