소 핵이식란의 이식 후 생존성에 관한 연구 (Viability of Nuclear Transfer Bovine Embryos after Embryo Transfer)

  • This study was conducted to examine the viability of nuclear transfer bovine embryos following embryo transfer. Donor embryos were treated with nocodazole to arrest their cell-cycle-stage at mitotic(M) phase. After releasing from nocodazole blastomeres were separated and transferred into the enucleated oocytes(BC), or cultured in medium with aphidicolin. Freshly cleaved blastomeres within 1.5h after cleavage(AC) and non-cleaved ones up to 3h after releasing from nocodazole(NC) were transferred into the enucleated oocytes. Blastocysts derived from nuclear transfer were transferred to Day 7~8 recipient cows. Some blastocysts were vitrified and thawed before embryo transfer. Developmental rates to the blastocyst stage were higher in AC(18.1%, P<0.05) than BC(8.6%) and NC(5.1%). Blastocyst development slightly enhanced with aphidicolin(1~2$\mu\textrm{g}$/ml) treatment(16.9~22.6%) compared to non treated control(11.1%). Survival rate fo vitrified nuclear transfer embryos after thawing was 75%(24/32). Twnety-three vitrified nuclear transfer embryos and 3 fresh ones were transferred to 23 recipients, 6 heads were pregnant and 1 male calf(24 kg) was born from a recipient cow recevied one vitrifiedthawed nuclear transfer embryo at 277 days after embryo transfer. This result suggests that the nuclear transfer embryos can developed to term after vitrification andembryo transfer.

Embryo Collection, Transfer and Pregnancy of Riding Horses : First Successful Case in Korea

  • Embryo transfer (ET) could be a relevant tool for genetic improvement programs in horses similar to those already underway in other species and produce multiple foals from the same mare in one breeding season. However, there have been no reports describing equine embryo transfer performed in Korea. In the present study, we performed an equine embryo collection and transfer procedure for the first time. We examined the embryo collection and pregnancy, size of embryo during the incubation period after collection, and progesterone (P4) and estradiol-$17{\beta}$ (E2) concentrations in mare's serum at embryo collection and transfer. A total of 16 donors responded to estrus synchronization; estrus was induced in 12 donors and 4 recipients, and artificial insemination was successful in 10 donors and six blastocysts were collected from donors. Of these blastocysts, we monitored the size of blastocysts for 3 day during incubation and transferred 2 blastocysts to a recipient, with 1 successful pregnancy and foal achieved. The dimensions of equine embryo at day 7 to day 9 were $409{\mu}m$, $814{\mu}m$ and $1,200{\mu}m$. The serum P4 and E2 concentrations were $7.91{\pm}0.37ng/{\mu}L$ and $45.45{\pm}12.65ng/{\mu}L$ in the donor mare, and 1$6.06{\pm}3.27ng/{\mu}L$ and $49.13{\pm}10.09ng/{\mu}L$ in the recipient mare.

  • Problems in the embryo transfer techniques and their practical a, pp.ication have been settle to some extent. However, with theorganic cooperation between universities and the related institutes for more active works on the training of embryo transfer technicians, quarantine procedures of embryos when they are imported and exported and the future researches related to the embryo transfer, the embryo transfer techniques in Korea will be more practically, a, pp.ied to animal industries. Universalization of embryo transfer techniques to the extent of artificial insemination will greatly contribute to the improvement of farm animals and multiplication of superior stocks and will save money by the import of relatively cheap embryos instead of expensive live animals.

돼지 수정란이식의 현황과 문제점 (Advances and Problems of Embryo Transfer in Swine)

  • Remarkable progress has recently been made in embryo transfer technology, resulting in the birth of IVF and nuclear transfer offsprings in swine. However, further progress of the technology to (I) make a safe, effective and economic estrual-cycle synchronization compound, (2) regulate each step of sperm capacitation (3) induce monospermic fertilization, (4) in vitro grow and mature oocytes, (5) fertilize the oocytes efficently, (6) culture the oocytes to the blastocyst stage in defined media, (7) produce multiply copies of embryos with superior genetic merit, (8) preselect the sex of these superior offsprings, and (9) preserve embryos by freezing and storage in liquid nitrogen is required before this promising technology is applied routinely to swine for practical use.
