  • Membranes are widely used to separate target solute molecules such as proteins on the basis of their size in cell broth mixture to minimize the loss of target compounds. In this study, membrane separation system using ultrafilters of MWCO 100 K and 50 K, was operated for concentration and purification of microbial transglutaminase. Fermentation broth containing MTGase was prefiltered by using pore size 1.6 and $0.7\;{\mu}m$ pre-filter made of cellulose fiber and $0.45\;{\mu}m$ microfilter made of cellulose acetate. The prefiltered solution was concentrated by 100 K and 50 K ultrafilter. The final enzyme concentration was 1.29 units/ml and enzyme specific activity was 0.2 units/mg protein. This specific activity were 3.7 times higher than that of initial cell broth mixture. Membrane separation process of microfiltration and ultrafiltration was proved to be very economic, energy efficient and effective separation method used to concentrate MTGase.

  • Numerical solutions were obtained to the model equations for various of the parameters characterizing the pore structure, effective internal diffusion, and the chemical reaction constant. The conversion was decreased with the cause of pore closure at the surface of reacting particles, reduction of porosity, surface area of reaction and effective diffusion coefficient in the solid with the progress of reaction. Total conversion was strongly dependent on the local conversion at surface. According to the decreasing of impregnated concentration of the copper oxide and the increase of the flue gases concentration, total conversion was increased. The conversion was affected by gas flow rate and pore size distribution in the reacting solid.

Estimation of the Permeability Variation in Saturated Sand Deposits Subjected to Shaking Load Using 1-g Stinking Table Test (1-g 진동대시험을 이용한 진동하중을 받는 포화된 모래지반의 투수계수 변화 추정)

  • The purpose of this study is to understand the dissipation pattern of excess pore pressure after liquefaction and to estimate the variation in permeability during shaking load, which should be known for settlement predictions of the ground undergoing liquefaction. In this study, 1-g shaking table tests were carried out for 5 different kinds of sands, all of which had high liquefaction potentials. During the tests excess pore pressure at various depths, and surface settlements were measured. The measured dissipation curve of the excess pore pressure after liquefaction was linearly simulated using the solidification theory, and from the analysis of the slopes of linearly simulated curves, the correlation between dissipation velocity and the gradation characteristics was obtained. By substituting this correlation and the measured settlement to the dissipation velocity equation recommended in solidification theory, the permeability during dissipation was calculated, which was used for estimating the permeability variation during shaking load. The dissipation velocity of excess pore pressure after liquefaction had a linear correlation with the effective grain size divided by the coefficient of uniformity. The permeability during dissipation and liquefaction increased by 1.1∼2.8 times and 1.4∼5 times compared to the initial permeability of the original ground, respectively. And the amount of increase became greater as the effective grain size of the test sand increased and the coefficient of uniformity decreased.

  • An attempt was made to analyze green microstructure of AIN samples prepared by slip casting and dry pressing through Hg-porosimetry. Slip cast samples with narrow pore size distribation and high packing density showed higher sinterability and homogeneous distribution of second phase(s). Hg-porosimetry is and effective way to determine pore structure if "ink bottle" phenomenon does not occur. A comparison study with porosity measurement by quantitative microscopy showed that the effectiveness of Hg-porosimetry measurement could be extended to higher sintered density as long as pores remained open.

  • The modified statistical-mechanical theory for dense fluid mixtures of rigid spheres has been applied to rigid sphere fluids in the nonoverlapping pore model. The resulting expressions for the partition coefficient and diffusivity illustrate the influence of steric hindrance on the thermodynamic and transport properties in such systems. The open membrane model without the size-exclusion and shielding effects shows considerable overestimation of the diffusion flux when the effective mean pore radii of the order of $20{\AA}$ or less are involved. Theoretical predictions investigated here were also compared with experimental data for hydrogen gases in inorganic porous membranes and it was observed a qualitative agreement in the low pressure limit.

Surface Milling for the Study of Pore Structure in Shale Reservoirs (셰일 저류층 내 공극 구조 연구를 위한 표면 밀링)

  • Understanding the pore structure including pore shape and connectivity in unconventional reservoirs is essential to increase the recovery rate of unconventional energy resources such as shale gas and oil. In this study, we found analysis condition to probe the nanoscale pore structure in shale reservoirs using Focused Ion Beam (FIB) and Ion Milling System (IMS). A-068 core samples from Liard Basin are used to probe the pore structure in shale reservoirs. The pore structure is analyzed with different pretreatment methods and analysis condition because each sample has different characteristics. The results show that surface milling by FIB is effective to obtain pore images of several micrometers local area while milling a large-area by IMS is efficient to observe various pore structure in a short time. Especially, it was confirmed that the pore structure of rocks with high content of carbonate minerals and high strength can be observed with milling by IMS. In this study, the analysis condition and process for observing the pore structure in the shale reservoirs is established. Further studies are needed to perform for probing the effect of pore size and shape on the enhancement of shale gas recovery.

Filter Plate Micro Trap as a Device for in situ Cultivation for Environmental Microorganisms (환경시료에 존재하는 미생물 배양을 위한 filter plate micro trap의 개발)

  • Filter plate microbial trap (FPMT) was invented as an in situ cultivation device for the isolation of bacteria from natural environments. FPMT consists of a medium and membrane filters (0.45 ${\mu}m$ pore size) and microorganisms and compounds can be moved freely moved into the medium. This device was applied to two soil samples of Greenland. The microbial diversity of both soil samples by FPMT was higher than that by the conventional Petri dish-based method. Moreover, novel bacterial species were isolated by FPMT. The new FPMT is effective for in situ cultivation of natural samples and could be applicable to the isolation of uncultivable microorganism.

A Volatile Organic Compound Sensor Using Porous Co3O4 Spheres

  • Porous $Co_3O_4$ spheres with bimodal pore distribution (size: 2-3 nm and ~ 30 nm) were prepared by ultrasonic spray pyrolysis of aqueous droplets containing Co-acetate and polyethylene glycol (PEG), while dense $Co_3O_4$ secondary particles with monomodal pore distribution (size: 2-3 nm) were prepared from the spray solution without PEG. The formation of mesopores (~ 30 nm) was attributed to the decomposition of PEG. The responses of a porous $Co_3O_4$ sensor to various indoor air pollutants such as 5 ppm $C_2H_5OH$, xylene, toluene, benzene, and HCHO at $200^{\circ}C$ were found to be significantly higher than those of a commercial sensor using $Co_3O_4$ and dense $Co_3O_4$ secondary particles. Enhanced gas response of porous $Co_3O_4$ sensor was attributed to high surface area and the effective diffusion of analyte gas through mesopores (~ 30 nm). Highly sensitive porous $Co_3O_4$ sensor can be used to monitor various indoor air pollutants.

Effect of Foaming Temperature on Cell Structure of 606X Series Aluminum Alloy Metallic Foams (Foaming 온도에 따른 606X계 발포 알루미늄의 제조 특성)

  • Metal foam is one of the most interesting materials with various multi-functional properties such as light weight, energy absorption, high stiffness and damping capability. Among them, energy absorption property has keen interests in the field of automotives for passenger protection. Nowadays, researches about pore size and porosity control of the foam are increased to correspond them. However, though energy absorption properties are improved, these results are not cost-effective process. In present research, however, as a part of improving the energy absorption property of metallic foams, 606X aluminum alloy was used for cell wall material which has higher strength than pure aluminum. And its morphological features are characterized. As a results, porosity and pore size are uniformity distribution with increasing foaming temperature in the case of 6061 alloy foams. 6063 alloy foam specimens have opposite tendency because of the influence of alloying element and viscosity of the molten melt.

Changes in the Constituents of Citrus Juice by Ultrafiltration (한외여과에 의한 온주 밀감주스의 성분 변화)

  • The citrus juice obtained from Jeju mandarines, Citru unshiu, was filtered to remove 20% of its original volume through the hollow fiber ultrafiltration systems equipped with various pore sizes of membranes. As the results of ultrafiltration, the contents of total acids, ascorbic acid, ascorbic acid, free sugars and neohesperidin in retentate showed the tendencies of gradual decreases with the increase of membrane pore sizes from 10K to 100K daltons, but tendencies were inverted when the membrane with 500k was used. The changes of color, soluble solids, total nitrogen, amino-nitrogen, naringin and hesperidin were not consistent with the membrane pore size. Considering all the data obtained using various pen sizes of membranes, the filtration system with NMWC 500K daltons was the most effective to produce citrus juices with hither quality.
