Development of Vivorium, a new indoor horticultural ornamental plants via plant tissue culture techniques (식물조직배양 기술을 이용한 새로운 실내 원예 장식품인 비보리움(Vivorium)의 개발)

  • Indoor gardening includes wall greening, terrariums, and flower arrangements. Among these types of indoor gardens, the terrarium is easy to access for the general public, but in Korea, because of the focus on esthetics, the original purpose of creating terrariums, which was to grow plants sustainably in an enclosed space, has been lost. In addition, miniaturization of plants is required to grow plants in an enclosed space. Since the available plant species suitable for a terrarium are limited, only plants such as succulents, cacti, and moss have been used. In this study, Bronze (X Graptosedum) was used, and these problems were solved using the following three methods: placement and growth of virus-free plants in the terrarium; extending the diversity of plants with minimal size that can be planted in terrariums; and reducing the price of in vitro plants with minimal size by achieving large-scale production. In particular, tissue-cultured succulents were developed into a Vivorium by replacing the tissue culture container and renewing the composition of the plant. This paper suggests a new indoor horticultural field, Vivorium, that can improve the current limitations of terrariums and make them more accessible to the general public. The introduction and popularization of new indoor gardening fields with the increase in single-person households and indoor activities in the Pandemic era can also improve psychological stability among people and in the society.

The Storage Stability of Semi-Salted and Dried Mackerel by Free-Oxygen Absorber (탈산소제에 의한 반염건고등어 저장중의 품질안정성)

  • The preservative effect of modified-atmosphere storage on the shelf-life of packed semi-salted and dried mackerel, Scomber japonicus, was examined. The semi-salted and dried mackerel fillets were packed in laminated plastic film bags (polyester/nylon/casted polypropylene: $12{\mu}m/15{\mu}m/60{\mu}m,\;15{\times}16cm$) filled with air (control, C), nitrogen gas (N), deoxygenized air (O) prepared by using free-oxygen absorber enclosed in the bag, in vacuum(V), and stored at $5^{\circ}C$. The quality of packed sample during the storage were examined in terms of viable cell counts of bacteria, thiobarbituric acid(TBA) value, perozide value(POV), volatile basic nitrogen(VBN), trimethylamine(TMA), adenosine triphosphate(ATP) and its related compounds and sensory evaluation. The results obtained are as follows: The pH of all the samples was in the range of $6.1{\pm}0.2$, and the contents of VBN and amino nitrogen of them increased during storage. In color values, L value(lightness) decreased while a and b values (red and yellow) revealed a tendency to increase during storage. The viable cell counts of the control sample(C) increased to $3.0{\times}10^6/g$ after 15 days storage but those of the other samples(V, N and O)were in the range of $2{\sim}6{\times}10^5/g$ after 20 days storage. The content of TMA increased during storage, but the histamine content showed a little change during storage and its content of all samples were less than 16 mg/100g. The inosinic acid(IMP) was rapidly degraded while inosine and hypoxanthine increased during storage. The TBA value of the control(C) reached a peak in 9 days and then decreased gradually while that of the sample(O) showed a little change during storage. The changes in POV of all the samples during storage showed a similar tendency to the TBA value. Fatty acid composition of raw mackerel consists of $35.6\%$ of saturated acid, $30.3\%$ of monoenoic acid and $34.2\%$ of polyenoic acid. The major fatty acid of the sample products were oleic acid($C_{18:1}$), palmitic acid($C_{16:0}$), docosahexaenoic acid($C_{22:6}$). The contents of polyenoic acid such as $C_{22:6},\;C_{20:5}$ decreased during storage while the other fatty acids showed a little change. From the results of sensory evaluation, the shelf-life of the control sample(C) was about 7 days and that of sample(V), (N) and (O) was about 15 days. It was concluded that deoxygenized atmosphere(free-oxygen absorber enclosed in the bag) was a good condition for preserving the quality of semi-salted and dried mackerel.

Effects of Weaning and Spatial Enrichment on Behavior of Turkish Saanen Goat Kids

  • As is in all economic activities, the highest yield per unit area is the main goal in animal production, while addressing the temperamental needs of animals often is ignored. Animal welfare is not only an ethical fact; it also has an economic value. Spatial environmental enrichment contributes positively to animal welfare by addressing their behavioral and mental requirements. The present study was conducted to determine the effects of weaning and spatial environmental arrangements on behaviors of goat-kids. Experimental groups were arranged in structured and unstructured spatial environments. Roughage feeder, semi-automatic concentrate feeder, bunk, bridge, and wood block were placed in the structured environment. No equipment was placed in the unstructured environment and paddock sides were enclosed with an iron sheet to prevent bipedal stance and to provide environmental isolation. In the study 10 male and 10 female Turkish Saanen goat kids were used in each group. Spatial environmental arrangements did not have significant impacts on the growth performance of kids (p>0.05). All objects in the structured group were accepted by the kids. Average use ratios of roughage feeder, semi-automatic concentrate feeder, bunk, bridge and wood block were observed as 19.3%, 14.0%, 12.6%, 3.8%, and 0.7%, respectively. There were significant differences between before- and after-weaning in use of all objects except for underneath bridge ($p{\leq}0.05$). Concentrate feed consumption, locomotion, and resting behaviors in kids showed significant differences by structural group and growth period. Roughage consumption was similar between groups, while it differed by growth period ($p{\leq}0.05$). Interaction frequency was significantly higher in structured group (p = 0.0023). Playing behavior significantly differentiated based on the growth period rather than on groups ($p{\leq}0.05$). Playing behavior significantly decreased after weaning. Abnormal oral activity was significantly higher in the structured group before weaning ($p{\leq}0.05$). Despite there being no installations facilitating climbing and bipedal stance, the kids of the unstructured group were able to exhibit 1/3 as much bipedal stance behavior as the kids of the structured group through leaning over slippery paddock wall or over their groupmates. Bipedal stance behavior of unstructured group was similar before and after weaning, while bipedal stance behavior before weaning was about 2 times that of after weaning in structured group. It was concluded that unstructured environmental arrangement limited the behavior repertoire of the goat kids.

Field Survey on the Structure and Manure Treatment of Two-Storey and Sawdust Pig Houses in Korea (국내 2층 돈사와 톱밥돈사의 구조 및 분뇨처리 실태조사)

  • The farm house structure, ventilation system and manure treatment of two-storey buildings and sawdust pig houses were surveyed and analyzed. Based on the data for ten selected farms in five provinces during eight months, the goal is to eventually establish a standard two-storey pig house. Manure treatments were composting, slurry and activated sludge in two-storey pig houses, while fermentation method was done in sawdust pig house. The depth of sawdust as a litter material were 10 to 60cm, with a duration of 1/2, 1, 3 and 6 months, respectively. The ventilation systems were the mechanical type in two-storey pig houses and natural system in the sawdust pig house. Side wall in the two-storey pig house was enclosed with insulation materials such as block, colored metal sheet and sandwich panels. The minimum ceiling height in the first floor of the two-storey pig house was 2.0m and the maximum was 3.0m. On the second floor, ceiling height ranged from 2.0 to 2.7m. The construction cost in the two-storey systems were $700\~140$, and sidewall curtain systems were $30\~40$ thousand Won/pyung.

Effect of Melatonin on the Maturation of Mouse Germinal Vesicle(GV)-Stage Oocytes and Apoptosis of Cumulus Cells In Vitro (멜라토닌이 생쥐 미성숙 난자의 체외성숙과 난구세포의 세포자연사에 미치는 영향)

  • Melatonin (N-acetyl-5-methoxytryptamine), a major hormone of pineal gland in vertebrates, is known to be associated with regulation of the dynamic physiological functions in general and has some functions on reproduction in the ovarian follicles in particular. And its antioxidant properties as a scavenger are also reported. The aim of this study was to investigate the effect of melatonin on the in vitro maturation of mouse germinal vesicle (GV)-stage oocytes. Oocyte maturation, apoptosis, and mRNA expression of melatonin receptor were analyzed in the cumulus cell-enclosed oocytes (CEOs) cultured with melatonin for 18 h. The CEOs were obtained from 3 wk-old ICR female mice cultured in media with 0, 0.1 nM, 10 nM, or 1,000 nM melatonin for 18 h. And then the extrusion of the first polar body was assessed to evaluate the maturation rate. The apoptosis and mRNA expression of melatonin receptor (Mtnr1-a and Mtnr1-b) in cumulus cells of each group were measured by TUNEL assay, ELISA, and real time RT-PCR after in vitro maturation(IVM). The addition of melatonin in the IVM medium significantly improved nuclear maturation of the mouse GV oocytes and the highest maturation rate were obtained from the group treated with 1,000 nM melatonin. Apoptosis was not detected in IVM oocytes, but detected in cumulus cells. And cumulus cells treated with 1,000 nM melatonin exhibited significantly lower apoptosis. In the group treated with 1,000 nM melatonin, the expression of melatonin receptor mRNA was decreased in CEOs. In conclusion, melatonin has a potentially important role for regulating oocyte maturation and reduces the apoptosis of cumulus cells in vitro.

  • The typical Japanese house has the characteristics of open dwellings to make them cool in the hot and humid summer. And then the traditional town house 'Machiya', being built very closely to each other and walled up both sides, it has taken the open-oriented characteristics in itself. The purpose of this study is to clarify that the open-oriented of traditional 'Machiya' has been succeeded to the new 'Machiya' in the latest. The new 'Machiya' shows the open-oriented, taking the ventilative 'Huki-nuke' space of traditional 'Machiya' as the new spatial formal elements, in the changes such as scale material space organization. The characteristics of 'Huki-nuke' space are represented as follows ; 1. The facade of the traditional 'Machiya', which has taken on a semitransparent qualty, has been generally changed to the closing qualty except for the open parts of shop and garage. This facade of the new 'Machiya' has been taken to be in keeping with the existing town as much as possible. 2. A series of three rooms, composed of shop/living dining kitchen/room from the road, have been dispersed to every floors in a building with a very extensive scale. But this serial and linear type remains as the loosefit space, and the long dwellings of the upper stories are divided by type each dwelling unit. 3. 'Tori-niwa', which is a consecutive and penetrating space, connects the road with the rooms of dwelling and functions as the circulation of man thing energy, The new 'Machiya' changed to the multi-story, the corridor and the stair have been fumed up as the elements in the place of 'Tori-niwa' The 'Huki-nuke' space was locted in the hall, stairwell, living dining kitchen room, and so on. 4. The small court yard 'Tsubo-niwa' and back yard 'Ura-niwa' at the both ends of living spaces would be made a hole in a series of rooms and enclosed by the neighboring 'Machiya'. On the contrary the new 'Machiya' at present takes in the private and closing organization enclosing the innercourt. 5. The open-oriented ${\ulcorner}$In${\lrcorner}$ or ${\ulcorner}$Out${\lrcorner}$ is not brought out because of the delicate spatial formal configuration in the traditional 'Machiya'. But the open-oriented ${\ulcorner}$In${\lrcorner}$, all sides being closed by walls, is well brought out in the new 'Machiya'.

Investigation on Optimal Aeration Rate for Minimizing Odor Emission during Composting of Poultry Manure with Sawdust (계분톱밥 퇴비화시 악취발생의 최소화를 위한 적정 공기주입을 구명)

  • This study was conducted to find out the optimal aeration rates for minimizing odor emission and for increasing biological activities during composting of livestock manure in the enclosed bench-scale reactor system. It was treated with the mixture of poultry manure and sawdust controlled the initial water content of 60%, then aerated continuously at four different aeration rates (0.1, 0.2, 0.4 and 0.6 L/min/kg dry-solids). The average emitted concentration of ammonia in 0.6 L/min/kg dry-solids during composting reached the level of 40% in comparison with that of 0.2 L/min/kg dry-solids. In cases of sulfur compounds such as hydrogen sulfide, methylmercaptan and ethylmercaptan, their concentrations decreased with increasing aeration rates and the emission time was shortened. But they didn't detect in the treatment of 0.6 L/min/kg dry-solids. The biological activity for composting showed a trend of increasing as aeration rates increased. The treatment of 0.6 L/min/kg dry-solids gave the highest biological activity and the best compost quality.

  • The origins and the functions of the multi vesicular bodies and the various structures of the membranes related to the cytolysomes were studied in the mycelium cells of Rhizopus nigricans, Aspergillus niger and A. ochraceus, in the hymenium and basidium cells of Agricus bisporus sand Rhizopogon rubesecens, in the cells of assimilation tissue of Marchtantia polymorpha and Pogonalum inflexum and in the mesophyll cells of Pteridium aqiulinum, Pinus densiflora, Ginkgo biloba and Panax ginseng fixed with glutaraldehyde-paraformaldehyde-$ OsO_4$. In Rhizopus nigricans, Aspergillus niger, A. ochraceus, Agricus bisporus sand Rhizopogon rubescens, the concentric multilamellar, multivesicular, myelin-vesicle-tubular and concentric parallel-lamellar complexes were originated from the plasmalemma, while in Marehantia polymorpha, Pogonatum inflexum, Pteridium aquilinum, Pinus densiflora, Ginkgo biloba and Panax ginseng, they were originated from plasmalemma and the cytoplasm. The structures originated from the plasmalemma may be grouped into multi vesicular body and myelin-like structure, both forming the secondary vacuoles or protruding into the central vacuoles and finally degrading, In some cases, endoplasmic reticulum within the cytoplasm encloses some part of the cytoplasm to form a circle where the membranous lamellae increase in number, while the enclosed cytoplasm decrease to be eventually replaced by the multilamellar structure which is released into the vacuoles and subsquently degraded. The structures originated from the cytoplasm are believed to be the cytosegresomes or cytolysomes closely related to the differentiation of the vacuoles. The possible fate of these structures are also discussed.

A Survey of Plastid Crystals and Microtubules in Flowering Plants (꽃피는식물 색소체 내 결정구조와 미세소관의 발달양상 조사 연구)

  • The plastid inclusion has long been known to exist in leaves of numerous plant species, especially in those of flowering plants. Among the inclusions, crystalline bodies are the most frequently distinguished structures of the foliar plastids, however, microtubules and phytoferritins are also reported occasionally. The crystalline inclusions vary in shape, and are located either in the stroma or within intrathylakoidal spaces, whereas microtubules and phytoferritins are more uniform in shape and are formed in the stroma. In crystalline structures, the composing elements exhibit a lattice pattern and/or paralleled tubules that are either bounded by membranes or exist without membrane enclosing. Other types of inclusions have not been shown to be enclosed by any membranous structures. According to the current survey, the plastid inclusion, with the exception of phytoferritins, has been shown to exhibit a crystalline or tubular pattern, and has been reported in more than 56 species of various families. Their occurrence is not restricted to any photosynthetic pathway, but is found to be randomly distributed among C-3, C-4 and CAM species, without phylogenetic relationships. The progress in plastid inclusion research reveals more information about the function and complexity, but the need for characterizing the 3-D structure of the crystalline inclusions also has been acknowledged in previous studies. A 3-D characterization would utilize tilting and tomography of serial sections with appropriate image processing that would provide valuable information on the sub-structures of the crystalline inclusions. In fact, recent studies performed on 3-D reconstruction of the plastid inclusions revealed important information about their comprising elements. In this article, the crystals and microtubules that have been reported in various types of plastids have been reviewed, with special consideration given to their possible sub-cellular function within the plastids.

Freshness Monitoring of Raw Salmon Filet Using a Colorimetric Sensor that is Sensitive to Volatile Nitrogen Compounds (휘발성 질소화합물 감응형 색변환 센서를 활용한 연어 신선도 모니터링)

  • A colorimetric paper sensor was used to detect volatile nitrogen-containing compounds emitted from spoiled salmon filets to determine their freshness. The sensing mechanism was based on acid-base reactions between acidic pH-indicating dyes and basic volatile ammonia and amines. A sensing layer was simply fabricated by drop-casting a dye solution of bromocresol green (BCG) on a polyvinylidene fluoride substrate, and its color-change response was enhanced by optimizing the amounts of additive chemicals, such as polyethylene glycol, p-toluene sulfonic acid, and graphene oxide in the dye solution. To avoid the adverse effects of water vapor, both faces of the sensing layer were enclosed by using a polyethylene terephthalate film and a gas-permeable microporous polytetrafluoroethylene sheet, respectively. When exposed to basic gas analytes, the paper-like sensor distinctly exhibited a color change from initially yellow, then to green, and finally to blue due to the deprotonation of BCG via the Brønsted acid-base reaction. The use of ammonia analyte as a test gas confirmed that the sensing performance of the optimized sensor was reversible and excellent (detection time of < 15 min, sensitive naked-eye detection at 0.25 ppm, good selectivity to common volatile organic gases, and good stability against thermal stress). Finally, the coloration intensity of the sensor was quantified as a function of the storage time of the salmon filet at 28℃ to evaluate its usefulness in monitoring of the food freshness with the measurement of the total viable count (TVC) of microorganisms in the food. The TVC value increased from 3.2 × 105 to 3.1 × 109 cfu/g in 28 h and then became stable, whereas the sensor response abruptly changed in the first 8 h and slightly increased thereafter. This result suggests that the colorimetric response could be used as an indicator for evaluating the degree of decay of salmon induced by microorganisms.
