Correlation between Deoxycytidineuria and CdR-aminohydrolase Activity following X-Irradiation (X線照射에 따르는 Deoxycytidineuria와 CdR-aminohydrolase의 活性變化와의 連關性)

  • This work was undertaken to elucidate some aspects of mechanisms underlying the increased deoxycytidineuria following irradiation in the mice, by observing Dische-positive substances liberated from thissues, the activity of CdR-aminohydrolase of tissues and the CdR excreted in the urine at various times after single whole-body exposure to 400 and 800 R of X-rays. The activity of CdR-aminohydrolase declined markedly at 1 hour in the small intestine and liver, followed by a gradual rise reaching a maximum at 3 days after irradiation. In the case of the spleen and blood, however, only a trace of activity was observed in the control and irradiated animals. The amount of Dische-positive substance liberated from the small intestine postirradiation was elevated from 3 to 12 hours, showing a maximum during 6 to 9 hours after irradiation. On the contrary, the activity of the enzyme in the liver, spleen and kidney was less than one twentieth that of the small intestine, suggesting a prediction that these organs are not attributable to the increased deoxycytidineuria. A maximum deoxycytidineuria was exhibited at 9-12 hours period, attributed a large amount of CdR to the small intestine, which might correlate with the change in the CdR-aminohydrolase activity. Radiation-induced CdR seems to be liberated from the small intestine into the blood when the CdR-aminohydrolase activity declines abruptly. Then, the CdR is rapidly subjected to a filtration in the kidney without undergoing a further degradation pathway in the blood.

  • Bone remodeling is a complex process regulated by various mediators. Cytokines are known to be associated with the mechanically induced response in orthodontic tooth movement. In particular, IL-$1\beta$ stimulates bone resorption and induces osteoclast proliferation. The purpose of this study was to identify and quantify IL-$1\beta$ in human gingival crevicular fluid(GCF), and to investigate the changes in its level during orthodontic tooth movement. Twelve patients(mean age of 19.2 years) were used as the subjects. An upper canine of each patient haying treatment lot distal movements served as the experimental tooth, whereas the contralateral was used as the control. The GCF of compression and tension side of the experimental teeth and the GCF of mesial side of control teeth was taken from the each subject immediately before activation, and at 1, 24, and 168 hr after initiation tooth movement. IL-$1\beta$ amount was detected by ELISA. The concentration of IL-$1\beta$ was higher in experimental group than in the control group after treatment. Its level was elevated after initiation of tooth movement and it was the highest level at 24 hr in compression side of experimental group. But there was no significant change in control group. The results indicate that the change in IL-$1\beta$ level in GCF is associated with orthodontic tooth movement.

$TNF{\alpha}$ Increases the Expression of ${\beta}2$ Adrenergic Receptors in Osteoblasts

  • Tumor necrosis factor alpha ($TNF{\alpha}$) is a multifunctional cytokine that is elevated in inflammatory diseases such as atherosclerosis, diabetes and rheumatoid arthritis. Recent evidence has suggested that ${\beta}2$ adrenergic receptor (${\beta}2AR$) activation in osteoblasts suppresses osteogenic activity. In the present study, we explored whether $TNF{\alpha}$ modulates ${\beta}AR$ expression in osteoblastic cells and whether this regulation is associated with the inhibition of osteoblast differentiation by $TNF{\alpha}$. In the experiments, we used C2C12 cells, MC3T3-E1 cells and primary cultured mouse bone marrow stromal cells. Among the three subtypes of ${\beta}AR$, ${\beta}2$ and ${\beta}3AR$ were found in our analysis to be upregulated by $TNF{\alpha}$. Moreover, isoproterenol-induced cAMP production was observed to be significantly enhanced in $TNF{\alpha}$-primed C2C12 cells, indicating that $TNF{\alpha}$ enhances ${\beta}2AR$ signaling in osteoblasts. $TNF{\alpha}$ was further found in C2C12 cells to suppress bone morphogenetic protein 2-induced alkaline phosphatase (ALP) activity and the expression of osteogenic marker genes including Runx2, ALP and osteocalcin. Propranolol, a ${\beta}2AR$ antagonist, attenuated this $TNF{\alpha}$ suppression of osteogenic differentiation. $TNF{\alpha}$ increased the expression of receptor activator of NF-${\kappa}B$ ligand (RANKL), an essential osteoclastogenic factor, in C2C12 cells which was again blocked by propranolol. In summary, our data show that $TNF{\alpha}$ increases ${\beta}2AR$ expression in osteoblasts and that a blockade of ${\beta}2AR$ attenuates the suppression of osteogenic differentiation and stimulation of RANKL expression by $TNF{\alpha}$. These findings imply that a crosstalk between $TNF{\alpha}$ and ${\beta}2AR$ signaling pathways might occur in osteoblasts to modulate their function.

Protective Effect of Propofol against Hypoxia-reoxygenation Injury in HaCaT Human Keratinocytes

  • The aim of this study was to determine the beneficial effect of propofol on human keratinocytes that have undergone hypoxia reoxygenation (H/R) injury and to investigate whether autophagy is associated with the protective mechanism. Thus, we evaluated how propofol influences the intracellular autophagy and apoptosis during the H/R process in the HaCaT cells. The cultured human keratinocyte cells were exposed to 24 h of hypoxia (5% $CO_2$, 1% $O_2$, 94% $N_2$) followed by 12 h of reoxygenation (5% $CO_2$, 21% $O_2$, 74% $N_2$). The experiment was divided into 4 groups: (1) Control=Normoxia ; (2) H/R=Hypoxia Reoxygenation ; (3) PPC+H/R=Propofol Preconditioning+Hypoxia Reoxygenation; (4) 3-MA+PPC+ H/R=3-MA-Methyladenine+Propofol Preconditioning+ Hypoxia Reoxygenation. In addition, Western blot analysis was performed to identify the expression of apoptotic pathway parameters, including Bcl-2, Bax, and caspase 3 involved in mitochondrial-dependent pathway. Autophagy was determined by fluorescence microscopy, MDC staining, AO staining, and western blot. The H/R produced dramatic injuries in keratinocyte cells. In our study, the viability of Propofol in H/R induced HaCaT cells was first studied by MTT assay. The treatment with 25, 50, and $100{\mu}M$ Propofol in H/R induced HaCaT cells enhanced cell viability in a dose-dependent manner and $100{\mu}M$ was the most effective dose. The Atg5, Becline-1, LC3-II, and p62 were elevated in PPC group cells, but H/R-induced group showed significant reduction in HaCaT cells. The Atg5 were increased when autophagy was induced by Propofol, and they were decreased when autophagy was suppressed by 3-MA. These data provided evidence that propofol preconditioning induced autophagy and reduced apoptotic cell death in an H/R model of HaCaT cells, which was in agreement with autophagy playing a very important role in cell protection.

Anti-inflammatory effect of seed oil of Schisandra chinensis in the LPS-treated RAW 264.7 macrophages (LPS로 자극된 Raw 264.7 대식세포에서 오미자 씨앗오일의 항염증 효과)

  • Objectives : This study was designed to investigate of the anti-inflammatory effects of Schisandra chinensis seed oil(SSO) on the production of pro-inflammatory substances in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages.Methods : SSO was measured the production of pro-inflammatory factor (NO, PGE2, IL-1β iNOS and, COX-2) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. we used the following methods : cell viability assay, Griess reagent assay, enzyme-linked immunosorbent assay, Western blotting analysis.Results : The cell viability of SSO(0∼500 μl/mL) processing group was 96.9% and the processing of SSO didn't have an effect on the cytotoxicity. The inhibitory effect of the nitric oxide (no) production of SSO(500 μg/mL, 50 μg/mL, 10 μg/mL) was each 70.3%, 37.6% and 26.5%. IL-1β production inhibition ability of SSO(500 μg/mL, 100 μg/mL) was each 49.88% and 48.8%. PGE2 production inhibition ability of SSO(500 μg/mL, 100 μg/mL) was each 49.88% and 73.1%, 70.5%. By using SSO, it experimented about iNOS protein expression inhibition ability, that is the NO production enzyme. iNOS protein expression increased in the group processing LPS independently. iNOS protein expression decreased in the group processing SSO together. The expression of the COX-2 protein decreased 89.6%, 81.8% in the group processing SSO. The significance was in the relationship with NO formation inhibition with the relationship with the PGE2 formation inhibition and iNOS protein, it confirmed in SSO with the COX-2 protein.Conclusions : Stimulation of the RAW 264.7 cells with LPS caused an elevated production of nitric oxide (NO), IL-1β and PGE2 which was markedly inhibited by the pretreatment with SSO without causing any cytotoxic effects. The reduced expressions of iNOS protein were consistent with the reductions in NO production in the culture media. SSO may be useful for the treatment of various inflammatory diseases.

Seasonal Ground Water Table Changes Following Forest Harvesting in Small Headwater Riparian Areas (산지계류 수변지역에서 산림벌채 후 지하수위의 계절 변화)

  • This study addressed the influence of forest harvesting on seasonal water table dynamics in small headwater riparian areas. Four treatments including potential Best Management Practices(BMPs) for ephemeral and intermittent streams were implemented(BMP1, BMP2, clearcut and reference). Water table measurements were obtained at bi-monthly intervals for 3 years including one year of pre- and two years of post-harvest observations. Overall, water table responses affected largely by rainfall amount. In addition, significant increases in water table levels following harvesting occurred throughout the two post-harvest years. Water table levels increased up to 28.2cm in the clearcut treatment during 2008 and up to 54.2cm in BMP2 during 2009. However, increase in water table elevation was not directly related to basal area removal despite considerable differences in basal area removed between BMP2 and clearcut treatments. Water table rises were apparent in that water table were more elevated during dry season(June through November) than during wet season(December through May). These seasonal fluctuations were presumably driven by changes in evapotranspiration caused by differences in leaf area of overstory canopy and understory following harvest.

A Study on the Literature Therapy Using Sijo (시조를 통한 문학치료 연구)

  • This technical research paper attempts to examine literature therapeutic significance of Sijo. This research analyzes how the therapeutic mechanisms of literature or sentence can be physiologically applied to human nerve system. To study such interactions between therapeutic mechanisms of literature and human physiological mechanisms, diverse on- and offline Sijo activities, such as a Facebook group "Sijoya Nolja", were studied, as well as various publications in literature and medicine. For the three years this research was being conducted, 27 out of 488 Sijo poets debuted through Sinchunmunye, and many have gone professional through various contests. 19 of them became members of Sijo Poet Associations of Korea, with globalization of Sijo in mind. Their aggressive activity proves that the quality of live was elevated through literature therapy. The therapeutic effects are especially dramatic for older authors, and authors with traumatic experience. Through this research, Sijo turned out to be a form of literature with the quickest effect on elevating quality of life. To solve problems such as depression, emotional isolation, and other mental conditions, literature therapy through Sijo should be implemented, and therapeutic approach of literature should be more professionalized.

Effect of Soil Salinity and Soil-wetting by Summer-Rising of Water Table on the Growth of Fruit Trees Transplanted at the Saemangeum Reclaimed Tidal Land in Korea (새만금간척지의 토양염농도와 지하수위의 하계 상승이 이식한 과수의 생육에 미치는 영향)

  • The effect of soil salinity and soil-wetting by rise of water table on the growth of fruit trees was studied to obtain information for orchard establishment in the Saemangeum reclaimed tidal land. Survival ratio of trees was 85%for grape, 31%for fig, 15%for apple and pear, and near zero for peach and blueberry. Wet injury induced by water-logged or flooded condition, rather than salt injury(soil EC was lower than 3.0dS $m^{-1}$ during growing period) is thought to be more responsible for low survival ratio of fruit trees transplanted in Saemangeum area. During the summer raining season in the reclaimed areas, the soil salinity tends to be decreased by natural rainfall effect, and the rainfall acceptable capacity(RAC) of soils dramatically is reduced(10-24 mm) as rainfall is continued to occur. In spite of high hydraulic conductivity(121 cm $day^{-1}$) of soils across the area, low RAC of soils might be due to high soil saturation and elevated water table during summer raining season. Therefore, the installation of effective drainage system should be the primary factor determining successful establishment of orchard in the Saemangeum reclaimed tidal land.

Relationship between Nutrients Intakes, Dietary Quality, and Serum Concentrations of Inflammatory Markers in Metabolic Syndrome Patients (대사증후군 환자의 영양소 섭취상태 및 식사의 질과 염증지표 농도의 상관성)

  • Elevated serum concentration of inflammation markers is known as an independent risk factor of metabolic syndrome (MS) and dietary intake is an important factor to control MS. The purpose of this study was to investigated the hypothesis that inflammatory indices are associated with dietary intake and diet quality index-international (DQI-I) in subjects with MS. A cross-sectional study was conducted on 156 men and 73 postmenopausal women with MS, defined by three or more risk factors of the modified Adult Treatment Panel III criteria. Serum levels of high sensitive C-reactive protein (hs-CRP), adiponectin were examined and nutrients intake and DQI-I were assessed using a semiquantitative food frequency questionnaire. The total DQI-I score was significantly higher in female subjects ($65.87{\pm}9.86$) than in male subjects ($62.60{\pm}8.95$). There was a positive association between hs-CRP and polyunsaturated fatty acid intake (p < 0.05) and a negative association between adiponectin and lipid (p < 0.05), total sugar (p < 0.01), and total fatty acids (p < 0.05). When the subjects were divided into 5 groups by quintile according to serum adiponectin and hs-CRP level, there was no association between DQI-I score and hs-CRP levels. Moderation score of DQI-I was significantly higher in highest quintile group than the lower quintile groups. Therefore, our results provide some evidence that dietary intake and diet quality are associated with inflammation markers and dietary modification might be a predictor to decrease risk for metabolic syndrome complications. However further research is needed to develop the dietary quality index reflecting the inflammatory change by considering the dietary habit and pattern of Koreans.

Study on Temperature Dependence of Molecular Structure in Stearic Acid LB Films Using FTIR-RAS (FTIR-RA 분광법을 이용한 스테아르산 단분자막에서 분자구조의 온도의존성 고찰)

  • 1-, 3-, 9-, and 21-Monolayer Langmuir-Blodgett(LB) films of stearic acid were deposited on silver-coated glass slides at the surface pressure of 30 mN/m. Fourier transform infrared(FTIR) reflection-absorption spectra (RAS) of these LB films were recorded at various temperatures from 31 to $72^{\circ}C.$ The spectra at $31^{\circ}C$ exhibited characteristic features of highly perpendicular orientation of the hydrocarbon chain. In the 1-monolayer LB film, the C=O stretching band was not observed, presumably due to the image dipole effect on the silver surface. In the 1-and 3-monolayer LB films, the trans isomer of stearic acid was prominent, but the cis isomer was dominant in the 21-monolayer LB film. FTIR-RAS measurements at an elevated temperature indicated that the chain melting temperature increases and approached to the bulk melting point with increasing the number of monolayer, except for the 1-monolayer LB film which has a higher melting temperature than the 3-monolayer film due to the strong interaction with the silver substrate.
