The formation of insoluble aggregation of the recombinant kringle fragment of human apolipoprotein(a), rhLK8, in endoplasmic reticulum was identified as the rate-limiting step in the rhLK8 secretion in Saccharomyces cerevisiae. To analyze the protein secretion pathway, some of yeast genes closely related to protein secretion was rationally selected and their oligomer DNA were arrayed on the chip. The expression profiling of these genes during the induction of rhLK8 in fermentor fed-batch cultures revealed that several foldases including pdi1 gene were up-regulated in the early induction phase, whereas protein transport-related genes were up-regulated in the late induction phase. The coexpression of pdi1 gene increased rhLK8-folding capacity. Hence, the secretion efficiency of rhLK8 in the strain overexpressing pdi1 gene increased by 2-fold comparing in its parental strain. The oligomer DNA chip arrayed with minimum number of the genes selected in this study could be generally applicable to the monitoring system for the heterologous protein secretion and expression in Saccharomyces cerevisiae. With the optimization of fed-batch culture conditions and the alteration of genetic background of host, we obtained extracellular rhLK8 at higher yields than with Pichia pastoris systems, which was a 25-fold increased secretion level of rhLK8 compared to the secretion level at the initiation of this study.

We study the manufacturers of the astronomical instruments invented as part of the so-called Ganui-Dae (astronomical platform) project in the reign of King Sejong (1418-1450) of the Joseon dynasty. The main purpose of this project was to compile the Chiljeongsan-Naepyeon, a Korean calendar, that lasted for 5.5 years from 1432 to 1438. Through this project, a total of 17 astronomical instruments such as the armillary sphere, celestial globe, and auto-striking clepsydra were developed. It is generally known that most of them were manufactured by Lee Cheon (李蕆, 1376-1451) and Jang Yeong-Sil (蔣英實, fl. 1423-1442). In this study, we investigate the accounts of the Veritable Records of King Sejong, Munjong, and Sejo, focusing on the inscription written out to memorize the completion of the Ganui-Dae project. We found that at least 12 persons took part in manufacturing the astronomical instruments of the Ganui-Dae project. Lee Cheon was involved in the production of four instruments, while Jang Yeong-Sil was involved in two; therefore, it seems that the achievement of Jang Yeong-Sil was overestimated. We also found that Jeong Cho (鄭招, ?-1434) was a scientist and Lee Cheon was an engineer in the early phase of the Ganui-Dae project, while King Sejong and Yi Sun-Ji (李純之, 1406-1465) played major role in the late phase. According to our study, King Sejong and Yi Sun-Ji were involved in the production of at least seven and five instruments, respectively. In conclusion, we believe that this study will be helpful to understand the practical manufacturers of the astronomical instruments invented during the Ganui-Dae project in the Joseon dynasty.
Objectives : This study was performed to evaluate the general distribution and the efficacy of oriental medical treatment for low back pain. Methods: We have selected of the moxa bucket moxibustion. We make a comparative study of the thermodynamic characteristics of moxa bucket moxibustion. We examined combustion times, temperatures, temperature gradients in each period during a combustion of moxa bucket moxibustion made by oak wood. Results : The 80 cases of low back pain patients was analyzed according to the distribution of sex, age, the period of disease, contributing factor, the patient-condition on admission, the symptom on admission, the duration of admission, the reading of the X-ray and the treatment efficacy was evaluated respectively. 1. Female was more than male in the ratio of 1:1.2, and thirties and seventies 25% the most, the acutest phase 46.3% the most, reason unknown 33.8% the most, Grade III 50% the most, L.B.P. only 51.3% the most, the day of 6-10 30% the most, compression Fx 30% the most. 2. In the total treatment result, the good was 51.3%, the excellent 22.5%, the fair 17.5%, the poor 8.8% in order. About 'the effective rate'(the percentage of positive effective treatment cases) of each distribution, the fifties 100% the most, the acutest phase 97.3% the most, the slip down, weight lifting, overuse 100% the most respectively, Grade III 97.5% the most, L.B.P. only 95.1% the most, the day of 11-15 100% the most, HN.P. 93.3% the most. Conclusions : We have known the efficacy of oriental medical treatment for L.B.P. was good and early treatment was better than late treatment and main cause of L.B.P. was degenerative change.
To elucidate the role of tow environmental factors, temperatures and photoperiod, in diapause induction and development of the dark grey cutworm, Agrotis tokionis B., field and laboratory experiments were implemented. As larvae entered diapause later, the diapause duration became shorter resulting in the synchronization of pupation of the test population. The summer diapause of this insect was assumed to be obligatory because every mature larvae had to experience summer dormant period at all experimental conditions of temperature and photoperiod. It was suggested that the diapause stage could be divided into two different phases in relation to temperature reaction. as the temperatures examined, the duration of diapause was shortened by higher temperature in the early phase, while it was shortened by lower temperature in the late phase. Th diapause period was the shortest under short-day condition (LD 8:16)
Insects are a valuable natural source that can produce a variety of bioactive compounds due to their increasing species diversity. CopA3 is an antimicrobial peptide derived from Copris tripartitus (i.e., the dung beetle). It is known to increase the proliferation of colonic epithelial and neuronal stem cells by regulating cell cycle. This research hypothesized that CopA3 can promote the proliferation of porcine muscle satellite cells (MSCs). The effects of CopA3 on porcine MSCs, which are important for muscle growth and regeneration, remain unclear. Here, we investigated the effects of CopA3 on porcine MSCs. According to viability results, we designed four groups: control (without CopA3) and three treatment groups (treated with 5,10, and 25 ㎍/mL of CopA3). At a CopA3 concentration of 5 ㎍/mL and 10 ㎍/mL, the proliferation of MSCs increased more than that observed in the control group. Furthermore, compared to that in the control, CopA3 treatment increased the S phase but decreased the G0/G1 phase ratio. Additionally, early and late apoptotic cells were found to be decreased in the 5 ㎍/mL group. The expressions of the myogenesis-related transcription factor PAX7 and MYOD proteins were significantly upregulated in the 5 ㎍/mL and 10 ㎍/mL groups, whereas the MYOG protein remained undetected in all group. This study suggested that CopA3 promotes muscle cell proliferation by regulating the cell cycle of MSCs and can regulate the activity of MSCs by increasing the expressions of PAX7 and MYOD.

To establish a culture system with enhanced cellular nutrition, we investigated the effects of light quality (blue, 450 nm; yellow, 590 nm; and red, 630 nm) of a light-emitting diode (LED) on the biochemical composition of Tetraselmis suecica and T. tetrathele. The protein content of both species was higher (42-69%) than the content of other biochemical substances under all wavelengths. Carbohydrate, protein, and lipid contents were higher under the yellow wavelength, which showed a low growth rate, than those under other wavelengths. The contents of all biochemical substances were low under the red wavelength, which showed a high growth rate. These results indicated that protein synthesis occurs in response to decreased cell division rate, while lipid and carbohydrate synthesis occurs owing to altered chemical composition and enzymatic activity. Therefore, we suggested a two-phase LED culture system, which emitted red LED during the early-middle exponential phase and yellow LED during the late exponential and stationary phases, to increase the yield of useful biochemical substances of T. suecica and T. tetrathele.

Although the migration of hepatic stellate cells (HSCs) is important for hepatic fibrosis, the regulation of this migration is poorly understood. Notably, transforming growth factor (TGF)-β1 induces monocyte migration to sites of injury or inflammation during the early phase, but inhibits cell migration during the late phase. In the present study, the role of transforming protein RhoA signaling in TGF-β1-induced HSC migration was investigated. TGF-β1 was found to increase the protein and mRNA levels of smooth muscle actin and collagen type I in HSC-T6 cells. The level of RhoA-GTP in TGF-β1-stimulated cells was significantly higher than that in control cells. Furthermore, the phosphorylation of cofilin and formation of filamentous actin (F-actin) were more marked in TGF-β1-stimulated cells than in control cells. Additionally, TGF-β1 induced the activation of nuclear factor-κB, and the expression of extracellular matrix proteins and several cytokines in HSC-T6 cells. The active form of Rap1 (Rap1 V12) suppressed RhoA-GTP levels, whereas the dominant-negative form of Rap1 (Rap1 N17) augmented RhoA-GTP levels. Therefore, the data confirmed that Rap1 regulated the activation of RhoA in TGF-β1-stimulated HSC-T6 cells. These findings suggest that TGF-β1 regulates Rap1, resulting in the suppression of RhoA, activation of and formation of F-actin during the migration of HSCs.
The Geumhwa Au-Ag deposit is located within the Cretaceous Gyeongsang basin. Mineral paragenesis can be divided into two stages (stage I and II) by major tectonic fracturing. Stage II is economically barren. Stage I, at which the precipitation of major ore minerals occurred, is further divided into three substages(early, middle and late) with paragenetic time based on minor fractures and discernible mineral assemblages: early substage, marked by deposition of pyrite with minor wolframite; middle substage, characterized by introduction of electrum and base-metal sulfides with Cu-As and/or Cu-Sb sulfosalts; late substage, marked by hematite and Bi-sulfosalts with secondary minerals. Changes in vein mineralogy reflect decreases in temperature and sulfur fugacity with a concomitant increase in oxygen fugacity. Fluid inclusion data indicate progressive decreases in temperature and salinity within each substage with increasing paragenetic time. During the early portion of stage I, high-temperature (≥410℃), high-salinity fluids (up to ≈44 equiv. wt. % NaCl) formed by condensation during decompression of a magmatic vapor phase. During waning of early substage, high-temperature, high-salinity fluids gave way to progressively cooler, more dilute fluids associated with main Au-Ag mineralization (middle) and finally to ≈180℃ and ≥0.7 equiv. wt. % NaCl fluids associated with hematite and sulfosalts (± secondary) mineralization (late substage). These trends are interpreted to indicate progressive mixing of high- and medium to low-salinity hydrothermal fluids with cooler, more dilute, oxidizing meteoric waters. The Geumhwa Au-Ag deposit may represent a vein-type system transitional between porphyry-type and epithermal-type.

The global navigation satellite system (GNSS), which is the core technique for the location based service, adopts the direct sequence/spread spectrum (DS/SS) as its modulation method. The success of a DS/SS system depends on the synchronization between the received and locally generated pseudo noise (PN) signals. As a step in the synchronization process, the tacking scheme performs fine adjustment to bring the phase difference between the two PN signals to zero. The most widely used tracking scheme is the delay locked loop with early minus late discriminator (EL-DLL). In the ideal case, the EL-DLL is the best estimator among various DLL. However, in the band-limited multipath environment, the EL-DLL has tracking bias. In this paper, the timing offset range of correlation function is divided into advanced offset range (AOR) and delayed offset range (DOR) centering around the correct synchronization time point. The tracking bias results from the following two reasons: symmetry distortion between correlation values in AOR and DOR, and mismatch between the time point corresponding to the maximum correlation value and the synchronization time point. The former and latter are named as the type I and type II tracking bias, respectively. In this paper, when the receiver has finite bandwidth in the presence of multipath signals, it is shown that the type II tracking bias becomes a more dominant error factor than the type I tracking bias, and the correlation values in AOR are not almost changed. Exploiting these characteristics, we propose a novel tracking bias mitigation scheme and demonstrate that the tracking accuracy of the proposed scheme is higher than that of the conventional scheme, both in the presence and absence of noise.
This experiment evaluated the effects of dietary lysine restriction and energy density on growth performance, nutrient digestibility and meat quality of finishing pigs. A $2{\times}2$ factorial arrangement of treatments was utilized in a randomized complete block (RCB) design, and factor 1 was lysine restriction and factor 2 was energy density. The control diet was formulated to contain 3.265 Mcal of ME/kg, 0.75% lysine in the early-finishing phase and 3.265 Mcal of ME/kg, 0.60% lysine in the late-finishing phase and other nutrients met or exceeded NRC (1998) standards. Compared to the control diet (CON), lysine levels of experimental diets were restricted to 15% (treatment EL, EEL) or 30% (treatment ELL, EELL), whereas energy level of experimental diets was increased by 0.100 or 0.200 Mcal of ME/kg. A total of 100 crossbred pigs ([Yorkshire${\times}$Landrace]${\times}$Duroc), with average initial body weight of $58.47{\pm}1.42\;kg$, were allotted to 5 dietary treatments based on sex and body weight. Each treatment had 5 replicates with 4 pigs (two barrows and two gilts) per pen. ADG, ADFI and feed efficiency were calculated in an 8-week growth trial. In the late finishing period (5-8 weeks), pigs fed ELL or EELL diets had decreased ADG and feed efficiency (p<0.01), however, when the EEL diet was provided, a similar growth performance was observed compared to those fed the CON diet during the whole experimental period (p>0.05). In a metabolic trial, 15 pigs were used to evaluate the effect of dietary lysine restriction and energy density on nutrient digestibility. The digestibility of dry matter, crude fat and crude ash was not improved by restricting dietary lysine or energy density. However, crude protein digestibility was decreased (p<0.05) as dietary lysine was restricted. When dietary lysine was restricted, fecal nitrogen was increased whereas nitrogen retention was decreased. BUN concentration was affected by dietary lysine restriction; treatments ELL and EELL had higher BUN values than other treatments (p<0.01). Carcass characteristics and meat quality were measured when average body weight of pigs reached $107.83{\pm}1.50\;kg$. Treatment ELL had higher last rib backfat depth (p<0.05) than treatment CON, but ELL and EEL did not differ significantly. The ELL and EEL treatments had higher (p<0.05) subjective marbling score than treatment CON. Treatment EEL showed higher longissimus fat content than treatment EL and CON (p<0.01). The results indicated that finishing pigs fed a diet with 15% lysine restriction and 3.465 Mcal of ME/kg energy density had no detrimental effects on growth performance and N utilization, and could achieve substantial increases in marbling and longissimus fat content of pork.
