Lipopolysaccharide로 처리 된 RAW264.7 세포에서 고마리 추출물의 항염증 효과 (The Anti-Inflammatory Effects of Persicaria thunbergii Extracts on Lipopolysaccharide-Stimulated RAW264.7 Cells)

  • 본 연구는 고마리 추출물이 가지는 항염증 활성을 알아보기 위하여 쥐의 대식세포(RAW264.7 cell)에 Lipopolysaccharide (LPS)를 처리하여 염증반응을 유도하고 이때 발생되는 Nitric oxide (NO)의 생성 억제를 확인하였다. 또한 염증에서 중요하게 알려져 있는 Inducible nitric oxide synthase (iNOS), Cyclooxygenase-2 (COX-2), Nuclear factor-kappa B (NF-${\kappa}B$) 단백질들의 발현을 비교하였고, 추가적으로 NF-${\kappa}B$ 단백질의 핵 내부로의 이전 및 활성을 확인하였다. 메탄올 추출물은 NO 생성 및 iNOS, COX-2, NF-${\kappa}B$ 단백질의 발현을 억제하고, 세포를 보호하는 효과를 가지는 Heme oxygenase-1 (HO-1) 단백질의 발현을 증가시켰다. 위 결과를 바탕으로 하여 n-butanol, hexane, ethyl acetate 용매를 이용한 추가적인 분획을 실시하였다. 이들 분획 중 고마리의 ethyl acetate 추출물은 Prostaglandin $E_2$ ($PGE_2$), NO 생성을 억제 하였으며, iNOS, COX-2 단백질들의 발현을 감소, NF-${\kappa}B$의 핵 내부로의 이동을 억제하는 효과가 높다는 것을 확인하였다. 이러한 연구결과는 고마리 식물이 좋은 항염증 활성을 가지고 있음을 나타내며, 지속적인 분획으로 고마리 식물이 가지는 항염증 활성 물질을 선별하여 그 작용기작을 규명하는 연구가 필요하다.

Eryngium foetidum Suppresses Inflammatory Mediators Produced by Macrophages

  • Objective: This study assessed anti-inflammatory and antioxidant activities of $E.$ $foetidum$ leaf extract on LPS-activated murine macrophages. Methods: RAW264.7 cells were pretreated with or without $E.$ $foetidum$ extract for 1 h prior to incubation with LPS for 24 h. Anti-inflammatory activity was evaluated with reference to iNOS, COX-2, TNF-${\alpha}$ and IL-6 gene expression. In addition, NO and intracellular ROS generation were determined by Griess method and fluorescence intensity and activation of MAPKs and $I{\kappa}B$ by Western blotting. Results: Prior treatment with $E.$ $foetidum$ leaf extract inhibited elevation of IL-6, TNF-${\alpha}$, iNOS and COX-2, together with their cognate mRNAs in a dose-dependent manner. NO and intracellular ROS contents were similarly reduced. These effects were due to inhibition of LPS-induced phosphorylation of JNK and p38 as well as $I{\kappa}B$. $E.$ $foetidum$ ethanol extract were shown to contain lutein, ${\beta}$-carotene, chlorogenic acid, kaempferol and caffeic acid, compounds known to exert these bioactive properties. Conclusions: $E.$ $foetidum$ leaf extract possesses suppressive effects against pro-inflammatory mediators. Thus, $E.$ $foetidum$ has a high potential to be used as a food supplement to reduce risk of cancer associated with inflammation.

큰포식세포에서 퉁퉁마디 추출물의 아라키돈산 대사효소조절 및 항염증 활성 (Modulation of arachidonic acid metabolism and inflammatory process in macrophages by different solvent fractions of Glasswort (Salicornia herbacea L.) extract)

  • 퉁퉁마디는 식용 염생식물로서 이에 대한 유용 기능성 탐색 및 생리활성 평가에 대한 연구가 지속적으로 진행되고 있다. 본 연구에서는 퉁퉁마디 70% 메탄올 추출물을 극성에 따라 분획하여 얻은 Fr.H, Fr.E, Fr.EA, Fr.B 및 Fr.W의 5분획의 NO제거 및 iNOS 발현, 아라키돈산 대사에 관련된 효소 $cPLA_2$, COX-2, 5-LOX 등의 발현과 활성화에 대한 효과를 분석하여 항염작용과 메커니즘을 검토하였다. 이 중 Fr.EA가 가장 높은 폴리페놀 및 플라보노이드 성분 함량을 나타내었고 nitrite/아질산의 제거에 있어서도 가장 뛰어난 활성을 보였다. 하지만 LPS로 자극한 RAW264.7 큰포식세포 배양계에서는 Fr.E가 가장 우수한 NO 제거활성과 iNOS발현억제 활성을 나타내었고 이어 Fr.H와 Fr.EA의 활성 순을 나타냈다. Fr.E는 또한 LPS로 자극한 RAW264.7 배양 세포계에서 $cPLA_2$와 COX-2의 발현억제에 가장 큰 효과를 나타내었으며 이들의 억제를 위한 $IC_{50}$는 각각 33.4과 $27.9{\mu}g/mL$로 나타났다. Fr.E는 $cPLA_2$의 활성화에 중요한 영향을 미치는 ERK1/2의 인산화도 농도의존적으로 저해하였다. Fr.H와 Fr.EA도 $80{\mu}g/mL$ 이하 농도에서 iNOS와 아라키돈산 대사효소들의 발현을 농도의존적으로 억제하였다. 하지만 가장 극성 분획인 Fr.W는 모든 활성평가 시스템에서 거의 효과를 나타내지 않았다. 본 연구는 퉁퉁마디가 iNOS와 아라키돈산 대사효소계를 효과적으로 억제하여 항염증작용을 할 수 있다는 것을 보고하고 있으며, 특히 Fr.E와 Fr.H와 같은 소수성 분획들이 우수한 활성을 나타내어 향후 이를 타깃으로 하는 관련 기능성 소재로서의 활용이 기대된다.

  • 숙주 면역 반응과 면역 체계는 치주 질환에 대한 개인의 감수성의 주요 원인이다. 세균 감염과 흡연은 치주 조직의 파괴의 원인과 진행에 관여하는 중요한 환경 위험 요인이다. 따라서, 본 연구는 사람 치주인대세포에서 LPS와 니코틴이 전염증성 사이토카인인 iNOS/COX-2의 발현과 NO/$PGE_2$ 생산에 미치는 영향을 알아보고 NFATc1가 어떤 기전으로 항염작용을 하는지 밝히고자 하였다. LPS와 니코틴을 처리한 사람 치주인대세포에서 iNOS/COX-2의 발현과 함께 NO/$PGE_2$ 생산은 증가되었다. NFATc1 inhibitor인 CsA는 LPS와 니코틴에 의해 유도되는 iNOS/COX-2의 발현과 함께 NO/$PGE_2$ 생산을 감소시켰다. 이러한 연구 결과로 볼 때, NFAT signaling pathway가 LPS와 니코틴에 의한 iNOS/COX-2의 발현을 조절하여 NO/$PGE_2$ 매개 염증에 대해 방어할 수 있다고 생각된다.

고구마 소주 주박에 의한 RAW 264.7 세포주에서 lipopolysaccharide로 유도된 염증 반응의 억제 효과 (Suppressive Effects of Lees from Sweet Potato Soju on LPS-induced Inflammatory Responses in RAW 264.7 Cells)

  • 본 연구에서는 고구마 소주로부터 획득한 에탄올 추출물 및 순차적 유기용매 분획물을 제조하고, 이들 시료가 LPS로 활성화된 마우스 대식세포 RAW 264.7 세포주에서 nitric oxide (NO) 생산과 세포생존율에 미치는 영향을 연구하였다. 처리한 시료 중 ethyl acetate 분획물(KSD-E8-3) 처리군에서 LPS로 염증 유도된 RAW 264.7 세포주의 생존율에는 영향 없이 NO 생성을 현저하게 감소 시킴을 확인하였다. 또한 분획물 KSD-E8-3 처리에 의해서 염증매개 인자인 COX-2, iNOS 그리고 TNF-alpha의 유전자의 발현이 감소되었으며, iNOS 단백질의 발현이 현저히 감소됨을 Western blot으로 확인하였다. 또한, 분획물 KSD-E8-3 처리에 의해 세포내 대표적인 신호 전달 단백질인 3가지의 MAPK 단백질(p38, JNK, ERK1/2)의 인산화가 모두 억제됨을 확인하였다. 종합적으로, 이러한 연구결과는 고구마 소주 주박에 함유된 생리활성 물질에 의한 항염증 활성 및 작용 기전을 이해하는데 도움을 줄 것으로 기대된다.

고지혈증 흰쥐에서 생약제제 KH-204의 발기부전 치료연구 (The Effect of Herbal Formula KH-204 on Erectile Dysfunction in Hyperlipidemic Rat)

  • This study was designed to investigate the effects KH-204 on erectile dysfunction in hyperlipidemic rat. KH-204 has been evaluated antihyperlipidemic and antierectile dysfunction effects on experimental hyperlipidemic rats induced by high fat diet. After oral administration of the water extract KH-204 (50, 100, 200, 300 mg/kg) to hyperlipidemic rats for 8 weeks, the variables including body weight, total cholesterol, HDL and LDL levels in serum, the expression of eNOS and nNOS in penis were measured. And erectile function was determined by measurement of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve. Oral administration of KH-204 significantly inhibited the increases of serum total cholesterol and LDL-cholesterol levels and the decreased of serum HDL-cholesterol levels in hyperlipidemic rats induced by high fat diet. The penile expression level of the two enzyme (eNOS, nNOS) were increased significantly after oral administration of the KH-204 50 mg/kg. Erectile function after 10 volts stimulation was significantly decreased in the hyperlipidemic rat compared with the normal rat, but increased in KH-204 group compared with hyperlipidemic group. These results suggest that KH-204 is effective for erectile dysfunction in hyperlipidemia.

Inhibitory Effects of Eutigosides Isolated from Eurya emarginata on the Inflammatory Mediators in RAW264.7 Cells

  • The anti-inflammatory activity of Eurya emarginata (Thumb) Makino, of which leaves have been traditionally used to treat ulcers or diuretic in Jeju Island, has been investigated in the present study. Through the phytochemical study from the methanol extract of E. emaginiata, eutigosides Band C were isolated as the active components. Sseveral inflammatory markers including TNF-$\alpha$, IL-1$\beta$, IL-6, NO, iNOS, and COX-2 were examined. Eutigosides Band C potentially inhibited production of pro-inflammatory cytokines (IL-6 and TNF-$\alpha$) in a dose-dependent manner. Additionally, the intracellular contents of iNOS protein were markedly decreased after treatment with eutigosides Band C. The inhibition of iNOS activity was correlated with the decrease in nitrite levels. These results suggest that eutigoside Band C from E. emarginata may have anti-inflammatory activity through the inhibition of pro-inflammatory cytokines (TNF-$\alpha$ and IL-6), iNOS and COX-2.

Effect of Trolox on Altered Vasoregulatory Gene Expression in Hepatic Ischemia/Reperfusion

  • This study was designed to investigate the effect of Trolox, a hydrophilic analogue of vitamin E, on the alteration of vasoregulatory gene expression during hepatic ischemia and reperfusion (I/R). Rats were subjected to 60 min of hepatic ischemia in vivo. The rats were treated intravenously with Trolox (2.5 mg/kg) or the vehicle as a control 5 min before reperfusion. Liver samples were obtained 5 h after reperfusion for a RT-PCR analysis on the mRNA for the genes of interest. These mRNA peptides are endothelin-1 (ET -1), potent vasoconstrictor peptide, its receptor $ET_A$ and $ET_B$, vasodilator endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS), heme oxygenase-1 (HO-1), tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and cyclooxygenase-2 (COX-2). It was seen that serum alanine aminotransferase and lipid peroxi-dation levels were markedly increased after I/R and Trolox significantly suppressed this increase. In contrast, the glutathione concentration decreased in the I/R group, and this decrease was inhibited by Trolox. ET-1 mRNA expression was increased by I/R, an increase which was prevented by Trolox. The mRNA levels for $ET_A$ receptor was significantly decreased, whereas ET$_{B}$ receptor transcript increased in the I/R group. The increase in $ET_A$ was prevented by Trolox. The mRNA levels for iNOS and HO-1 significantly increased in the I/R group and Trolox attenuated this increase. There were no significant differences in eNOS mRNA expression among any of the experimental groups. The mRNA levels for COX-2 and TNF-$\alpha$ significantly increased in I/R group and Trolox also attenuated this increase. Our findings suggest that I/R induces an imbalanced hepatic vasoregulatory gene expression and Trolox ameliorates this change through its free radical scavenging activity.y.

오배자의 생리활성 연구 (Studies on the Cytoprotective and Anti-inflammatory Effects of Isolate from Galla Rhois)

  • Galla Rhois is a nest of parasitic bug, has been traditionally used for the treatment of the therapy of diarrhea, peptic ulcer, hemauria, etc., that showed various anti-inflammatory activity, and other biological properties. We studied the effect of Galla Rhois ethanol extract. we investigated whether compounds isolated from the ethanol extract of Galla Rhois, could modulate iNOS and COX-2 expression in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). We found compounds that suppressed LPS-induced iNOS and COX-2 expression. Suppression of the expression of iNOS and COX-2 was in parallel with the comparable inhibition of the production of nitric oxide (NO) and prostaglandin E2 (PGE2). Our results suggest that compounds can inhibit NO and PGE2 productions through suppression of LPS-induced iNOS and COX-2 expression. Because COX-2- or iNOS-dependent mechanisms are involved in inflammation and tumor progression, our findings provide a new uncovering mechanism responsible for anti-inflammatory and antitumor effects of Galla Rhois.

Upregulation of Nitric Oxide Synthase Activity by All-trans Retinoic Acid and 13-cis Retinoic Acid in Human Malignant Keratinocytes

  • Effect of retinoids, i.e., all-trans retinoic acid and 13-cis retinoic acid, on the activity of nitric oxide synthase (NOS) was evaluated in human malignant keratinocytes to examine the possible correlation of retinoids with NOS activities. All-trans retinoic acid and 13-cis retinoic acid did not alter the nitric oxide (NO) production. However, in the presence of lipopolysaccharide (LPS, $1{\mu}g/mL$), they significantly increased NO release in a dose-dependent manner until 48 h at concentrations of $50{\sim}100{\mu}M$. The degree of upregulation of NO by all-trans retinoic acid and 13-cis retinoic acid increased up to 35% and 37%, respectively, compared to that by the control, which demonstrated the upregulation of LPS-inducible nitric oxide synthase (iNOS)-dependent generation of NO as well as showing a crucial link between retinoids-induced activity and NOS. Findings of this study now suggest that the upregulation of LPS-iNOS activity may be associated with modulation of retinoids-induced control of cellular developmental processes, which may produce new therapeutics of retinoids in the complexity of how NO affects human keratinocytes.
