Comparison of Upgraded Methods for Detecting Pathogenic Escherichia coli in Foods Using Centrifugation or Filtration

  • In the present study, centrifugation and filtration pretreatments were evaluated to decrease sample preparation time and to improve the sensitivity and specificity of multiplex polymerase chain reaction (PCR) for the detection of low levels of pathogenic Escherichia coli in various foods. Pathogenic E. coli (E. coli NCCP11142, E. coli NCCP14037, E. coli NCCP 14038, E. coli NCCP14039, and E. coli NCCP15661) was inoculated into pork, beef, and baby leafy vegetables at 1, 2, and 3 Log CFU/g. The samples were shaken 30 times (control), then centrifuged or filtered. DNA extracts from the samples were subjected to PCR using the $Powerchek^{TM}$ Diarrheal E. coli 8-plex Detection Kit. In the pork samples, no E. coli was detected in the control samples, while E. coli were detected in 100% of 3-Log CFU/g inoculated and centrifuged samples, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In the beef samples, all control samples appeared to be E. coli-negative, while E. coli was detected in 50-75% of centrifuged samples, regardless of inoculated level, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In baby leafy vegetables, E. coli were not detected in 25-50% of the control samples, while E. coli were detected in 0-25% of the centrifuged samples, and 75-100% of the filtered samples, depending on the inoculum amount. In conclusion, filtration pretreatment can be used to minimize sample preparation time, and improve the sensitivity and specificity of rapid detection of pathogenic E. coli in various foods.

식중독 세균 검출에 있어서 리포좀의 이용 가능성 (Feasibility Study on the Use of Liposomes for Detecting Food-borne Pathogenic Bacteria)

  • 식중독 세균 검출에 있어서 리포좀의 이용 가능성을 병원성 식중독 세균인 E. coli O157:H7을 대상으로 검토하였다. 리포좀의 표면에 E. coli O157:H7을 인식하는 항체를 공유결합시키고 리포좀의 내부 수용액상에는 형광 표지물질인 sulforhodamine B를 포집시켰다. 이렇게 합성된 면역리포좀이 진단시약으로써의 기능을 하는지 알아보기 위해 두 가지 분석 방법을 적용하였다. 첫번째 방법으로써, test-strip을 이용한 분석은 E. coli O157:H7의 존재 유무를 test-strip 상에 나타난 분석 신호을 육안으로 관찰하는 것으로써 E. coli O157:H7 존재시 분석 신호가 저하되는 것을 원리로하였다. 이 방법은 고가의 실험 장비가 필요하지 않아 현장적용성이 용이한 장점이 있다. 두번째 방법은, IMS 후 목표 세균에 결합되어 있는 리포좀을 파괴시켜 나오는 형광도의 세기를 측정하는 방법으로 이때는 형광도의 강도와 E. coli O157:H7 세균 수와 비례적 관계가 나타났다. 상기의 두 방법에서 얻은 결과는, 리포좀을 E. coli O157:H7 검출에 응용할 수 있는 가능성을 제시하며 추후에 식품 적용 실험이 요구된다.

Hovenia Monofloral Honey can Attenuate Enterococcus faecalis Mediated Biofilm Formation and Inflammation

  • We evaluated the anti-biofilm formation and anti-inflammatory activity of Hovenia monofloral honey (HMH) against Enterococcus faecalis. Co-culture of HMH with E. faecalis attenuated the biofilm formation of E. faecalis on a polystyrene surface. In addition, HMH effectively eradicated the established E. faecalis biofilm. HMH significantly attenuated E. faecalis growth but did not affect the production of extracellular polymeric substances on E. faecalis, indicating that reduction of E. faecalis biofilm is a result of HMH-mediated killing of E. faecalis. Furthermore, we found that HMH can effectively attenuate E. faecalis-induced expression of a proinflammatory interleukin-8 (IL- 8) in HT-29 cells. Interestingly, treatment of HMH significantly attenuated the E. faecalis-mediated expression of Toll-like receptor-2 (TLR-2) and its adaptor molecules, myeloid differentiation primary response 88 (MyD88), in HT-29 cells. In addition, E. faecalis-induced mitogen-activated protein kinases (MAPKs) phosphorylation was significantly attenuated by HMH administration. Furthermore, HMH-mediated antiinflammatory efficacy (0.2 mg/mL of HMHs) had an equal extent of inhibitory efficacy as 5 μM of MyD88 inhibitor to attenuate E. faecalis-mediated IL-8 expression in HT-29 cells. These results suggest that HMH could effectively inhibit E. faecalis-mediated gastrointestinal inflammation through regulating the TLR-2/MyD88/MAPKs signaling pathways. Collectively, our data suggest that HMH could be developed as a potential natural agent to control E. faecalis-mediated biofilm formation and inflammation.

Global Transcriptome-Wide Association Studies (TWAS) Reveal a Gene Regulation Network of Eating and Cooking Quality Traits in Rice

  • Eating and cooking quality (ECQ) is one of the most complex quantitative traits in rice. The understanding of genetic regulation of transcript expression levels attributing to phenotypic variation in ECQ traits is limited. We integrated whole-genome resequencing, transcriptome, and phenotypic variation data from 84 Japonica accessions to build a transcriptome-wide association study (TWAS) based regulatory network. All ECQ traits showed a large phenotypic variation and significant phenotypic correlations among the traits. TWAS analysis identified a total of 285 transcripts significantly associated with six ECQ traits. Genome-wide mapping of ECQ-associated transcripts revealed 66,905 quantitative expression traits (eQTLs), including 21,747 local eQTLs, and 45,158 trans-eQTLs, regulating the expression of 43 genes. The starch synthesis-related genes (SSRGs), starch synthase IV-1 (SSIV-1), starch branching enzyme 1 (SBE1), granule-bound starch synthase 2 (GBSS2), and ADP-glucose pyrophosphorylase small subunit 2a (OsAGPS2a) were found to have eQTLs regulating the expression of ECQ associated transcripts. Further, in co-expression analysis, 130 genes produced at least one network with 22 master regulators. In addition, we developed CRISPR/Cas9-edited glbl mutant lines that confirmed the role of alpha-globulin (glbl) in starch synthesis to validate the co-expression analysis. This study provided novel insights into the genetic regulation of ECQ traits, and transcripts associated with these traits were discovered that could be used in further rice breeding.

LPS 유도된 HCT116 인간 대장암세포에서 cordycepin의 prostaglandin E2-EP4 receptor 감소 조절을 통한 세포의 이동과 전이 억제 효과 (Cordycepin Inhibits LPS-induced Cell Migration and Invasion in Human Colorectal Carcinoma HCT116 cells through Down-regulation of Prostaglandin E2-EP4 Receptor)

  • Prostaglandin E2(PGE2), a major product of cyclooxygenase-2 (COX-2), plays an important role in the carcinogenesis of many solid tumors, including colorectal cancer. Because PGE2 functions by signaling through PGE2 receptors (Eps), which regulate tumor cell growth, invasion, and migration, there has been a growing amount of interest in the therapeutic potential of targeting Eps. In the present study, we investigated the role of EP4 on the effectiveness of cordycepin in inhibititing the migration and invasion of HCT116 human colorectal carcinoma cells. Our data indicate that cordycepin suppressed lipopolysaccharide (LPS)-enhanced cell migration and invasion through the inactivation of matrix metalloproteinases (MMP)-9 as well as the down-regulation of COX-2 expression and PGE2 production. These events were shown to be associated with the inactivation of EP4 and activation of AMP-activated protein kinase (AMPK). Moreover, the AMPK inhibitor, compound C, as well as AMPK knockdown via siRNA, attenuated the cordycepin-induced inhibition of EP4 expression. Cordycepin treatment also reduced the activation of CREB. These findings indicate that cordycepin suppresses the migration and invasion of HCT116 cells. Through modulating EP4 expression and the AMPK-CREB signaling pathway. Therefore, cordycepin has the potential to serve as a potent anti-cancer agent in therapeutic strategies against colorectal cancer metastasis.

사료내 비타민 E의 추가급여가 돈육질에 미치는 영향 (Effect of Vitamin E Supplementation in Diets on Pork Quality)

  • 본 연구는 비타민 E의 추가급여가 돈육질에 미치는 영향을 규명하기 위해 비육돈 60두 (75.0$\pm$0.7kg)를 공시하였다. 처리구는 대조구로 일반사료 급여구와 일반사료에 비타민 E 200 IU 추가 급여구로 되어 있다. 각 처리간 정육율과 도체길이는 유의적인 차이가 나타나지 않았다. 두 처리간 등심의 색상에서는 유의적인 차이를 보였으며(P<0.05), 이와 같은 차이는 등심의 경도에서도 나타났다(P<0.05). 하지만 등심의 마블링이나 등심면적에서는 비타민 E의 고수준 첨가 급여로 인한 차이는 나타나지 않았다. 또한 등지방 두께에서도 비타민 E의 고수준 첨가 급여로 인한 유의적인 효과는 없었다. 보수력의 경우 비타민 E의 고수준 첨가로 인해 증가하는 경향(P<0.05)을 보이고 있었다. 하지만 전단력, 해동감량 및 조리감량에서는 처리간의 차이를 보이지 않았다. 시간 경과에 따른 L*, a*, b*와 TBA값 역시 처리간의 차이는 볼수 없었다. 결론적으로 비타민 E 추가 급여는 등심의 색상에 보수력에 영향을 미치는 것으로 사료된다.

모색발현 유전자의 DNA Marker를 이용한 쇠고기 품종 판별

  • 본 연구는 축우의 모색발현을 조절하는 MCIR, MGF 및 TYRP1 3종류의 모색 유전자를 이용하여 한우육 판별기술을 개발하고자 PCR-RFLP 기법으로 이들 모색유전자 좌위의 대립유전자를 검출하고 각 품종 간 RFLP 유전자형 출현빈도를 비교 분석하였다. MCIR 유전자의 RFLP 유전자형 출현빈도에서 한우는 e/e과 E+/e형이 출현되었고 이외의 다른 유전자형의 출현은 전혀 인정되지 않았다. 그러나, Holstein종 젖소는 $E^D/E^D$$E^D/e$ 2종류의 유전자형 그리고 Angus종에서는 $E^D/E^D$, $E^D/E^++$$E^D/e$ 3종류의 유전자형이 각각 출현하여 한우와 이들 두 품종간의 MCIR유전자형 출현빈도에 뚜렷한 차이가 인정되었다. MGF 유전자의 RFLP 유전자형 출현빈도에서 한우는 R/r과 r/r형이 각각 25%와 75%로 rr형의 출현율이 비교적 높았으며 Holstein종과 Angus 종은 R/r형이 100% 출현했으며, Charolais 종은 rr형이 100% 출현하였고 이외의 다른 유전자형은 인정되지 않았으며 Hereford종은 RR형이 80% 그리고 R/r형이 20%의 출현율을 보여 RR형의 출현율이 매우 높아 한우와 Holstein 및 육우 품종간의 MGF 유전자형 출현빈도에 명백한 차이가 인정되었다. 따라서, 소 모색관련 MCIR과 MGF 유전자의 품종 특이적 PCR-RFLP 유전자형은 한우육과 국내산 Hostein 젖소육 및 도입육우 품종을 식별하는데 매우 유용한 DNA marker로 이용될 수 있음이 확인되었다.

Significant Genotype Difference in the CYP2E1 PstI Polymorphism of Indigenous Groups in Sabah, Malaysia with Asian and Non-Asian Populations

  • CYP2E1 PstI polymorphism G-1259C (rs3813867) genotype distributions vary significantly among different populations and are associated with both diseases, like cancer, and adverse drug effects. To date, there have been limited genotype distributions and allele frequencies of this polymorphism reported in the three major indigenous ethnic groups (KadazanDusun, Bajau, and Rungus) in Sabah, also known as North Borneo. The aim of this study was to investigate the genotype distributions and allele frequencies of the CYP2E1 PstI polymorphism G-1259C in these three major indigenous peoples in Sabah. A total of 640 healthy individuals from the three dominant indigenous groups were recruited for this study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) at G-1259C polymorphic site of CYP2E1 gene was performed using the Pst I restriction enzyme. Fragments were analyzed using agarose gel electrophoresis and confirmed by direct sequencing. Overall, the allele frequencies were 90.3% for c1 allele and 9.7% for c2 allele. The genotype frequencies for c1/c1, c1/c2 and c2/c2 were observed as 80.9%, 18.8%, and 0.3%, respectively. A highly statistical significant difference (p<0.001) was observed in the genotype distributions between indigenous groups in Sabah with all Asian and non-Asian populations. However, among these three indigenous groups, there was no statistical significant difference (p>0.001) in their genotype distributions. The three major indigenous ethnic groups in Sabah show unique genotype distributions when compared with other populations. This finding indicates the importance of establishing the genotype distributions of CYP2E1 PstI polymorphism in the indigenous populations.
