Intratumoral distribution of 64Cu-ATSM and 18F-FDG in VX2tumor xenografted rabbit

  • $^{64}Cu$-labeled diacetyl-bis($N^4$-methylthiosemicarbazone) is a promising agent for internal radiation therapy and imaging of hypoxic tissues. In the study, we confirmed hypoxia regions in VX2 tumor implanted rabbits with injection $^{64}Cu$-ATSM and $^{18}F$-FDG using positron emission tomography (PET)/computed tomography (CT). PET images with $^{18}F$-FDG and $^{64}Cu$-ATSM were obtained for 40 min by dynamic scan and additional delayed PET images of $^{64}Cu$-ATSM the acquired up to 48 hours. Correlation between intratumoral $O_2$ level and $^{64}Cu$-ATSM PET image was analyzed. $^{64}Cu$-ATSM and $^{18}F$-FDG were intravenously co-injected and the tumor was dissected and cut into slices for a dual-tracer autoradiographic analysis. In the PET imaging, $^{64}Cu$-ATSM in VX2 tumors displayed a specific uptake in hypoxic region for48 h. The uptake pattern of $^{64}Cu$-ATSM in VX2 tumor at 24 and 48 h did not match to the $^{18}F$-FDG. Through ROI analysis, in the early phase (dynamic scan), $^{18}F$-FDG has positive correlation with $^{64}Cu$-ATSM but late phase (24 and 48 h) of the $^{64}Cu$-ATSM showed negative correlation with $^{18}F$-FDG. High uptake of $^{64}Cu$-ATSM in hypoxic region was responded with significant decrease of oxygen pressure, which confirmed by $^{64}Cu$-ATSM PET imaging and autoradiographic analysis. In conclusion, $^{64}Cu$-ATSM can utilize for specific targeting of hypoxic region in tumor, and discrimination between necrotic- and viable hypoxic tissue.

  • Geopolymer is an eco-friendly construction material that has various advantages such as reduced $CO_2$ emission, fire resistance and low thermal conductivity compared to cement. However, it has not been many studies on the thermal behavior of the surface of the geopolymer panel when flame is applied to the surface. In this study, surface characteristics of hardened geopolymer on flame exposure was investigated to observe its characteristics as heat-resistant architectural materials. External structure changes and crack due to the heat shock were not observed during the exposure on flame. According to the residue of calcite and halo pattern of aluminosilicate gel, decarboxylation and dehydration were extremely limited to the surface and, therefore, it is thought that durability of hardened geopolymer was sustained. Gehlenite and calcium silicate portion was inversely proportional to quartz and calcite and significantly directly proportional to BFS replacement ratio. Microstructure changes due to the thermal shock caused decarboxylation and dehydration of crystallization and it was developed the pore and new crystalline phase like calcium silicate and gehlenite. It is thought that those crystalline phase worked as a densification and strengthening mechanism on geopolymer panel surface.

Synthesis and Optical Property of (GaN)1-x(ZnO)x Nanoparticles Using an Ultrasonic Spray Pyrolysis Process and Subsequent Chemical Transformation (초음파 분무 열분해와 화학적 변환 공정을 이용한 (GaN)1-x(ZnO)x 나노입자의 합성과 광학적 성질)

  • In this study, (GaN)1-x(ZnO)x solid solution nanoparticles with a high zinc content are prepared by ultrasonic spray pyrolysis and subsequent nitridation. The structure and morphology of the samples are investigated by X-ray diffraction (XRD), field-emission scanning electron microscopy, and energy-dispersive X-ray spectroscopy. The characterization results show a phase transition from the Zn and Ga-based oxides (ZnO or ZnGa2O4) to a (GaN)1-x(ZnO)x solid solution under an NH3 atmosphere. The effect of the precursor solution concentration and nitridation temperature on the final products are systematically investigated to obtain (GaN)1-x(ZnO)x nanoparticles with a high Zn concentration. It is confirmed that the powder synthesized from the solution in which the ratio of Zn and Ga was set to 0.8:0.2, as the initial precursor composition was composed of about 0.8-mole fraction of Zn, similar to the initially set one, through nitriding treatment at 700℃. Besides, the synthesized nanoparticles exhibited the typical XRD pattern of (GaN)1-x(ZnO)x, and a strong absorption of visible light with a bandgap energy of approximately 2.78 eV, confirming their potential use as a hydrogen production photocatalyst.

Fluorescence Anisotropy Study on the Effect of Phellodendri Cortex's Berberine on Regulation of the Function of DNA (황백(黃柏)의 berberine이 DNA의 기능조절에 미치는 영향에 관한 형광이방성 연구)

  • Objectives : We tried to observe the fluorescence anisotropy and intensity of ethidium ion in the intercalating binding interaction between DNA and ethidium ions in the presence of berberine, and then tried to explain the effect of berberine on the intercalating interaction of ethidium ion with DNA. Methods : DNA(calf thymus DNA), berberine and ethidium bromide(EtBr) were purchased from Sigma-Aldrich Co. Proper amount of each compound was dissolved in 20 mM sodium phosphate buffer(pH 7.0) containing 100 mM of NaCl to prepare stock solutions. Collections of the fluorescence anisotropy and intensity data were performed on JASCO FP-8300 spectrofluorometer equipped with a polarizer and a Peltier temperature controller. The excitation of ethidium ion was done at 550 nm and the emission data were collected at 600 nm. For Stern-Volmer plot, the fluorescence data were collected at $18^{\circ}C$ and $30^{\circ}C$. Results : According to the results of this research, the weak competitive binding pattern between ethidium ion and berberine appeared in binding with DNA at low ratio of DNA to ethidium ion. But at high ratio of DNA to ethidium ion, this weak competition disappeared. Instead, berberine might bind to DNA by intercalating way. In other words, berberine could de-intercalate ethidium ion from DNA at low concentration of DNA relative to ethidium ion, but could not at high concentration of DNA relative to ethidium ion. In addition, the mechanism of fluorescence quenching of ethidium ion could also proceed differently, depending on the ratio of the amount of DNA to that of ethidium ion. Conclusions : The effect of berberine on the DNA-ethidium ion intercalating interaction could work differently, depending on the relative ratio of the amount of DNA to that of ethidium ion. This study also showed that fluorescence anisotropy analysis is very useful method to obtain detailed information for investigation of the complex binding interactions. In order to fully understand the mechanism of action of the pharmacological effect by berberine, studies on the effect of berberine on the action of proteins such as various enzymes closely related to berberine-induced medicinal effects should be continued.

Atmospheric Circulation Patterns Associated with Particulate Matter over South Korea and Their Future Projection (한반도 미세먼지 발생과 연관된 대기패턴 그리고 미래 전망)

  • Particulate matter air pollution is a serious problem affecting human health and visibility. The variations in $PM_{10}$ concentrations are influenced by not only local emission sources, but also atmospheric circulation conditions. In this study, we investigate the temporal features of $PM_{10}$ concentrations in South Korea and the atmospheric circulation patterns associated with high concentration episodes of $PM_{10}$ during winter (December-January-February) 2001-2016. Based on those analyses, a Korea Particulate matter Index (KPI) is developed to represent the large-scale atmospheric pattern associated with high concentration episodes of $PM_{10}$. The atmospheric patterns are characterized by persistent high-pressure anomalies, weakened lower-level north-westerly anomalies, and northward shift of the upper-level meridional wind anomalies near the Korean Peninsula. To evaluate the change in occurrence of high concentration episodes of $PM_{10}$ under a possible future warmer climate, we apply KPI analysis to CMIP5 climate simulations. Here, historical and two representative concentration pathway (RCP) scenarios (RCP 4.5 and RCP 8.5) are used. It is found that the occurrence of atmospheric conditions favorable for high $PM_{10}$ concentration episodes tends to increase over South Korea in response to climate change. This suggests that large-scale atmospheric circulation changes under future warmer climate can contribute to increasing high $PM_{10}$ concentration episodes in South Korea.

Evaluation of Reasonable $^{18}F$-FDG Injected Dose for Maintaining the Image Quality in 3D WB PET/CT (PET/CT 검사에서 영상의 질을 유지하기 위한 적정한 $^{18}F$-FDG 투여량의 평가)

  • Purpose: $^{18}F$-FDG injected dose to the patient is quite different between the recommended dose from manufacturer and the actual dose applied to each of hospitals. injection of inappropriate $^{18}F$-FDG dose may not only increase the exposed dose to patients but also reduce the image quality. we thus evaluated the proper $^{18}F$-FDG injected dose to decrease the exposed dose to patients considering the image quality. Materials And Methods: NEMA Nu2-1994 phantom was filled with $^{18}F$-FDG increasing hot cylinder radioactivity concentration to 1, 3, 5, 7, 9 MBq/kg based on the ratio of 4:1 between the hot cylinder and background activity. after completing the transmission scan using ct, emission scan was acquired in 3D mode for 2 minutes 30 seconds/bed. ROI was set up on hot cylinder and background radioactivity region. after measuring $SUV_{max}$ those regions, then analyzed SNR at the points. clinical experiment has been conducted the object of patients who have came to smc from november 2009 to august 2010, 97 patients without having a hepatic lesions were selected. ROI was set up in the liver and thigh area. after measuring $SUV_{max}$, the image quality was compared following the injected dose. Results: in phantom study, as the injected radioactivity concentration per unit mass was 1, 3, 5, 7, 9 MBq/kg, $SUV_{max}$ was 23.1, 24.1, 24.3, 22.8, 23.6 and SNR was shown 0.48, 0.54, 0.56, 0.55, 0.55. according to increment of the injected dose, $SUV_{max}$ and SNR was increased under 5 MBq/kg but they were decreased over 7 MBq/kg. in case of clinical experiment, as increased the injected radioactivity concentration per unit mass was 4.72, 5.34, 6.16, 7.41, 8.68 MBq/kg, $SUV_{max}$ was 2.68, 2.67, 2.26, 1.88, 1.95 and SNR was shown 0.52, 0.53, 0.46, 0.46, 0.44. if the injected dose exceeds 5 MBq/kg, showed a decrease pattern as phantom study. Conclusion: increasing $^{18}F$-FDG injected dose considered patient's body weight improve image quality within a certain range. if it exceeds the range, it can be reduced image quality due to random and scatter coincidences. this study indicates that the optimal injected dose was 5 MBq/kg per unit mass the injected radioactivity concentration in 3d wb pet/ct.

Influence of N Fertilization Level, Rainfall, and Temperature on the Emission of N2O in the Jeju Black Volcanic Ash Soil with Carrot Cultivation (당근 재배 화산회토양에서 질소시비 수준 및 강우, 온도 환경 변화에 따른 N2O 배출 특성)

  • This study was conducted to obtain investigated characteristic factors which has an influence on nitrous oxide ($N_2O$) emissions related to the environment change of nitrogen application level, rainfall and temperature. It was done by the carrot cultivation at black volcanic ashes soil in the experimental field of Jeju Special Self-governing Province Agricultural Research and Extension Services from 2010 to 2011. During the carrot cultivation period, the more amount of nitrogen fertilizer applied, the more amount of $N_2O$ emissions were released. Generally $N_2O$ emissions were so deeply released to climate as that in the first and middle of cultivation with heavy rainfall released amount is high, otherwise it was released very low at the end of cultivation and drought season. $N_2O$ emissions type was considered to relate with the rainfall pattern and soil water content. We obtained the result correlated with $N_2O$ emissions, in 2010, as the soil water and soil temperature were significant to $0.5718^{**}$ ($r$) and $0.4908^{**}$ ($r$) respectively, but soil EC was not significant to 0.2704 ($r$). In 2011, soil water was significant to $0.3394^*$ ($r$), but soil temperature and soil EC were not significant to 0.2138 ($r$) and 0.2462 ($r$) respectively. Also, $NO_3$-N and soil nitrogen ($NO_3-N+NH_4-N$) were not significant to 0.0575 ($r$) and 0.0787 ($r$) respectively. During the carrot cultivation period, the average emissions factor released by the nitrogen fertilizer application for 2 years was presumed to be 0.0025 $N_2O$-N kg / N kg. This factor was 4 times than the IPCC (0.0100 $N_2O$-N kg / N kg) factor.

Assessment of Metabolic Impairment in Alzheimer's Disease with [$^{18}F$]FDG PET: Validity and Role of Simplified Tissue Radioactivity Ratio Analysis (알쯔하이머병에서 양전자방출단층촬영을 이용한 국소뇌포도당대사의 변화에 관한 연구)

  • The purpose of the present study was to validate the use of tissue radioactivity ratios instead of regional metabolic rates for the assessment of regional metabolic changes in Alzheimer's disease(AD) with [$^{18}F$]FDG PET and to examine the correlation of ratio indices with the severity of cognitive impairment in AD. Thirty-seven AD Patients(age $68{\pm}9 yrs$, $mean{\pm}s.d.$; 36 probable and 1 definite AD), 28 patients with dementia of non-Alzheimer type(age $66{\pm}7 yrs$), and 17 healthy controls(age $66{\pm}4 yrs$) underwent [$^{18}F$]FDG PET imaging. Two simplified radioactivity ratio indices were calculated from 37-66 min image: region-to-cerebellar radioactivity ratio(RCR) and a composite radioactivity ratio(a ratio of radioactivity in the most typically affected regions over the least typically affected regions: CRR). Local cerebral metabolic rate for glucose(LCMRglu) was also measured using a three-compartment, five-parameter tracer kinetic model. The ratio indices were significantly lower in AD patients than in controls(RCR in temporoparietal cortex, $0.949{\pm}0.136$ vs. $1.238{\pm}0.129$, p=0.0004; RCR in frontal cortex, $1.027{\pm}0.128$ vs. $1.361{\pm}0.151$, p<0.0001; CRR, $0.886{\pm}0.096$ vs. $1.032{\pm}0.042$. p=0.0024). On the RCR analysis, 86% of AD patients showed a pattern of bilateral temporoparietal hypometabolism with or without frontal involvement; hypometabolism was unilateral in 11% of the patients. When bilateral temporoparietal hypometabolism was considered to be suggestive of AD, the sensitivity and specificity of the RCR analysis for the differential diagnosis of AD were 86% and 73%, respectively. The RCR was correlated significantly with the macroparameter K [$K_1k_3/(k_2+k_3)$] (r=0.775, p<0.0001) and LCMRglu(r=0.633, p=0.0002) measured using the kinetic model. In patients with AD, both average RCR of cortical association areas and CRR were correlated with Mini-Mental Status Examination(r=0.565, p=0.0145; r=0.642, p=0.0031, respectively), Clinical Dementia Rating(r=-0.576, p=0.0124; r=-0.591, p=0.0077), and total score of Mattis Dementia Rating Scale (r=0.574, p=0.0648; r=0.737, p=0.0096). There were also significant correlations between memory and language impairments and corresponding regional RCRs. The results suggest that the [$^{18}F$]FDG PET ratio indices, RCR and CRR, reflect global and regional metabolic rates and correlate with the severity of cognitive impairment in AD. The simplified ratio analysis may be clinically useful for the differential diagnosis and serial monitoring of the disease.

Review of Policy Direction and Coupled Model Development between Groundwater Recharge Quantity and Climate Change (기후변화 연동 지하수 함양량 산정 모델 개발 및 정책방향 고찰)

  • Global climate change is destroying the water circulation balance by changing rates of precipitation, recharge and discharge, and evapotranspiration. The Intergovernmental Panel on Climate Change (IPCC 2007) makes "changes in rainfall pattern due to climate system changes and consequent shortage of available water resource" a high priority as the weakest part among the effects of human environment caused by future climate changes. Groundwater, which occupies a considerable portion of the world's water resources, is related to climate change via surface water such as rivers, lakes, and marshes, and "direct" interactions, being indirectly affected through recharge. Therefore, in order to quantify the effects of climate change on groundwater resources, it is necessary to not only predict the main variables of climate change but to also accurately predict the underground rainfall recharge quantity. In this paper, the authors selected a relevant climate change scenario, In this context, the authors selected A1B from the Special Report on Emission Scenario (SRES) which is distributed at Korea Meteorological Administration. By using data on temperature, rainfall, soil, and land use, the groundwater recharge rate for the research area was estimated by period and embodied as geographic information system (GIS). In order to calculate the groundwater recharge quantity, Visual HELP3 was used as main model for groundwater recharge, and the physical properties of weather, temperature, and soil layers were used as main input data. General changes to water circulation due to climate change have already been predicted. In order to systematically solve problems associated with how the groundwater resource circulation system should be reflected in future policies pertaining to groundwater resources, it may be urgent to recalculate the groundwater recharge quantity and consequent quantity for using via prediction of climate change in Korea in the future and then reflection of the results. The space-time calculation of changes to the groundwater recharge quantity in the study area may serve as a foundation to present additional measures for the improved management of domestic groundwater resources.

Evaluation of Airborne Pb Sources in an Industrialized City by Applying Pb Isotope Ratios and Concentrations in PM10 (PM10 내 납의 동위원소와 농도를 활용한 산업도시지역 대기 중 납 오염원 평가)

  • The present study evaluated the major lead sources in a steel metallurgy industrialized city by measuring lead isotopes/lead concentrations of ambient air and potential sources in an industrial area and residential areas according to relative distance. The quality control program obtained during the measurement procedure for lead isotopes and concentrations exhibited $0.5ng/m^3$ for method detection limit, more than 90% for recoveries of standard particulate matters, and lower than 0.2% for reproducibility errors of four lead isotopes ($^{204}Pb$, $^{206}Pb$, $^{207}Pb$, $^{208}Pb$). For all three lead isotope ratios ($^{206}Pb/^{204}Pb$, $^{207}Pb/^{206}Pb$, $^{208}Pb/^{206}Pb$), the ratios were obtained in the industrial area were closer to nearby residential area than those of a residential area far away from the industrial area, thereby suggesting that lead sources were more similar each other in the industrial and nearby residential area. Furthermore, for both summer and winter seasons ambient lead concentrations were more than four times higher in the industrial area than in the residential areas and in turn, they were higher in the nearby residential area compared with the far-away residential area. As a result, it was suggested that lead emitted from the industrial area would influence more the ambient lead in the nearby residential area than the far-away residential area. Both slag and traffic emissions are likely to be major lead sources in the industrial and nearby residential areas, since their three lead isotope ratios ($^{206}Pb/^{204}Pb$, $^{207}Pb/^{206}Pb$, $^{208}Pb/^{206}Pb$) were similar to the ratios obtained from ambient air of these two areas. In addition, the lead isotope ratios revealed different pattern between seasons, and the ambient lead concentrations were higher for winter than for summer.
