Environmentally-friendly Control Methods and Forecasting the Hatching Time Lycorma delicatula (Hemiptera: Fulgoridae) in Jeonnam Province (전남지역에서 꽃매미 부화시기 예측과 친환경 방제방법)

  • This study was conducted to predict the hatching time of eggs of Lycorma delicatula, to select an effective environmentally-friendly agriculture material (EFAM) and to evaluate the attraction effect of brown sticky traps for controling of Lycorma delicatula nymph and adults. Eggs hatched 55.9, 26.8, 21.6 days after incubation at 15, 20, $25^{\circ}C$ with 14L:10D condition and the hatching rates of egg were 61.9, 57.8, 30.4%, respectively. At high temperature conditions, egg development periods were shorter and the hatching rate was lower. The relationship between temperature and developmental rate was expressed by the linear equation Y=0.0028X-0.0228, $R^2$=0.9561. The low temperature threshold of eggs was $8.14^{\circ}C$ and the thermal constant required to reach larva was 355.4 DD. According to this relationship, the mean estimated hatching date was $22^{nd}$ May. The effective EFAM was natural plant extract, sophora extract, derris extract to nymph and natural plant extract, pyrethrum extract, sophora extract to adult. Among three colors of sticky trap : brown, blue and yellow, the brown sticky trap was the most attractive to nymphs and adults of L. delicatula over a 2 weeks trial period. It suggested that the brown sticky trap could be a very useful and environment-friendly control method for nymphs and adults of L. delicatula.

  • Root-knot disease caused by Meloidogyne incognita is major soil pathogen and cause severe economic damages to vegetable crops. In this study, we selected rhizobacteria for biocontrol of the root-knot nematode, M. incognita, and identified; performed bioassay of the bacterial extracts in cucumber seedlings. The crude extracts of strains CT16 and JC05 out of 180 strains inhibited egg hatching and increased juvenile mortality in vitro assay; based on 16S rRNA sequences analysis, the two strains were identified as Bacillus sp. CT16, and Neobacillus sp. JC05. After extracting the bacterial supernatants by using various organic solvents, n-butanol and n-hexane extracts of strain CT16 and n-butanol extract of strain JC05 showed inhibitory activity of egg hatching depending on concentrations. Subsequently, n-butanol extracts of two strains significantly suppressed formation of egg masses in cucumber seedling. Therefore, these results indicated that strains CT16 and JC05 could be used as potential biocontrol agents against M. incognita.

Inhibitory Effects of Natural Antimicrobial Agenton Postharvest Decay in Fruits and Vegetables under Natural Low Temperature (천연 항균제처리를 병용한 과채류의 자연 저온저장기술 개발에 관한 연구)

  • In order to prevent the postharvest decay and to promote the freshness retention of fruits and vegetables grapefruit seed extract(GFSE), natural microorganism control agent, was applied to the preservation of fresh fruits and vegetables. Freshfruits and vegetables treated with GFSE and stored in polyethylene film (0.1mm) at 1$0^{\circ}C$-15$^{\circ}C$ of natural low temperature low kept better qualities in color and texture than the GFSE -not- treated control. The treatment using GFSE ina 250ppm to 500ppm concentration seemed to be an effective one for the control of Botrytis cinerea isolated in red wine grapes. After 4 weeks of storage the firmness rate of cucumbers treated with the dilute GFSE was four times higher than that of non-treated ones. GFSE showed effective inhibitory action towards plant pathological bacteria and fungi which were involved in the decay of fruits and vegetables. Minimum inhibitory concentrations of GFSE towards them were in the range of 250ppm to 500ppm .Direct visualization of microbial cells and spores using electron microscopy showed microbial cells and fungal spores the function of which was destroyed by treating with the dilute solutions of GFSE. It was observed that GFSE would reduced disease damages and have bactericide & fungicide properties during the storage of such fruits and vegetables as egg plant, wild edible greens , kumquat, and kiwi fruit.

Effect of Supplementary Herbs and Plant Extracts on the Performance of Laying Hens (허브 및 식물 추출물의 급여가 산란계의 생산성에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of herbs and plant extracts (PE) on the performance, small intestinal microflora and immune response in laying hens. A total of 1,440 Hy-Line Brown laying hens of 67 wks old were assigned to one of the following 9 dietary treatments : T1 : Control (C), T2 : C + Avilamycine 6 ppm, T3 : C + Herb $Mix^{(R)}$ 0.2%, T4 : C + Biostrong $510^{(R)}$ 0.02%, T5 : C + $APEX^{(R)}$ 0.02%, T6 : C + $Digestarom^{(R)}$ 0.02%, T7 : C + $Phellozyme^{(R)}$ 0.1%, T8 : C + $Galicin^{(R)}$ 0.05%, T9 : C + CRINA $Poultry^{(R)}$ 0.05%. Each treatment was replicated 8 times with twenty birds housed in 2 bird cages. Twenty bird units were arranged according to completely randomized block design. Feeding trial lasted 6 wks under 16 hours lighting regimen. Hen-day egg production was not significantly different among the treatments, but that of supplemented groups tended to be higher than the control. There were significant differences among treatments in feed intake and feed conversion ratio. Feed intake was higher in the supplemented groups than the control. Feed conversion ratio was higher in T8 than other treatments. Egg shell color index and egg yolk color index were significantly different among treatments. Egg shell color was affected by supplements and egg yolk color index of T9 (PE-CRINA) was significantly higher than the control. Haugh unit was not significantly different among treatments. There were significant differences in leukocytes and erythrocytes parameters. The level of serum WBC and stress index (heterophil/lymphocyte) were higher in supplemented groups than the control. The level of RBC tended to be lower in the herb or PE groups than the control. The concentration of serum IgG was not significantly different among the treatments, but all those of the supplemented groups were higher than the control. The number of Lactobacilli spp. tended to increase and that of Cl. perfrigens decrease in the supplemented groups. The number of E. coli significantly decreased in the supplemented groups. The results of this experiment showed that feeding herbs and plant extracts to laying hens tended to improve the egg production and affect positively on the level of blood parameters and small intestinal microflora.

In vivo Angiogenic Activity of Dichloromethane Extracts of Aloe vera Gel

  • The angiogenic activity of Aloe vera (Aloe baradensis), known as a good healing plant, was investigated. We have extracted and fractionated dichloromethane extract (G1M1D1) and methanol soluble fraction of dichloromethane extract (G1M1D1M1) which contain low-molecular weight substances of Aloe vera gel. G1M1D1 and G1D1M1 fractions induced a radially arranged, spoke-sheel-like vasculature in chick embryo chorioallantoic membrane (CAM) assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay was very similar to that of phorbol 12-myristate-13-acetate (PMA) used as a positive control. The modified CAM assay, e simple and accurate quantitating method, was used to quntitate the angiogenic activity of G1D1M1 fraction. Application of G1M1D1M1 fraction ($100\mug/egg$) resulted in much more intense angiogenesis than in control while slightly less intense angiogenesis than in PMA (100 ng/egg).

Developmental characteristics of Henosepilachna vigintioctomaculata under different temperatures and control effect of eco-friendly agricultural materials on Lycium chinense (구기자에서 큰이십팔점박이무당벌레(Henosepilachna vigintioctomaculata) 의 온도조건에 따른 발육특성과 친환경제재에 의한 방제효과)

  • Host plants of the Henosepilachna vigintioctomaculata are known as Solanaceae plants such as potato, tomato, eggplant and Lycium chinense. Henosepilachna vigintioctomaculata (Motschulsky) cause continuous damage from early June to mid-August in leaves of L. chinense under environmental-friendly management in Cheongyang, Chungnam. The developmental period of egg and larva of H. vigintioctomaculata were investigated. H. vigintioctomaculata were reared on L. chinense at several temperatures, for example, 15, 20, 25 and $30^{\circ}C$ in the laboratory. In addition, the control effect of 5 eco-friendly agricultural materials were examined. As results, hatching rate is the highest to 91% at $25^{\circ}C$. Egg periods were 10.8, 7.7, 5.1 and 3.7 days at each tested temperature, respectively. And the total developmental periods from egg to adult were 42.7, 26.3, 18.4 and 19.4 days at each tested temperature, respectively. Otherwise, their eclosion rates from pupa to adult were showed 58%, 77%, 85%, 65% at each tested temperature, respectively. As a result of the control effect, both adults and larvae of H. vigintioctomaculata were shown a high mortality to mixture with extract of Sophora flavescens, Chenopodium ambrosioides and Melia azedarach. Additional, larvae is shown high mortality to extract of Azadirachta indica and Cymbopogon nardus. Thus H. vigintioctomaculata might be controled by using plant extract of eco-friendly agricultural materials in due season.

Nematicidal Efficacy of Herbal Extracts against Meloidogyne hapla (당근뿌리혹선충에 대한 식물추출물의 살선충 효과)

  • The nematicidal and egg haching inhibitory effects of extracts from 30 herbal plants (total 32 samples) against Meloidogyne hapla J2 juveniles and eggs was tested using the dipping method. At 1,000 ppm, extracts of Daphne genkwa flower buds, Eugenia caryophyllata flowers, Quisqualis indica fruits, and Zingiber officinale rhizomes produced > 80% mortality in J2 juveniles. At 125 ppm, extracts of D. genkwa and Q. indica produced 91 and 99% mortality, respectively. The toxicity of 5 selected plant extracts to M. hapla differed depending on the solvent used (i.e. hexane, methanol, hot water, or cold water). Hot water extracts of Z. officinale and Q. indica produced nematicidal efficacies of 99 and 99%, compared to 36 and 98%, respectively, with cold water extraction. Q. indica extract was highly active against M. hapla regardless of extraction method. The inhibitory effects of Areca catechu, D. genkwa, Desmodium caudatum, Pharbitis nil, Q. indica, and Z. officinale extracts on egg hatching of M. hapla was evaluated. At 1,000 ppm, D. genkwa, P. nil, and Q. indica extracts significantly reduced hatching at 7, 14 and 21 days after treatment. Numbers of juveniles in soil treated with the methanol extract D. genkwa (1,000 ppm) were significantly lower than in untreated soil in trials in pots and in a ginseng (Phanax ginseng) field. These results indicate that Q. indica extracts could be used as an environmental friendly control agent of M. hapla.

Development of a Bioassay Method Using Aluminium Foil Sheet for Screening Ovicidal Activity Against Diamondback Moth Egg, Plutella xylostella L. and Selection of Plant Extracts with High Ovicidal Activity (알루미늄호일 이용 배추좀나방(Plutella xylostella L.) 살란활성 검정법 개발 및 살란활성 식물추출물의 선발)

  • Diamondback moth (DBM, Plutella xylostella L.) is known as the most destructive pest of cruciferous crops worldwide. As most insecticides targeted to mainly larval stage, new insecticides which have hatching-inhibitory or ovicidal activity could be more efficient to control DBM. Therefore, we developed an easy and efficient method for screening ovicidal activity of DBM eggs using aluminum foil. The aluminum foil ($4{\times}12$ cm) coated with filtered juice of chinese cabbage leave (hereinafter called oviposition foil) exposed to 300 newly-emerged adults for 24 hours inside the rearing container. The oviposition foils were replaced every 4 days consecutively after mating, but it was better to discarded over then. Oviposition foil were divided into 6 to 12 pieces depending on egg mass volume. After dipping into test solutions for 10 seconds using faucet, oviposition foil pieces were placed into common petri dish, and then investigated hatchability. The effect of methanol solvent (50%) for 10 seconds dipping on the toxicity against DBM eggs was negligible. In addition, whether covering the petri dish or not should be dependent on nature of active compounds tested. With applying the new bioassay method, methanol extracts from 50 plants were tested the ovicidal activity to DBM eggs. Among them, four plant extracts; Angelica tenuissima root, Lycium chinense root, Cnidium officinale root and Polygala tenuifolia root, showed high ovicidal activity of over 90% control efficacy, against DBM eggs.

Biological Control of Root-knot Nematode by Streptomyces sampsonii KK1024 (Streptomyces sampsonii KK1024를 이용한 뿌리혹선충 (Root-knot nematode)의 생물학적 방제)

  • Streptomyces sampsonii KK1024 having strong chitinolytic activity was isolated from crab-shell rich soil at Muan, Jeolanamdo. The KK1024 produced chitinase, protease, gelatinase and lipase. When 50% of KK1024 culture broth was treated to juveniles and eggs of root-knot nematode, juvenile mortality at 3 days was 81.67% and egg hatch rate at 5 days was 2.00%. When $183.7{\mu}g\;mL^{-1}$ of crude enzyme produced by KK1024 was treated, juvenile mortality at 3 days was 96.00% and egg hatch rate at 5 days was 5.33%. At 1% of butanol extract from KK1024, juvenile mortality was highest with 90.00% and egg hatch rate was lowest with 0%. The comparison of the effect of KK1024 culture broth with only medium, synthetic fertilizer, and commercial nematicide on tomato growth and nematode infection was examined in pot trials. KK1024 culture broth showed lower number of egg mass and gall in plant, and population of juveniles in soil compared with only medium and synthetic fertilizer treatment, but not in commercial nematicide. However, the highest shoot weight and length was discovered in KK1024 culture broth. These results suggest that Streptomyces sampsonii KK1024 producing lytic enzymes and nematicidal compounds can be one of candidates for biocontrol agents against root-knot nematodes.

Comparison of an Automated Most-Probable-Number Technique TEMPO®TVC with Traditional Plating Methods PetrifilmTM for Estimating Populations of Total Aerobic Bacteria with Livestock Products (축산물가공품에서 건조필름법과 TEMPO®TVC 검사법의 총세균수 비교분석)

  • We compared between an automated most-probable-number technique $TEMPO^{(R)}$TVC and traditional plating methods $Petrifilm^{TM}$ for estimating populations of total aerobic bacteria in various livestock products. 257 samples randomly selected in local retail stores and 87 samples inoculated with $E.$ $coli$ ATCC 25922, $Staphylococcus$ $aureus$ ATCC 12868 were tested in this study. The degree of agreement was estimated according to the CCFRA (Campden and Chorleywood Food Research Association Group) Guideline 29 and the agreement indicates the difference of two kinds methods is lower than 1 log base 10($log_{10}$). The samples of hams, jerky products, ground meat products, milks, ice creams, infant formulas, and egg heat formed products were showed above 95% in the agreement of methods. In contrast, proportion of agreement on meat extract products, cheeses and sausages were 93.1%, 92.1%, 89.1%, respectively. One press ham and five sausages containing spice and seasoning, two pork cutlets containing spice and bread crumbs, two meat extract product and two natural cheeses and one processing cheese with a high fat content, and one ice cream containing chocolate of all samples showed the discrepancy. Our result suggest that $TEMPO^{(R)}$TVC system is efficient to analyses total aerobic bacteria to compare manual method in time-consuming and laborious process except livestock products having limit of detection.
