Identification of Genes for Growth with Oxygen in Escherichia coli by Operon Fusion and Southern Blot Techniques

  • Seven Escherichia coli cells defective with aerobic growth were isolated by the insertion of ${\lambda}placMu53$, a hybrid bacteriophage of ${\lambda}$ and Mu, which created a transcriptional fusion to lacZY. These insertion mutant cells were tested on an XG ($5-bromo-4-chloro-3-indolyl-{\beta}-D-galactopyranoside$) medium for anaerobic expression of lacZ by fusion to a promoter. The chromosomal DNA from these strains were digested by EcoRI, and the EcoRI fragments that contained the fused gene and lacZ sequence were identified by Southern hybridization, using lacZ containing plasmid as a probe. The EcoRI fragment from each strain was cloned and sequenced. The sequence data were compared with the GenBank database. The mutated gene of three strains, CYT4, CYT5, and OS11, was found to be identical, and it was nrdAB that encoded ribonucleoside diphosphate reductase. The gene nrdAB was at min 50.5 on the Escherichia coli linkage map and 2,348,084 on the physical map, and is involved in hemAe-related reduction-oxidation reaction. OS6 and OS14 mutant strains had insertion at min 8.3 and the mutated gene was hemB. The hemB encodes 5-aminolevulinate dehydratase or porphobilinogen synthase. The OS3 mutant had insertion in cydB at min 16.6. The cydAB encodes cytochrome d oxidase. In the case of OS1, the fusion was made with sucA, the E1 component of ${\alpha}-ketoglutarate$ dehydrogenase.

  • Over the years, there have been many school construction projects resulting from the activation of the Build Transfer Lease(BTL). Furthermore, due to the need for the qualitative expansion of educational facilities, as well as the attempt to fulfill social requirements and the need for eco-friendly learning environments, these projects have become a major priority. As a result, the government has established a proper standard for school construction based on a certain level of green certification that has been mandated. The aim of this study is to assess the status of schools as relates to the composition of the ecological environment surrounding them. These elements of eco-friendly school facilities are larger than the educational environment and are directly related to the environmental and educational effects on the students. Looking at the overall results of this study, the areas of ecological environment outside school are found to be inadequate. Therefore, this field needs to adopt more realistic goals, and focus on the areas where the most improvement is needed.

Energy-saving optimization on active disturbance rejection decoupling multivariable control

  • An industrial control process multiple-input multiple-output (MIMO) coupled system is analyzed in this study as an example of a Loss of Coolant Accident (LOCA) simulation system. Ordinary control algorithms can complete the steady state of the control system and even reduce the response time to some extent, but the entire system still consumes a large amount of energy after reaching the steady state. So a multivariable decoupled energy-saving control method is proposed, and a novel energy-saving function (economic function, Eco-Function) is specially designed based on the active disturbance rejection control algorithm. Simulations and LOCA simulation system tests show that the Eco-function algorithm can cope with the uncertainty of the multivariable system's internal parameters and external disturbances, and it can save up to 67% of energy consumption in maintaining the parameter steady state.

The Management Methods of Multi-Purpose Ecological Reservoir by System Thinking - Focused on Anteo Eco Park - (시스템 사고를 통한 다기능 생태저류지의 관리방안 - 광명 안터생태공원을 중심으로 -)

  • Ecological reservoir is a multifunctional space where provides the functions of retention, animal habitat and improvement of ecosystem health and landscape. The ecological reservoir of Anteo Eco Park located in Gwangmyeong-si has established to functions for water purification, maintenance of healthy aquatic ecosystem. Because the Anteo Eco Park is located in the site where nonpoint pollutant materials flow in, Anteo Eco Park has potential factors which aquatic ecosystem health deteriorates and damages the habitat of golden frog(Rana plancyi chosenica) which is restoration target species. Therefore, the purpose of this study is to suggest the plan to manage the variables which impede the right functions of aquatic ecosystem by understanding the causal loop diagram for the change of water quality environment and the interaction of predator-prey through system thinking. The results are as follows. First, the study showed that the individual number of golden frog which is an indicator species of Anteo Eco Park is threatened by snakeheaded fish, which is an upper predator. Therefore, balanced food chain should be hold to protect golden frog by capturing the snakeheaded fish which is individual number's density is high, and the monitoring management of the individual number for predator(snakeheaded fish)-prey(golden frog) should be performed. Second, the study represented that water pollution and carnification is caused by the sediment as the dead body of the large emergent vegetation in the winter cumulates as sediment. Ecological reservoir in Anteo Eco Park has been managed by eliminating the dead body of the large emergent vegetation, but the guideline for the proper density maintenance of vegetation community is additionally needed. Lastly, the study showed that aquatic ecosystem of Anteo Eco Park where is contaminated from the inflow of nonpoint pollutants affects the individual number's decline of golden frog and snakeheaded fish. Accordingly, the creation of a buffer area and a substitution wetland is needed in the periphery of the Anteo Eco Park to control the inflow of nonpoint pollutants including organic matters, nutrients and heavy metals. This study will be helpful that Anteo Eco Park improves the regional landscape and maintain healthy aquatic ecosystem space for the park visitors including local residents.

New Record of the Genus Oncholaimus Nematode Species (Nematoda: Oncholaimidae) from the East Sea of Korea

  • Oncholaimid nematode, Oncholaimus secundicollis Shimada, Kajihara and Mawatari, 2009, is firstly reported based on a comparative morphological study. The present specimens of O. secundicollis are collected from rocky intertidal seagrass on the eastern coast of Korea. Oncholaimus secundicollis is distinguished from all of its congeners by the following combination of morphological characteristics: buccal cavity large with thick cuticular wall and three teeth, left subventral tooth larger than right subventral or dorsal teeth, and the presence of two remarkable precloacal papillae with nine pairs of cloacal setae and single ventral tail papilla with two pairs of short setae. In this study, we provide taxonomic description and illustration of unrecorded species of the genus Oncholaimus.

Action Plans of Green r-Learning Services based on UCR(User Created Robots) (창작로봇(UCR) 기반 친환경 r-러닝 서비스 실천방안)

  • Expectation for improvement of creativity and problem-solving capability has increased the creative robotics classes in the form of after-school activity in more than half of total elementary schools. While Ministry of Education, Science and Technology has promoted 'Green IT Guidelines' as a part of 'Eco-friendly Green School Development Project', the Green issues have not been considered enough in those creative robotics classes. In this paper, we would like to address the Green issues, especially in the r-Learning services based on UCR (User Created Robots). First, trend of green IT education, r-Learning services and UCR are reviewed. And the current status of eco-related operations and teachers' perception in the robotics classes of elementary schools is investigated. Examples of Green UCR are also searched and green programs based on the three kinds of UCR, UC-TR, UC-AR, UC-CR, are explored. Finally, we propose action plans to promote the UCR-based r-Learning service reflecting green issues.

Triadenum breviflorum (Hypericaceae): First report in Korea

  • Triadenum breviflorum (Wallich ex Dyer) Y. Kimura in Nakai & Honda (Hypericaceae), previously known only in northeast India, China and Taiwan, was found in a few wetlands of Jeju Island in 2008. Additionally, some images online of T. breviflorum suggest that it exists in other places on the Korean peninsula but that it has been misidentified as Triadenum japonicum. This species is distinguished from the latter by white petals, inflorescence from nodes 6-11, linear to narrowly elliptic leaves, and an attenuated base. The local name Huin-kkot-mul-go-chu-na-mul implies a white flower rather than the flesh-pink color of T. japonicum. The morphological characteristics of the species are provided with photographs of the habitat.

  • Fossil fuel, the energy source of internal combustion engine automobiles, is limited in resource and has caused environmental issues for decades. Accordingly, automobile manufacturers from many countries around the world are developing or producing eco-friendly vehicles that utilize alternative sources of energy. These vehicles are equipped with many electronic and electrical components which operate on high voltage and/or large current that were not used in conventional combustion engine automobiles. In this paper, in order to analyze the electro-magnetic interference noise, electric vehicles and fuel cell electric vehicles are tested under the guidelines of KMVSS (Korean Motor Vehicle Safety Standards) as well as under test modes that are not stipulated under the guidelines.

Knowledge and Consciousness on Environment and Eco-friendly Behavior related to dietary life of Middle School Students (중학생의 식생활 관련 환경지식과 환경의식 및 환경친화적 식생활행동)

  • In this study, environmental knowledge and consciousness and eco-friendly behavior related to dietary life of middle school students were examined. Male and female students in Gyeongsan and Daegu Metropolitan City were selected for the study. Total of 490 questionnaires were delivered and collected, and 445 of them were analyzed by using SPSS Win Ver 12.0. Percentages of students who checked' don't know the answer' in 20 environmental knowledge assessment questions were $14.4\~41.8\%$, and those who gave correct answer to these responded to how the answer were $40.1\~93.3\%$. The average score of environmental consciousness was 3.45 and that of eco-friendly dietary behavior was 2.9 out of 5. When dietary behavior was divided by three categories, such as food purchasing, eating. and dish washing, the score of food purchasing behavior was 2.7, those of eating behavior and dish washing behavior were 3.0, 3.1, respectively. In the relationship between environmental knowledge. consciousness. and eco-friendly dietary behavior, there was significant positive correlation between howledge and consciousness, but no significant relation between consciousness and behavior. It showed that environmental knowledge and consciousness didn't always lead to eco-friendly dietary behavior. Therefore, education for practical betlavior must be emphasized even though the knowledge and consciousness education on environment are important.
