Proteus sp. ST-1에 의한 Azo계 색소의 분해에 관한 연구 (The Study on Degradation of Azo Dyes by Proteus sp. ST-1)

  • Direct Sky Blue-5B is an Azo dye known as general for staining of textile and leather, etc., and as materials which are difficult to be biodegraded in nature. The bacterium strain which could degrade direct Sky Blue-5B was isolated from activated sludge of dyeing factory and identified as Proteus sp. by experiment on morphological, cultural and biochemical characteristics, and so named Proteus sp. ST-1. The optimum condition of the strain for degradation of Sky Blue-5B were at about 35$^{\circ}C$ and PH 7~8. The strain had been capable of degradation with organic nitrogen effectively and had completely degraded 200mg/1 of the dye within 12hrs at 37$^{\circ}C$. The enzyme system related to degradation of Azo dye may be intracellular, and so degraded the dye after absorption into cell. The degradation products of Sky Blue-5B by Proton sp. 57-1 were analyzed by Gas Chromatography /Mass Spectrometry and Spectrophotomer, from this observation, it may be infered that the strain degraded the dye directly without any mediate.

울금의 생육, 수량 및 품질에 미치는 퇴비의 효과 (Effects of Composts on the Growth, Yield and Effective Components of Turmeric (Curcuma longa L.))

  • Curcuma longa L. (turmeric) is an important medicinal plant that has been historically used in herbal medicine and in the health food throughout Asia etc. Recently, the demands on rhizome of turmeric are increasing greatly by well-being boom, but there is not enough to meet the demands. To fulfill increasing demands, cultivation system strategies using the organic fertilizers are required to produce a greater amount of rhizome with good quality and yield. A field experiment was conducted to determine the effect of composts, NPK fertilizer(control, F), NPK + swine manure(SM) and NPK + fermentation manure from the wasted oriental medicine materials(OMWM), on rhizome yield and the content of bioactive components for quality. Our results showed that two compost applications can increase both rhizomes(24.1-25.9%) and curcumin(21.7-41.0%) yields, respectively, compared to F control. The content of amino acids increased significantly by SM and OMWM treatments. SM and OMWM application also increased the total phenol yields 7.8 and 8.7 g/10a compared with control 6.3 g/10a, the flavonoid yields 6.3 and 7.3 g/10a compared with control 5.3 g/10a, and also antioxidant activity 21.7 and 41%, respectively, as compared to the control. Especially, OMWM was more effective in total rhizomes yields and bioactivities and in the biosynthesis of curcumin and bioactive components than SM treatments, but the biological pathway was not clear, still. This experiment suggests that curcumin or bioactive components affected by adding SM and OMWM could increase the yields and quality of turmeric.

전통 염색 재료를 활용한 기능성 종이 연구 (Research on Functional Paper by using Traditional Dyestuffs)

  • 전통 천연 염색지를 이용한 보존 보관용 재료 개발을 위하여 전통적으로 사용되어온 15종의 염재에서 염료를 추출하여 세균 3종과 진균 2종에 대하여 paper disk 방법으로 항균성을 조사하였다. 동일한 방법으로 매염제 3종을 평가하고, 매염제와 염재 추출물을 혼합하여 항균성을 조사하였다. 위 결과로부터 항균성이 우수한 9종의 염료를 매염제와 혼합하여 염색지를 제작하였다. 염색지 중 안개나무, 황련, 황벽, 오배자 신나무 복합매염지에서 진균에 대한 항균성이 확인되었으며, 흰개미를 이용한 방충력 실험에서는 황련, 황벽 염색지에서 방충력이 확인되었다. 또한 염색지를 이용하여 금속과 같은 타재질에 미치는 영향을 확인한 결과, 황련이 상대적으로 안정적이었다. 그리고 안개나무, 황련, 황벽3종의 염색지에서 유해가스 제거능력이 확인되었다. 이러한 염색지를 활용하여 문화재 보존용 보관상자로 제작할 경우 다양한 유기물 문화재의 생물피해 방제에 사용이 가능할 뿐만 아니라 다양한 공간에서 기능성 마감재로서의 활용성이 기대된다.

견의 탄닌가공에 관한 연구 (A Study on the Tannin Weighting of Silk)

  • 견에 대한 오배자탄닌과 합성탄닌의 처리조임이 견의 탄닌증량율에 미치는 영향과 탄닌증량견의 염색조건 및 실용적 특성을 구명하기 위하여 시험결과 1. 견의 증량율을 기안한 합리적인 탄닌처리 농도는 오배자 30g/l, 합성탄닌산 15g/l이었고, 처리온도 및 시간은 8$0^{\circ}C$에서 60분 내외 처리용액의 산도는 pH 2-3이었다. 2. 탄닌처리 견의 염색조건에 있어서 부착된 탄닌의 탈락을 적게하며 염착농도(K/S)를 향상시킬 수 있는 OrangeII에 의한 합리적 염색온도 및 시간은 6$0^{\circ}C$, 90분이었다. 3. 탄닌염색 가공견포의 Soaping에 의한 색착($\Delta$E)는 미가공 견포에 비하여 현저히 감소됨으로서 세탁견도가 2급이나 향상되었다. 4. 탄닌 염색가공 견포의 자외선 조사에 의한 dyeloss율은 무처리염색견포의 1/6정도로 감소됨으로서 탄니처리 견포의 내광성이 현저히 개선되었다. 5. 탄닌 염색가공 견포의 마찰견도와 발수성은 미가공 견포와 같은 수준이었으나 Drape계수는 감소됨으로서 의복의 착용감이 향상되었다.

가미강활산(加味羌活散)이 집먼지 진드기 추출물로 유도된 NC/Nga mouse의 아토피 피부염에 미치는 영향 (Effect of Kami-kanghwalsan (KKHS) on Atopic Dermatitis-like Skin Lesions Induced in NC/Nga Mouse by Mite Antigen Stimulation)

  • Objectives The purpose of this study is to investigate the effect of KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to this condition in humans. Methods To investigate the effect of KKHS on atopic dermatitis (AD), we evaluated atopic dermatitis-like skin lesions by clinical skin index and analyzed immunological parameters in peripheral blood mononuclear cells(PBMCs), splenocytes, draining lymph node(DLN) and performed skin histology in ears and dorsal skin of atopic dermatitis of NC/Nga mouse in vivo. Results In vivo, clinical skin severity score was significantly lower in the KKHS group than in the control group. IgE, IL-6, TNF-${\alpha}$, IgM, IgG2a and IgG2b levels in serum decreased remarkably in the KKHS group than in the control group, and the level of IFN-${\gamma}$ production which is secreted from Th1 cell was increased by KKHS. After this experiment we analyzed immunological cells ($CD3^+$, $CD19^+$, $CD4^+$, $CD8^+$, $CD3^+CD69^+$, $CD4^+CD25^+$ and $CD49b^+$) by flow cytometry. It results that the total absolute number of $CD3^+$, $CD19^+$, $CD4^+$ and $CD8^+$ cells were recovered as much as normal state, and the level of $CD3^+CD69^+$ in isolated DLN and PBMCs were significantly decreased, and total absolute number of $Gr-1^+$, $CD11b^+$ and $CD3^+$ in dorsal skin of NC/Nga mouse were decreased by KKHS. We analyzed ear, DLN, and neck-back skin after biopsy and dyeing by hematoxyline/eosin(H&E), toluidine staining (mast cells marker). KKHS were very effective to the histological symptoms which are in dermal and epidermal thickening, hyperkeratosis and inflammatory cell infiltration. Ear thickness was significantly decreased compared with the control group and the size of inflammatory lymphocytes cells (ILC) and plasma cells (PC) in DLN were also decreased. Conclusions KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse was very effectiveness to the atopy dermatitis treatment.

구풍제습탕(驅風除濕湯)이 DNCB로 유도된 NC/Nga mice의 아토피 피부염에 미치는 영향 (Effect of Gupoongjeseuptang (GPJST) on DNCB (dinitrochlorobenzene)-induced Atopic Dermatitis-like Model NC/Nga Mice)

We analyzed ear and neck-back skin after biopsy and dyeing by hematoxyline/eosin (H&E) and toluidine staining (mast cells marker) and obtained results that GPJST are very effective to histological symptoms (dermal and epidermal thickening, hyperkeratosis and inflammatory cell (CD4, $CCR3^+$) infiltration). Conclusions : This study demonstrates immunological activity of GPJST on atopic dermatitis-like model mice.

양모.폴리에스터 혼방직물의 효소가공 시 활성제 복합사용의 효과 (Effects of Mixed Activators on Enzymatic Activation for Wool.polyester Blend Fabrics)

  • This study provides effects of mixed activators on enzymatic activation and determines optimum mixture ratio for enzymatic treatment. Wool 80% and polyester 20% blend fabric and papain from carica papaya are used in this experiment. L-cysteine and sodium sulfite are used as activators for papain treatment process. The treatment condition is pH 7.5, $70^{\circ}$, papain concentration 10%(o.w.f), 60 minutes. L-cysteine and sodium sulfite are added in enzyme solution with various concentrations($0{\sim}50mM$). The optimum treatment condition is determined by measuring weight loss, tensile strength, whiteness, water contact angle(WCA), dyeability and surface micrographs. The results are as follow; The optimum mixture ratio of activators is L-cysteine 2mM and sodium sulfite 10mM. Mixed activators assists in improving the activation of papain. WCA of papain treated fabrics is decreased since papain treatment with activator mixture makes wool polyester blend fabrics more hydrophilic. Dyeing property of papain-treated fabrics more improves by the treatment with mixed activators than with single activator. It means that this method can save time and lower cost. After papain treatment in the presence of mixed activator, the surface of fabrics is modified. The surface of wool fiber shows to be descaled and hydrolyzed, and that of polyester fiber shows to be cracked.

배지의 혼입비율과 관비 양액 농도가 오이 플러그묘의 초기생장에 미치는 영향 (Effects of Mixing Ratio of Substrates and Concentration of Fertigation in Nutrient Solutions on the Early Growth of Cucumber Plug Seedlings)

  • 오이['장형흑진주' 오이; (주)서울종묘]를 공시작물로 하여 양질의 플러그묘 생산을 위한 혼합배지 개발과 적정 관비 양액 농도를 규명하기 위해 수행하였다. 혼합배지는 피트모스를 기본으로 왕겨, 훈탄, 부숙톱밥, 펄라이트 및 입상압면 등을 상이한 비율로 혼합하여 5처리로 하였고, 그 중 양호한 혼합배지 3종을 선발하여 관비 양액 농도(EC)를 대조구(EC $0.1dS{\cdot}m^{-1}$), 0.5, 1.0 및 $1.5dS{\cdot}m^{-1}$등으로 처리하여 오이 유묘의 생장반응(27일째)을 검토하였다. 오이 플러그묘 생장에 양호한 혼합배지는 피트모스:왕겨:훈탄:부숙톱밥:펄라이트=25:10:25:20:20(v/v), 피트모스 왕겨:부숙 톱밥:입상암면=30:25:20:25(v/v) 및 피트모스:왕겨:부숙톱밥=40:40:20(v/v)였다. 대조구 (EC $0.1 dS\;m^{-l}$)에 비해 관비 양액의 농도가 높아질수록 초장, 엽면적 및 총건물생산량 등이 현저히 높아 EC $1.5dS{\cdot}m^{-1}$로 두상관수 2-3회 관비하였을 때 가장 좋은 플러그묘를 생산할 수 있었다. 관비 양액 농도와 혼합배지 종류 처리간에는 유의차가 인정되지 않았다.

양액농도가 추식 수경재배 나물자원 3종의 생장과 무기물 함량에 미치는 영향 (Effects of the Different Concentration of the Nutrient Solution on the Growth and the Inorganic Matter Contents of Three Kinds of Fall Planting Namul Resources in Water Culture)

  • 나물자원의 수경재배시 양액농도가 생장 및 무기물함량에 미치는 영향을 조사하고자 벌개미취, 원추리 및 질경이를 펄라이트 배지에 추식하여 70일간 재배 한 후 생장정도와 무기물 함량을 분석하였다. 벌개미취와 원추리의 초장, 줄기직경, 잎의 수, 신선중 및 건물중은 1.5배액 처리구에서 확연하게 우수한 결과를 나타내었다. 질경이의 지상부와 지하부 생체중 및 건물중 등은 양액의 농도가 0.2재액에서 1.5배액으로 증가할수록 증가한 반면, 엽수와 근장은 0.5배액과 표준농도에서 가장 높은 것으로 나타났다. Ca Mg 및 Na 함유량은 벌개미취의 경우 1.5배액에서, 원추리와 질경이는 0.25배액에서 가장 많았다. K 함량은 벌개미취와 원추리는 0.5배액에서, 질경이는 1.5배액에서 가장 많았다. $P_2O_5$는 나물종류 및 양액의 농도에 따른 차이가 거의 없었다.
