Genetic Diversity and Morphological Variations of Goosegrass [Eleusine indica (L.) Gaertn] Ecotypes in Malaysia

  • Goosegrass [Eleusine indica (L.) Gaertn] has been a nuisance to growers in Malaysia due to its increased resistance to commercial herbicides, rapid growth and dissemination, and interference with agricultural practices. In the course of developing an apt integrated management to control goosegrass, more information of this weed is needed. The aim of this study was to look into variations among the goosegrass ecotypes sampled throughout Malaysia from the aspects of genotype and phenotype. Sequence-related amplified polymorphism (SRAP) markers were employed in investigating the genetic diversity and relationships among the 18 goosegrass ecotypes. Consequently, 5 primer combinations amplified 13 fragments with the polymorphism rate of 69.23%. At 74% similarity, the ecotypes were clustered into 6 groups. Phenotypic variability of the goosegrass ecotypes was assessed by observing their morphology, growth and seed traits. Goosegrass ecotypes were sorted into 3 major groups at the genetic distance (DIST) of 0.37. Concurrences of the evaluated genetic distance, ecotypes with the closest and most distant relationships were assembled together in Group I which showed high variation even among ecotypes in the same group. Results obtained thus implied high molecular and morphological variations of the goosegrass ecotypes in Malaysia.

Differential Competitiveness of Echinochloa colona Ecotypes (ECHINOCHLOA COLONA 생태형(生態型)의 경합력(競合力) 차이(差異))

  • The ability of three Echinochloa colona (L.) Link ecotypes to compete with rice (Oryza sativa L.) was evaluated. For all the ecotypes, E. colona was shorter and produced less leaf area and dry matter than rice at 15 days after seeding (DAS), whereas the reverse was observed from 30 to 60 DAS. Neither E. colona nor rice tillered during the first 15 DAS, but E. colona had greater accumulative tiller length than rice from 15 DAS. Rice absorbed more nitrogen than E. colona at 15 DAS. E. colona absorbed greater amounts of nitrogen, phosphorus, and potassium than did rice between 30 and 60 DAS and increase in its density decreased the nutrients uptake of rice. Differences in the competitive ability of the ecotypes were related to the growth characteristics of the ecotypes and the period of competition between the two species. At the early growth stages the Pangasinan ecotype, which increased plant size rapidly and had a shorter life cycle, was more competitive against rice than the Leyte and South Cotabato ecotypes, which had a longer vegetative growth period. However, the reverse was observed at the later growth stages of the ecotypes.

Studies on the soybean varieties in Korea -1. Classificiatons of Ecotypes and Maturity groups- (대두의 품종에 관한 연구 -제1보 생태형과 성숙군의 분류-)

  • Experiments were carried out to classify the ecotypes and maturity groups of soybean varieties, and to make clear the relationships among these ecotypes, maturity groups and some characteristics in the growing and ripening process of soybean varieties in Korea. Soybean varieties used as the material were 138, 57 varieties collected from Japan, America and Canada and 81 local varieties of Korea. These varieties were grown in the Experimental Farms, College of Agriculture, Seoul National University, Suwon, Korea and Chinju Agricultural College, Kyungsang Namdo, Korea. Seed sowing was conducted at 8 times from April15 to July 29, at 15 day-intervals, in 1962. The classification methods of ecotypes and maturity groups applied in this study, and the results obtained are summarized as follows; 1. Ecotypes were classified from the standpoint of the periods from emergence to flowering, the shortened ratio of days from sowing to flowering by delayed sowing, and the periods from flowering to maturity(as shown in Table A). 2. All varieties wer classified into 10 ecotypes of Ia$_1$, Ib$_1$, IIa$_2$, IIb$_1$, IIb$_2$, IIIb$_1$IIIb$_2$, IIIc$_1$and IIIc$_2$ symbols. Korean local varieties, however, belong to 6 ecotypes of IIb$_1$, IIb$_2$, IIIb$_1$, IIIc$_2$, IIIc$_1$ and IIIc$_2$, respectively(table 1). 3. Durations from sowing to maturity were from 120 to 190 days in April sowing plots of all varieties. From the standpoint of the durations, maturity groups were classified into 9 groups from group I, extreme early, to group IX, extreme late, but our local varieties belong to 5 maturity groups of III, IV, V, VI and VII symbols, respectively(Table 2). 4. Kinds of ecotypes and maturity groups, and the number of soybean varieties belonging to those are as shown in Table B. 5. In this study, it was observed that there were some close relations between these ecotypes and maturity groups (Table 3,4), and among the ecotypes, maturity groups and some other characteristics in the growing and ripening process of the soybean varieties(Table 5). 6. Furthermore, it was also observed that the kinds of ecotypes and maturity groups of recommended varieties in the southern Korean provinces are more numerous than those of the northern provinces in Korea.

Molecular markers based on chloroplast and nuclear ribosomal DNA regions which distinguish Korean-specific ecotypes of the medicinal plant Cudrania tricuspidata Bureau

  • Cudrania tricuspidata Bureau is a widely-used, medicinal, perennial and woody plant. Obtaining information about the genetic diversity of plant populations is highly important with regard toconservation and germplasm utilization. Although C. tricuspidata is an important medicinal plant species registered in South Korea, no molecular markers are currently available to distinguish Korean-specific ecotypes from other ecotypes from different countries. In this study, we developed single nucleotide polymorphism (SNP) markers derived from the chloroplast and nuclear genomic sequences, which serve to to identify distinct Korean-specific ecotypes of C. tricuspidata via amplification refractory mutation system (ARMS)-PCR and high resolution melting (HRM) curve analyses. We performed molecular authentication of twelve C. tricuspidata ecotypes from different regions using DNA sequences in the maturaseK (MatK) chloroplast intergenic region and nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The SNP markers developed in this study are useful for rapidly identifying specific C. tricuspidata ecotypes from different regions.

Changes in Antioxidant Enzyme Activities of Two Contrasting Ecotypes of Arundinella hirta to Drought Stress

  • To understand antioxidant enzyme response of two contrasting Arundinella hirta ecotypes to drought stress, drought-tolerant Youngduk and drought-sensitive Jinju-1, were comparatively analyzed changes in the enzymatic activities of peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), superoxide dismutase (SOD), and glutathione reductase (GR). Two ecotypes, drought-tolerant Youngduk and drought-sensitive Jinju-1 were subjected to drought stress by withholding water for 12 days. ROS accumulation level and electrolytic leakage were significantly increased in both A. hirta ecotypes by drought stress treatment but less in Youngduk than Jinju-1. The RWC significantly decreased in both the drought stress-treated ecotypes as compared to control, but less in Youngduk than Jinju-1. Soluble sugar and protein content were increased more in drought stress-treated Youngduk as compared to Jinju-1. The activities of antioxidant enzymes such as SOD, CAT, POD, APX, and GR increased significantly in both the drought stress-treated ecotypes Youngduk and Jinju-1 as compared to control. The increase in antioxidant enzyme activity level was more prominent in drought stress-treated Youngduk as compared to Jinju-1. Taken together, these results suggest that Youngduk was more tolerant to drought stress than Jinju-1, and seem to indicate that tolerance of A. hirta to drought stress is associated with increased activity of antioxidant enzymes.

Blooming and Morphological Characteristics of Korean Native Kentucky bluegrass(Poa pratensis L.) Ecotypes (국내에서 수집된 주요 왕포아풀(Poa pratensis L.)의 개화 및 형태적 특성)

  • In this study, about 80 Kentucky bluegrass (Poa pratensis L.) ecotypes native to Korea were collected annually to analyze their morphology and growth characteristics from 1995 to 2007. Kentucky bluegrass ecotypes with superior characteristics of 'Pureundle' (Trial No. : P38), 'Sewon' (P77), 'Inje' (00034), 'Donggang' (00052), 'Hwasun' (01001), 'Pyengchang' (01071), 'Kongju' (02006), 'Chungsong' (02077), and 'Kumsan' (03005) and three foreign cultivars were transplanted to flowerpots in May 8, 2006 in order to analyze the blooming and morphological characteristics. Out of the selected superior ecotyes, six were collected on the roadsides, two were collected around paddy or dry fields, and one was collected at riverside. Length of 1st internode of 'Chungsong' was the longest (30.7cm), that of 'Pyengchang' was the shortest (12.8cm), and ecotypes showed variation in length. The 1st angle of branches of 'Donggang' was the narrowest ($141.7^{\circ}$), that of 'Hwasun' was the widest ($188.3^{\circ}$), and that of the introduced foreign cultivars were between $159.3^{\circ}$ and $166.7^{\circ}$. The number of nodes of the introduced cultivars were same (six), while that of Korean ecotypes were four to six. Length of flowering culm of 'Chungsong' was the longest (50.7cm) and while that of 'Pyengchang' was the shortest (19.2cm). 'Donggang' with the highest seed yield showed relatively high (36.8cm) inflorescence height when compared to other ecotypes. The five ecotypes of 'Sewon', 'Inje', 'Donggang', 'Pyengchang', and 'Kongju' were investigated to have relatively wide leaf width. Especially 'Sewon' and 'Inje' were investigated to have wide leaves of 4.5mm and over.

Geographical Distribution and Characteristics of Kentucky Bluegrass(Poa pratensis L..) Native to Korea (우리 나라 자생 왕포아풀의 수집지 분포 및 특성)

  • This research has been conducted to collect regional ecotypes of Poa pratensis throughout the southern part of korean peninsula during 2000~2002. 227 local ecotypes of Poa pratensis were collected mostly from roadsides, riversides and rice paddies. The $35^{\circ}$ latitude was supposed to be the southern limit of native Poa pratensis habitat. 130 ecotypes of Poa pratensis found from the poor environmental condition such as roadsides showed Poa pratensis naturally grew throughout the country. When surveyed the habitat environment, it was concluded that native Poa pratensis were mostly found on open fields or half shaded areas, on sandy soil or sandy loam soil, and in the individual patch type.

  • The intraspecific susceptibility of twelve Echinochloa colona(L.) Link ecotypes to herbicides was studied at three different stages of growth. At germination and post-germination stage, the Batangas ecotype was most susceptible to both butachlor (N-butoxymethyl-2-chloro-2', 6'-diethylacetanilide) and thiobencarb (S-4-dichlorobenzyldiethylthiocarbamate), whereas the Bukidnon ecotype was the most tolerant. The susceptibility of the ecotypes used was not associated with the seed weight and maturity. When the preemergence herbicides were applied at the 2-leaf stage of the ecotypes, the Nueva Ecija ecotype was least susceptible to butachlor at 1 ppm, but it was most susceptible to thiobencarb. The reverse was exhibited by the Cagayan ecotype. Differential susceptibility of the ecotypes to propanil (3', 4'-dichloropropionanilide) as determined by the necrotic length of the third leaf of 20-day-old seedlings was pronounced in the Camarines Sur, Iloilo, and IRRI (red) ecotypes. However, no necrotic tip appeared in the Cagayan and Batangas ecotypes. The differential responses varied with the type and the concentration of the herbicide studied and the growth stage of the ecotypes.

  • Three ecotypes of the freshwater red alga Compsopogon coeruleus (Balbis) Montagne were recorded from different freshwater and brackish water habitats of Orissa state in the east coast of India. These three had persistent differences in their branching pattern, e.g. (i) acute angle between main axis and lateral branch, (ii) equal or near to right angle between main axis and lateral branch, and (iii) short spine-like outgrowth instead of a branch in older filaments, besides having differences in the length, breadth and thickness of cortex of the thallus. Morphological observation of these taxa, and the ecological characteristics of the habitat of their occurrence is presented.
