It is widely recognized that the embryonic or fetal loss after breeding is common in the cattle and that it is an important factor affecting reproductive efficiency. The causes of this loss have been subject of extensive researches and the results indicate that the embryonic mortality may he primary factor responsible for low pregnancy rates in non-embryo transfer bovine populations as well as embryo transfer programs. However, it's causes are still not clearly understood. The embryonic mortality or pregnancy rate has been influenced by various embryonic and maternal effects related to genetic and environmental factors. The timing and extent of embryonic mortality vanes greatly according to authors and estimating methods, because it is difficult to make direct measurements. The major important factors that may influence the embryonic losses or pregnancy rates after embryo transfer can be summeirized. 1.When an embryo is transferred to unmated recipients, the contralateral transfer to corpus luteum results in a lower survival rate than ipsilateral deposition. When the embryos are transferred for the production of twin calves, their survivals and twin pregnancies have quite inconsistent according to the transfer methods either to the unmated-synchronized or already mated recipients and more works are needed to accurrately clarify the previous results. 2.Although embryos can be cultured in vitro some hours without the great declines in pregnancy rates, the rates differ markedly among culture times and media but may be improved by co-transfer systems. 3.Embryo developmental stages and quality grades clearly affect the survival rate following freezing and the pregnancy rate after transfer and the selection of embryos without chromosome abnormalities and of high fertile semen may also be considered to increase the pregnancy rates. 4.Many researches have attempted to relate the plasma progesterone levels to pregnancy rates and others have done either direct progesterone supplementation or luteal stimulation by hCG treatment in order to increase the pregnancy rates. However, these effects on pregnancy rates are inconsistent and also contradictory. 5.The asynchrony between donors or embryos and recipients may he a major cause of embryo death and low pregnancy rate and the sensitivity to uterine asynchyony differs in according to the quality and stages of embryos. 6.The extremes of poor or over nutrition during early pregnancy in the recipients are detrimental to the survival of embryos and the good body condition is required to prevent a reduejion of pregnancy rates. The uterine pathogens in embryonic mortality or fertility have been questioned but the infection of C.pyogenes and Campylobacter fetus is still important pathogens. 7.The heat stress during early pregnancy may reduce conceptus weight and possibly increase the embryonic mortality.
To evaluate possible teratogenicity of food additives such as sodium propionate and monosodium glutamate, 300 haching eggs were subjected to potential mutagenicity assay by administration of low and high doses of the materials via york and air sac. In vitro embryo culture techniques provide significant contributions not only for a basic research of fertilization and early embryogenesis, but also for a low cost mass production of bovine embryos for transfer, embryo diagnosis, nuclear cloning and the production of transgenic cows. This presentation introduces newly developed serum-free media (IVD101 and IVMD101) that are effective far high yields of transferable embryos of excellent quality from in vitro-matured and fertilized oocytes. Both serum-free media are superior to a conventional serum-containing medium on the increased rates of blastocyst formation, post-thaw embryo viability, and pregnancy after transfer. Furthermore, reduced risks of calf mortality and large calf syndrome are also observed for the serum-free-derived embryos. Serum-derived embryos contain a large number of lipid droplets and immature mitochondria in their cytoplasm that may account for the lower production of transferable embryos and poor embryo quality.
In this experiment, embryos of Asian toad, Bufo gargarizans, were investigated to evaluate toxicity of chemicals along FETAX(Frog Embryo Teratogenesis Assay-Xenopus) protocol. Asian toad, Bufo gargarizans, embryos incubated and investigation of Zn and Benomyl effect by probit analysis. As a result, depends on the concentrations of Zn and Benomyl, mortality and malformation rates were increases and larval body length were decreased. The teratogenic concentration($EC_{50}$) of Zn and Benomyl were 2.3, $1.0mg/{\ell}$, respectively and the embryo lethal concentration($LC_{50}$) Zn and Benomyl were 10.3, 6.9, respectively. The teratogenic index(TI) were 4.4 in Zn and 6.7 in Benomyl, thus showed teratogenicity in embryonic development of B. gargarizans. These results reveal that Zn and Benomyl in this experiment suppressed the development of embryos at relatively low concentration. Much of B. gargarizans embryos can be secured, and easy to incubate. In addition, mortality, malformation ratios, malformation patterns and growth rates are similar to the results from the other assay systems. Therefore, the B. gargarizans embryo teratogenesis assay system could be a useful tool to evaluate toxicity of pollutants in environment.
Duck viral hepatitis is an acutic, highly infectious viral disease of young ducklings. The most practical means for controlling duck viral hepatitis is the vaccination of ducklings or of a breeding stock. We attempted to develop a vaccine strain of duck hepatitis virus (DHV) using a Korean isolate by serial chicken embryo passages. The propagation of DHV in chicken embryos was carried 140 passages. After the $50^{th}$ passage, of which the virus was non-pathogenic for ducklings, approximately every $20^{th}$ passage of the virus was tested for vaccinal efficacy. Both the $70^{th}$ and $90^{th}$ passage of the virus gave good protection against challenge infection to a DHV-DRL reference strain(type 1) and a virulent Korean isolate. The $110^{th}$, $125^{th}$ and $140^{th}$ passage of the virus were less protective than the $70^{th}$ and $90^{th}$ passage, which means that more than $110^{th}$ passage may lead to over-attenuation of the virus. Ducklings vaccinated with the chicken-embryo-adapted virus by oral, intramuscular or eye drop administration showed earlier resistance to challenge infection from 3 to 7 days postvaccination. Of the above methods, ducklings vaccinated intramuscularly presented the most rapid resistance against challenge. The minimum immune dose of the chicken-embryo-adapted virus in ducklings was also studied. Ducklings inoculated with a dose of $10^{2.0}\;ELD_{50}$ and below were not fully protected against challenge with a virulent DHV, showing a protection rate of 67% to 73%, but ducklings inoculated with a dose of $10^{3.0}\;ELD_{50}$ and over were completely protected. The virus yield of the chicken-embryo-adapted DHV was examined at 24hrs and 48hrs of the incubation time in the allantoic fluid, embryo head and embryo minus head of the embryonating egg. In all three components, the titer of the virus was higher at 48 hours than that at 24 hours after incubation. And the titer of the virus was higher in the embryo minus head, embryo head and the allantoic fluid, in order. Field trials for evaluating the efficacy of the attenuated DHV as a live vaccine were done in duck farms with about 25% mortality of flocks resulting from duck viral hepatitis. After the use of the experimental vaccine, the mortality due to duck viral hepatitis was dramatically reduced in the farms. These results indicated that the attenuated DHV using a Korean isolate could be a good candidate as a live vaccine strain of DHV in Korea.
In this experiment, we investigated the toxicity of tebuconazole (fungicide) using domestic frog embryos, along the FETAX (Frog Embryo Teratogenesis Assay-Xenopus) protocol. Bufo gargarizans, Hyla japonica, and Pelophylax nigromaculatus embryos were incubated, and investigation of the tebuconazole effect was performed by the probit analysis. As a result, depending on the concentrations of tebuconazole, the mortality and malformation rates were increased and larval body length was decreased. The teratogenic concentrations (EC50) of tebuconazole were 34.4mg L-1, 10.6mg L-1, and 14.9mg L-1, respectively, and the embryo lethal concentrations(LC50) of tebuconazole were 74.7 mg L-1, 38.5 mg L-1, and 39.1 mg L-1, respectively. The teratogenic index (TI) valuesof tebuconazole were 2.19, 3.58, and 2.65; thus, it showed teratogenicity in embryonic development of these three frogs. These results revealed that in this experiment, tebuconazole suppressed the development of embryos at a relatively low concentration. In addition, mortality, malformation ratios, malformation patterns, and growth rates were similar to the results from the other assay systems. Therefore, tebuconazole was thought to have an effect on the embryo development of domestic frogs. In future, it will be necessary to identify species specificity in order to the clarify the causes of differences in mortality, malformation rate, and malformation patterns depending on the species.
Reproductive traits of Palaemon gravieri such as embryo size, number of embryo (fecundity), incubation period, larval development mode, larval development period, larval survival and larval growth were described and compared to analyze the correlation among those traits. Embryo volume is a primary factor determining other ensuing reproductive features. Egg volume was $0.042mm^3$ in the first developmental stage. Embryo volume in P. gravieri was comparatively small which is indicative of great number of embryo (y = 3.0161x + 0.0185 $R^2$ = 0.74 positive isometric relationship) and relatively long incubation period. Larvae survived from zoea 1 to post-larvae and it took 45 days at $22^{\circ}C$. Survival rate of the larvae was rather great in the early stage and thereafter steadily decreased. Daily growth rate of larvae in P. gravieri at $22^{\circ}C$ was 0.0195 mm on average. They grew steadily as time went by. Incubation period was between 10-14 days at $22^{\circ}C$. Larval development mode was almost complete planktotrophic. PNR (point of no return) appeared to be the third day on average. Survival rate of larvae without feeding declined rapidly between 3 and 4 days. Larval development period and stage frequency were 23-30 days and 11 stages which imply prolonged larval period and high mortality. The pattern of brood production followed fast successive parturial pattern. Most ovigerous female had mature ovary when they performed parturial molt soon after hatching (larval release).

We determined the risk factors for late embryonic mortality in dairy cows. We diagnosed pregnancy at 31 days and then confirmed the diagnosis at 45 days after artificial insemination (AI) via ultrasonography. The presence of an embryo with a heartbeat was the criterion for a positive pregnancy diagnosis. A diagnosis of late embryonic mortality was made when there was no positive sign of pregnancy in cows previously diagnosed as pregnant. The overall incidence of late embryonic mortality among 3,695 pregnancies was 6.9%. Logistic regression analysis revealed that herd size, AI month, synchronization protocol, and postpartum disease were important risk factors for late embryonic mortality. Herd size > 100 (odds ratio [OR]: 0.66, p < 0.05) and 50-100 lactating cows (OR: 0.63, p < 0.01) had lower risks of late embryonic mortality than herd size < 50 lactating cows. Cows inseminated during May-July had a higher risk (OR: 1.49, p < 0.05) of late embryonic mortality than cows inseminated during February-April. Cows inseminated after estrus following $PGF_{2{\alpha}}$ treatment also had a higher risk (OR: 1.77, p < 0.001) of late embryonic mortality than cows inseminated following natural estrus. Lastly, cows with postpartum disease tended to have a higher risk (OR: 1.26, p < 0.1) of late embryonic mortality than cows without postpartum disease. In conclusion, late embryonic mortality associated with the herd size, AI month, synchronization protocol, and postpartum disease in dairy cows.
Attempts to isolate chicken anemia agent (CAA) were made by inoculating tissue homogenates into MDCC-MSBl or LSCC-1104B1 cell lines and passaging the cells serially. CAA was isolated from the liver and thymus of 11 weeks old layer chickens and from the liver of 10 weeks old broiler breeder chickens. The layer flock experienced approximately 45% mortality during 9 to 14 week of age from gangrenous dermatitis and lymphoid organs of affected chickens were severely atrophied. The broiler breeder flock experienced approximately 7% mortality during 7 to 9 weeks of age and affected birds showed lesions of colibacillosis, staphylococcal arthritis, and coccidiosis together with atrophied lymphoid organs. The isolated viruses were identified as CAA by the indirect fluorescent antibody test and virus neutralization test using CAA immune sera including one to Gifu-1 strain of CAA. The CAA isolate 89-69, when inoculated into susceptible 1 day old SPF chicks, induced anemia 14 to 16 days after inoculation. It did not induce any cytopathic effects in chicken embryo liver and chicken embryo fibroblast cell cultures. Infectivity of the isolate was not affected by the treatment of chloroform or heat ($70^{\circ}C$ for 15 minutes).
Effect of cadmium on the early amphibian development was analyzed through FETAX (Frog Embryo Teratogenesis Assay: Xenopus). Embryos manifested concentration-dependent mortality and realformations; shortage of anterior-posterior axis gut realformation, ocular anomalies, bent notochord, misshapen dorsal fin, and derreal blisters. The treatment with 1.5ppm cadmium solution caused 100% mortality and concentration of lppm did not kill the embryos that caused 100% anomaly. The teratogenic index (TI = LC50 /EC50) was 2.8 indicating that $CdCl_2$ is teratogenic for Xenopus laevis. Embryos that were pulsetreated with at early to late blastula stage (St. 3-9) and mid to late blastula stage (St. 6-10) showed relatively strong resistance to cadmium, but the embryos treated at gastrula stage (St. 10-13) showed high mortality. And the embryos treated at tailbud stage (after St. 25) showed highest mortality of any other early stages. Effects of temperature were studied through pulse- treatment during gastrula stage at $20^{\circ}C$ and $30^{\circ}C$. The embryos treated with 7.5ppm at $30^{\circ}C$ and 15ppm at $20^{\circ}C$ caused 100% mortality respectively, indicating that higher temperature had more severe toxic effect. One of the most peculiar effect of cadmium at gastrulation was distortion of the tail. The probable cause of toxic effect of Cd was discussed.
