Left ventricular dysfunction measured by tissue Doppler imaging and strain rate imaging in hypertensive adolescents (고혈압 청소년에서 tissue Doppler imaging과 strain rate imaging을 이용한 좌심실 기능 이상에 대한 연구)

  • Purpose: Left ventricular (LV) hypertrophy and impaired diastolic function may occur early in systemic hypertension. Diastolic dysfunction is associated with increased cardiovascular risk. Tissue Doppler imaging (TDI)-derived tissue velocity and strain rate are new parameters for assessing diastolic dysfunction. The aim of this study is to determine whether TDI and strain rate imaging (SRI) would improve the ability to recognize early impaired diastolic and systolic functions compared with conventional echocardiography in hypertensive adolescents. Methods: We included 38 hypertensive patients with systolic blood pressure above 140 mmHg or diastolic blood pressure above 90 mmHg. Ejection fraction and myocardial performance index (MPI) were estimated by conventional echocardiography. Peak systolic myocardial velocity, early diastolic myocardial velocity (Em), and peak late diastolic myocardial velocity (Am) were obtained by using TDI and SRI. Results: In the hypertensive group, interventricular septal thickness was significantly increased on M-mode echocardiography. Em/Am was significantly decreased at the mitral valve annulus. Among hypertensive subjects, the E strain rate at basal, mid, and apex was significantly decreased. Systolic strain was significantly decreased at the septum in the hypertensive group. Conclusion: Strain rate might be a useful new parameter for the quantification of both regional and global LV functions and could be used in long-term follow up in hypertensive patients. Early identification by SRI of subjects at risk for hypertensive and ventricular dysfunction may help to stratify risk and guide therapy. Further studies, including serial assessment of LV structure and function in a larger number of adolescents with hypertension, is necessary.

  • 1. In the tortoise, Amyda japonica, a cold-blooded animal readily available in this country, the role of catecholamines in the regulation of free fatty acids(FFA) metabolism was investigated in both in vivo and in vitro studies. 2. Norepinephrine elevated both FFA and glucose levels in plasma. 3. When $50{\mu}g/kg$ of Epinephrine, Norepinephrine and Isopropylarterenol were administered intravenously, the relative effectiveness of mobilizing FFA was in the descending order of potency-Epinephrine, Norepinephrine and Isopropylarterenol. 4. In order to exclude the 'tonic influence of the endogenous catecholamines', reserpine was given to some animals. Two days after the reserpine-treatment, glucose showed a significant increase over the solely vehicle treated controls, FFA but an insignificant one. Excised auricles from those animals showed a diminished response to tyramine. Seven days after the treatment, however, when the depletion of catecholamines from the tissue stores seemed to be complete, judged from the absence of the response of isolated auricles to tyramine, both FFA and glucose levels were definitely lowered. 5. In in vitro experiments Epinephrine enhanced the FFA-release from the adipose tissue. The effect increased proportionately with the concentration until a maximal effect was attained at a concentration of 1x $10^5$ g/ml. 6. The order of potency in releasing FFA from adipose tissue in vitro was the same as in vitro, i.e., Epinephrine, Norepinephrine and Isopropylarterenol, but the differences were much less marked. 7. Ergotamine exerted no lipolytic action, but inhibited the lipolytic effect ef Epinephrine significantly. 8. Nethalide showed a slight lipolytic effect per se but inhibited the Epinephrine-induced lipolysis significantly. 9. Catecholamines play an important role in regulating FFA metabolism in the cold-blooded animal, just as in the warm-blooded animals, and the tortoise, Amyda japonica, may be used in the studies of fat metabolism as well as the rat.

  • 5-Hydroxytryptamine (5-HT), a monoamine, as a local regulator in the mammary gland is a chemical signal produced by the mammary epithelium cell. In cows, studies have shown that 5-HT is associated with epithelial cell apoptosis during the degenerative phase of the mammary gland. However, studies in other tissues have shown that 5-HT can effectively promote cell viability. Whether 5-HT could have an effect on mammary cell viability in dairy cows is still unknown. The purpose of this study was to determine: (1) effect of 5-HT on the viability of bovine mammary epithelial cells and its related signaling pathways, (2) interaction between prolactin (PRL) and 5-HT on the cell viability. The bovine mammary alveolar cell-T (MAC-T) were cultured with different concentrations of 5-HT for 12, 24, 48 or 72 hours, and then were assayed using cell counting kit-8, polymerase chain reaction (PCR) and immunobloting. The results suggested that 20 μM 5-HT treatment for 12 or 24 h promote cell viability, which was mainly induced by the activation of 5-HT receptor (5-HTR) 1B and 4, because the increase caused by 5-HT vanished when 5-HTR 1B and 4 was blocked by SB224289 and SB204070. And protein expression of mammalian target of rapamycin (mTOR), eukaryotic translation elongation factor 2 (eEF2), janus kinase 2 (JAK2) and signal transducer and activator of transcription 5 (STAT5) were decreased after blocking 5-HT 1B and 4 receptors. When MAC-T cells were treated with 5-HT and PRL simultaneously for 24 h, both the cell viability and the level of mTOR protein were significantly higher than that cultured with 5-HT or PRL alone. In conclusion, our study suggested that 5-HT promotes the viability of MAC-T cells by 5-HTR 1B and/or 4. Furthermore, there is a reciprocal relationship between PRL and 5-HT.

De Novo Assembly and Comparative Analysis of the Enterococcus faecalis Genome (KACC 91532) from a Korean Neonate

  • Using a newly constructed de novo assembly pipeline, finished genome level assembly had been conducted for the probiotic candidate strain E. faecalis KACC 91532 isolated from a stool samples of Korean neonates. Our gene prediction identified 3,061 genes in the assembled genome of the strain. Among these, nine genes were specific only for the E. faecalis KACC 91532, compared with all of the four known reference genomes (EF62, D32, V583, OG1RF). We identified genes related to phenotypic characters and detected E. faecalis KACC 91532-specific evolutionarily accelerated genes using dN/dS analysis. From these results, we found the potential risk of KACC 91532 as a useful probiotic strain and identified some candidate genetic variations that could affect the function of enzymes.

Effect of Hemodialysis on Left and Right Ventricular Volume and Function (말기신질환에서 혈액투석에 따르는 좌우심실용적 및 기능변화에 관한 연구)

  • With the improvement of hemodialysis, the course of the disease in patient with endstage renal disease has been clearly improved. Nevertheless, among several shortcomings to our present mode of renal replacement therapy, cardiovascular complications have been the leading cause of morbidity and mortality. Several factors such as anemia, arteriovenous shunting of blood, intermittent extracorporeal circulation and hypertension may be contributing. But little is known about the quantitative cardiac hemodynamic characteristics occurred during hemodialysis. The purpose of this study is to observe the sequential hemodynamic changes before, during and after the hemodialysis and to investigate reliable parameters in the detection of ventricular dysfunction. In the present study, equilibrium radionuclide cardiac angiography was performed and left and right ventricular volume indices, ejection phase indices of both ventricular, performance were measured in the 16 stable patients with chronic renal failure treated with maintenance hemodialysis sequentially i.e. before, during (early and late phase) and after the hemodialysis. The results obtained were as follows; 1) The indices of the left ventricular function were not changed during the hemodialysis but increased after the hemodialysis. 2) The indices of the right ventricular function(EF, SVI) were significantly decreased in the early phase (15, 30 minutes after starting extracorporeal circulation) but recovered after the hemodialysis. 3) The ratio of right ventricular to left ventricular ejection fraction was significantly decreased in the early phase and the lung volume indices were significantly increased at the same phase. As a conclusion, hemodialysis improves left ventricular function maybe due to increased contractility, and effects on the right ventricular function maybe due to the increased lung volume in the early phase of hemodialysis.

LETM1 Promotes Gastric Cancer Cell Proliferation, Migration, and Invasion via the PI3K/Akt Signaling Pathway

  • Purpose: Globally, there is a high incidence of gastric cancer (GC). Leucine zipper-EF-hand containing transmembrane protein 1 (LETM1) is reported to play a vital role in several human malignancies. However, there is limited understanding of the role of LETM1 in GC. This study aims to investigate the effects of LETM1 on proliferation, migration, and invasion of GC cells. Materials and Methods: The expression levels of LETM1 in the normal gastric mucosal epithelial cells (GES-1) and GC cells were analyzed by quantitative real-time polymerase chain reaction and western blotting. CCK-8, wound healing, and Transwell invasion assays were performed to evaluate the effect of LETM1 knockdown or overexpression on the proliferation, migration, and invasion of the GC cells, respectively. Additionally, the effect of LETM1 knockdown or overexpression on GC cell apoptosis was determined by flow cytometry. Furthermore, the effect of LETM1 knockdown or overexpression on the expression levels of PI3K/Akt signaling pathway-related proteins was evaluated by western blotting. Results: The GC cells exhibited markedly higher mRNA and protein expression levels of LETM1 than the GES-1 cells. Additionally, the knockdown of LETM1 remarkably suppressed the GC cell proliferation, migration, and invasion, and promoted the apoptosis of GC cells, which were reversed upon LETM1 overexpression. Furthermore, the western blotting analysis indicated that LETM1 facilitates GC progression via the PI3K/Akt signaling pathway. Conclusions: LETM1 acts as an oncogenic gene to promote GC cell proliferation, migration, and invasion via the PI3K/Akt signaling pathway. Therefore, LETM1 may be a potential target for GC diagnosis and treatment.

Potentiation of Antitumor Effect of Radiotherapy by Recombinant Tumor Necrosis Factor-$\alpha$ (방사선의 항암작용에 대한 재조합 TNF-$\alpha$의 효과)

  • Purpose : To determine whether TNF-$\alpha$ increases the antitumor effect of radiotherapy in murine syngeneic tumor system. Materials and Methods : Syngeneic murine tumors of MCa-K or MCa-4 (mammary carcinoma), OCa-I (ovarian carcinoma), or HCa-I(hepatocarcinoma were grown in hind legs of C3Hf/HeJ mice. When tumors were grown to 6 mm in mean diameter mice were treated with TNF-$\alpha$, radiation, or combination of the both. Gamma-radiation was given as a single dose of 30 Gy for HCa-I and 15 Gy for other tumors using Cobalt-60 teletherapy unit. A novel TNF-$\alpha$ mutein developed in Korea, was intraperitoneally administered daily at a dose of 10 ug per mouse for 7 days. In combination of radiation and TNF-$\alpha$, the drug was started 1 hour after radiation. Tumor growth delay assay was used to measure the tumor response to the treatment. Results : Among 4 tested tumors, TNF-$\alpha$ alone showed significant antitumor activity in MCa-K and OCa-I tumors, which showed absolute growth delay (AGD) of 5.0 days and 6.5 days, respectively. In combination with radiation, TNF-$\alpha$ showed significant delay of AGD (41.1 days) in OCA-I compared to AGDs of TNF-$\alpha$ alone and radiation, i.e., 6.5 days and 26.9 days, respectively(p<0.05). Enhancement factor was 1.29 in OCa-I, which showed supraadditive effect. TNF-$\alpha$ did not show significant delay of AGDs in the remaining 3 tumors compared to AGDs of TNF-$\alpha$ alone and radiation. Conclusions: TNF-$\alpha$ alone showed antitumor effects in MCa-K and OCa-I. In combination with radiation, TNF-$\alpha$ acted in supraadditive way in OCa-I only. The results of this study imply that the combination of TNF-$\alpha$ and radiation has different therapeutic potential depending on tumor model and further study is advocated.

Prediction of Forest Fire Danger Rating over the Korean Peninsula with the Digital Forecast Data and Daily Weather Index (DWI) Model (디지털예보자료와 Daily Weather Index (DWI) 모델을 적용한 한반도의 산불발생위험 예측)

  • Digital Forecast of the Korea Meteorological Administration (KMA) represents 5 km gridded weather forecast over the Korean Peninsula and the surrounding oceanic regions in Korean territory. Digital Forecast provides 12 weather forecast elements such as three-hour interval temperature, sky condition, wind direction, wind speed, relative humidity, wave height, probability of precipitation, 12 hour accumulated rain and snow, as well as daily minimum and maximum temperatures. These forecast elements are updated every three-hour for the next 48 hours regularly. The objective of this study was to construct Forest Fire Danger Rating Systems on the Korean Peninsula (FFDRS_KORP) based on the daily weather index (DWI) and to improve the accuracy using the digital forecast data. We produced the thematic maps of temperature, humidity, and wind speed over the Korean Peninsula to analyze DWI. To calculate DWI of the Korean Peninsula it was applied forest fire occurrence probability model by logistic regression analysis, i.e. $[1+{\exp}\{-(2.494+(0.004{\times}T_{max})-(0.008{\times}EF))\}]^{-1}$. The result of verification test among the real-time observatory data, digital forecast and RDAPS data showed that predicting values of the digital forecast advanced more than those of RDAPS data. The results of the comparison with the average forest fire danger rating index (sampled at 233 administrative districts) and those with the digital weather showed higher relative accuracy than those with the RDAPS data. The coefficient of determination of forest fire danger rating was shown as $R^2$=0.854. There was a difference of 0.5 between the national mean fire danger rating index (70) with the application of the real-time observatory data and that with the digital forecast (70.5).

Genetic studies of blood markers in Cheju horses II. Blood protein types (제주마의 혈액형에 관한 연구 II. 혈액 단백질형)

  • The present study was carried out to investigate the blood markers of Cheju horses. The blood protein types (biochemical polymorphism) were tested from 73 Cheju native horses (CNH) and 118 Cheju racehorses(CRH) by horizontal polyacrylamide gel electrophoresis (HPAGE), isoelectric focusing (IEF) and starch gel electrophoresis (SGE). At the same time, their phenotypes and gene frequencies were studied. The biochemical polymorphism phenotypes observed with high frequency were A1B-KK(97.3%), ALB-AB(49.3%), AP-SS(100%), ES-II(30.1%), GC-FF(87.7%), HB-BIBI(49.3%), TF-F2R(41.1%), TF-EF2(8.2%), PGD-FF(97.3%), PGM-SS(50.7%), GPI-II(74.0%) in CNH, While A1B-KK(99.2%), ALB-BB(50.8%), AP-SS(99.2%), ES-II(42.4%), ES-IS(14.4%), GC-FF(95.8%), HBB-IB II(39.8%), TF-F2R(21.2%), PGD-FF(77.1%), PGD-SS(4.3%), PGM-SS(72.9%), GPI-II(90.7%) in CRH. Alleles observed with high frequency were $AlB^{K}$(0.986), $ALB^{B}$(0.616), $AP^{S}$(1.000), $ES^{I}$(0.479), $ES^{F}$(0.274), $GC^{F}$(0.938), $GPI^{I}$(0.856), $HB^{BI}$(0.685), $PGD^{F}$(0.993), $PGM^{S}$(0.753), $TF^{F2}$(0.404), $TF^{R}$(0.397) in CNH and $AlB^{K}$(0.996), $ALB^{B}$(0.720), $AP^{S}$(0.996), $ES^{I}$(0.661), $ES^{F}$(0.203), $GC^{F}$(0.979), $GPI^{I}$(0.936), $HB^{BI}$(0.534), $PGD^{F}$(0.864), $PGM^{S}$(0.852), $TF^{F2}$(0.428), $TF^{R}$(0.272) in CRH. $TF^{E}$(0.041) allele and silent gene($ES^{I{^*}}$ : 0.014) were observed in CNH. The mean heterozygosity in CNH and CRH was observed 0.2974 and 0.2864, respectively.

Effect of the Polychaete Antimicrobial Peptide as feed Additives on Olive Flounder and Black Rockfish Immune Activity (넙치 및 조피볼락 면역 활성에 대한 사료첨가제로서 갯지렁이 항균펩타이드의 효과)

  • In this study, the effect of the polychaete antimicrobial peptide as feed additives on fish, olive flounder (Paralichythys olivaceus) and black rockfish (Sebastes schlegelii), immune activity was described. The antimicrobial peptide of the polychaete was induced by peptidoglycan from Micrococcus luteus. The fish were fed an experimental diet supplemented with 0%, 0.05%, 0.1%, 0.5% or 1% of immune induced the polychaete to a commercial diet. Haematological parameters, nonspecific immunes and stress were evaluated 2, 4, 6 and 8 weeks during fed. The resistance against bacteria, Edwardsiella tarda and Streptococcus iniae, were analysed on after 8 weeks. The haematological parameters were not significantly changed among tested groups. But the lysozyme activities were significantly high in the 0.1% and 0.5% supplement group of olive flounder and black rockfish, respectively. Additionally, cortisol in plasma was low in the 0.1% and 0.5% supplement group of olive flounder and black rockfish, respectively. And resistance of these supplement groups were significantly induced against bacterial injection.
