Studies on the Electrophoretic Variation in Tetrazolium Oxidase Isozyme of Nicotiana Species (Nicotiana속에 있어서 Tetrazolium Oxidase Isozyme의 전기영동적 변이에 관한 연구)

  • Four kinds of Nicotiana species and five varieties belonging to N. tabacum were used as materials for electrophoretic analysis of the tetrazolium oxidase isozyme to examine the taxonomic affinity among them based on the biochemical property. All the five verieties of N. tabacum showed same isozyme bands despite the fact that these varieties had notably varied characteristics including morphological traits. The band patterns were more or less different among the four species. Although N. rustica and N. tabacum were of the same genome group of AABB, their isozyme bands showed considerable difference. N. sylvestris, genome A donor of Nicotiana species, was found to be markedly different from N. tatacum in band pattern, including the absence of system 2 in N. sylvestris.

  • These researches were carried out to investigate the morphology of different species of Trichoderma and the possibilities of differentiation of the species of Trichoderma by electrophoretic methods. Variations between the isolates of a species of Trichoderma indicate the genetical differences, also isozyme and protein patterns will be useful to investigate genetical variations betweens the isolates. It might be possible that distinct bands of isozymes of esterase, phosphotase, catalase, catalase differentiate species of Trichoderma.

  • The electrophoretic banding patterns of 6-phosphogluconate dehydrogenase (PGD) in the two different chromosomal ploidy groups of Gyraulus were compared. The monomeric or dimeric banding patterns or allelic variations in a locus of PGD were observed in four diploid populations (Osan, Sohre, Kimpo and Kangwha) of G. convexiusculus occurring in Korea, whereas the isozyme banding patterns encoded by at least 3 different loci were shown in the tetraploid populations of G. (Torquis) groups collected from Michigan, the U.S.A. Of 3 different tetraploid groups, G. (T.) circumstriatus group showed 3 monomorphic isozyme banding patterns, and the other 2 populations showed some allelic variations. Such results provided good evidence to differentiate tetraploid subgenus Torquis group from the diploid Gyraulus populations.

Electrophoresis of Reptilian Blood Proteins (파충류 혈액 단백질의 전기영동)

  • The blood proteins of ten reptilian species were studied by cellulose acetate elctrophoresis. Three members examined of the genus Agkistrodon have unusually similar patterns in plasma protein, hemoglobin, lactate dehydrogenase and malate dehydrogenase. On the basis of their electrophoretic patterns, it was concluded that Agkistrodon blomhoffii brevicaudus was closely related to the others. In general the plasma protein patterns reflect species specificity. Under the conditions employed, all snakes had a single hemoglobin band except Dinodon rufozonatum rufozonatum which showing two component patterns. Two members of the Chelonia showed four bands of hemoglobin. The zymograms indicated a distinct divergence in blood proteins of the Squamata from those of the Chelonia. The results reflected superficially the established phylogenies of these groups.

  • The purpose of this study was to analyze the electrophoretic patterns of soluble proteins from ginseng roots and to compare the protein patterns from Korean ginseng and American quinquefolium. The size difference was found in the major protein bands of a molecular weight of about 27,000 between Korean ginseng and American quinquefolium. The protein band of a molecular weight of 22,000 showed a quantitative difference in its amount. The major 27 K proteins appeared to form a complex heterodimer of 66,000 and to have internal bisulfide bonds, from band shifting studies under non-denaturing conditions. Three peaks appeared when the protein extract from root homogenates was purified using gel filtration and DEAE ion exchange chromatography. The examination of physiological activity and further purification of these fractions are underway.

  • The concentrations of serum proteins fractions and their electrophoretical patterns were investigated in 135 patients with coal workers' pneumoconiosis who participated in confirmative examination for pneumoconiosis in December 1989. Their radiographical profusions were classified as 1/0 or more. Agarose film and phosphoric acid-sodium hydroxide buffer(pH 9.6) were used for electrophoresis. Concentration of each protein fractions and electrophoretical patterns seemd to be equivalent to reference values. Serum ${\alpha}_1$- and $\beta$-globulin concentrations, however, were significantly different (p<0.50) among categories of small opacity profusions and showed the lowest level in the group of category 1. Albumin concentratins decreased and ${\alpha}_2$- globulin concentrations increased significantly (p<0.05) in the group of complicated with pulmonary tuberculosis. $\gamma$-globulin concentrations were not varied by category of profusions nor by pulmonary tuberculosis complication.

  • Taxonomic relations among three species of some edible fleshy fungi(Lentinus edodes, Pleurotus ostreatus, Flammulina velutipes) in the family Tricholomataceae were examined by using polyacrylamied gel disc electrophoresis. The soluble, crude extract of mycelium grown on potato sucrose broth was subjected to electrophoresis. Similarities in the protein bands for each isolated of one species were compared with those for others. In the banding patterns there was a closer relationship between isolates within one species than among isolates of different species. However, the isozyme patterns obtained from each isolate of Peurotus ostreatus (esterase, peroxidase, tyrosinase) were appeared to represent the degree of geographical variability within one species.

  • In order to evaluate the applicability of enzyme electrophoresis for the identification of intra/interspecific hybride obtained by the protoplast fusion in Trichoderma, soluble proteins, intracellular soluble enzymes and extracellular $\beta$-glucosidase were analyzed by polyacrylamide gel electrophorsis. As the results, patterns of soluble protein, and isozyme patterns of peroxidase, malate dehydrogenase, and $\beta$-glucosidase in hydrids were defferent from those in parental and wild type strains. Therefore, it was established that the analysis of protein pattern by electrophoresis could be applied to isolate and identify the hybrids from the protoplast fusion.

  • To determine the prevalence of bovine rotavirus Infection in Chungbuk area, fecal specimen were collected from calves nth diarrhea and tested using ELISA. The positive rates were 53.8%(1 to 30days old), 19.0%(31 to 60days old), and 3.2%(over 60days old). Electrophoretic migration patterns of genomic RNA from field isolates were similar to that of NCDV strain, prototype of bovine rotavirus. Bovine rotavirus field isolate showed characteristic morphology of rotavirus particle with 80nm in diameter, using EM.

  • Strains of Fusarium oxysporum f. sp. lycopersici isolated from Korea, Japan and U.S.A. were used for electrophoretic karyotype (EK) analysis. Chromosome separations on FastLane agarose gels (FMC BioProducts, Rockland, ME), called pulsed field gel electrophoresis (PFGE), were performed by CHEF-DRII apparatus (Bio-Rad Laboratories, Melville, NY) using TAE as a running buffer. To obtain optimal condition for separation of chromosome sized DNAs, variable running conditions such as field strengths, swithching intervals, and running time were applied in CHEF gel electrophoresis. We were able to resolve 9 to 11 chromosome sized DNAs ranging in size from 0.76 to 6.41 Mb in isolates from Korea and estimate that the total genome size was ranging from 35.29 to 38.92 Mb. Distinct differences in length range and genome size exist among isolates from different countries. Isolates from Japan and U.S.A. were resolved 9 to 11 chromosome sized DNAs ranging in size from 1.24 to 6.85 Mb and estimated that the total genome size was ranging from 35.32 to 43.87 Mb. Isolates from variable provinces in Korea had the same or similar chromosomal polymorphism and showed different chromosomal DNA patterns compared to isolates from the other countries.
