Consequences of Post-grazing Residues Control and Birth Season on the Body Traits, Reproductive Performance and Offspring's Growth of Suckling Goats and Ewes Reared at Pasture in Guadeloupe (FWI)

  • In Guadeloupe small ruminants (SR) are reared for meat production under pasture conditions. Intensive rotational grazing systems (irrigated, fertilised and high stocked) allow reasonable levels of production but generate high post-grazing residues. Experiments were designed to control them. A system in which residuals were mown (RM) was tested in comparison to the control system (Residuals Remained, RR). The same design was carried out for two years with Creole goat (G) and Martinik sheep (S). An accelerated reproductive rate (3 parturitions over 2 years) was carried out. Systems were compared at three parturition seasons per year(dry, intermediate and rainy seasons). Each group was composed of 20 goats ($36.0{\pm}2.5kg$) or 20 ewes ($46.8{\pm}2.4kg$). The female body traits did not vary according to pasture management and seasons. The stocking rate averaged 1,400 kg LW/ha. The mean fertility rate for does varied significantly (p<0.05) within the kidding season, from $80.4{\pm}0.5%$ to $93.7{\pm}2.9%$ while the mean litter size was $2.30{\pm}0.07$ total kids born. No effect of pasture system was observed. Corresponding values for ewes were $83.2{\pm}12.8%$ vs. $75.6{\pm}12.5%$ (p<0.05) and $2.43{\pm}0.24$ vs. $2.03{\pm}0.29$ (p<0.01) total lambs born for SRM and SRR ewes, respectively. A seasonal effect was observed upon ewe performances. The preweaning mortality of kids and lambs averaged 16.3% and 14.4%, respectively. It was 7 and 9 percentage points more (p<0.01) for RR than for RM kids and lambs, respectively. For both species, weaning took place at an average age of $81.4{\pm}3.6days$. In Creole kids, live weight at birth and at weaning were $1.9{\pm}0.2kg$ and $8.9{\pm}0.8kg$, respectively. In the Martinik sheep, the traits averaged $2.9{\pm}0.2kg$ and $18.9{\pm}0.9kg$. For both traits in both species, significant (p<0.05) group${\times}$season interactions were recorded. The consequences of elimination of post-grazing residues varied according to the SR species, the environmental conditions and the animal physiological status. The forage characteristics were not limiting factors since forage availability in the RM systems (2,300 and 2,600 kg DM/ha, respectively) and chemical composition were at satisfactory levels (CP content averaged 12 and 10%). It is recommended to develop new grazing system which would allow the use of post-grazing residues instead of mowing the refusals.

Effect of Dietary Antimicrobials on Immune Status in Broiler Chickens

  • This study evaluated the effects of dietary anticoccidial drugs plus antibiotic growth promoters (AGPs) on parameters of immunity in commercial broiler chickens. Day-old chicks were raised on used litter from a farm with endemic gangrenous dermatitis to simulate natural pathogen exposure and provided with diets containing decoquinate (DECX) or monensin (COBN) as anticoccidials plus bacitracin methylene disalicylate and roxarsone as AGPs. As a negative control, the chickens were fed with a non-supplemented diet. Immune parameters examined were concanavalin A (ConA)-stimulated spleen cell proliferation, intestine intraepithelial lymphocyte (IEL) and spleen cell subpopulations, and cytokine/chemokine mRNA levels in IELs and spleen cells. ConA-induced proliferation was decreased at 14 d post-hatch in DECX-treated chickens, and increased at 25 and 43 d in COBN-treated animals, compared with untreated controls. In DECX-treated birds, increased percentages of $MHC2^+$ and $CD4^+$ IELS were detected at 14 d, but decreased percentages of these cells were seen at 43 d, compared with untreated controls, while increased $TCR2^+$ IELs were evident at the latter time. Dietary COBN was associated with decreased fractions of $MHC2^+$ and $CD4^+$ IELs and reduced percentages of $MHC2^+$, $BU1^+$, and $TCR1^+$ spleen cells compared with controls. The levels of transcripts for interleukin-4 (IL-4), IL-6, IL-17F, IL-13, CXCLi2, interferon-${\gamma}$ (IFN-${\gamma}$), and transforming growth factor${\beta}$4 were elevated in IELs, and those for IL-13, IL-17D, CXCLi2, and IFN-${\gamma}$ were increased in spleen cells, of DECX- and/or COBN-treated chickens compared with untreated controls. By contrast, IL-2 and IL-12 mRNAs in IELs, and IL-4, IL-12, and IL-17F transcripts in spleen cells, were decreased in DECX- and/or COBN-treated chickens compared with controls. These results suggest that DECX or COBN, in combination with bacitracin and roxarsone, modulate the development of the chicken post-hatch immune system.

Protein Profile in Corpus Luteum during Pregnancy in Korean Native Cows

  • Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism, but the profile of proteins associated with progesterone synthesis in cyclic and pregnant corpus luteum (CL) is not well-known in cattle. In Experiment 1, plasma progesterone level was monitored in cyclic cows (n = 5) and pregnant cows (n = 6; until d-90). A significant decline in the plasma progesterone level occurred at d-19 of cyclic cows. Progesterone level in abbatoir-derived luteal tissues was also determined at d 1 to 5, 6 to 13 and 14 to 20 of cyclic cows, and d-60 and -90 of pregnant cows (n = 5 each). Progesterone level in d-60 CL was not different from those in d 6 to 13 CL and d-90 CL, although the difference between d 6 to 13 and d-90 was significant. In Experiment 2, protein expression pattern in CL at d-90 (n = 4) was compared with that in CL of cyclic cows at d 6 to 13 (n = 5). Significant changes in the level of protein expression were detected in 32 protein spots by two-dimensional polyacrylamide gel electrophoresis (2-DE), and 23 of them were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Six proteins were found only in pregnant CL, while the other 17 proteins were found only in cyclic CL. Among the above 6 proteins, vimentin which is involved in the regulation of post-implantation development was included. Thus, the protein expression pattern in CL was disorientated from cyclic luteal phase to mid pregnancy, and alterations in specific CL protein expression may contribute to the maintenance of pregnancy in Korean native cows.

The study on enhanced micro climate of the oyster mushroom cultivation house with multi-layered shelves by using CFD analysis (CFD 분석에 의한 느타리버섯 재배사 환경균일성 향상 연구)

  • The oyster mushroom cultivation house typically has multiple layers of growing shelves that cause the disturbance of air circulation inside the mushroom house. Due to this instability in the internal environment, growth distinction occurs according to the area of the growing shelves. It is known that minimal air circulation around the mushroom cap facilitates the metabolism of mushrooms and improves their quality. For the purpose of this study, a CFD analysis FLUENT R16 has been carried out to improve the internal environment uniformity of the oyster mushroom cultivation house. It is found that installing a section of the working passage towards the ceiling is to maintain the internal environment uniformity of the oyster mushroom cultivation house. When all the environment control equipment - including a unit cooler, an inlet fan, an outlet fan, an air circulation fan, and a humidifier - were operated simultaneously, the reported Root Mean Square (RMS) valuation the growing shelves were as follows: velocity 23.86%, temperature 6.08%, and humidity 2.72%. However, when only a unit cooler and an air circulation fan operated, improved RMS values on the growing shelves were reported as follows: velocity 23.54%, temperature 0.51%, and humidity 0.41%. Therefore, in order to maintain the internal environment uniformity of the mushroom cultivation house, it is essential to reduce the overall operating time of the inlet fan, outlet fan, and humidifier, while simultaneously appropriately manage the internal environment by using a unit cooler and an air circulation fan.

Characteristics of the SrBi2Nb2O9 Thin Films Deposited by RF Magnetron Sputtering with Controlling of Bi Contents (RF마그네트론 스퍼터링 법에 의해 증착된 SrBi2Nb2O9 박막의 Bi 량의 조절에 따른 특성분석)

  • The $SrBi_2$$Nb_2$$O_{9}$ (SBN) thin films were deposited with $SrNb_2$$O_{6}$ / (SNO) and $Bi_2$$O_3$ targets by co-sputtering method. For the growth of SBN thin films, we adopted the various power ratios of two targets; the power ratios of the SNO target to $Bi_2$$O_3$ target were 100 W : 20 W, 100 W : 25 W, and 100 W : 30 W during sputtering the SBN films. We found that the electrical properties of SBN films were greatly dependent on Bi content in films. The $Bi_2$Pt and $Bi_2$$O_3$ phase as second phases occurred at the films with excess Bi content greater than 2.4, resulting in poor ferroelectric properties. The best growth condition of the SBN films was obtained at the power ratio of 100 W : 25 W for the two targets. At this condition, the crystallinity and electrical properties of the films were improved at even low annealing temperature as $700^{\circ}C$ for 1h in oxygen ambient and the Sr, Bi and Nb component in the SBN films were about 0.9, 2.4, and 1.8 respectively. From the P-E and I-V curves for the specimen, the remnant polarization value ($2P_{r}$) of the SBN films was obtained about 6 $\mu$C/c $m^2$ at 250 kV/cm and the leakage current density of this thin film was $2.45$\times$10^{-7}$ $A/cm^2$ at an applied voltage of 3 V.V.

EFFECT OF NITROGEN AND AGE OF ALFALFA (Medicago sativa L. ) SEEDING ON GROWTH AND NODULATION WHEN GROWN UNDER A HOT ENVIRONMENT (고온하에서 질소시비가 근류균을 접종한 Alfalfa의 생육부위에 미치는 영향에 관하여)

  • There are differences in opinion as to whether nitrogen fertilizer should be used when establishing alfalfa (Medicago sativa L.). Various reports show that under a hot environment, rhizobia (Rhizobium meliloti) are not as effective in fixing atmospheric nitrogen as they are under moderate temperatures. It is also believed that the addition of nitrogen fertilizer inhibits nodulation of alfalfa seedlings. A replicated experiment was conducted under controlled environmental conditions at the University of Nevada-Reno, Reno, Nevada, USA, to determine the effects of nitrogen application on seedling growth and nodulation of alfalfa grown in a hot environment. Sterile sand was used as the growing media to which a complete nutrient solution minus nitrogen was applied volumetrically to each pot daily. In addition, half of the pots received NH4-$NO_3$, at the rate of 11.2 kg per ha at seeding and at two and four weeks after planting giving a total nitrogen application rate of 33.6 kg per ha during the seven-week experimental period. Rhizobia inoculant (R-12) consisted of a mixture of strains 171-15a, 1682c and 80 PI 265 of (Rhizobium meliloti). Inoculant was applied to the seeds prior to planting and to the sand media at two and four weeks after seeding. Twenty seeds were planted in pots 14.0 cm in diameter and 11.5 cm deep. Plants were thinned to ten plants per pot after emergence and were grown in a controlled environment chamber with a 16-hour light period. Soil temperature at 6 cm depth ranged from 17.4^{\circ}C.$ to 31.1^{\circ}C.$ and had a daily mean of 26.5^{\circ}C.$. Plants were harvested at weekly intervals for seven weeks. Root, shoot and total length, dry weight, volume and number of nodules per plant were determined. Root, shoot, and total length were greater in seedlings grown in soil where nigrogen was applied than that grown in soil to which no nitrogen was applied. The average size of the seedlings as determined by volume and weight was more than two times greater where plants were fertilized with nitrogen. Nodule number per seedling was also greater when nitrogen was applied compared to those which received no nitrogen. The differences were greater as the plants became older. The rhizobia did not fix enough nitrogen for adequate growth of seedlings. This is probably due to high temperature growing conditions that caused the rhizobia to become relatively ineffective as compared to cooler growing conditions. Data suggests it would be desirable to apply nitrogen at seeding when alfalfa is established under hot conditions that occur in mid- or late summer.

Influence of Filling Density in the Positive Active-material on the Cycle-life Performance of the Tubular Type Gelled Valve Regulated Lead Acid Batteries (튜브식 겔형 납축전지에 있어서 활물질 밀도에 따른 싸이클 수명 특성)

  • The characteristics of VRLA (valve regulated Iead-acid) battery with the tubular positive plate and gel type electrolyte were examined as a function of active material filling density. The filling density of positive plate was 3.2 g/mL, 3.4 g/mL, and 3.6 g/mL, respectively. These VRLA batteries were cycled with 100% DOD (depth of discharge) at the $C_5/5$ rate, followed by IU-type recharge with $I_{max}=0.2C_{10}/10$ and a final voltage V=2.40 V/cell. The test was performed in a thermostatic room at $25{\pm}1^{\circ}C$. The result indicated that the initial capacity was independent of active material filling density, i.e., the highest initial capacity was 3.4 g/mL of filling density and the lowest was 3.6 g/mL. On aspect of the cycle-life performance of the VRLA battery, the filling density of 3.6 g/mL was similar to that of 3.4 g/mL in the positive plate, and both were higher than that of 3.2 g/mL. Water-loss and degradation of the VRLA battery were decreased according to an increase of the filling density in the positive plate. The optimum filling density of the active material was 3.4~3.6 g/mL.

A ubiquitin-proteasome system as a determination factor involved in methylmercury toxicity

  • The methylmercury (MeHg) is a toxic environmental pollutant, causing serious neurological and developmental effects in humans. Recent epidemiological studies have indicated that ingestion of MeHg in fish during pregnancy can result in neuroethological effects in the offspring. However, the mechanism underlying the MeHg-toxicity is not fully understood. To elucidate the mechanisms of toxicity of MeHg and of defense against MeHg, we searched for factors that determine the sensitivity of yeast cells to MeHg, and found that overexpression of Cdc34, a ubiquitin-conjugating enzyme (E2) that is a component of the ubiquitin-proteasome (UP) system, induces a resistance to MeHg toxicity in both yeast and human cells. The UP system is involved in the intracellular degradation of proteins. When Cdc34 is overexpressed in cells, ubiquitination reactions are activated and the degradation of certain proteins by the UP system is enhanced. Therefore, it seems likely that certain as-yet-unidentified proteins that increase MeHg toxicity might exist in cons and that toxicity might be reduced by the enhanced degradation of such proteins, mediated by the UP system, when Cdc34 is overexpressed. SCF ubiquitin-ligase is a component of UP system and consists of Skpl, the scaffold protein Cdc53, the RING-finger protein Hrt1, and one member of the family of F-box proteins. The F-box proteins directly bind to the substrates and are the determinants of substrate specificity of SCF. Therefore, we searched for the f-box protein that cofers resistance to MeHg, and found that overexpression of Hrt3 or Yi1224w induced resistance to MeHg toxicity in yeast cells. Since the protein(5) that enhance toxicity of MeHg might plausibly be induced in substrates of both f-box proteins, we next searched for substrate proteins that are recognized by Hrt3 or Y1r224w using two-hybrid screen. We found that Did3 or Crsl interacts with Hrt3; and Eno2 interacts with Yir224w. The yeast cells that overexpressed each those proteins showed hypersensitivity to MeHg, respectively, indicating that those proteins enhance the MeHg toxicity. Both Dld3 and Eno2 are proteins involved in the synthesis of pyruvate, and overexpression of both proteins might induce increase in interacellular levels of pyruvate. Deletion of Yi1006w that transports pyruvate into the mitochondria induced aresistance to MeHg. These results suggest that the promotion of the pyruvate irdlowinto the mitochondria might enhance MeHg toxicity. This study providesimportant keyfor the elucidauon of the molecular mechanism of MeHg toxicity.

Prediction of SWAT Stream Flow Using Only Future Precipitation Data (미래 강수량 자료만을 이용한 SWAT모형의 유출 예측)

  • Much attention has been needed in water resource management at the watershed due to drought and flooding issues caused by climate change in recent years. Increase in air temperature and changes in precipitation patterns due to climate change are affecting hydrologic cycles, such as evaporation and soil moisture. Thus, these phenomena result in increased runoff at the watershed. The Soil and Water Assessment Tool (SWAT) model has been used to evaluate rainfall-runoff at the watershed reflecting effects on hydrology of various weather data such as rainfall, temperature, humidity, solar radiation, wind speed. For bias-correction of RCP data, at least 30 year data are needed. However, for most gaging stations, only precipitation data have been recorded and very little stations have recorded other weather data. In addition, the RCP scenario does not provide all weather data for the SWAT model. In this study, two scenarios were made to evaluate whether it would be possible to estimate streamflow using measured precipitation and long-term average values of other weather data required for running the SWAT. With measured long-term weather data (scenario 1) and with long-term average values of weather data except precipitation (scenario 2), the estimate streamflow values were almost the same with NSE value of 0.99. Increase/decrease by ${\pm}2%$, ${\pm}4%$ in temperature and humidity data did not affect streamflow. Thus, the RCP precipitation data for Hongcheon watershed were bias-corrected with measured long-term precipitation data to evaluate effects of climate change on streamflow. The results revealed that estimated streamflow for 2055s was the greatest among data for 2025s, 2055s, and 2085s. However, estimated streamflow for 2085s decreased by 9%. In addition, streamflow for Spring would be expected to increase compared with current data and streamflow for Summer will be decreased with RCP data. The results obtained in this study indicate that the streamflow could be estimated with long-term precipitation data only and effects of climate change could be evaluated using precipitation data as shown in this study.

Applied Analysis for Metabolic Profiling of Trace-level Amino Acid in Biological Fluid (생체시료 중 미량 아미노산 대사 프로필을 위한 분석법 응용)

  • The universality of low molecular weight metabolites (i.e. amino acids, steroid hormones) allows rapid and straightforward investigation of biochemistry of genetically un-characterized species. Thus in vivo metabolic profiling of amino acid in combination with multivariate data analysis (metabolomics) offers great potential in comparative biology. In this paper, amino acid profiles in biological fluid (media) were studied by using HPLC/FLD. HPLC procedure for amino acids require the formation of derivatives due to the low absorption of the free compounds. o-Phthalaldehyde (OPA) used in association with a thiol, such as 3-mercaptopropionic acid (3-MPA), is one of the most popular and sensitive reagents, which yield quickly fluorescent iso-indoles at room temperature. To improve unstability of OPA/3-MPA derivatization, we optimized injector programs for fixed injection times. Linear regressions for the standard curves were linear in the range 0.5 - 100.0 ppb, giving correlation coefficents above 0.99. The detection limit were 1.70 pmol(GLU) - 23.81 pmol(SER). It is practically useful when the amount of sample is very low on single cells.
