한국산 Rotifer, Brachionus plicatilis (L과 S-type)의 내구란 대량생산 (Mass Production of Resting Egg of Korean Rotifer, Brachionus plicatilis (L and S-type))

  • 한국산 B. plicatilis, L-type rotifer와 S-type rotifer의 내구란 대량생산은 L-type rotifer의 경우 15일 동안 농축 Chlorella + 빵효모 혼합구와 빵효모 단독구로 하였고 S-type rotifer는 7일동안 1 $m^3$ 수조에서 농축 Chlorella로, $6{\~}8$일 동안 4 $m^3$ 수조에서 냉동 농축 Chlorella + 빵효모로 대량생산하였다. L-type rotifer 내구란 대량생산 실험에서 농축 Chlorella + 빵효모 혼합구가 총 내구란 생산 $54.5{\times}10^6$개, $10^{8}$ rotifers 당 내구란 생산 $30.5{\times}10^6$개, 먹이 건조 중량 1g 당 내구란 생산 $100{\times}10^3$개로 빵효모 단독구 보다 높게 나타났다. S-type rotifer의 내구란 대량생산은 4 $m^3$ 수조에서 냉동 농축 Chlorella + 빵효모를 먹이로 총 내구란 생산 $149{\~}567{\times}10^6$개, $10^{8}$ rotifers당 내구란 생산 $36{\~}123{\times}10^6$개, 먹이 건조 중량 1 g당 내구란 생산 $131{\~}338{\times}10^3$개를 생산하여 1 $m^3$ 수조에서 농축 Chlorella를 단독 먹이로 생산한 경우 보다 높았다.

Growth and Photosynthetic Responses of Cuttings of a Hybrid Larch (Larix gmelinii var. japonica x L. kaempferi) to Elevated Ozone and/or Carbon Dioxide

  • We studied the effects of elevated ozone ([$O_3$]) and $CO_2$ concentrations ([$CO_2$]) on the growth and photosynthesis of the hybrid larch $F_1(F_1)$ and on its parents (the Dahurian larch and Japanese larch). $F_1$ is a promising species for timber production in northeast Asia. Seedlings of the three species were grown in 16 open top chambers and were exposed to two levels of $O_3$ (<10 ppb and 60 ppb for 7 h per day) in combination with two levels of $CO_2$ (ambient and 600 ppm for daytime) over an entire growing season. Ozone reduced the growth as measured by height and diameter, and reduced the needle dry mass and net photosynthetic rate of $F_1$, but had almost no effect on the Dahurian larch or Japanese larch. There was a significant increase in whole-plant dry mass induced by elevated [$CO_2$] in $F_1$ but not in the other two species. Photosynthetic acclimation to elevated [$CO_2$] was observed in all species. The net photosynthetic rate measured at the growing [$CO_2$] (i.e. 380 ppm for ambient treatment and 600 ppm for elevated $CO_2$ treatment) was nevertheless greater in the seedlings of all species grown at elevated [$CO_2$]. The high [$CO_2$] partly compensated for the reduction of stem diameter growth of $F_1$ at high [$O_3$]; no similar trend was found in the other growth and photosynthetic parameters, or in the other species.

  • Proteins accumulated in dry, stratified Arabidopsis seeds or young seedlings, totaled 1100 to 1300 depending on the time of sampling, were analyzed by using immobilized pH gradient 2-DE gel electrophoresis. The molecular identities of 437 polypeptides, encoded by 355 independent genes, were determined by MALDI-TOF or TOF-TOF mass spectrometry. In the sum, 293 were present at all stages and 95 were accumulated during the time of radicle protrusion while another 18 appeared in later stages. Further analysis showed that 226 of the identified polypeptides could be located in different metabolic pathways. Proteins involved in carbohydrate, energy and amino acid metabolism constituted to about 1/4, and those involved in metabolism of vitamins and cofactors constituted for about 3% of the total signal intensity in gels prepared from 72 h seedlings. Enzymes related to genetic information processing increased very quickly during early imbibition and reached highest level around 30 h of germination.

Extraction Conditions for Phenolic Compounds with Antioxidant Activities from White Rose Petals

  • The extract of white rose petals has an antioxidant effect and can be used to treat allergic disease. The purpose of this study was to identify optimal conditions for extracting antioxidative compounds from white rose petals with 2,2-diphenyl-1-picrylhydrazyl scavenging activities. A response surface methodology based on a central composite design was used to investigate the effects of three independent variables: ethanol concentration ($X_1$), extraction temperature ($X_2$), and extraction time ($X_3$). The estimated optimal conditions for obtaining phenolic compounds with antioxidant activities were as follows: ethanol concentration of 42% ($X_1$), extraction time of 80 min ($X_3$), and extraction temperature of $75^{\circ}C$ ($X_2$). The estimated optimal conditions for obtaining flavonoid compounds with antioxidant effects were an ethanol concentration of 41% ($X_1$), extraction time of 119 min ($X_3$), and an extraction temperature of $75^{\circ}C$ ($X_2$). Under these conditions, predicted response values for the phenolic and flavonoid contents were 243.5 mg gallic acid equivalent/g dry mass and 19.93 mg catechin equivalent (CE)/g dry mass, respectively.

해송엽(海松葉) Litter의 분해(分解)와 N, P 및 K의 동태(動態) (Decomposition and, Nitrogen, Phosphorus and Potassium Dynamics of Pinus thundbergii Needle Litter)

  • 해송엽(海松葉) litter의 분해(分解)와 그에 따른 양분방출(養分放出)의 동태(動態)를 일본지방(日本地方)의 해안(海岸) 해송림(海松林)에서 litterbag법(法)으로 조사(調査)하였다. litter의 분해(分解)에 따른 건물중(乾物重)의 소실(消失)은 초기(初期) 1년간(年間)에 빨랐고 그후는 보다 둔화(鈍化)하였으며, 분해(分解) 1년동안의 소실율(消失率)은 약(約) 40%였다. 분해계수(分解係數)는 0.5-6.0, 반감기(半減期)(년(年))는 l.1-l.4의 범위였다. 분해(分解)에 따른 잔존(殘存) litter내의 N과 P의 양분농도(養分濃度)는 경시적(經時的)으로 증가(增加)하는 경향(傾向)이었으나 K의 농도(濃度)는 감소(減少)하였다. K의 양분(養分)의 절대량(絶對量)은 분해(分解)가 진행(進行)하는 동안 현저히 감소하였으나 N의 절대량(絶對量)은 증가하는 경향을 보였다. 분해(分解)에 따른 방출(放出)은 K>P>N의 순(順)이었다. 실험기간중 N의 무기화(無機化) 단계는 나타나지 않았다.

Carboxylated Alginic Acid 및 이를 함유한 세정용 화장품의 납 흡착 효과 (Lead Adsorption by Carboxylated Alginic Acid and Its Application in Cleansing Cosmetics)

  • This study investigated lead adsorption by carboxylated alginic acid and its application in cleansing cosmetics. Carboxylated alginic acid showed the highest lead adsorptivity after oxidation in a 4-6 mM hydrogen peroxide solution at $20-30^{\circ}C$ for 30-40 min. Carboxylated alginic acid adsorbed $648.1{\pm}2.8-653.0{\pm}2.9$ mg/g of lead dry mass at pH 4-6. Carboxylated alginic acid modified by hydrogen peroxide and potassium permanganate adsorbed $651.3{\pm}3.8$ and $639.9{\pm}4.0$ mg/g of lead dry mass, respectively. Carboxylated alginic acid showed higher lead adsorptivity after modification by hydrogen peroxide than by potassium permanganate, with an increase of ~30% compared with raw alginic acid. To access the potential application of carboxylated alginic acid in cleansing cosmetics, we investigated the lead adsorptivity, conditions of the cosmetics procedure, and cytotoxicity of various concentrations of cleansing cosmetics added to 5% carboxylated alginic acid. The ideal cosmetic concentrations combined with 5% carboxylated alginic acid were 70% for peeling gel, 20% for massage cream, 20% for foam cleansing and 40% for cleansing cream. There was no cytotoxicity in cleansing cosmetics combined with 5% carboxylated alginic acid.

눈꽃동충하초 배지별 인공재배법과 성분분석에 관한연구 (Study on analysis of components and artificial cultural practice on several culture media of Paecilomyces japonica)

  • 1. 누에를 이용한 눈꽃동충하초 재배에서 종균접종은 누에 5령1일째 실시하였으며, 감염율은 72% 되었으며 2. 눈꽃동충하초 대량생산을 위한 인공재배 최적배지는 "현미80g+번데기가루20g" 처리구에서 자실체 길이 11.4cm,무게 51.6g, 짙노랑색으로 아주 양호했으며, 현미 처리구, 번데기+PDA처리구에서도 양호했음. 3.누에동충하초에서 발생된 눈꽃동충하초는 번데기 1마리당 1.2g(번데기포함), 길이 3.0~3.5cm정도 되었으며, 일부는 누에상태에서도 동충하초가 발생하였음. 4. $\beta$-glucan함량은 누에 5령 1일 접종 처리구가 40.5%로 가장 높고 저비용 배지인 현미 16.4%, 번데기 20.7%, 현미+번데기가루 23.1%, 소나무 톱밥+밀기울 28.7%로 $\beta$-glucan 함량이 높았다. 5. 지네, 굼벵이, 누에가루처리구에서는 균사생육이 부진하였고 분생포자만 발생하였다.

Decomposition and Nutrient Dynamics of Aquatic Macrophytes in Lake Paldang

  • This study examined the decomposition of blades and culms of aquatic emergent plant species, Zizania latifolia, Phragmites communis and Typha angustata, and changes in nutrient contents during decomposition. Z. latifolia, P. communis and T. angustata were the most frequently occurring species in Lake Paldang of Han River, Korea. Experiments were carried out from July 27 to December 14, 2005 in Lake Paldang using the litter bag method. The remaining masses of blade litter of each species at the end of experimental period were 21.2% of initial dry weight in Z. latifolia, 32.5% in P. communis, and 44.7% in T. angustata. In addition, the remaining mass of culm was 22.6% of initial dry mass in Z. latifolia, 56.4% in P. communis, and 38.1% in T. angustata. During the litter decomposition period, P, K, Na, and Mg concentration decreased rapidly within 10 days, but Ca and Mg concentration declined slowly. K contents remained below 10% of initial values in all litter samples retrieved during decomposition, whereas Ca and Mg concentration remained above 40% and 50% during decomposition in all three species. Na, P and Mn contents in litter varied among species and plant parts. P concentration in culms of P. communis remained at about 60% of initial concentration throughout the study, but the remaining P content in culms of Z. latifolia was only 10% of the original value at the end of the study period. The Mn concentration in blades of P. communis increased about 15-fold relative to the initial content by the end of experiment.

국내산 죽종의 화학성분 및 섬유소 이용에 관한 연구 (제2보) - 대나무 섬유의 제조 및 특성 - (A Study on the Utilization of Ingredients and Fibers from Korean Bamboo Species in Value-added Industry: Part 2. Preparation and Characterization of Bamboo Fibers)

  • The bamboo fibers from three Korean bamboo species, Moso, Henon and Timber bamboo, were prepared by disintegration after thermal treatment. The samples were prepared according to the age of growth; bamboo shoots, 20 days and 50 days of growth. The fiber quality was also investigated by morphological and chemical analyses. There was no big difference in the dry mass of fibers among the three bamboo species. However, the dry mass of fibers from 50 days of growth was increased by 34%, whereas it was ranged in 9-20% in the case of fibers from bamboo shoots and 20 days of growth. In the results of morphological analysis, the fibers could be classified as the fibers from metaxylem, the fibers from parenchyma and the fines produced during disintegration. The fibers from 50 days of growth were separated from metaxylem, whereas the fibers from bamboo shoots and 20 days of growth were mainly consisted of fibers from metaxylem and parenchyma. The chemical analysis of fibers showed that the contents of carbohydrates, lignin and extractives were not much affected by thermal treatment.

항독성 소간추출물의 생산을 위한 효소분해의 최적화 (Optimization of Enzymatic Hydrolysis for the Production of Antitoxic Bovine Hepatic Extract)

  • 소간추출물은 각종 간질환에 해독효과를 가진 것으로 알려져있다. 항독성 소간추출물의 대량생산 공정을 개발하고자 효소의 선발 및 가수분해의 최적조건을 탐색하였고, 확립된 분해조건으로 항독성 소간추출물의 시제품을 제조하여 일반성분, 미생물, 그리고 비타민 $B_{12}$함량을 분석하였다. Bromelain, papain, ficin, pancreatin, 그리고 protease NP등의 효소 중 추출물의 건물량과아미노태 질소의 수율이 양호하고 경제성이 뛰어난 papain을 최적효소로 선발하였다. Papain(1%)을 소간 분쇄물에 첨가하여 65$^{\circ}C$에서 24시간 동안 가수분해하는 조건을 확립하였다. 분무건조한 항독성 소간추출물의 시제품은 원료 간 대비 11%의 수율을 보였고 건물량은 95%, 총질소 함량은 11.8%이었다. 또한 병원성 미생물은 검출되지 않았고 비타민 $B_{12}$ 함량은 4.1 ${\mu}$g/g이었다. 본 연구에서 확립한 가수분해조건은 높은 수율의 항독성 소간추출물 생산에 적용할 수 있을 것으로 생각한다.
