Spawning Period Characteristics and Early Life History of the Eight Barbel Loach, Lefua costata (Pisces: Balitoridae) (쌀미꾸리(Lefua costata)의 산란기 특징 및 초기생활사)

  • This study was conducted to elucidate spawning period characteristics and early life development of eight barbel loach, Lefua costata (Balitoridae) at the Jusucheon stream, Okgye-myeon, Gangneung-si, Gangwon-do, Korea between January and December in 2018. The spawning period was estimated to be from May to August, considering the change in the gonad-somatic index, the appearance of young fry, and frequency distribution in egg diameter. It was a multi-spawning type. The gender ratio was 1:0.79 with 1,117 females and 879 males collected. The egg size was 0.24-0.93 mm, with mature and immature eggs found during the spawning period. The size of mature eggs was 0.71±0.02 mm, and the average number of fecundity was 1,786±818 (n=31). Observation of the egg development showed that the fertilized eggs were the sticky, gray, segregated, and demersal type with 0.76±0.03 mm in diameter. The hatching of the embryos began at about 34 hours (hatching rate 50%) after fertilization underwater temperature of 25℃. The average length of the newly hatched pre-larvae was 2.7±0.11 mm. The average length of pre-larvae at 4 days after hatching was 4.5±0.16 mm, and the yolk sac was completely absorbed and entered the post-larvae stage. At 20 days after hatching, the average length of post-larvae was 11.5±0.67 mm, and their fin rays were formed before they transitioned to the juvenile stage. At 100 days after hatching, the average length reached 49.8±2.60 mm, and the appearance and the lateral sideband patterns were similar to those of the adult fish.

  • The ripe unfertilized eggs of Acheilognathus signifer were elliptical shape and baggily swollen toward animal pole. The size of fertilized eggs were $2.19{\pm}0.01$ mm in long axis and $1.85{\pm}0.08$ mm in short axis and oil globe were not observed. The zona radiata of A. signifer had the structure of regular fibrilar net and the diameter of outer opening micropyle was about $2.5{\mu}m$ and that of inner opening micropyle was about $1.5\sim2.0{\mu}m$. Development of fertilized eggs in A. signifer was proceeded in inclining position of animal pole and hatched in 49 hours under $20{\pm}1^{\circ}C$. The yolk sac was absolutely absorbed in 21 days after hatching.

  • This study was carried out to investigate feeding of bacteria by Tigripus japonicus (Copepoda : Harpacticoida) under axenic culture. The ovigerous females and nauplii were grown with feed of aquatic bacteria. Growth of RT bacteria strain was suppressed by feeding of co-existing T. japonicus. T. japonicus of non-axenic culture was observed with oil bead in the egg sac. On the other hand, early nhauplius stage did not develop to the next stage without stage took bacteria as food. And the adult of T. japonicus may utilize the baxteria as nutrient source for egg development.

Spawning Behavior and Morphological Development of Larvae and Juvenile of the Nake-Headed Goby, Favonigobius gymnauchen (Bleeker) (날개망둑 (Favonigobius gymnauchen)의 산란습성, 난 및 자치어의 형태 발달)

  • The eggs of Favonigobius gymnauchen attached on the under side of a small stone were collected off Seongsan-eup Cheju-do in August, 2000 to investigate their development of egg, larvae and juveniles. The fertilized eggs were elliptical in shape (mean long axis: 1.50 mm; mean short axis: 0.57 mm) and transparent. There were filaments on one side of the egg membrane. Larvae hatched at 48 hrs 50 mins after morula stage with 25-26 myotomes in $22.8-28.5^{\circ}C\;(mean\;24.7^{\circ}C).$ The newly hatched larvae were 2.31-2.49 mm (mean 2.37 mm n=10) in total length (TL) and their mouth and anus were already opened. Their melanophores were appeared on the over gas globule, around anus and the part of caudal peduncle with 24-25 myotomes. At 4-5 days after hatching. larvae attained 3.81-4.07 mm (mean 3.96 mm, n=10) in TL and their yolk sac was completely absorbed. They began to eat rotifer and transformed to postlarvae stage. At 14 days after hatching, postlarvae attained 6.17-6.31 mm (mean 6.21 mm, n=10) in TL and their caudal notocord was flexed $45^{\circ}$ upward. At 24 days after hatching, postlarvae attained 8.69-9.10 mm (mean 8.87 mm, n=10) in TL had reached the juvenile stage. All fins were formed with the complete set of fin rays with the following counts: dorsal fin rays IV-I, 9-10; anal fin rays I, 9; pectoral fin rays 17; ventral fin rays: I, 5; caudal fin rays: 9+8= 17.

Egg Development and Morphology of Larva and Juvenile of Liparis tanakae in the Coastal Waters off Yeosu (여수 연안산 꼼치(Liparis tanakae)의 난발생 및 자치어 형태발달)

  • This study aims to contribute to the research on resource recovery for the rapidly declining population of Liparis tanakae by observing the larval development process and the morphology of juveniles based on their growth. Natural spawning eggs collected in Yeosu were used for observing the process of egg development and larval morphology. The water temperature during the rearing process was maintained at 12.3~13.5℃ (average 12.7℃). The fertilized eggs had an egg diameter ranging from 1.57 to 1.79 mm (average 1.71 mm) and were spherical and adhesive. Within 4 hours 35 minutes after fertilization, they reached the two-cell stage, and after 74 hours 10 minutes, the formation of the yolk sac began. At 106 hours post-fertilization, a caudal fin appeared at the tail tip. Hatching began at 526 hours, and the larvae developed with the yolk sac positioned just behind the eyes. The newly hatched larvae had both the mouth and anus open. Melanophores appeared inside the lower jaw and around the tail on the third day after hatching. By the 16th day after hatching, most of the yolk was absorbed, and melanophores were visible in the head region. Finally, on the 63rd day after hatching, the head region significantly developed, and the body shape and mouth were similar to those of an adult fish, signifying the transition to the juvenile stage. This study will serve as valuable data for aquaculture techniques related to the conservation and restoration of fish species based on the hatching and juvenile morphology of Liparis tanakae.

Diethylnitrosamine Induced Tissue Damage and Change of Lipid Components in the Chick Embryo Liver (Diethylnitrosamine에 의한 계배 간 조직 손상 및 지질 성분의 변화)

  • Diethylnitrosamine (DEN) is known as a potential hepatic carcinogen by single administration. This study was designed to measure the effects of DEN-induced cell damage on the triglyceride and cholesterol concentration in the liver, excluding dietary effects. Fertilized chicken eggs, 10 days before hatching, were randomly divided into three groups (n=20) and each egg was injected 10 ${mu}ell$ of corn oil (vehicle control), 5 $\mu\textrm{g}$ of DEN/10 ${mu}ell$ of DEN/10 ${mu}ell$ into yolk via air sac. After 48 hr and 96 hr incubation, the damage of the chick-embryo liver cell was investigated by electron microscopy and by measuring the concentration of lipid components (total cholesterol, free cholesterol, phospholipid and triglyceride). For eggs administered 10 $\mu\textrm{g}$ of DEN and incuvated 96 hr, in hepatocyte, the nucleus membrane was roughed, the size of nucleolus was apparently increased and euchromatin was accumulated. Mitochondria were condensed and cristae, located mitochondiral inner membrane, were obscured. Additionally, the leaves of triglyceride and cholesterol classes were significantly increased depend on the amount treated with 10 $\mu\textrm{g}$ DEN at 96 hr, but phospholipids component of cell membrane, were decreased with significance. As a conclusion, carcinogen induced hepatic lesion was correlated with the changes in lipid component of liver.

Early Life Stage Toxicity of 2,3,7,8,-Tetrachlorodibenzop-Dioxin(TCDD) in Goldfish(Carassius auratus) (금붕어 수정난의 2,3,7,8-TCDD 노출에 따른 다이옥신 관련 유전자의 발현 및 형태학적 변화)

  • In this study, we obtained the fertilized eggs from goldfish(Carassius auratus) and observed normal developmental stage(from fertilized eggs to larvae) in non-exposed groups. Goldfish embryos at 3 h postfertilization(hpf) were statically exposed for 1 h to either dimethylsufoxide(DMSO, 0.1%, v/v) or TCDD($0.5{\mu}g/L$). Toxicity and morphological changes were characterized from 3 to 120 h postfertilization(hpf). Egg mortality($0{\sim}48$ hpf) and hatching ratio($72{\sim}83$ hpf) in TCDD-exposed group were significantly different from control groups. However, pericardial edema was first observed at 72 hpf, followed by the onset of yolk sac edema and mortality. In addition, goldfish embryos-larvae exposed to TCDD significantly increased TCDD-related gene such as CYP1A($24{\sim}72$ hpf) and AhR2(72 hpf). This is the first study about in-depth characterization of TCDD-induced developmental toxicity in goldfish(Carassius auratus).

  • Meloidogyne marylandi Jepson & Golden, 1987 is described from Zoysia grass collected in Cheonan, Korea. The nematode did not produce galls, and females were generally surrounded by a massive egg sac. Perineal patterns of female were a high to rounded arch with shoulders, wavy striae usually forming a rough. Vulva and anus were sunken. Second-stage juvenile body length was 390.3 $\mu\textrm{m}$ (290-430), tail length 55.1 $\mu\textrm{m}$ (49-59) and lateral field with 4 incisures. Male stylet length was 18.1 $\mu\textrm{m}$ (15-24), spicules length 26.2 $\mu\textrm{m}$ (23-29) and lateral field with 4-5 incisures.

Cross-generational Effect of Bisphenol A on the Harpacticoid Copepod Tigriopus west: A Full Life Cycle Toxicity Test

  • The purpose of this study was to assess cross-generational effects of bisphenol A exposure in benthic copepods, Tigriopus west. Nauplii (<24 hours old) were exposed to graded concentrations of bisphenol A, and toxicity end-points such as survival, development, sex ratio, and fecundity were measured. $F_1$ generations were grown under innoxious conditions, and similarly assessed. Significant differences were observed in development of nauplii and copepodites, between exposed and non-exposed copepods; however, there were no differences in survival of nauplii or copepodites, sex ratio, or brooding rate in parental generation. In contrast, in the $F_1$ generation, there were significant differences between the control group and exposed group in survival and development of nauplii. Length, width, and biomass of parental and $F_1$ generations were reduced in the exposed group compared to the control group. In addition, some deformities, such as swelling of the prosome, abnormally shaped egg sac, and dwarfism were observed after exposure to bisphenol A. So, our study demonstrates that a cross-generation toxicity test and monitoring of morphological deformities in harpacticoid copepods, can be useful for development of potential bioindicators for environmental monitoring, and assessment of chemical impact.

  • Egg development and early life history of the Korean spine loach, Cobitis tetralineata, were investigated in the present study. Adult fish were sampled using spoon nets at the Seomjin River in Jeongsan-ri, Bokheong-myeon, Sunchang-gun, Jeollanbuk-do, Korea, July 2013. Eggs were obtained after injecting Ovarprim into females. Eggs were then artificially fertilized using the Dry method in the laboratory. Mature eggs were slightly adhesive and transparent with grey coloring, and measured $1.04{\pm}0.03mm$ ($mean{\pm}SD$) in diameter. Hatching of the embryo occurred approximately 56 hours after fertilization at $25^{\circ}C$ of water temperature, and the average size of newly hatched larvae was about $4.6{\pm}0.11mm$ in total length. At fifth day after hatching, the larval full length reached $6.8{\pm}0.28mm$ on average and their yolk sac had been completely absorbed. At 17th day after hatching, larva entered the juvenile stage and reached $9.8{\pm}0.50mm$ in total length. At 100th day after hatching, the formation of Gambetta's zone of four line was complete and juveniles were similar in general appearance to adults, and they averaged $34.7{\pm}5.77mm$ in total length.
