Superoxide Dismutase Profiles in the Mesophilic Deinococcus Species

  • Electrophoretic resolution of superoxide dismutase (SOD) from the highly UV-resistant bacteria, Deinococcus species revealed multiple forms of superoxide dismutases (SODs) in D. radiodurans, D. grandis, and D. proteolyticus, as judged from electrophoretic properties and metal cofactors. A single SOD occurred in both D. radiophilus and D. radiopugnans. Deinococcal SODs were either MnSOD, FeSOD or cambialistic Mn/FeSOD. The unique SOD profile of each mesophilic Deinococcus species, multiplicity and metal cofactors would be valuable in identifying Deinococcus species.

Iso-catalase Profiles of Deinococcus spp.

  • The obligate aerobic Deinococcus are highly resistant against lethal effect of UV-and ionizing-radiation. Only five mesophilic Deinococcus species, i. e. D. radiodurans, D. radiophilus, D. proteolyticus, D. radiopugnans, and D. grandis are known. Since an indispensable role of catalase has been suggested in protecting cells against oxidative stress and UV radiation, Deinococcal catalase activity of each species and electrophoretic profiles of catalases were investigated on gel. Total catalase activity was varied among the species in the aerobically grown culture at stationary phase. The occurrence of multiple forms of catalases with different molecular weights in four species of Deinococcus and of a single catalase in D. radiopugnans suggests that each species shows the unique catalase profiles on gel. Some Deinococcal catalases also exhibit peroxidase activity. Since Deinococcus spp. are less-distinct to each other in their morphology, biochemical and physiological properties, the catalase profiles on PAGE would be useful in identifying the species of Deinococcus.

Fusarium속에서 PFGE를 이용한 Electrophoretic Karyotyping (Electrophoretic Karyotyping by PFGE in the Genus Fusarium)

  • CHEF (Contour-Clamped homogeneous electric field) gel electrophoresis를 이용하여 Fusarium section Sporotrichiella, Liseola, Gibbosum, Discolor와 Martiella에 속하는 10종의 electrophoretic karyotype을 비교하였다. Intact chromosomal DNA는 균류의 원형질체로부터 추출하였으며, 크기에 따라 다양한 조건을 주어 DNA 분자를 분리시켰다. Fusarium속에 속하는 종의 염색체는 0.78Mb에서 7.20Mb의 크기를 가진 염색체가 종에 따라 $5{\sim}13$개였다. 각 종의 total genome 크기는 18.32Mb에서 48.20Mb였다. Electrophoretic karyotype을 비교한 후 F. oxysporum formae speciales lilii로부터 무작위로 선택하여 만든 genomic DNA를 probe로 하여 Southern hybridization 분석을 수행하였다.

A New Strategy for Determining Optimum pH of Isozymes

  • A hydrogenperoxide sensor containing peroxidase extracted from horseradish was constructed and pH effect on its sensing ability was investigated. Current profiles of the biosensor with pH and the electrophoretic analysis showed that horseradish peroxidase consists of two isozymes. Assuming that it is a hypothetical twoisozyme mixture, the current profiles were deconvoluted into two Gaussians. Application of the new Michaelis-Menten equation connoting pH concept to this system enabled to find all the related dissociation constants of the isozyme-substrates and the isozyme-proton complexes and to determine pHs for the maximal isozyme activities.

  • A carbon paste electrode was constructed with peroxidase extracted from Horseradish and the variation of the response of the sensor with pH was investigated. Current profiles showed two highest sensitivities at two pH values respectively. In addition, two bands were observed in the electrophoretic expansion. A coincidence of the two experimental results added support to the possibility that the biosensor has two different isozymes. Assuming that current profiles are the sum of two gaussians, we deconvoluted them and determined the optimum pH of peroxidase isozymes.

  • Doping effects and semiconductor behaviors of the dispersed p- and n-Si, p- and n- GaAs particles in the aqueous electrolyte have been studied using microelectrophoretic, voltammetric and chronoamperometric techniques. The cations (K$^{+}$) are adsorbed on both the p- and n- Si particle surfaces regardless of the sign of space charges in the depletion layers, i.e. doping profiles. The surface states are negatively charged acceptor states. On the other hand, the anions (CI$^{-}$) are adsorbed on both the p- and n- GaAs particle surfaces regardless of the sign of space charges in the depletion layers, i.e. doping profiles. The surface states are positively charged donor states. Under the same conditions, electrophoretic mobilities, electrochemical processes, doping effects and related semiconductor behaviors of the Si and the GaAs particles are similar regardless of the doping profiles, i. e. dopants and doping concentrations. The doping effects and related semiconductor behaviors of the dispersed p- and n- type semiconductor particles are gradually lost with decreasing dimensions.

경주 적혈구막 단백의 전기영동법에 의한 분석 -빠른 적혈구 침강 속도와의 관계- (Electrophoretic analysis of the major proteins of race horse erythrocyte membrane: Their relation to fast erythrocyte sedimentation rate)

  • The proteins of the race horse erythrocyte membrane were analysed by polyacrylamide gel electrophoresis in sodium dodecyl sulfate(SDS-PAGE), and their relations to the fast erythrocyte sedimentation rate(ESR) of the race horse were investigated. The erythrocyte sedimentation rate of race horse were very fast compared with the human one(33 times <$90^{\circ}-plastic-ESR/30m$> and 25 times <$90^{\circ}-micro-ESR/30m$> as fast as the human one) are reported previously. Although the general protein profiles of the race horse erythrocyte membranes were almost similar to that of human, band 3 content was showing higher in race horse (34.7%) than in human (25.3%). The glycoprotein profiles of the race horse erythrocyte membranes revealed by periodic acid Schiff's(PAS) stain showed a marked difference from that of human. The PAS-1(glycophorin) and PAS-2(sialoglycoprotein) present in human erythrocyte memo brane were almost absent from the Holstein and race horse erythrocyte membranes, but PAS-2 was more in only race horse from that of human. Instead, the bovine erythrocyte membranes showed a strong PAS-B near the origin of the electrophorograms and the race horse erythrocyte membranes showed a strong PAS-negative band near the end of the electrophorograms, which is named as PAS-E in this study. These results suggest that the fast sedimentation rate of race horse erythrocyte is due in part to the presence of more band 3 protein fraction and PAS-E glycoproteins in the race horse erythrocytes.
