  • Networks are growing faster than ever before causing a multi-domain complexity. The diversity, variety and dynamic nature of network traffic and services require enhanced orchestration and management approaches. While many standard orchestrators and network operators are resulting in an increase of complexity for handling E2E slice orchestration. Besides, there are multiple domains involved in E2E slice orchestration including access, edge, transport and core network each having their specific challenges. Hence, handling of multi-domain, multi-platform and multi-operator based networking environments manually requires specified experts and using this approach it is impossible to handle the dynamic changes in the network at runtime. Also, the manual approaches towards handling such complexity is always error-prone and tedious. Hence, this work proposes an automated and abstracted solution for handling E2E slice orchestration using an intent-based approach. It abstracts the domains from the operators and enable them to provide their orchestration intention in the form of high-level intents. Besides, it actively monitors the orchestrated resources and based on current monitoring stats using the machine learning it predicts future utilization of resources for updating the system states. Resulting in a closed-loop automated E2E network orchestration and management system.

  • This study was carried out to construct a DNA profile database for 102 watermelon cultivars through the comparison of polymorphism level and genetic relatedness using genomic microsatellite (gMS) and expressed sequence tag (EST)-microsatellite (eMS) markers. Sixteen gMS and 10 eMS primers showed hyper-variability and were able to represent the genetic variation within 102 watermelon cultivars. With gMS markers, an average of 3.63 alleles per marker were detected with a polymorphism information content (PIC) value of 0.479, whereas with eMS markers, the average number of alleles per marker was 2.50 and the PIC value was 0.425, indicating that eMS detects a lower polymorphism level compared to gMS. Cluster analysis and Jaccard's genetic distance coefficients using the unweighted pair group method with arithmetic average (UPGMA) based on the gMS, eMS, and combined data sets showed that 102 commercial watermelon cultivars could be categorized into 6 to 8 major groups corresponding to phenotypic traits. Moreover, this method was sufficient to identify 78 out of 102 cultivars. Correlation analysis with Mantel tests for those clusters using 3 data sets showed high correlation ($r{\geq}0.80$). Therefore, the microsatellite markers used in this study may serve as a useful tool for germplasm evaluation, genetic purity assessment, and fingerprinting of watermelon cultivars.

  • It is now widely recognized that new Information and Communication Technologies (ICTs) have been changing the way tv which firms conduct their businesses. Examples are the application of enterprise resources planning (ERP) and business process reengineering (BPR) to increase the efficiency of internal resources management, adoption of Business-to-Business e-commerce (B2B e-com) to integrate supply chain, and invention of new marketing channels such as Business-to-Customer (B2C) e-com. These new ways of conducting businesses are believed to help firms to reduce transaction costs and increase productivity. As a result, new ICTs have played an important role in recent growth of many small firms into multi-functional and multi-product corporations and in their spatial expansion towards internationalization as well. This paper takes Haier in China as a case to study the role of new ICTs in the growth of firms and reveal how the new technologies have facilitated the expansion of Haier into a transnational corporation (TNC) by examining the internationalization process of the firm in relation to its adoption of new ICTs in the period from 1990 to 2002. The study reveals that the adoption of new Ins has helped Haier to integrate its functional units located in dozens of places across the world, which is essential to the internationalization of a firm, and to link closely together its worldwide suppliers and customers to achieve just-in-time (JIT) production and delivery. As such, the authors of the paper argue that, without the facilitation of new ICTS, Haier could not have developed into a TNC in less than ten years. It is now widely recognized that new Information and Communication Technologies (ICTs) have been changing the way tv which firms conduct their businesses. • The vector pCW5 with plasmid pC7, originally isolated in Lactobacillus paraplantarum C7 derived from kimchi, was constructed using a p32 strong promoter, the pC7 replicon, and green fluorescent protein (GFP) as the reporter. The constructed vector was transformed into E. coli and Leuconostoc mesenteroides, and GFP expression detected using a Western blot analysis. GFP fluorescence was recognized in E. coli and Leuconostoc mesenteroides using a confocal microscope. In addition, GFP fluorescence was also clearly detected in several industrially important lactic acid bacteria (LAB), including Lactobacillus bulgaricus, Lactobacillus paraplantarum, and Lactobacillus plantarum. Thus, pCW5 was shown to be effective for Leuconostoc mesenteroides when using GFP as the reporter, and it can also be used as a broad-host-range vector for other lactic acid bacteria.

  • To isolate probiotic lactic acid bacteria for animal, we have screened the microorganisms from chicken feces, by random selection and agar well diffusion assay. Among them, CPM-7 strain showing superior inhibitory activity against Escherichia coli was selected. By examining carbohydrates utilization, morphologic property and 16S rRNA gene sequence, CPM-7 strain was identified as Lactobacillus salivarius, then named L. salivarius CPM-7. L. salivarius CPM-7 produced thirteen enzymes in the test using API ZYM kit, and showed resistance to low pH and bile salts. It survived at pH 2 for 30 min. and pH 3 for 6 hr. And, it was able to grow in MRS medium containing 0.2% (w/v) bile salts. L. salivarius CPM-7 adhered to the jejunal epithelium cells of pig. Both the supernatant of L. salivarius CPM-7 and the its neutralized one showed high inhibitory activity against E. coli K88.

  • An endolichenic fungus Xylaria grammica EL000614 produces grammicin, a potent nematicidal pyrone derivative that can serve as a new control option for root-knot nematodes. We optimized an Agrobacterium tumefaciens-mediated transformation (ATMT) protocol for X. grammica to support genetic studies. Transformants were successfully generated after co-cultivation of homogenized young mycelia of X. grammica with A. tumefaciens strain AGL-1 carrying a binary vector that contains the bacterial hygromycin B phosphotransferase (hph) gene and the eGFP gene in T-DNA. The resulting transformants were mitotically stable, and PCR analysis showed the integratin of both genes in the genome of transformants. Expression of eGFP was confirmed via fluorescence microscopy. Southern analysis showed that 131 (78.9%) out of 166 transformants contained a single T-DNA insertion. Crucial factors for producing predominantly single T-DNA transformants include 48 h of co-cultivation, pretreatment of A. tumefaciens cells with acetosyringone before co-cultivation, and using freshly prepared mycelia. The established ATMT protocol offers an efficient tool for random insertional mutagenesis and gene transfer in studying the biology and ecology of X. grammica.

  • A non-destructive environmental DNA protocol for the genetic analysis of sea cucumber (Apostichopus japonicus) resources DNA was established. Among the several commercial DNA extraction kits, the DNeasy® Plant Mini Kit was selected as the best choice to obtain the high-quality genomic DNAs from the mucous sea cucumber. As the temperature and incubation time increased, the amount of extracted environmental DNA was also large, but it was judged that the increased amount did not affect as much as 2-3 times. Therefore, these conditions were not considered to be the main factors to consider in actual environmental DNA extraction. However, the amount of seawater relative to the size of the sample was judged as a major consideration, and a sufficient amount of environmental DNA for analysis was secured when stored within 1 min while stirring the volume of seawater corresponding to the total sea cucumber weight (g). In securing the environmental DNA of sea cucumbers, the mortality rate of sea cucumbers in all experiments was 0, and it was judged that the effects of sea cucumbers were not significant through this treatment. Through the results of this study, sea cucumber DNA research, which has been conducted in a destructive method, can be conducted non-destructively through environmental DNA analysis. Through this study, we have secured a standard protocol that can successfully extract the sea cucumber DNA through environmental DNA. It is not only excellent in terms of time and cost of traditional DNA analysis method currently used, but it is completely non-destructive in the ecosystem of the survey area. It is believed that the system can be transformed in a way that does not affect it. However, it is thought that various standard protocols should be established considering the characteristics of each type.
