2, 3, 7, 8-Tetrachlorodibenzo-p-dioxin Induces Recruitment of Shc/Cbl/Grb2/Sos Conplex in Early Signaling Pathway of CYP1A1 Induction in the Primary Culture of Hepatocytes

  • 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) is known to induce cytochrome p450 1A1 and to activate c-Src kinase and p21 Ras. This study examined the molecular interactions of adaptor proteins including Shc, Grb2, and Sos in rat primary hepatocytes and their relationship to the induction of CYP1A1 by TCDD. TCDD induced CYP1A1 level and EROD activity in a dose-dependent mode. Sos/Grb2 association isincreased by TCDDㅑㅜ a dose dependent mode. Tyrosine phosphorylated Shc, mainly p152, onloads to Grb2/Sos complex upon TCDD stimulation. The electrophoretic mobility shift of Sos is showed by TCDD. These results indicate that TCDD modulated the molecular interaction features of adaptor compoes proteins including Shc, Grb2, and Cnl in early signaling pathway of TCDD-mediated CYP 1A1 induction of rat primary hepatocyte.

  • The human baby appears to be born with preexistent knowledge of language. This specialized neural structure in the brain awaits auditory experience with language to trigger it into functioning. The auditory-linked acquisition of language is a time-locked function related to early maturational periods in the infant's life. The longer auditory language stimulation is delayed, the less efficient the language facility will be. It is for these reasons that it is urgent to fight the hearing problems of children with all the skill, knowledge and insights of which we are capable, the so called 'rehabilitative process'. An understanding of the timetable and the origin of prenatal to early life development of auditory mechanism will help in planning the aural rehabilitation. Further interests and studies are needed to establish the systematic structure of rehabilitative audiology.

The Evaluation of Improvement of Bell's Palsy by Yanagihara's System (Yanagihara's System을 이용한 구안와사의 호전도 평가)

  • Objective : The aim of this study was to assess the improvement of Bell's Palsy by Yanagihara's System. Methods : Early admission care, western medicine, acupuncture and electrical stimulation was applied to the 22 patients who dignosisd as Bell's palsy. The effects of these treatment was evaluated by Yanagihara's unweighted grading system. Results : After 25 day treatment the mean values of the Yanagihara's score(26.90±9.53) showed stastically significant improvement in comparison with pre-treatment(8.68±5.13). Conclusion : These results provided that the early admission care, western medicine, acupuncture and electrical stimulation is a valuable treatment for Bell's palsy. Further case control study is need to confirm the effect of above treatment on Bell's palsy.

The Effects of Service Quality and Sensibility Quality on Brand Fanship - For Parents of Early Childhood Education Institutions (서비스품질과 감성품질이 브랜드팬십에 미치는 영향 - 영유아 교육기관 학부모를 대상으로)

  • Purpose: The purpose of this study is to analyze whether service quality and sensibility quality have an effect on securing passionate fans of educational institutions as a management marketing strategy that can maximize parent satisfaction in order to secure a competitive advantage for infant and toddler educational institutions. Methods: In this study, a questionnaire survey was conducted on parents attending educational institutions for infants and toddlers, and hypotheses were verified through frequency analysis, reliability and validity correlation analysis using SPSS 22.0. Results: The results of this study are as follows. First, it was found that service quality has a significant effect on rapport formation. Second, among the sub-factors of emotional quality, stimulation and identity had a significant effect on rapport formation. Third, among the sub-factors of service quality, sensiblity and responsiveness had a significant effect on brand fanship. Fourth, among the sub-factors of sensibility quality, stimulation and identity had a significant effect on brand fanship. Fifth, rapport formation was found to have a significant effect on brand fanship. Conclusion: This study is meaningful as a service and sensibility quality study focused on educational institutions at the present time when research on service quality and sensiblity quality targeting early childhood educational institutions is insufficient. In addition, in terms of management of educational institutions for infants and young children, by paying attention to the formation of a careful and pleasant relationship with the fans of educational institutions, an empirical example of a theoretical research model was shown that strong attachment to educational institutions and positive participation lead to loyal fans.

Changes in Early Postmortem Contents of ATP and Other Nucleotides in Normal and Poor Quality-simulated Pork (정상돈육과 모의 열등돈육의 사후 초기 ATP 및 관련 뉴클레오타이드 함량 변화)

  • Early postmortem ATP level is known as a good predictor of pork quality. Pork carcasses were divided into two; one was electrically stimulated (ES) to simulate poor quality pork and the other was left untreated and served as a control normal pork. Fractions of longissimus were excised from carcasses regularly for 2 hours after death and deep-frozen (80C) until analyses. The ATP level of normal untreated control pork decreased from 5.00 to 2.04 μmole/g within 2 hours postmortem. The decrement of ATP was approximately 60% of its initial content. In the meantime, ES poor quality pork had a more drastic rate of ATP decrease. Electrical stimulation itself decreased ATP level from 4.70 to 3.50 μmole/g, by approximately 25%. ATP level of ES pork dropped to 1.71 μmole/g within 1 hour postmortem and was further plunged to 0.26 μmole/g and almost exhausted during the next hour. The level of IMP increased from 0.49 to 3.17 μmole/g and it became the dominant nucleotide within 2 hours postmortem. Electrical stimulation prompted the increase of IMP from 0.69 to 3.19 μmole/g and its level went up to 6.64 μmole/g within 2 hours postmortem. The level of ADP also decreased from 1.45 to 0.67 μmole/g for 2 hours after death and ES also accelerated ADP breakdown. The AMP levels were lower than those of other nucleotides and increased from 0.16 to 0.31 μmole/g within 2 hours postmortem. The increase of AMP was accelerated between 60 and 90 minutes after electrical stimulation. Early postmortem electrical stimulation prompted a drastic rate of changes in contents of 4 nucleotides during 2 hours postmortem. In the meantime, the ATP levels for ES poor quality pork were much lower than those of normal pork.

The Influence of the Application of NEES to GV20 and L14, on HSP27 and HSP70, after the Ischemic Brain Injury

  • This research was attempted to seek for a positive approach within the framework of physical therapy instead of the drug treatment in the past, with regard to the ischemic brain injury in the early stage. Accordingly, the aim of this research is to observe the change of HSP27 and HSP70, the genes that are expressed in the early stage of brain injury and to investigate the effects of needle electrode electrical stimulation(NEES), upon applying NEES after ischemia. The experimental method is to give rise to global ischemia and apply NEES to 27 SD-Pat rats with the particulars of being eight-week-old, male, around 300g, and adapted to laboratory environment for more than a week, and divide them into three groups, that is, GV20 NEES group(n=9), L14 NEES group(n=9), no applied NEES global ischemia(GI) group(n=9), and then observe their changes of HSP27 and HSP70 at the time lapse of 6, 9 hr and 12 hr after ischemia, using immunohistochemistry methods. Upon observing through the immunohistochemistry method, it was noticed that there was a significant difference between the GV20 NEES group and the L14 NEES group as for HSP27 and there were significant differences among all groups as for HSP70(p<.05). Accordingly, it is supposed that the application of NEES after the outbreak of cerebral ischemia delay the apoptosis in the early ischemic part of forebrain or protect neurons against apoptosis.

Effects of Various Stimulants on Spawning Induction and Early Development at Different Water Temperatures in the Noble Scallop (흔한가리비, Chlamys nobilis의 자극방법별 산란유발 효과와 난 발생에 미치는 수온의 영향)

  • We examined the effects of stimulants including sunlight and UV-irradiation on the spawning induction and early development of the noble scallop, Chlamys nobilis. The sunlight stimulation resulted in nuch faster spawning induction (100% success within 40 minutes) compared to UV-irradiation (100% success within 70 minutes). Early development of the scallop larva took place between 15C to 30C. The time to reach the early D-shaped stage was 63.5, 31.5, 18.5 and 17.0 hours at 15, 20, 25 and 30C, respectively. The correlations between the water temperature-(WT) regimes and the time (t) required for each developmental stage are as follows. 2 cell stage: 1/t=0.0606WT-0.6194 (r2=0.9791) 8 cell stage: 1/t=0.0304WT-0.3453 (r2=0.9941) Morula: 1/t=0.0100WT-0.1049 (r2=0.9663) Trochophore: 1/t=0.0058WT-0.0618 (r2=0.9848) D-shaped larva: 1/t=0.0030WT-0.0282 (r2=0.9731) These correlations indicated that the biological minimum temperature of the species is around 10.44C. The highest survival rate up to D-shaped larva at different water temperature was observed at 25C.

  • Mammalian eggs are ovulated arrested at meiotic metaphase II until fertilization. Generally in mammals, fertilization results in a series of intracellular calcium oscillations that are mediated by inositol triphosphate (IP3) or cyclic adenosine diphosphoribose (cADPr). The high levels of maturation promotion factor (MPF) within the cell decrease, pronuclei form, the cytoskeleton is reorganized and proteins are post-translationally modified. If all is normal, the newly formed embryo initiates the developmental program specific to that species. Artificial methods of producing these effects in pig oocytes are discussed. One potential mechanism mediated via a signal transduction pathway is present in pig oocytes. Stimulation of this pathway leads to the early events following fertilization, and electrical stimulation leads to apparently normal de v velopment to day 12. Further studies are needed to determine which mechanism(s) the sperm uses to initiate development.

  • Spinal cord stimulation (SCS) is used to manage chronic pain syndromes and it is accepted a cost-effective therapy. Child-bearing women who had SCS become or choose to become pregnant despite these policies pregnancy is a relative contraindication. A 32-year-old woman had SCS as a treatment for the CRPS I of the left lower extremity, During various check up tests, we happen to find out that her serum beta-hCG was positive and confirmed pregnancy. SCS is not recommended in pregnancy because the effects of SCS on pregnancy and nursing mothers had not been confirmed. However, many female patients suffering from chronic pain may expect future pregnancy and we think that they must be informed about the possibility of pregnancy and the effects of SCS device implantation in the course of pregnancy. First of all, a good outcome requires a multidisciplinary team approach, including obstetrics, neonatology, pain medicine and anesthesia, as was used from an early pregnancy. Unfortunately, she had a misabortrion after 6 weeks.

The Effect of NEES on the Occurrence of Caspase-3 in the Cerebellum of Rats with Transient Global Ischemia

  • The cerebellum is known to control balance, equilibrium, and muscle tone. If the cerebellum becomes damaged, the body is unable to retain its balancing functions or involuntary muscle movement. This is why, in stroke patients, there is a high risk of functional disability, as well as a myriad of other disabilities secondary to stroke. Ischemia was induced in SD mice by occluding the common carotid artery for 5 minutes, after which blood was reperfused. Needle electrode electrical stimulation(NEES) was applied to acupuncture points, at 12, 24, and 48 hours post-ischemia on the joksamri. Protein expression was investigated through caspase-3 antibody immuno-reactive cells in the cerebral nerve cells and Western blotting. The results were as follows: The number of caspase-3 reactive cells in the corpus cerebellum 12 and 24 hours post-ischemia was significantly (p<.05) smaller in the NEES group compared to the GI group. caspase-3 expression 12 and 24 hours post-ischemia was significantly(p<.05) smaller in the NEES group compared to the GI group. Based on these results, NEES seems to have a significant effect on Caspase-3 in the cerebellum in an ischemic state at 12 and 24 hours post ischemia, NEES delays the occurrence of early stage apoptosis-inducing Caspase-3, delaying and inhibiting apoptosis. Further systematic studies will have to be conducted in relation to the application of this study's results on stroke patients.
