Identification of the Marine Microorganisms Producing Bioactives -I. Isolation and Cultural Conditions of the Marine Actinomycetes No. 101 Producing Antimicrobial compounds- (생리활성물질을 생성하는 해양미생물의 동정 -I. 항미생물 물질을 생산하는 해양방선균 분리균주 No. 101의 분리 및 배양조건-)

  • Marine organisms were investigated to identify the marine actinomycetes that produced noble bioactive compounds. Microorganism counts range from $2.1{\times}10^3\;to\;1.2{\times}10\;CFU/g$ of marine organisms. Actinomycetes constituted 0.01 to $0.5\%$ of culturable microbial community. We identified the marine actinomycetes that produced novel bioactive compounds. During the course of screening for bioactives from the marine microorganisms, we found that the strain in sponge had antimicrobial activities. From the morphological, cultural and various physiological characteristics, this strain was identified for Actinomycetes No. 101. The optimal compositions of culture medium for Actinomycetes No. 101 were starch 30g/l as carbon source, casamino acid 10g/l as nitrogen source. The optimal pH of medium and fermentation temperature were $6.5{\sim}7.0$ and $30^{\circ}C$, respectively. Fermentation has been conducted in the marine broth at $30^{\circ}C$ for 72 hour. The yield of fermentation got about 3g as dry weight(per liter of broth). The distribution of antimicrobial activity of Actinomycetes No. 101 was screened by paper disc. The extract of cultured cell and broth inhibited the growth of Staphylococcus aureus and Bacillus subtilis, but the inhibition action was week against yeast and mold.

  • Bacillus subtilis KMU-13 was isolated from the Lillehammer forest soils at Norway and shown a strong antifungal activity on cucumber scab, Cladosporium cucumerinum KACC 40576. B. subtilis KMU-13 produced a maximum level of antifungal substance under incubation aerobically at $30^{\circ}C$, 180 rpm for 48 hours in LB broth containing 0.5% maltose and 0.5% bactopeptone and initial pH adjusted to 6.0. Butanol extract of cultured broth was confirmed inhibitory zone by plate assay and Rf 0.64 value substance by thin layer chromatography (TLC) represented high antifungal activity against C. cucumerinum KACC 40576 and also shown fungal growth inhibitory activity against Botytis cinerea KACC 40573, C. gloeosporioides KACC 40804, D. byoniae KACC 40669, F. oxysporum KACC 40037, F. oxysporum KACC 40052, F. oxysporum f. sp. radicis-lycopersici KACC 40537, F. oxysporum KACC 40902, M. cannonballus KACC 40940, P. cambivora KACC 40160, R. soiani AG-1 KACC 40101, R. solani AG-4 KACC 40142, and S. scleotiorum KACC by agar diffusion method.

Preparation and Its Taste-Active Components of Grass puffer (Takifugu niphobles) Extracts (복섬 엑스분의 추출 및 정미발현성분의 조성)

  • To develop natural intermediate flavoring substances, optimal hydrolysis conditions and taste compounds for two step enzyme hydrolysate(TSEH) using Grass puffer(Takifugu niphobles) were investigated. The optimal conditions for TSEH method were revealed in temperature at $55^{\circ}C$ 3 hour digestion with Alcalase (pH 7.5) at the 1st step and 2 hour at $45^{\circ}C$ digestion with Flavourzyme(pH 6.0~6.5) at the 2nd step. TSEH method was superior to hot-water extraction on the aspect of yield, nitrogen contents and organoleptic taste quality such as umami and control of bitter taste formation. In taste active-components in Glass puffer TSEH, total free amino acid content was 4,502 mg%, major free amino acids were Pro, Leu, Lys, Hypro, Tau, Arg, Phe, Ala, Glu and Val in ordor. As for nucleotides, IMP(372 mg%) was the principal component and also contents of TMAO, total creatinine, and betaine were 43, 278 and 41 mg% in Glass puffer TSEH, respectively. The major inorganic ions in TSEH were Na(949 mg%), K(222 mg%), Cl(1,180 mg%) and $PO_4$(1,081 mg%).

Changes of Physicochemical Components and Antioxidant Activity of Garlic During its Processing (흑마늘 가공 중 이화학적 성분 및 항산화 활성의 변화)

  • This study was performed to confirm of biological function of black garlic, it heated for 11 days at $40{\sim}90^{\circ}C$ (first step: heate for 2 days at $90^{\circ}C$, second step: heated for 4 days at $80^{\circ}C$, third step: heated for 4 days at $60^{\circ}C$ and fourth step: heated for 1 day at $40^{\circ}C$). Samples were analyzed physico-chemical characteristics and antioxidant activity. Hunter L, a and b values were decreased during processing, and then inner part Hunter values were highly decreased at the second step. The moisture contents were decreased to 58.48${\pm}$0.41 g/100 g at fourth step. pH was also acidified to pH 4.22${\pm}$0.02, but O.D. value at 420 nm was increased during processing of black garlic. At fourth step, total phenolics and flavonoids contents were increased about 1.9 and 2.6 folds than first step sample. Also, total pyruvate and thiosulfinate contents were increased about 1.6 and 5.8 folds as change of total phenolic and flavonoid contents, respectively. Fructose contents were the highest level among free sugars and its contents increased to 2,454.45${\pm}$4.20 mg/100 g. Contents of sucrose and maltose were decreased during processing of black garlic. The contents of total minerals were the highest at fourth step (1,009.20${\pm}$6.91 mg/100 g) during its processing. Contents of glutamic acid, proline and aspartic acid were detected higher than other composition amino acids. Taurine and ethanolamine were not detected in the first step sample, but they were detected 0.88${\pm}0.60{\sim}1.06{\pm}$0.04 and 0.28${\pm}0.4{\sim}0.5{\pm}$0.09 mg/100 g in next processing step, respectively. DPPH radical scavenging ability of water and ethanol extracts from black garlic was increased during its processing. Abilities of DPPH radical scavenging were the highest in fourth step sample, its abilities were 67.40${\pm}$0.21% in 1,000 ${\mu}g/ml$ of water extracts. Reducing power was also significantly higher in water extract than ethanol extract on the whole.

Studies on Brewing of Apple Wine -Apple wine containing lower concentration of alcohol- (사과주(果實酒)양조(釀造)에 관(關)한 연구(硏究) -저농도주정함유(低濃度酒精含有) 사과주에 관(關)하여-)

  • This study aims to brew apple wine containing lower concentration of alcohol by fermentation and to retain $CO_2$ gas in apple wine, and investigation for the possibility of storage at room temperature was performed. A Saccharomyces sp. was proved to be acceptable for production of base wine as its higher fermentation rate at $20{\sim}25^{\circ}C$. However, B-2 was most reasonable for post-fermentation of apple wine as this strain strongly ferments sugars at low temperature $(4^{\circ}C)$. The yield of apple juice increased by maceration of apple pulps. The yield was about 5 % more than that of the unmacerated juice, whereas acid content was decreased by 10% compared with control. When stored apple wine containing 9% alcobol was introduced $1{\sim}3%$ sucrose at $7{\sim}8^{\circ}C$ for 100 days or more, the $CO_2$ pressure of apple wine in bottle shows $3kg/cm^2$ by bottle-pressure meter. It showed good storage of the wine at room temperature. $CO_2$ gas pressure in apple wine containing 6% alcohol, $5{\sim}10%$ hop extract, and 2% sugar was $2kg/cm^2$, he result also showed possibility of storage. Whereas 6% concentration of alcoholic apple wine without hop extract caused unusual fermentation during storage at the same condition. The desirable conditions for high quality apple wine should have $CO_2$ pressure of $2kg/cm^2$ or more and should be added $1{\sim}2% sugar to base wine. From these results, it can be concluded that the brewing of lower alcoholic apple wine is possible.

Antioxidative Effect and Neuraminidase Inhibitory Activity of Polyphenols Isolated from a New Korean Red Waxy Sorghum (Sorghum bicolor L. cv. Hwanggeumchalsusu) (황금찰수수(Sorghum bicolor L. cv. Hwanggeumchalsusu) 유래 에탄올 추출물 및 폴리페놀계 화합물의 항산화 활성 및 뉴라미니데이즈 억제 효과)

  • To identify nutritional and therapeutic properties of the new Korean red waxy sorghum cultivar ‘Hwanggeumchalsusu (HGC)’, we assayed the antioxidative effects and neuraminidase inhibitory activity. A methanol and 70% ethanol extract of HGC exhibited strong antioxidative effects (IC50 values of 83.2±2.7 for DPPH) and 85.6±2.4 μg/ml for ABTS) and neuraminidase (ND) inhibitory activity (IC50 values of 1.8±0.1 from extracted with methanol and 3.4±0.1 μg/ml from extracted with 70% ethanol) compared with that of the control, noncolored sorghum cultivar ‘Huinchalsusu (HC)’ (IC50> 200 μg/ml). We isolated nine polyphenols, Gallic acid (1), Protocatecuic acid (2), p-Hydroxy benzoic acid (3), Vanillic acid (4), Caffeic acid (5), Ferulic acid (6), Luteolinidin (7), Apigeninidin (8), Luteolin (9), from the HGC - methanol extract, to determine whether they were the active components Luteolinidin of one kind of polyphenols from the HGC, exhibited significant antioxidative effects (IC50 values of 10.9±0.5 μM for DPPH and 8.6 0.6 μM for ABTS) and neuraminidase (ND) inhibitory activity (IC50 values of 26.3±0.6) showed noncompetitive inhibition model. The binding affinity of the ND inhibitors in molecular docking experiments correlated with their ND inhibitory activities. These results suggest that HGC may be utilized to serve as a potential effective antioxidant and inhibitor of ND.

Antimicrobial and Antioxidant Activity of Grapefruit and Seed Extract on Fishery Products (수산물에 대한 Grapefruit 종자추출물의 항균 및 항산화효과)

  • The antimicrobial and antioxidant activities of grapefruit seed extract(GFSE), which was extracted with glycerine in the special schematic extraction apparatus, were investigated for handling and processing of fishery products. The effectivity of GFSE has been tried on sardine, mackerel and shrimp divided into six lots for each fishery product: control(no treatment) and five GFSE-treated samples. Samples were inoculated with Salmenella typhi, incubated for 24hrs at $30^{\circ}C$ in dextrose-tryptone broth medium and prepared for microbiological 8f chemical analysis and organoleptic assessment. The bacteriological analytical results with GFSE(250ppm) showed the reduction of $1.8\times10^6\to2.0\times10^4,\;1.9\times10^6\to1.8\times10^4$ and $1.6\times10^6\to2.7\times10^3$ in total bacterial count for sardine, mackerel and shrimp, respectively. The test results with GFSE(500ppm) showed a $100\%$ reduction of bacterial mackerel treated with GFSE(500ppm) was reduced to $1.1\times10^4$ and $9.0\times10^3$ respectively. Antioxidant effect of treatment with GFSE at 500ppm level for three products was significant. LSD test results on organoleptic parameter for the samples treated with various showed a significant influence on the appearance, odor and texture in which at concentration 500ppm level give the excellent scours compared to each control.

Changes on physio-chemical properties of oak sawdust during fermentation (참나무류 톱밥의 발효기간 중 물리화학적 특성 변화)

  • Changes in physical and chemical properties of oak sawdust were investigated by depth and time for 46 days during the fermentation process of 33 tons of the sawdust for oak mushroom cultivation. The degrees of change in the properties of the sawdust differed depending on the depth and fermentation period. Most of the physical-chemical properties except temperature and pH gradually changed during the fermentation. The temperature change was highly sensitive to the environment at the surface sawdust to 20 cm depth, while it gradually increased to the maximum $58.9^{\circ}C$ at 40~100 cm depths in 12 days and slowly to the maximum at 150 cm depth in 24 days. The moisture content of the sawdust decreased gradually from 31% to 26.5~28.0% in 24 days. Of the chemical properties during the fermentation, pH generally rose from 5.2 to 5.6, but it decreased to 4.4~4.7 at 150 cm depth in 16 days. While the carbon content of the sawdust was 68~70% without significant change, nitrogen content increased from 0.22% to 0.25% and thus C/N ratio gradually lowered from 320 to 280. P content in the sawdust gradually increased from 0.005% to 0.022% for 46 days. Osmotic concentration of the hot water extract of the sawdust varied 41.5~44.2 mmol/kg without significant change by the depth and time. The starch particles within initial ray parenchyma cells of sawdust decreased and fungal hyphae formed on the surface of the sawdust granules and within xylem vessel cells in 35 days. The effect of the sawdust fermentation on oak mushroom cultivation needs continued research.

Anti-Obese Effects of Mixture Contained Pine needle, Black Tea and Green Tea Extracts (솔잎, 홍차 및 녹차 추출물 함유 조성물의 항비만 효과)

  • The aims of this study were to evaluate the anti-obese effects of pine needle, black tea and green tea in rats and overweight people. Supplementation of 1, 2, 4 and 8% amount to the control diet of pine needle extract and mixture groups significantly decreased body weight gain and visceral fat mass compared with that of control diet group. Supplementation of 1, 2 and 4% amount to the control diet of black and green tea extracts groups significantly decreased body weight gain and visceral fat mass compared with that of control diet group. Anti-obese effect in body weight gain and visceral fat mass of mixture group was higher than in other groups. In human study, extracts mixture supplementation to overweight subjects significantly decreased both body weight and body fat compared with placebo control group. In vitro study, black and green tea extracts significantly inhibited both the pancreatic lipase and ${\alpha}-amylase$ activities dose dependently. In conclusion, the anti-obese effects of pine needle, black, and green teas in rats were found. In overweight human subjects, extracts mixture decreased body weight and body fat compared with placebo control group. Anti-obese effect in black and green tea groups might be from an decrease in carbohydrate and fat digestions via inhibition of pancreatic ${\alpha}-amylase$ and lipase activities in part.

Heavy Metal Contents in Dfferent Bran Layers of Rice (미곡중(米穀中) 도정부위별(搗精部位別) 중금속함량(重金屬含量))

  • Contents of heavy metals in various bran layers of rice collected from Kyeong Nam province in 1978, were determined. The components of crude prote in, crude fat, crude cellulose, and ash in all the rice samples were found to be most concentrated in the rice of each layer of brown, 70% polished, and 90% polished, but the contents of crude protein (9.22%), crude fat (1.16%), crude cellulose (1.01%), and ash (0.38%) in polished rice decreased rapidly. On the other hand, the amounts of nitrogenfree extract in the rice were showed increasingly toward the inner layer and those of polished rice were 75.54%. The contents of lead, zinc, nickel, cadmium, and arsenic in bran layer of rice decreased in order of that of 90% polished, 70% polished, brown, and polished rice. Their amounts in polished rice were lead, $0.054{\sim}0.610ppm$; zinc, $9.830{\sim}19.093ppm$; nickel, trace-1.776ppm; cadmium, trace-0.039ppm; arsenic, ND-0.170ppm. It was shown that copper was more concentrated in the layer of 90% polished rice $(0.581{\sim}1.476ppm)$, and iron more in brown rice $(27.971{\sim}66.569ppm)$ than in any other portion, whereas mercury $(0.006{\sim}0.072ppm)$, chromium $(1.538{\sim}7.822ppm)$, and manganese $(57.371{\sim}179.252ppm)$ were much contained in the layer of 70% polished rice. Their amounts in polished rice were copper, $0.218{\sim}0.858ppm$; iron, $1.480{\sim}9.573ppm$; mercury, $0.006{\sim}0.027ppm$; chromium, ND-1.520ppm; and manganese, $6.730{\sim}11.562ppm$.
