Phosphorylation of Eukaryotic Elongation Factor 2 Can Be Regulated by Phosphoinositide 3-Kinase in the Early Stages of Myoblast Differentiation

  • We have previously reported that phosphorylation of eukaryotic elongation factor 2 (eEF2) is related to the differentiation of chick embryonic muscle cells in culture. In the present study, we found that eEF2 phosphorylation declined shortly after induction of differentiation of L6 myoblasts, when the cells prepare for terminal differentiation by withdrawing from the cell cycle. This decrease in phosphorylation was prevented by inhibitors of phosphoinositide 3-kinase (PI3-kinase) that strongly inhibit myoblast differentiation. We hypothesized that PI3-kinase plays an important role in myoblast differentiation by regulating eEF2 phosphorylation in the early stages of differentiation. To test this hypothesis, myoblasts were synchronized at in $G_2/M$ and cultured in fresh differentiation medium (DM) or growth medium (GM). In DM the released cells accumulated in $G_0$/$G_1$ while in GM they progressed to S phase. In addition, cyclin D1 was more rapidly degraded in DM than in GM, and eEF2 phosphorylation decreased more. Inhibitors of PI3-kinase increased eEF2 phosphorylation, but PI3-kinase became more activated when eEF2 phosphorylation declined. These results suggest that the regulation of L6 myoblast differentiation by PI3-kinase is related to eEF2 phosphorylation.

  • Relationships between yield and various nitrogen efficiencies, between efficiencies and between efficiency and nitrogen uptake amount of rice plant were proposed and tested using data from N.P.K simple trials about 30 to 50 locations, for three years. Established relationships are well in accordance with experimental results by showing highly significant correlations between them. The overall indications are that high yielding capacity of fields with fertilizer application, depends primarily on high fertilizer nitrogen uptake by increasing fertilizer use efficiency (Eu), secondly the efficiency (Ef) of absorbed fertilizer nitrogen (Nf) and fertilization efficiency (Fe) and also depends much on nitrogen efficiency for grain yield (E) to great extend and that the efficiency (Es) of soil nitrogen (Ns) contributes to E more than Ef does. All nitrogen efficiencies are negatively correlated with the uptake amount of corresponding nitrogen and counterpart efficiency. Es and Ef could be determined firstly by difference method and secondly E versus Cs (Cs=Ns/Ns+Nf) plotting and thirdly E-Cs plotting with labelled fertilizermethod using the equation E=Es Cs+B where B=Ef Cf but a constant under the given condition and at last Y-Ns plotting with labelled fertilizer using Eq Y=$Es{\cdot}Ns+B$ where B=$Ef{\cdot}Nf$. Es which seems not much variable from field to field is mostly greater (about 80% of tested fields) than Ef which is much variable and depends much on fertilizer form. The relationships tested and well agreed are as follows: 1. Y=$Es{\cdot}Ns+Ef{\cdot}Nf$ (Y is yield) 2. E=$Es{\cdot}Cs+Ef{\cdot}Cf$ where Cf=Nf/Nf+Ns 3. E=b-aN where E=E, Es or Ef and N=N, Ns or Nf respectively, (E=Y/N, N=Nf+Ns), b is theoretical maximum under the given system and a is tangent at N=O of the curve, Y=EN. 4. Fe=Ef Eu and Se=$Es{\cdot}Eu$ where Se is efficiency of soil available nitrogen. 5. E=$(Se{\cdot}Cs+Fe{\cdot}Cf)/Eu$ 6. Y=$Es{\cdot}Eu{\cdot}Sf+Ef{\cdot}Eu{\cdot}Fn$or Y=$Es{\cdot}Eu{\cdot}Ea{\cdot}Sn+Ef{\cdot}Eu{\cdot}Fn $where Sf=$Ea{\cdot}Sn$, Ea is soil available nitrogen equivalent to fertilizer(Sf) divided by total soil nitrogen (Sn).

Phosphorylation of Elongation Factor-2 And Activity Of Ca2+/Calmodulin-Dependent Protein Kinase III During The Cell Cycle

  • Phosphorylation of the eukaryotic elongation factor 2 (eEF-2) blocks the elongation step of translation and stops overall protein synthesis. Although the overall rate of protein synthesis in mitosis reduces to 20% of that in S phase, it is unclear how the protein translation procedure is regulated during the cell cycle, especially in the stage of peptide elongation. To delineate the regulation of the elongation step through eEF-2 function, the changes in phosphorylation of eEF-2, and in activity of corresponding $Ca^{2+}$/calmodulin (CaM)-dependent protein kinase III (CaMK-III) during the cell cycle of NIH 3T3 cells, were determined. The in vivo level of phosphorylated eEF-2 showed an 80% and 40% increase in the cells arrested at G1 and M, respectively. The activity of CaMK-III also changed in a similar pattern, more than a 2-fold increase when arrested at G1 and M. The activity change of the kinase during one turn of the cell cycle also demonstrated the activation at G1 and M phases. The activity change of cAMP-dependent protein kinase (PKA) was reciprocal to that of CaMK-III. These results indicated: (1) the activity of CaMK-III was cell cycle-dependent and (2) the level of eEF-2 phosphorylation followed the kinase activity change. Therefore, the elongation step of protein synthesis might be cell cycle dependently regulated.

Clinical Value of Eukaryotic Elongation Factor 2 (eEF2) in Non-small Cell Lung Cancer Patients

  • Background: The purpose of this study was to evaluate a new type of tumor biomarker, eukaryotic elongation factor 2 (eEF2), in serum for the early diagnosis, confirmative diagnosis as well as assessment of treatment of non-small cell lung cancer (NSCLC). Methods: 130 patients with NSCLC and 50 healthy individuals undergoing physical examination in our hospital provided the observation and healthy control groups. An enzyme linked immune sorbent assay (ELISA) method was applied to determine serum eEF2 levels. Serum neuron specific enolase (NSE) and squamous cell carcinoma antigen (SCC) levels in the observation group were assessed with an automatic biochemical analyzer. Results: The median levels of eEF2 in the serum of NSCLC patients was found to be significantly higher than the healthy control group (p < 0.01) and it was markedly higher in stages III, IV than stages I, II (p < 0.05). eEF2 was higher with tumor size ${\geq}2$ cm than <2 cm (P< 0.01). Furthermore, two weeks after surgery patients showed a significant trend for eEF2 decrease (p < 0.05). Conclusions: The eukaryotic elongation factor 2 (eEF2) has certain clinical values for early diagnosis, verification, and prognosis as well as classification of lung cancer patients.

Various Nitrogen Efficiencies and their Interrelation Among Rice Varieties (수도품종간(水稻品種間) 여러 질소효율(窒素効率)의 상호관계(相互關係))

  • The relationships between various nitrogen efficiencies among 57 rice cultivars grouped into three developmental categories were investigated by simple correlation with (12kg/10a) and without nitrogen fertilizer under the field condition. The relationship showed similar in a group or among all varieties of three groups. Yield among varieties showed highly significant correlation with Fe (fertilization efficiency: yield increment/nitrogen applied), E (nitrogen efficiency for yield), Ef(efficiency of nitrogen derived from fertilizer) and Eu (fertilizer use efficiency: nitrogen derived from fertilizer divided by total nitrogen applied). The E was correlated significantly and positively with harvest index (HI), percent translocation (T) of nitrogen to ear and negatively with nitrogen uptake amount (N), nitrogen concentration in grain (GN%) and in straw (SN%). The E depends almost on Ef and only inTongil group partly on Es (efficiency of nitrogen derived from soil). The Ef contributes to Fe more than Eu does (Fe = $Ef{\cdot}Eu$). It appears that Ef and Eu increased from the old group to the recommended local varieties, but only EF and Es increased markedly when rice was developed from recommended local group to Tongil line ($indica{\times}japonica$ hybrid selction). The fact that E and Fe depend more on Ef among rice varieties is very good contrast to the result that E and Fe depend more on Es and Eu respectively among soils in the previous investigatigation. The Ef appears as the most important parameter for rice varietal selection under fertilizer application system.

Anti-Inflammatory Effects of Fermented Milk Supplemented with Heat-Killed Enterococcus faecalis EF-2001 Probiotics (Enterococcus faecalis EF-2001 유산균 사균체 첨가 발효유의 항염증 효과)

  • This study was conducted to verify the physiological activity of heat-killed Enterococcus faecalis EF-2001 probiotics in fermented milk. The anti-inflammatory effects of fermented milk supplemented with different concentrations (0, 100, and 500 ㎍/mL) of E. faecalis EF-2001 were determined using MTT assay and nitric oxide inhibition assay. The MTT assay was performed using RAW 264.7 cells. Results revealed that the rates of cytotoxicity and cell survival decreased significantly with increase in the concentration of heat-killed probiotics (p<0.05). Moreover, fermented milk supplemented with 100 ㎍/mL EF-2001 (EFM1) and the fermented milk supplemented with 500 ㎍/mL EF-2001 (EFM2) exhibited higher nitric oxide inhibition than normal fermented milk (NFM). Additionally, EFM2 significantly reduced the ratio of prostaglandin E2 compared to NFM (p<0.05). In conclusion, the treatment sample showed higher anti-inflammatory activity than NFM. The findings of this study could be used as a basic guideline for manufacturing of NFM supplemented with heat-killed probiotics.

Translation elongation factor-1A1 (eEF1A1) localizes to the spine by domain III

  • In vertebrates, there are two variants of eukaryotic peptide elongation factor 1A (eEF1A; formerly eEF-$1{\alpha}$), eEF1A1 and eEF1A2, which have three well-conserved domains ($D_I$, $D_{II}$, and $D_{III}$). In neurons, eEF1A1 is the embryonic type, which is expressed during embryonic development as well as the first two postnatal weeks. In the present study, EGFP-tagged eEF1A1 truncates were expressed in cortical neurons isolated from rat embryo (E18-19). Live cell images of transfected neurons showed that $D_{III}$-containing EGFP-fusion proteins (EGFP-$D_{III}$, -$D_{II-III}$, -$D_{I-III}$) formed clusters that were confined within somatodendritic domains, while $D_{III}$-missing ones (EGFP-$D_I$, -$D_{II}$, -$D_{I-II}$) and control EGFP were homogeneously dispersed throughout the neuron including axons. In dendrites, EGFP-$D_{III}$ was targeted to the heads of spine- and filopodia-like protrusions, where it was colocalized with $SynGAP{\alpha}$, a postsynaptic marker. Our data indicate that $D_{III}$ of eEF1A1 mediates formation of clusters and localization to spines.

Hepatitis B Virus DNA Polymerase Displays an Anti-Apoptotic Effect by Interacting with Elongation Factor-1 Alpha-2 in Hepatoma Cells

  • Hepatitis B virus (HBV) genome P-encoded protein HBV DNA polymerase (Pol) has long been known as a reverse transcriptase during HBV replication. In this study, we investigated the impact of HBV Pol on host cellular processes, mainly apoptosis, and the underlying mechanisms. We showed a marked reduction in apoptotic rates in the HBV Pol-expressed HepG2 cells compared to controls. Moreover, a series of assays, i.e., yeast two-hybrid, GST pull-down, co-immunoprecipitation, and confocal laser scanning microscopy, identified the host factor eEF1A2 to be associated with HBV Pol. Furthermore, knockdown of eEF1A2 gene by siRNA abrogated the HBV Pol-mediated anti-apoptotic effect with apoptosis induced by endoplasmatic reticulum (ER) stress-inducer thapsigargin (TG), thus suggesting that the host factor eEF1A2 is essential for HBV Pol's anti-apoptosis properties. Our findings have revealed a novel role for HBV Pol in its modulation of apoptosis through integrating with eEF1A2.

The Effects of Eriobotryae Folium Herbal-acupuncture at ST36 on ovalbumin-induced asthma in C57BL mouse (백서(白鼠)의 족삼리(足三里)에 시술한 비파엽약침(枇杷葉藥鍼)이 천식억제 및 면역조절효과에 미치는 영향)

  • To study the effects of EF(Eriobotryae Folium) herbal acupuncture on asthma, we injected EF-HAS into Jok-samni(ST36) of C57BL/6 mice Objective : The aim of this study was to investigate the effect of EF-HA(herbal acupuncture) at ST36 on ovalbumin-induced asthma in mice Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). Experimental groups were treated with concentrations(1%) of EF-HA at Jok-samni(ST36) for the later 8 weeks(3times/week). Result : 1. The weight and total cells of lung of the mice group treated with EF-HA decreased significantly compared with that of Control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with EF-HA decreased significantly compared with those of Control group. 4. The concentration of IgE, IL-4, IL-5 in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 5. The concentration of IL-4, IL-5, IL-13 in Serum of the mice group treated with EF-HA decreased significantly compared with those of Control group. 6. The number of $Gr-1^+/CD11b^+,\;CD3^-/CCR3^+,\;CD4^+,\;CD8^+,\;CD3e^+/CD69^+$ cells in the lungs of the mice group treated with EF-HA decreased significantly compared with those of Control group. 7. The cytokine's manifestation of mRNA of the mice group treated with EF-HA with RT-PCR decreased significantly compared with that of Control group. Conclusion : We conclude that EF-HA is effective on OVA-induced asthma of C57BL/6 mouse.

Basic Study of Behavior Characteristics of Emulsified Fuel with Fuel Design (연료설계에 의한 에멀젼연료의 거동특성에 관한 기초연구)

  • A compression ignition type of diesel engine makes fuel efficiency better and $CO_2$ in the exhaust gas lower. Also it is suitable to apply alternative fuels(blended fuel) to the engine. The objective of this study is the emissions reduction of diesel engine with EF(Emulsified fuel). The emulsified fuel consists of diesel and peroxide($H_2O_2$) and Soot reduction without worsening of NOx emissions can be achieved by using thermal decomposition of the peroxide, i.e. the chemical effect of the OH radical in actual engine. For manufacturing emulsified fuel, a surfactant which is comprised of span 80 and tween 80 mixed as 9:1, was mixed with a fixed with 3% of the total volume in the emulsion fuel. In addition, considering the mixing ratio of the surfactant, the mixing ratio of $H_2O_2$ in the emulsified fuel was set as EF0, EF2, EF12, EF22, EF32, and EF42, respectively. Consequently, this study aims to obtain the optimization of fuel design(mixing) for the emulsified fuel applying to the diesel engine.
